| 1 | Cycloastragenol upregulates SIRT1 expression,attenuates apoptosis and suppresses neuroinflammation after brain ischemia显示文摘Cycloastragenol(CAG)is the active form of astragaloside V isolated from Astragalus Radix,which displays multiple pharmacological effects.Silent information regulator 1(SIRT1),a classll histone deacetylase,has been shown to play an important role in neuroprotection against cerebral ischemia.In this study,we investigated whether CAG protected against ischemic brain injury and,if so,whether the beneficial effects were associated with the regulation of SIRT1 in the ischemic brain.Mice were subjected to 45 min of middle cerebral artery occlusion(MCAO)followed by reperfusion.CAG(5,10,20 mg/kg)was injected intraperitoneally at the onset of reperfusion,12h later and then twice daily for up to three days.CAG dose-dependently reduced brain infarct volume,significantly ameliorated functional deficits,and prevented neuronal cell oss in MCAO mice.Meanwhile,CAG signicantly reduced matrix metalloproteinase-9 activity,prevented tight junction degradation and subsequently ameliorated blood-brain barier disruption.Moreover,CAG significantly upregulated SIRT1 expression in the ischemic brain but did not directly activate its enzymatic activity.Concomitant with SIRT1 upregulation,CAG reduced p53 acetylation and the ratio of Bax to Bcl-2 in the ischemic brain.CAG also inhibited NF-κB p65 nuclear translocation.As a result,CAG suppressed the mRNA expression of pro-inflammatory cytokines,including TNF-α and IL-1β,and inhibited the activation of microglia and astrocytes in the ischemic brain.Our findings suggest that CAG is neuroprotective against ischemic brain injury in mice and that its beneficial effect may involve SIRT1 upregulation and the inhibition of apoptosis and neuroinflammation in the ischemic brain. | Man Li Shichun Li Bao-kai Dou Yving-xiang Zou Hao-zhen Han Dong-xiang Liu Zunji Ke Zhi-fei Wang | 2020 | Acta Pharmacologica Sinica2020,41,8: | 22 |
| 2 | Metformin promotes microglial cells to facilitate myelin debris clearance and accelerate nerve repairment after spinal cord injury显示文摘Spinal cord injury(SCI)is one kind of severe trauma for central nervous system.Myelin debris clearance and axon regeneration are essential for nerve regeneration after SCI.Metformin,a glucose-lowering drug,has been demonstrated to promote the locomotor functional recovery after SCI.In this study,we investigated the role and molecular mechanism of metformin on myelin preservation in a rat SCI model.SCI was induced in rats by compression at T9 level using a vascular clip.We showed that administration of metformin(50 mg·kg^(-1)·d^(-1),ip)for 28 days significantly improved locomotor function in SCI rats.Metformin also ameliorated SCI-induced neuronal apoptosis and promoted axon regeneration in the spinal cord.Using co-immunofluorescence of IBa-1 and MBP,and luxol fasting blue(LFB)staining,we demonstrated that metformin promoted the transformation of M1 to M2 phenotype polarization of microglial cells,then greatly facilitated myelin debris clearance and protected the myelin in SCI rats.Furthermore,metformin ameliorated SCI-induced blockade of autophagic flux in the spinal cord,and enhanced the fusion of autophagosome and lysosome by inhibiting the AMPK-mTOR signaling pathway.Moreover,metformin significantly attenuated inflammatory responses in the spinal cord.In LPS-treated BV2 cells,pretreatment with metformin(2 mM)significantly enhanced autophagy level,suppressed inflammation and cell apoptosis.The protective effects were blocked in the presence of an autophagy inhibitor 3-methyladenine(3-MA,5 mM),suggesting that the effect of metformin on autophagy in microglial cells is essential for the myelin preservation during nerve recovery.This study reveals a novel therapeutic effect of metformin in SCI recovery by regulating the activation of microglial cells and enhancing its autophagy level. | Yan-qing Wu Jun Xiong Zi-li He Yuan Yuan Bei-ni Wang Jing-yu Xu Man Wu Su-su Zhang Shu-fang Cai Jia-xin Zhao Ke Xu Hong-yu Zhang Jian Xiao | 2022 | Acta Pharmacologica Sinica2022,43,6: | 8 |

| 4 | Progress of circulating fluidized bed combustion technology in China:a review显示文摘Circulating fluidized bed(CFB)technology plays an important role in the utilization of low-grade coal in China.This article reviews CFB combustion technology development in China and summarizes recent achievements.Since 1990 Chinese engineers and researchers have been undertaking work to improve CFB boiler technology.A completely novel CFB boiler design theory was developed and used in the domestic manufacturing of CFB boilers with various capacities.China is the largest supplier and customer of CFB boilers in the world due to the widespread use of CFB boilers.In 2007,the lower energy consumption CFB technology was successfully developed by re-specifying the fluidization state,which reduced the power consumption of forced fans and solved the potential erosion problems on the water wall.Afterwards,in order to increase the electric power generation efficiency,the supercritical CFB(SCCFB)boiler was developed and the first 600 MW SCCFB boiler was demonstrated and put into commercial operation in 2013.The success of the technology is evident with over 80 SCCFB boilers on order with capacities of 350 MW to 660 MW.Chinese scientists and engineers are also developing technology to lower the emissions of CFB combustion to meet the requirements of China’s strict emission regulations.This emission reduction is through high-efficiency desulfurization by limestone injection into the furnace and low NOx combustion,ultra-low emission of SO2 and NOx in the furnace can be realized by improving the bed quality and increasing the solid circulation rate.There is currently further research and design development being undertaken to develop a 660 MW ultra-supercritical CFB(USCCFB)boiler.The new boiler is expected to be operational before 2020 resulting in higher efficiency and lower energy consumption and emissions. | Runxia Cai Xiwei Ke Junfu Lyu Hairui Yang Man Zhang Guangxi Yue Wen Ling | 2017 | Clean Energy2017,1,1: | 5 |
| 5 | Numerical study of the semi-circular bend dynamic fracture toughness test using discrete element models显示文摘The semi-circular bend(SCB) dynamic fracture toughness test is simulated using discrete element models. The influence of the frictional boundary condition, constitutive law and specimen thickness on the test measurements is investigated. It is found that friction between loading plates and the rock specimen affects the test results. Therefore, friction must be carefully considered to obtain accurate measurements. The simulation results also show that in contrast to the 2D model in which a rate-dependent cohesive law must be introduced, 3D models with a rate-independent law can produce good results. | 14 | A case of human infection with a novel Babesia species in China显示文摘Background:Babesiosis is an uncommon but emerging tick-borne disease caused by the genus Babesia.In this case study,we report a case of human infection with a novel Babesia sp.in China.Findings:The patient in question had been suffering from repetitive occurrences of mild fever of unknown origin and fatigue for 10 years.Ring forms,tetrads,and one or two dots of chromatin or trophozoite-like organisms were observed in the patient’s thin blood smears and bone marrow smears.Using a confocal laser-scanning microscope,it was observed that the patient’s serum had reactivity with the surface proteins of the B.microti strain.Electron microscopy revealed oval red blood cells with 1~2μm of knob protrusions in the cellular membrane.The results of the Babesia-specific nested PCR assay for 18S rRNA confirmed the presence of Babesia infection.The construction of a phylogenetic relationship showed clustering with B.microti and B.duncani,which was identified as a novel Babesia species and named as Babesia sp.XXB/HangZhou.Azithromycin,doxycycline,and moxifloxacin hydrochloride were shown to relieve symptoms but were not as effective after continuous usage.After atovaquone(Mepron®)administration,the patient recovered from fever and tested negative for detection of Babesia-specific genes.Conclusion:Babesia sp.XXB/HangZhou is a novel Babesia species,which causes mild babesiosis in an immunocompetent patient. | Su-Qin Man Ke Qiao Jie Cui Meng Feng Yong-Feng Fu Xun-Jia Cheng | 2016 | Infectious Diseases of Poverty2016,5,1: | 1 |

| 20 | FSAP aggravated endothelial dysfunction and neurological deficits in acute ischemic stroke due to large vessel occlusion显示文摘Revascularization and angiogenesis,as substrates of sustained collateral circulation,play a crucial role in determining the severity and clinical outcome of acute ischemic stroke(AIS)due to large vessel occlusion(LVO).Developing an adjunct biomarker to help identify and monitor collateral status would aid stroke diagnosis and prognosis.To screen the potential biomarkers,proteomic analysis was performed in this study to identify those distinct plasma protein profiles in AIS due to LVO with different collateral status.Interestingly,we found that levels of Plasma Factor VII Activating Protease(FSAP)significantly increased in those AIS patients with poor collaterals,and were correlated with worse neurological outcome.Furtherly,both in vitro and in vivo models of ischemic stroke were used to explore pathological mechanisms of FSAP in endothelial dysfunction.We demonstrated that the FSAP inhibitor,high-molecular-weight hyaluronan(HMW-HA),enhanced the pro-angiogenic vascular factors,improved the integrity of brain blood barrier,and promoted newly formed cerebral microvessels in the ischemic penumbra,consequently improving neurological function.To elucidate the pathways that might contribute to revascularization during LVO,we applied transcriptomic analysis via unbiased RNA sequencing and showed that Wnt signaling was highly involved in FSAP mediated endothelial dysfunction.Notably,inhibition of Wnt5a largely reversed the protective effects from HMW-HA treatment,implying that FSAP might aggravate endothelial dysfunction and neurological deficits by regulating Wnt5a signaling.Therefore,FSAP may represent a potential biomarker for collateral status after LVO and a promising therapeutic target to be explored in the treatment of stroke. | Dai-Shi Tian Chuan Qin Luo-Qi Zhou Sheng Yang Man Chen Jun Xiao Ke Shang Dale BBosco Long-Jun Wu Wei Wang | 2022 | Signal Transduction and Targeted Therapy2022,7,2: | 1 |
