| 1 | Effect of Boschniakia rossica on expression of GSTP,p53 and p21^(ras)proteins in early stage of chemical hepatocarcinogenesis and its anti-inflammatory activities in rats显示文摘AIM To investigate the effect of Boschniakiarossica(BR)extract on expression of GST-P,p53 and p21rasproteins in early stage of chemicalhepatocarcinogenesis in rats and its anti-inflammatory activities.METHODS The expression of tumor marker-placental form glutathione S-transferase(GST-P),p53 and p21rasproteins were investigated byimmunohistochemical techniques and ABCmethod.Anti-inflammatory activities of BR werestudied by xylene and croton oil-induced mouseear edema,carrageenin,histamine and hotscald-induced rat pow edema,adjuvant-inducedrat arthritis and cotton pellet-induced mousegranuloma formation methods.RESULTS The 500 mg/kg of BR-H2O extractfractionated from BR-Methanol extract hadinhibitory effect on the formation of DEN-inducedGST-P-positive foci in rat liver(GST-P stainingwas 78% positive in DEN+AAF group vs 20%positive in DEN+AAF+BR group,P<0.05)andthe expression of mutant p53 and p21rasproteinwas lower than that of hepatic preneoplasticlesions(33% and 22% positive respectively inDEN+AAF group vs negative in DEN+AAF+BRgroup).Both CH2Cl2 and H2O extracts from BRhad anti-inflamatory effect in xylene and crotonoil-induced mouse ear edema(inhibitory rateswere 26%-29% and 35%-59%,respectively). BR-H2O extract exhibited inhibitory effect incarrageenin,histamine and hot scald-inducedhind paw edema and adjuvant-induced arthritis inrats and cotton pellet-induced granulomaformation in mice.CONCLUSION BR extract exhibited inhibitory effect on formation of preneoplastic hepatic foci in early stage of rat chemical hepato-carcinogenesis. | 3 | Aberrant activation of latent transforming growth factor-β initiates the onset of temporomandibular joint osteoarthritis显示文摘There is currently no effective medical treatment for temporomandibular joint osteoarthritis(TMJ-OA) due to a limited understanding of its pathogenesis. This study was undertaken to investigate the key role of transforming growth factor-β(TGF-β)signalling in the cartilage and subchondral bone of the TMJ using a temporomandibular joint disorder(TMD) rat model, an ageing mouse model and a Camurati–Engelmann disease(CED) mouse model. In the three animal models, the subchondral bone phenotypes in the mandibular condyles were evaluated by μCT, and changes in TMJ condyles were examined by TRAP staining and immunohistochemical analysis of Osterix and p-Smad2/3. Condyle degradation was confirmed by Safranin O staining, the Mankin and OARSI scoring systems and type X collagen(Col X), p-Smad2/3 a and Osterix immunohistochemical analyses. We found apparent histological phenotypes of TMJ-OA in the TMD, ageing and CED animal models, with abnormal activation of TGF-βsignalling in the condylar cartilage and subchondral bone. Moreover, inhibition of TGF-β receptor I attenuated TMJ-OA progression in the TMD models. | 5 | Transcriptomic landscape regulated by the 14 types of bone morphogenetic proteins(BMPs)in lineage commitment and differentiation of mesenchymal stem cells(MSCs)显示文摘Mesenchymal stem cells(MSCs)are ubiquitously-existing multipotent progenitors that can self-renew and differentiate into multiple lineages including osteocytes,chondrocytes,adipocytes,tenocytes and myocytes.MSCs represent one of the most commonly-used adult progenitors and serve as excellent progenitor cell models for investigating lineagespecific differentiation regulated by various cellular signaling pathways,such as bone morphogenetic proteins(BMPs).As members of TGFb superfamily,BMPs play diverse and important roles in development and adult tissues.At least 14 BMPs have been identified in mammals.Different BMPs exert distinct but overlapping biological functions.Through a comprehensive analysis of 14 BMPs in MSCs,we demonstrated that BMP9 is one of the most potent BMPs in inducing osteogenic differentiation of MSCs.Nonetheless,a global mechanistic view of BMP signaling in regulating the proliferation and differentiation of MSCs remains to be fully elucidated.Here,we conducted a comprehensive transcriptomic profiling in the MSCs stimulated by 14 types of BMPs.Hierarchical clustering analysis classifies 14 BMPs into three subclusters:an osteo/chondrogenic/adipogenic cluster,a tenogenic cluster,and BMP3 cluster.We also demonstrate that six BMPs(e.g.,BMP2,BMP3,BMP4,BMP7,BMP8,and BMP9)can induce ISmads effectively,while BMP2,BMP3,BMP4,BMP7,and BMP11 up-regulate Smad-independent MAP kinase pathway.Furthermore,we show that many BMPs can upregulate the expression of the signal mediators of Wnt,Notch and PI3K/AKT/mTOR pathways.While the reported transcriptomic changes need to be further validated,our expression profiling represents the first-of-its-kind to interrogate a comprehensive transcriptomic landscape regulated by the 14 types of BMPs in MSCs. | Linghuan Zhang Qing Luo Yi Shu Zongyue Zeng Bo Huang Yixiao Feng Bo Zhang Xi Wang Yan Lei Zhenyu Ye Ling Zhao Daigui Cao Lijuan Yang Xian Chen Bin Liu William Wagstaff Russell R*Reid Hue H*Luu Rex C*Haydon Michael J*Lee Jennifer Moriatis Wolf Zhou Fu Tong-Chuan He Quan Kang | 2019 | Genes & Diseases2019,6,3: | 11 |

Herein, we provide a comprehensive overview of the state-of-the-art advances in liquid crystal-templated chiral nanomaterials. The chiroptical properties of chiral nanomaterials are touched, and their fundamental design principles and bottom-up synthesis strategies are discussed. Different chiral functional nanomaterials based on liquid-crystalline soft templates, including chiral plasmonic nanomaterials and chiral luminescent nanomaterials, are systematically introduced, and their underlying mechanisms, properties, and potential applications are emphasized. This review concludes with a perspective on the emerging applications, challenges, and future opportunities of such fascinating chiral nanomaterials. Some previous studies showed that SPs possessed cytotoxicity in some immune cells such as human lymphocytes and rat bone marrow. However, the cytotoxicity of SPs to macrophages, which are crucial to innate immunity, has not been explored. In the present report, we investigated a new pyrethroid insecticide, lambda-cyhalothrin (LCT), which may increase the generation of reactive oxygen species (ROS) and DNA damage levels and cause cytotoxicity in RAW 264.7 cells in dose- and time-dependent manners. The results for the first time implicated increased endogenous ROS and DNA damage as co-mediators of LCT-induced cytotoxicity in macrophages. Our results also suggested that macrophages were involved in synthetic pyrethroid-induced adverse immune effects. Considering the ubiquitous environmental presence of SPs, this study provided new information relative to the potential long-term physiological and immunological effects associated with chronic exposure to SPs. | 20 | Integrating morphological spatial pattern analysis and the minimal cumulative resistance model to optimize urban ecological networks: a case study in Shenzhen City, China显示文摘Background:With the increasing fragmentation of landscape induced by rapid urbanization,the construction of ecological networks is of great signifcance to alleviate the degradation of urban habitats and protect natural envi‑ronments.However,there is considerable uncertainty when constructing ecological networks,especially the difer‑ent approaches to selecting ecological sources.We used the southern Chinese city of Shenzhen as a study area to construct and optimize ecological networks using a coupling approach.Ecological source areas were extracted using morphological spatial pattern analysis(MSPA)and the landscape index method.Ecological networks were con‑structed using the minimal cumulative resistance(MCR)model and the gravity model.Stepping stones and ecological fault points were added in corridors to optimize the ecological network.Results:Ten core areas with maximum importance patch values were extracted by the landscape index method as ecological source areas according to MSPA,after which corridors between ecological sources were constructed based on the MCR model.The constructed ecological networks were optimized using 35 stepping stones and 17 ecologi‑cal fault points.The optimized ecological networks included 11 important corridors,34 general corridors,and seven potential corridors.The results of corridor landscape-type analysis showed that a suitable ecological corridor is 60 to 200 m wide.Conclusions:Overall,our results imply that ecological source areas can be identifed virtually,and that ecological networks can be signifcantly optimized by combining MSPA and MCR models.These results provide a methodologi‑cal reference for constructing ecological networks,and they will be useful for urban planning and biodiversity protec‑tion in Shenzhen and other similar regions around the world. | Yang‑Yang Li Yu‑Zhe Zhang Zhi‑Yun Jiang Cheng‑Xuan Guo Ming‑Yue Zhao Zhi‑Guang Yang Ming‑Yan Guo Bing‑Yue Wu Quan‑Ling Chen | 2021 | Ecological Processes2021,10,1: | 3 |
