| 2 | The current agonists and positive allosteric modulators ofα7 nAChR for CNS indications in clinical trials显示文摘The alpha-7 nicotinic acetylcholine receptor(α7 nAChR), consisting of homomeric α7 subunits, is a ligand-gated Ca^(2+)-permeable ion channel implicated in cognition and neuropsychiatric disorders. Enhancement of α7 nAChR function is considered to be a potential therapeutic strategy aiming at ameliorating cognitive deficits of neuropsychiatric disorders such as Alzheimer's disease(AD) and schizophrenia. Currently, a number of α7 nAChR modulators have been reported and several of them have advanced into clinical trials. | 3 | Preparation of TiC/Ni_3Al Composites by Upward Melt Infiltration显示文摘TiC/Ni3Al composites have been prepared using upward infiltration method. The densification was performed by both Ni3Al melt filling and TiC sintering during the infiltration. The dissolution of TiC in liquid Ni3Al has been evidenced by finding Ni3(Al,Ti)C after fast cooling in the TiC/Ni3Al composites. The dissolution may be responsible for the infiltration and sintering. Compared with downward infiltration, the upward infiltration brought about higher strength and fracture toughness and shorter infiltration time. TiC/20 vol. pct Ni3Al composite processed by upward infiltration had a flexural strength of 1476 MPa with a statistic Weibull modulus of 20.2 and a fracture toughness of 20.4 MPa . | 5 | Inhibition of temperature-sensitive TRPV3 channel by two natural isochlorogenic acid isomers for alleviation of dermatitis and chronic pruritus显示文摘Genetic gain-of-function mutations of warm temperature-sensitive transient receptor potential vanilloid 3(TRPV3)channel cause Olmsted syndrome characterized by severe itching and keratoderma,indicating that pharmacological inhibition of TRPV3 may hold promise for therapy of chronic pruritus and skin diseases.However,currently available TRPV3 tool inhibitors are either nonselective or less potent,thus impeding the validation of TRPV3 as therapeutic target.Using whole-cell patch-clamp and single-channel recordings,we report the identification of two natural dicaffeoylquinic acid isomers isochlorogenic acid A(IAA)and isochlorogenic acid B(IAB)that selectively inhibit TRPV3 currents with IC50 values of 2.7±1.3 and 0.9±0.3μmol/L,respectively,and reduce the channel open probability to 3.7±1.2%and 3.2±1.1%from 26.9±5.5%,respectively.In vivo evaluation confirms that both IAA and IAB significantly reverse the ear swelling of dermatitis and chronic pruritus.Furthermore,the isomer IAB is able to rescue the keratinocyte death induced by TRPV3 agonist carvacrol.Molecular docking combined with site-directed mutations reveals two residues T636 and F666 critical for the binding of the two isomers.Taken together,our identification of isochlorogenic acids A and B that act as specific TRPV3 channel inhibitors and gating modifiers not only provides an essential pharmacological tool for further investigation of the channel pharmacology and pathology,but also holds developmental potential for treatment of dermatitis and chronic pruritus. | Hang Qi Yuntao Shi Han Wu Canyang Niu Xiaoying Sun KeWei Wang | 2022 | Acta Pharmaceutica Sinica B2022,12,2: | 3 |

| 20 | Pharmacological characterization of JWX-A0108 as a novel type Ⅰ positive allosteric modulator of a7 nAChR that can reverse acoustic gating deficits in a mouse prepulse inhibition model显示文摘The a7 nicotinic acetylcholine receptor (a7 nAChR) is a ligand-gated Ca^2+-permeable homopentameric ion channel implicated in cognition and neuropsychiatric disorders. Pharmacological enhancement of a7 nAChR function has been suggested for improvement of cognitive deficits. In the present study, we characterized a thiazolyl heterocyclic derivative, 6-(2-chloro-6- methylphenyl)-2-((3-fluoro-4-methylphenyl)amino)thiazolo[4,5-cflpyrimiclin-7(6H)-one (JWX-A0108), as a novel type I a7 nAChR positive allosteric modulator (PAM), and evaluated its ability to reverse auditory gating and spatial working memory deficits in mice. In Xenopus oocytes expressing human nAChR channels, application of JWX-A0108 seleaively enhanced a7 nAChR-mediated inward current in the presence of the agonist ACh (EC50 value = 4.35 ±0.12 pM). In hippocampal slices, co-application of ACh and JWXA0108 (10 pM for each) markedly increased both the frequency and amplitude of spontaneous inhibitory postsynaptic currents (sIPSCs) recorded in pyramidal neurons, but JWX-A0108 did not affect GABA-induced current in oocytes expressing human GABAa receptor aip3y2 and a5(33Y2 subtypes. In mice with MK-801-induced deficits in auditory gating, administration of JWX-A0108 (1, 3, and 10 mg/kg, i.p.) dose-dependently attenuates MK-801-induced auditory gating deficits in five prepulse intensities (72, 76, 80,84, and 88 dB). Furthermore, administration of JWX-A0108 (0.03, 0.1, or 0.3 mg/kg, i.p.) significantly reversed MK-801-induced impaired spatial working memory in mice.
