1Brain-derived neurotrophic factor genes transfect rat bone marrow mesenchymal stem cells based on cationic polymer vector显示文摘BACKGROUND: Gene therapy is an effective expression of genes within target cells after transferring exogenous target genes. Both vector selection and transfection method are important factors for gene transfection. An ideal gene vector is required for a high transfusion of target gene and an exact introduction of target gene into specific target cells so as to express gene products. OBJECTIVE: To study the expression of mRNA and protein after transfecting rat bone marrow mesenchymal stem cells (BMSCs) with brain-derived neurotrophic factor (BDNF) genes based on cationic polymer vector. DESIGN, TIME AND SETTING: A randomized, controlled in vitro study using gene engineering, performed at the Neurobiology Laboratory, Xuzhou Medical College between October 2007 and April 2008. MATERIALS: PcDNA3.1 BDNF was obtained from Youbiai Biotechnological Company, Beijing and cationic polymer vector used was the SofastTM gene transfection reagent that was made by Taiyangma Biotechnological Co., Ltd., Xiamen. METHODS: BMSCs extracted from six Sprague Dawley (SD) rats aged 1 month were isolated and cultured in vitro. Third passage BMSCs were inoculated on a 6-well culture plate at the density of 1×106 cells/L. At about 80% confluence, BMSCs were transfected with PcDNA3.1-BDNF (2 μg) combined with SofastTM gene transfection reagent (6 μg) (BDNF group) or with PcDNA3.1 (2 μg) combined with SofastTM gene transfection reagent (6 μg) (blank vector group). Cells that were not transfected with any reagents but still cultured under primary culture conditions were used as a non-transfection group. MAIN OUTCOME MEASURES: Enzyme linked immunosorbent assay was used to measure time efficiency of BMSC-secreted BDNF protein. Twenty-four hours after gene transfection, RT-PCR was used to detect expression of BDNF mRNA in the BMSCs. Immunohistochemistry was used to determine expression of BDNF protein in the BMSCs. RESULTS: BDNF protein expression was detected at day 1 after gene transfection, rapidly increased after 5–9 days and gradually increased after 11–15 days in the BDNF group; moreover, BDNF protein expression was higher than that in the non-transfection group and the blank vector group at different time points (P < 0.01). Additionally, BDNF mRNA expression in the BDNF group was higher than that in the blank vector group and the non-transfection group (P < 0.01). CONCLUSION: A cationic polymer vector can effectively mediate the BDNF gene to transfect BMSCs; genetically modified BMSCs can express BDNF protein effectively for a long term.Zunsheng Zhang Kun Zan Yonghai Liu Xia Shen 2009Neural Regeneration Research2009,4,1:2
2Oxygen-glucose deprivation of neurons transfected with toll-like receptor 3-siRNA Determination of an optimal transfection sequence显示文摘Toll-like receptor 3 protein expression has been shown to be upregulated during cerebra ischemia/reperfusion injury in rats.In this study,rat primary cortical neurons were subjected to oxygen-glucose deprivation to simulate cerebral ischemia/reperfusion injury.Chemically synthesized small interfering RNA(siRNA)-1280,-1724 and-418 specific to toll-like receptor 3 were transfected into oxygen-glucose deprived cortical neurons to suppress the upregulation of toll-like receptor 3 protein expression.Western blotting demonstrated that after transfection with siRNA toll-like receptor 3 protein expression reduced,especially in the toll-like receptor 3-1724 group These results suggested that siRNA-1724 is an optimal sequence for inhibiting toll-like receptor 3expression in cortical neurons following oxygen-glucose deprivation.Guiyun Cui Xiaopeng Wang Xinchun Ye Jie Zu Kun Zan Fang Hua 2013Neural Regeneration Research2013,8,34:2
3Non-destructive study of fruits using grating-based X-ray imaging显示文摘Grating-based X-ray imaging can make use of conventional tube sources to provide absorption, refraction and scattering contrast images from a single set of projection images efficiently. In this paper, a fresh cherry tomato and a dried umeboshi are imaged by using X-ray Talbot–Lau interferometer. The seed distribution in the scattering image of the cherry tomato, and the wrinkles of epicarp in the refraction image of the umeboshi, are shown distinctly. The refraction and scattering images provide more information on subtle features than the absorption image. Also, the contrast-to-noise ratio values show distinguishing capacity of the three kinds of imaging techniques. 5137例MAID方案-线化疗转移软组织肉瘤的回顾性临床分析(英文)显示文摘Objective:The aim of this study was to evaluate the efficacy,adverse reaction and survival of MAID protocol as first-line treatment on 137 metastatic soft tissue sarcomas patients.Methods:One hundredand thirty-seven patients with metastatic soft tissue sarcoma were given MAID protocol chemotherapy which consists of ifosfamide at a total dose of 8-10 g/m2 in 4-5 days,adriamycin at a dose of 60 mg/m2 on first day and dacarbazine at a dose of 200 mg/m2/d in 4-5 days.Twenty one days were regarded as a cycle and 2-6 cycles were completed.The median cycle number was 4.Results:Four cases had complete remission(CR)(2.9%),thirteen cases achieved partial remission(PR)(9.5%),eighty-eight cases maintained a stable disease(SD)(64.2%),and thirty-two cases had progressive disease(PD)(23.4%).The overall response rate(RR) was 12.4%,disease control rate(DCR) was 76.6%.The progression-free survival(PFS) was 2-14 months and median PFS was 5.00 ± 1.12 months.The overall survival(OS) was 3-19 months and median OS was 8.00 ± 1.32 months.Three months PFS rate was 56.9%,six months PFS rate was 27.2% and one year PFS rate was 7.8%.Three months OS rate was 100.0%,six months OS rate was 41.5% and one year OS rate was 10.7%.The main adverse reactions were bone marrow supression,nausea/vomiting and alopecia.Other adverse reactions were seldom observed.Conclusion:The effect of MAID protocol as first-line treatment on metastatic soft tissue sarcomas patients was exact,the treatment can control disease progression effectively and the side effects can be tolerable.It was obviously very worthy in clinical use as one of the first-line chemotherapy protocols on advanced soft tissue sarcomas.Kun Han Yuanjue Sun Jianjun Zhang Lina Tan Shui'er Zheng Zhiyu Wang Hui Zhao Feng Lin Zan Shen Yang Yao 2012The Chinese-German Journal of Clinical Oncology2012,11,2:0
