| 1 | GSDMB promotes non-canonical pyroptosis by enhancing caspase-4 activity显示文摘Gasdermin B (GSDMB) has been reported to be associated with immune diseases in humans, but the detailed molecular mechanisms remain unsolved. The N-terminus of GSDMB by itself, unlike other gasdermin family proteins, does not induce cell death. Here, we show that GSDMB is highly expressed in the leukocytes of septic shock patients, which is associated with increased release of the gasdermin D (GSDMD) N-terminus. GSDMB expression and the accumulation of the N-terminal fragment of GSDMD are induced by the activation of the non-canonical pyroptosis pathway in a human monocyte cell line. The downregulation of GSDMB alleviates the cleavage of GSDMD and cell death. Consistently, the overexpression of GSDMB promotes GSDMD cleavage, accompanied by increased LDH release. We further found that GSDMB promotes caspase-4 activity, which is required for the cleavage of GSDMD in non-canonical pyroptosis, by directly binding to the CARD domain of caspase-4. | 2 | Mitochondrial ROS promote macrophage pyroptosis by inducing GSDMD oxidation显示文摘Disrupted mitochondrial membrane potential (MMP) and reactive oxygen species (ROS) generation are often associated with macrophage pyroptosis. It remains unclear how these forms of mitochondrial dysfunction relate to inflammasome activation and gasdermin-D (Gsdmd) cleavage, two central steps of the pyroptotic process. Here, we also found MMP collapse and ROS generation induced by Nlrp3 inflammasome activation as previous studies reported. The elimination of ROS alleviated the cleavage of Gsdmd, suggesting that Gsdmd cleavage occurs downstream of ROS release. Consistent with this result, hydrogen peroxide treatment augmented the cleavage of Gsdmd by caspase-1. Indeed, four amino acid residues of Gsdmd were oxidized under oxidative stress in macrophages. The efficiency of Gsdmd cleavage by inflammatory caspase-1 was dramatically reduced when oxidative modification was blocked by mutation of these amino acid residues. | 4 | An injectable and self-healing hydrogel with controlled release of curcumin to repair spinal cord injury显示文摘The harsh local micro-environment following spinal cord injury(SCI)remains a great challenge for neural regeneration.Local reconstitution of a favorable micro-environment by biocompatible scaffolds with desirable functions has thus been an area of concern.Herein,a hybrid hydrogel was developed using Fmoc-grafted chitosan(FC)and Fmoc peptide(FI).Dynamic reversibleπ-πstacking interactions of the fluorenyl rings enabled the FC/FI hybrid hydrogel to exhibit excellent injectable and self-healing properties,as characterized by visual appearances and rheological tests.Furthermore,the FC/FI hybrid hydrogel showed a slow and persistent release of curcumin(Cur),which was named as FC/FI-Cur hydrogel.In vitro studies confirmed that with the support of FC/FI-Cur hydrogel,neurite outgrowth was promoted,and Schwann cell(SC)migration away from dorsal root ganglia(DRG)spheres with enhanced myelination was substantiated.The FC/FI-Cur hydrogel well reassembled extracellular matrix at the lesion site of rat spinal cord and exerted outstanding effects in modulating local inflammatory reaction by regulating the phenotypes of infiltrated inflammatory cells.In addition,endogenous SCs were recruited in the FC/FI-Cur graft and participated in the remyelination process of the regenerated nerves.These outcomes favored functional recovery,as evidenced by improved hind limbs movement and enhanced electrophysiological properties.Thus,our study not only advanced the development of multifunctional hydrogels but also provided insights into comprehensive approaches for SCI repair. | Jinghua Luo Xueshuang Shi Liming Li Zan Tan Feng Feng Jun Li Mao Pang Xiaoying Wang Liumin He | 2021 | Bioactive Materials2021,6,12: | 4 |
| 5 | Oil production rate predictions for steam assisted gravity drainage based on high-pressure experiments显示文摘Dual-well steam assisted gravity drainage(SAGD) has significant potential for extra-heavy oil recovery.China is conducting two dual-well SAGD pilot projects in the Fengcheng extra-heavy oil reservoir.Quick,direct predictions of the oil production rate by algebraic models rather than complex numerical models are of great importance for designing and adjusting the SAGD operations.A low-pressure scaled physical simulation was previously used to develop two separate theoretical models corresponding to the two different growth stages observed in the SAGD steam chambers,which are the steam chamber rising stage and the steam chamber spreading stage.A high-pressure scaled model experiment is presented here for one dual-well SAGD pattern to further improve the prediction models to reasonably predict oil production rates for full production.Parameters that significantly affect the oil recovery during SAGD were scaled for the model size based on the reservoir characteristics of the Fengcheng reservoir in China.Experimental results show the relationship between the evolution of the steam chamber and the oil production rate during the entire production stage.High-pressure scaled model test was used to improve the gravity drainage models by modifying empirical factors for the rising model and the depletion model.A new division of the SAGD production regime was developed based on the relationship between the oil production rate and the evolution of steam chamber.A method was developed to couple the rising and depletion models to predict oil production rates during the SAGD production,especially during the transition period.The method was validated with experiment data and field data from the literature.The model was then used to predict the oil production rate in the Fengcheng reservoir in China and the Athabasca reservoir in Canada. | GUO Jia ZAN Cheng MA DeSheng SHI Lin | 2013 | Science China(Technological Sciences)2013,56,2: | 2 |

| 7 | Compound Gaoziban tablet(复方高滋斑片) alleviates depression via toll-like receptor 4/myeloid differentiation factor 88/nuclear factor-kappa B pathway显示文摘OBJECTIVE: To evaluate the effect of compound Gaoziban tablet(复方高滋斑片, CGZBT) on depression, and to investigate the underlying mechanism. METHODS: The components of CGZBT were analysed by high-performance liquid chromatography. Then, we assessed the effects of varying doses of CGZBT on an established chronic unpredictable mild stress(CUMS) model in rats. Whether animals were depressed was evaluated by sucrose preference test, open field test and forced swimming test. Neurotransmitters of hippocampus were detected by liquid chromatography-mass spectrometry. Serum levels of tumor necrosis factor-alpha(TNF-α), interleukin(IL)-1β, IL-6, IL-4, and IL-10 were measured by enzyme-linked immunosorbent assay. Expressions of toll-like receptor 4(TLR4), myeloid differentiation factor 88(My D88), phospho-nuclear factorkappa B(p-NF-κB), cyclooxygenase-2(COX-2), ionized calcium binding adapter molecule-1(IBA-1) were assessed by immunohistochemical staining and western blotting. RESULTS: Eight compounds were identified from CGZBT, moreover, our results showed that CGZBT effectively reversed the CUMS-induced decrease in sucrose preference, shortened the movement distance and prolonged immobility time. CGZBT significantly increased levels of 5-hydroxytryptamine, dopamine, norepinephrine, 5-hydroxyindoleacetic acid levels, and reduced the expression of TNF-α, IL-1β, IL-6, yet increased IL-4 and IL-10. Furthermore, the expressions of TLR4, My D88, COX-2, p-NF-κB and IBA-1 in hippocampus were effectively reversed after treatment with CGZBT.
