| 2 | T cells expressing a LMP1-specific chimeric antigen receptor mediate antitumor effects against LMP1-positive nasopharyngeal carcinoma cells in vitro and in vivo显示文摘T cells modified with chimeric antigen receptor are an attractive strategy to treat Epstein-Barr virus(EBV) associated malignancies.The EBV latent membrane protein 1(LMP1) is a 66-KD integral membrane protein encoded by EBV that consists of transmembrane-spanning loops.Previously,we have identified a functional signal chain variable fragment(scFv) that specifically recognizes LMP1 through phage library screening.Here,we constructed a LMP1 specific chimeric antigen receptor containing anti-LMP1 scFv,the CD28 signalling domain,and the CD3ζchain(HELA/CAR).We tested its functional ability to target LMP1 positive nasopharyngeal carcinoma cells.HELA/CAR cells were efficiently generated using lentivirus vector encoding the LMP1-specific chimeric antigen receptor to infect activated human CD3+ T cells.The HELA/CAR T cells displayed LMP1 specific cytolytic action and produced IFN-γ and IL-2 in response to nasopharyngeal carcinoma cells overexpressing LMP1.To demonstrate in vivo anti-tumor activity,we tested the HELA/CAR T cells in a xenograft model using an LMP1 overexpressing tumor.Intratumoral injection of anti-LMP1 HELA/CAR-T cells significantly reduced tumor growth in vivo.These results show that targeting LMP1 using HELA/CAR cells could represent an alternative therapeutic approach for patients with EBV-positive cancers. | Xiaojun Tang Yan Zhou Wenjie Li Qi Tang Renjie Chen Jin Zhu Zhenqing Feng | 2014 | The Journal of Biomedical Research2014,28,6: | 16 |
| 3 | Pore network modeling of water block in low permeability reservoirs显示文摘A pore network model was used in this paper to investigate the factors, in particular, throat radius, wettability and initial water saturation, causing water block in low permeability reservoirs. A new term–‘relative permeability number’ (RPN) was firstly defined, and then used to describe the degree of water block. Imbibition process simulations show that the RPN drops in accordance with the extension of the averaged pore throat radius from 0.05 to 1.5 μm, and yet once beyond that point of 1.5 μm, the RPN reaches a higher value, indicating the existence of a critical pore throat radius where water block is the maximum. When the wettability of the samples changes from water-wet to weakly water-wet, weakly gas-wet, or gas(oil)-wet, the gas RPN increases consistently, but this consistency is disturbed by the RPN dropping for weakly water-wet samples for water saturations less than 0.4, which means weakly water-wet media are more easily water blocked than water-wet systems. | 4 | Folate receptor-mediated boron-10 containing carbon nanoparticles as potential delivery vehicles for boron neutron capture therapy of nonfunctional pituitary adenomas显示文摘Invasive nonfunctional pituitary adenomas(NFPAs) are difficult to completely resect and often develop tumor recurrence after initial surgery.Currently,no medications are clinically effective in the control of NFPA.Although radiation therapy and radiosurgery are useful to prevent tumor regrowth,they are frequently withheld because of severe complications.Boron neutron capture therapy(BNCT) is a binary radiotherapy that selectively and maximally damages tumor cells without harming the surrounding normal tissue.Folate receptor(FR)-targeted boron-10 containing carbon nanoparticles is a novel boron delivery agent that can be selectively taken up by FR-expressing cells via FR-mediated endocytosis.In this study,FR-targeted boron-10 containing carbon nanoparticles were selectively taken up by NFPAs cells expressing FR but not other types of non-FR expressing pituitary adenomas.After incubation with boron-10 containing carbon nanoparticles and following irradiation with thermal neutrons,the cell viability of NFPAs was significantly decreased,while apoptotic cells were simultaneously increased.However,cells administered the same dose of FR-targeted boron-10 containing carbon nanoparticles without neutron irradiation or received the same neutron irradiation alone did not show significant decrease in cell viability or increase in apoptotic cells.The expression of Bcl-2 was down-regulated and the expression of Bax was up-regulated in NFPAs after treatment with FR-mediated BNCT.In conclusion,FR-targeted boron-10 containing carbon nanoparticles may be an ideal delivery system of boron to NFPAs cells for BNCT.Furthermore,our study also provides a novel insight into therapeutic strategies for invasive NFPA refractory to conventional therapy,while exploring these new applications of BNCT for tumors,especially benign tumors. | DAI CongXin CAI Feng HWANG Kuo Chu ZHOU YongMao ZHANG ZiZhu LIU XiaoHai MA SiHai YANG YaKun YAO Yong FENG Ming BAO XinJie LI GuiLin WEI JunJi JIAO YongHui WEI ZhenQing MA WenBin WANG RenZhi | 2013 | Science China(Life Sciences)2013,56,2: | 8 |

| 6 | Pollen flux and vertical dispersion in coniferous and deciduous broadleaved mixed forest in the Changbai Mountains显示文摘The pollen flux in coniferous and deciduous broadleaved mixed forest in the Changbai Mountains is presented in one year's experiments. The results indicate that arboreal pollen percentages are more than 65% and pollen flux is higher than 5000 grain·cm-2·a-1,while less than 2% and lower than 1000 grain·cm-2·a-1 for shrubby pollen,and less than 20% and lower than 3000 grain·cm-1·a-1 for herbaceous pollen for most samples at different heights. The pollen assemblages are similar to those in the samples under 8 m height where Pinus and Ouercus are dominant and followed by the few non-local pollen types,and Fraxinus percentages are high with a few non-local pollen types at 16 m to 32 m height as well as non-local pollen clearly increased at 40 m height. | 10 | Nucleoprotein-based indirect enzyme-linked immunosorbent assay(indirect ELISA) for detecting antibodies specific to Ebola virus and Marbug virus显示文摘Full-length nucleoproteins from Ebola and Marburg viruses were expressed as His-tagged recombinant proteins in Escherichia coli and nucleoprotein-based enzyme-linked immunosorbent assays(ELISAs) were established for the detection of antibodies specific to Ebola and Marburg viruses. The ELISAs were evaluated by testing antisera collected from rabbit immunized with Ebola and Marburg virus nucleoproteins. Although little cross-reactivity of antibodies was observed in antiEbola virus nucleoprotein rabbit antisera, the highest reactions to immunoglobulin G(Ig G) were uniformly detected against the nucleoprotein antigens of homologous viruses. We further evaluated the ELISA's ability to detect antibodies to Ebola and Marburg viruses using human sera samples collected from individuals passing through the Guangdong port of entry. With a threshold set at the mean plus three standard deviations of average optical densities of sera tested, the ELISA systems using these two recombinant nucleoproteins have good sensitivity and specificity. | 11 | The Zhaxikang Vein-type Pb-Zn-Ag-Sb Deposit in Himalayan Orogen, Tibet: Product by Overprinting and Remobilization Processes during Post-collisional Period显示文摘The Zhaxikang Pb-Zn-Ag-Sb deposit, the largest polymetallic deposit known in the Himalayan Orogen of southern Tibet, is characterized by vein-type mineralization that hosts multiple mineral assemblages and complicated metal associations. The deposit consists of at least six steeply dipping veintype orebodies that are hosted by Early Jurassic black carbonaceous slates and are controlled by a Cenozoic N–S-striking normal fault system. This deposit records multiple stages of mineralization that include an early period(A) of massive coarse-grained galena–sphalerite deposition and a later period(B) of Sb-bearing vein-type mineralization. Period A is only associated with galena–sphalerite mineralization, whereas period B can be subdivided into ferrous rhodochrosite–sphalerite–pyrite, quartz–sulfosalt–sphalerite, calcite–pyrite, quartz–stibnite, and quartz-only stages of mineralization. The formation of brecciated galena and sphalerite ores during period A implies reworking of pre-existing Pb–Zn sulfides by Cenozoic tectonic deformation, whereas period B mineralization records extensive openspace filling during ore formation. Fluid inclusion microthermometric data indicate that both periods A and B were associated with low–medium temperature(187–267°C) and low salinity(4.00–10.18% wt. Na Cl equivalent) ore-forming fluids, although variations in the physical–chemical nature of the period B fluids suggest that this phase of mineralization was characterized by variable water/rock ratios. Microprobe analyses indicate that Fe concentrations in sphalerite decrease from period A to period B, and can be divided into three groups with Fe S concentrations of 8.999–9.577, 7.125–9.109, 5.438–1.460 mol.%. The concentrations of Zn, Sb, Pb, and Ag within orebodies in the study area are normally distributed in both lateral and vertical directions, and Pb, Sb, and/or Ag concentrations are positive correlation within the central part of these orebodies, but negatively correlate in the margins. Sulfide S isotope compositions are highly variable(4‰–13‰), varying from 4‰ to 11‰ in period A and 10‰ to 13‰ in period B. The Pb isotope within these samples is highly radiogenic and defines linear trends in 206 Pb/204 Pb vs. 207 Pb/204 Pb and 206 Pb/204 Pb vs. 208 Pb/204 Pb diagrams, respectively. The S and Pb isotopic characteristics indicate that the period B orebodies formed by mixing of Pb–Zn sulfides and regional Sbbearing fluids. These features are indicative of overprinting and remobilization of pre-existing Pb–Zn sulfides by Sb-bearing ore-forming fluids during a post-collisional period of the Himalayan Orogeny. | 13 | A viral vector expressing hypoxia-inducible factor 1 alpha inhibits hippocampal neuronal apoptosis显示文摘Hypoxia-inducible factor 1(HIF-1) attenuates amyloid-beta protein neurotoxicity and decreases apoptosis induced by oxidative stress or hypoxia in cortical neurons. In this study, we constructed a recombinant adeno-associated virus(rAAV) vector expressing the human HIF-1α gene(rAAV-HIF-1α), and tested the assumption that rAAV-HIF-1α represses hippocampal neuronal apoptosis induced by amyloid-beta protein. Our results confirmed that rAAV-HIF-1α significantly reduces apoptosis induced by amyloid-beta protein in primary cultured hippocampal neurons. Direct intracerebral rAAV-HIF-1α administration also induced robust and prolonged HIF-1α production in rat hippocampus. Single rAAV-HIF-1α administration resulted in decreased apoptosis of hippocampal neurons in an Alzheimer's disease rat model established by intracerebroventricular injection of aggregated amyloid-beta protein(25–35). | 14 | Immobilization of insulin-like growth factor-1 onto thermosensitive hydrogels to enhance cardiac progenitor cell survival and differentiation under ischemic conditions显示文摘Stem cell therapy is a promising approach to treat myocardial infarction. However, direct delivery of stem cells into hearts experiences poor cell engraftment and differentiation, due to ischemic conditions(low nutrient and oxygen) in the infarct hearts. Development of suitable cell carriers capable of supporting cell survival and differentiation under these harsh conditions is critical for improving the efficacy of current stem cell therapy. In this work, we created a family of novel cell carriers based on thermosensitive hydrogels and insulin-like growth factor 1(IGF-1), and investigated if these cell carriers can improve cell survival and differentiation under ischemic conditions. The thermosensitive hydrogels were synthesized from N-isopropylacryla- mide, acrylic acid, acrylic acid N-hydroxysuccinicimide ester, and 2-hydroxyethyl methacrylate-oligo(hydroxybutyrate). The hydrogel solutions can be readily injected through 26G needles, and can quickly solidify at 37 oC to form highly flexible hydrogels. IGF-1 was immobilized into the hydrogels in order to support long-term cell survival and differentiation. Different amount of IGF-1 was immobilized by using hydrogels with different content of N-hydroxysuccinicimide ester groups. Cardiosphere derived cells were encapsulated in the hydrogels and cultured under ischemic conditions. The results demonstrated that a significant improvement of cell survival and differentiation was achieved after IGF-1 immobilization. | 15 | Mineralogy and Chemistry of Sulfides from the Longqi and Duanqiao Hydrothermal Fields in the Southwest Indian Ridge显示文摘Recent investigations found that hydrothermal activity and sulfide mineralization occurs along the Southwest Indian Ridge(SWIR). The Longqi and Duanqiao hydrothermal fields between 49° E and 53° E of the SWIR are two prospective mineralization areas discovered by Chinese scientists. With the aim to determine the mineralogical and chemical characteristics of sulfide minerals, we have conducted detailed studies for samples from the two areas using an optical microscope, X-ray diffractometer, scanning electron microscope, and electron microprobe. The mineralization processes in the Longqi area are divided into three main stages:(1) the low-medium-temperature stage: colloform pyrite(Py I) + marcasite → euhedral pyrite(Py II),(2) the high-temperature stage: isocubanite(±exsolved chalcopyrite) + pyrrhotite → coarse-grained chalcopyrite(Ccp I), and(3) the medium–low-temperature stage: sphalerite + fine-grained chalcopyrite inclusions(Ccp II) → aggregates of anhedral pyrite(Py III) ± marcasite → Fe-oxide(-hydroxide) + amorphous silica. The mineralization processes in the Duanqiao area are divided into two main stages:(1) the medium–high-temperature stage: subhedral and euhedral pyrite(Py I′) → coarse-grained chalcopyrite(Ccp I′) and(2) the medium–low-temperature stage: sphalerite → fine-grained chalcopyrite(Ccp II′) + chalcopyrite inclusions(Ccp II′) → silica-cemented pyrite(Py II′) + marcasite → Fe-oxide + amorphous silica. We suggest that the fine-grained chalcopyrite inclusions in sphalerite from Longqi and Duanqiao were formed by co-precipitation and replacement mechanisms, respectively. Primary sphalerites from both fields are enriched in Fe(avg. 5.84 wt% for the Longqi field vs. avg. 3.69 wt% for the Duanqiao field), Co(avg. 185.56 ppm for the Longqi field vs. 160.53 ppm for the Duanqiao field), and Cd(avg. 1950 ppm for the Longqi field vs. avg. 525.26 ppm for the Duanqiao field). Cu contents in pyrite from the Duanqiao field(Py I′: avg. 849.23 ppm and Py II′: avg. 1191.11 ppm) tend to be higher than those from the Longqi field(Py I: avg. 26.67 ppm, Py II: avg. 445 ppm, and Py III: avg. 179.29 ppm). Chalcopyrite from both fields is enriched in Zn(Ccp I: avg. 3226.67 ppm, Ccp II: avg. 9280 ppm, Ccp I′: avg. 848 ppm, Ccp II′(inclusions): avg. 1098 ppm, and Ccp II′(fine-grained): avg. 1795 ppm). The varying contents of Zn in the different pyrite and chalcopyrite generations may result from the zone refining process. | 16 | Relationship between increase rate of human plague in China and global climate index as revealed by cross-spectral and cross-wavelet analyses显示文摘Plague has caused the death of hundreds of millions of people throughout the human history.Today this disease is again re-emerging and hence is again becoming an increasing threat to human health in several parts of the world.However,impacts of global climate variation(e.g.El Nino and Southern Oscillation[ENSO])and global warming on plagues are largely unknown.Using cross-spectral analysis and cross-wavelet analysis,we have analyzed the relationship between increase rate of human plague in China during 1871–2003 and the following climate factors(as measured by the Southern Oscillation Index[SOI],Sea Surface Temperature of east Pacific equator[SST]and air Temperature of the Northern Hemisphere[NHT]).We found in the frequency domain that increase rate of human plague was closely associated with SOI and SST.Cross-spectral analysis reveals that significant coherencies between increase rate of human plague and ENSO were found over short periods(2–3 years),medium periods(6–7 years)and long periods(11–12 years,30–40 years).Cross-wavelet analysis reveals that increase rate of human plague oscillates in phase with SOI,but in anti-phase with SST over periods of 2–4 years and approximately 8 years(6–10 years).These results indicate that ENSO-driven climate variation may be important for occurrences of human plague in China.However,there is a need for a further analysis of the underlying mechanism between human plague in China and ENSO. | Zhibin ZHANG Zhenqing LI Yi TAO Min CHEN Xinyu WEN Lei XU Huidong TIAN Nils Chr.STENSETH | 2007 | Integrative Zoology2007,2,3: | 3 |

| 18 | Thiocyanate-induced labilization of schwertmannite: Impacts and mechanisms显示文摘Schwertmannite is an amorphous iron(III)-oxyhydroxysulfate that forms in acid mine drainage(AMD) environments. The characteristic of high heavy metal adsorption capability makes schwertmannite a potentially useful, environmentally friendly material in wastewater treatment. Unstable schwertmannite is prone to recrystallization.Understanding the mechanisms that induce schwertmannite labilization and affect its capacity to remove heavy metals are of great environmental and geochemical significance.Thiocyanate(SCNˉ) is a hazardous pseudohalide that is also normally found in AMD.However, little is known about the impact of Fe(III)-binding ligand SCNˉ on schwertmannite stability and its subsequent capacity to bind trace elements. Here, we investigated the adsorption of SCNˉ on schwertmannite and subsequent mineral transformation to characterize this little-known process. The appearance of Fe2+indicated that the interactions between schwertmannite and SCNˉ may involve complexation and reduction reactions. Results showed that the majority of the adsorbed-SCNˉ was immobilized on schwertmannite during the 60-days transformation. The transformation rates of schwertmannite increased with increasing concentrations of SCNˉ. Goethite was detected as the dominant transformation product with or without SCNˉ. The mechanisms of SCNˉ-promoted dissolution of schwertmannite can be described as follows:(1) formation of Fe(III)–NCS complexes on the schwertmannite surface and in solution, a process which increases the reactivity of solid phase Fe(III);(2) the extraction of Fe(III) from schwertmannite by SCNˉ and subsequent schwertmannite dissolution; and(3) the formation of secondary minerals from extracted Fe(III). | 19 | Tectonically-controlled Evolution of the Late Cenozoic Nihewan Basin, North China Craton: Constraints from Stratigraphy, Mineralogy, and Geochemistry显示文摘The Late Cenozoic basins in the Weihe–Shanxi Graben, North China Craton are delineated by northeast-striking faults. The faults have, since a long time, been related to the progressive uplift and northeastward expansion of the Tibetan Plateau. To show the relation between the basins and faults, two Pliocene–Pleistocene stratigraphic sections(Chengqiang and Hongyanangou) in the southern part of the Nihewan Basin at the northernmost parts of the graben are studied herein. Based on the sedimentary sequences and facies, the sections are divided into three evolutionary stages, such as alluvial fan-eolian red clay, fan delta, and fluvial, with boundaries at ~2.8 and ~1.8 Ma. Paleocurrent indicators, the composition of coarse clastics, heavy minerals, and the geochemistry of moderate–fine clastics are used to establish the temporal and spatial variations in the source areas. Based on features from the middlenorthern basin, we infer that the Nihewan Basin comprises an old NE–SW elongate geotectogene and a young NW–SE elongate subgeotectogene. The main geotectogene in the mid-north is a half-graben bounded by northeast-striking and northwest-dipping normal faults(e.g., Liulengshan Fault). This group of faults was mainly affected by the Pliocene(before ~2.8–2.6 Ma) NW–SE extension and controlled the deposition of sediments. In contrast, the subgeotectogene in the south was affected by northwest-striking normal faults(e.g., Huliuhe Fault) that were controlled by the subsequent weak NE–SW extension in the Pleistocene. The remarkable change in the sedimentary facies and provenance since ~1.8 Ma is possibly a signal of either weak or strong NE–SW extension.
