| 3 | A phase 3,double-blind,randomized placebo-controlled efficacy and safety study of abiraterone acetate in chemotherapynaı¨ve patients with mCRPC in China,Malaysia,Thailand and Russia显示文摘Objective:This double-blind,placebo-controlled phase 3 study was designed to compare efficacy and safety of abiraterone acetate+prednisone(abiraterone)to prednisone alone in chemotherapy-naı¨ve,asymptomatic or mildly symptomatic metastatic castrationresistant prostate cancer(mCRPC)patients from China,Malaysia,Thailand and Russia.Methods:Adult chemotherapy-naı¨ve patients with confirmed prostate adenocarcinoma,Eastern Cooperative Oncology Group(ECOG)performance status(PS)grade 0e1,ongoing androgen deprivation(serum testosterone<50 ng/dL)with prostate specific antigen(PSA)or radiographic progression were randomized to receive abiraterone acetate(1000 mg,QD)t prednisone(5 mg,BID)or placebo t prednisone(5 mg,BID),until disease progression,unacceptable toxicity or consent withdrawal.Primary endpoint was improvements in time to PSA progression(TTPP).Results:Totally,313 patients were randomized(abiraterone:n Z 157;prednisone:n Z 156);and baseline characteristics were balanced.At clinical cut-off(median follow-up time:3.9 months),80% patients received treatment(abiraterone:n Z 138,prednisone:n Z 112).Median time to PSA progression was not reached with abiraterone versus 3.8 months for prednisone,attaining 58%reduction in PSA progression risk(HR=0.418;p<0.0001).Abirateronetreated patients had higher confirmed PSA response rate(50%vs.21%;relative odds=2.4;p<0.0001)and were 5 times more likely to achieve radiographic response than prednisonetreated patients(22.9%vs.4.8%,p=0.0369).Median survival was not reached.Most common(≥10% abiraterone vs.prednisone-treated)adverse events:bone pain(7%vs.14%),pain in extremity(6%vs.12%),arthralgia(10%vs.8%),back pain(7%vs.11%),and hypertension(15%vs.14%).Conclusion:Interim analysis confirmed favorable benefit-to-risk ratio of abiraterone in chemotherapy-naı¨ve men with mCRPC,consistent with global study,thus supporting use of abiraterone in this patient population. | Dingwei Ye Yiran Huang Fangjian Zhou Keji Xie Vsevolod Matveev Changling Li Boris Alexeev Ye Tian Mingxing Qiu Hanzhong Li Tie Zhou Peter De Porre Margaret Yu Vahid Naini Hongchuan Liang Zhuli Wu Yinghao Sun | 2017 | Asian Journal of Urology2017,4,2: | 3 |
| 4 | Recovery of a Far-Eastern Strain of Tick-Borne Encephalitis Virus with a Full-Length Infectious cDNA Clone显示文摘Tick-borne encephalitis virus(TBEV) is a pathogenic virus known to cause central nervous system(CNS) diseases in humans, and has become an increasing public health threat nowadays. The rates of TBEV infection in the endemic countries are increasing. However, there is no effective antiviral against the disease. This underscores the urgent need for tools to study the emergence and pathogenesis of TBEV and to accelerate the development of vaccines and antivirals. In this study, we reported an infectious c DNA clone of TBEV that was isolated in China(the WH2012 strain). A beta-globin intron was inserted in the coding region of nonstructural protein 1(NS1) gene to improve the stability of viral genome in bacteria. In mammalian cells, the inserted intron was excised and spliced precisely, which did not lead to the generation of inserted mutants. High titers of infectious progeny viruses were generated after the transfection of the infectious clone. The cDNA-derived TBEV replicated efficiently, and caused typical cytopathic effect(CPE) and plaques in BHK-21 cells. In addition, the CPE and growth curve of cDNA-derived virus were similar to that of its parental isolate in cells. | 7 | Collagen fiber membrane-derived chemically and mechanically durable superhydrophobic membrane for high- performance emulsion separation显示文摘Developing high-performance separation membrane with good durability is a highly desired while challenging issue.Herein,we reported the successful fabrication of chemically and mechanically durable superhydrophobic membrane that was prepared by embedding UiO-66 as size-sieving sites within the supramolecular fiber structure of collagen fiber membrane(CFM),followed by the polydimethylsiloxane(PDMS)coating.The as-prepared CFM/UiO-66(12)/PDMS membrane featured capillary effect-enhanced separation flux and homogeneous porous channels guaranteed high separation efficiency.When utilized as double-layer separation membranes,this new type of composite membranes separated various surfactant stabilized water-in-oil microemulsions and nanoemulsions,with the separation efficiency high up to 99.993%and the flux as high as 973.3 L m−2 h−1.Compared with commercial polytetrafluoro ethylene(PTFE)membrane,the advantage of the double-layer CFM/UiO-66(12)/PDMS membranes in separation flux was evident,which exhibited one order of magnitude higher than that of commercial PTFE membrane.The CFM/UiO-66(12)/PDMS membrane was acid-alkali tolerant,UV-aging resistant and reusable for emulsion separation.Notably,the CFM/UiO-66(12)/PDMS membrane was mechanically durable against strong mechanical abrasion,which was still capable of separating diverse water-in-oil emulsions after the abrasion with sandpaper and assembled as double-layer separation membranes.We anticipate that the combination of CFM and metal organic frameworks(MOFs)is an effective strategy for fabricating high-performance separation membrane with high mechanical and chemical durability. | Huifang Li Wan Zheng Hanzhong Xiao Baicun Hao Yujia Wang Xin Huang Bi Shi | 2021 | Journal of Leather Science and Engineering2021,3,1: | 1 |

| 12 | Application of diatomite as an effective polysulfides adsorbent for lithium-sulfur batteries显示文摘The lithium–sulfur batteries show the great potential to be the most promising candidate for high energy applications. However, the shuttling of soluble polysulfides deteriorates the battery performance tremendously. To suppress the diffusion of soluble polysulfides, diatomite that has abundant natural three-dimensional ordered pores is incorporated into the cathode to trap polysulfides. The composite cathode material(S-DM-AB for short), including sulfur(S), diatomite(DM), and acetylene black(AB) is prepared by an impregnation method. For comparison, another composite cathode material(S-AB for short) including sulfur and acetylene black is also prepared by the same method. The battery with S-DMAB composite cathode material delivers a discharge capacity of 531.4 m Ah/g after 300 cycles at 2 C with a capacity retention of 51.6% at room temperature. By contrast, the battery with S-AB composite cathode material delivered a capacity of only 196.9 m Ah/g with a much lower capacity retention of 18.6% under the same condition. Urine samples of the RCCC patients and healthy controls were collected in the morning.Weak cation exchange (WCX) bead-based MALDI-TOF MS technique was applied in detecting differential protein peaks in the urine of RCCC. ClinProTools2.2 software was utilized to determine the characteristic proteins in the urine of RCCC patients for the predictive model of RCCC.Results The technique identified 160 protein peaks in the urine that were different between RCCC patients and health controls; and among them, there was one peak (molecular weight of 2221.71 Da) with statistical significance (P=0.0304). | 14 | Enhanced Protective Efficacy of H5 Subtype Influenza Vaccine with Modification of the Multibasic Cleavage Site of Hemagglutinin in Retroviral Pseudotypes显示文摘Traditionally, the multibasic cleavage site (MBCS) of surface protein H5-hemagglutinin (HA) is converted to a monobasic one so as to weaken the virulence of recombinant H5N1 influenza viruses and to produce inactivated and live attenuated vaccines. Whether such modification benefits new candidate vaccines has not been adequately investigated. We previously used retroviral vectors to generate wtH5N1 pseudotypes containing the wild-type HA (wtH5) from A/swine/Anhui/ca/2004 (H5N1) virus. Here, we generated mtH5N1 pseudotypes, which contained a mutant-type HA (mtH5) with a modified monobasic cleavage site. Groups of mice were subcutaneously injected with the two types of influenza pseudotypes. Compared to the group immunized with wtH5N1 pseudotypes, the inoculation of mtH5N1 pseudotypes induced significantly higher levels of HA specific IgG and IFN-γ in immunized mice, and enhanced protection against the challenge of mouse-adapted avian influenza virus A/Chicken/Henan/12/2004 (H5N1).
