| 2 | LncRNAs-directed PTEN enzymatic switch governs epithelial–mesenchymal transition显示文摘Despite the structural conservation of PTEN with dual-specificity phosphatases,there have been no reports regarding the regulatory mechanisms that underlie this potential dual-phosphatase activity.Here,we report that K27-linked polyubiquitination of PTEN at lysines 66 and 80 switches its phosphoinositide/protein tyrosine phosphatase activity to protein serine/threonine phosphatase activity.Mechanistically,high glucose,TGF-β,CTGF,SHH,and IL-6 induce the expression of a long non-coding RNA,GAEA(Glucose Aroused for EMT Activation),which associates with an RNA-binding E3 ligase,MEX3C,and enhances its enzymatic activity,leading to the K27-linked polyubiquitination of PTEN.The MEX3C-catalyzed PTEN^K27-polyUb activates its protein serine/threonine phosphatase activity and inhibits its phosphatidylinositol/protein tyrosine phosphatase activity.With this altered enzymatic activity,PTEN^K27-polyUb dephosphorylates the phosphoserine/threonine residues of TWIST1,SNAI1,and YAP1,leading to accumulation of these master regulators of EMT.Animals with genetic inhibition of PTEN^K27-polyUb,by a single nucleotide mutation generated using CRISPR/Cas9(Pten^K80R/K80R),exhibit inhibition of EMT markers during mammary gland morphogenesis in pregnancy/lactation and during cutaneous wound healing processes.Our findings illustrate an unexpected paradigm in which the lncRNA-dependent switch in PTEN protein serine/threonine phosphatase activity is important for physiological homeostasis and disease development. | Qingsong Hu Chunlai Li Shouyu Wang Yajuan Li Bo Wen Yanyan Zhang Ke Liang Jun Yao Youqiong Ye Heidi Hsiao Tina K.Nguyen Peter K.Park Sergey D.Egranov David H.Hawke Jeffrey R.Marks Leng Han Mien-Chie Hung Bing Zhang Chunru Lin Liuqing Yang | 2019 | Cell Research2019,29,4: | 8 |

| 5 | QTL Mapping for Rice RVA Properties Using High-Throughput Re-sequenced Chromosome Segment Substitution Lines显示文摘The rapid visco analyser(RVA) profile is an important factor for evaluation of the cooking and eating quality of rice. To improve rice quality, the identification of new quantitative trait loci(QTLs) for RVA profiling is of great significance. We used a japonica rice cultivar Nipponbare as the recipient and indica rice 9311 as the donor to develop a population containing 38 chromosome segment substitution lines(CSSLs) genotyped by a high-throughput re-sequencing strategy. In this study, the population and the parent lines, which contained similar apparent amylose contents, were used to map the QTLs of RVA properties including peak paste viscosity(PKV), hot paste viscosity(HPV), cool paste viscosity(CPV), breakdown viscosity(BKV), setback viscosity(SBV), consistency viscosity(CSV), peak time(PeT) and pasting temperature(PaT). QTL analysis was carried out using one-way analysis of variance and Dunnett's test, and stable QTLs were identified over two years and under two environments. We identified 10 stable QTLs: qPKV2-1, qSBV2-1; qPKV5-1, qHPV5-1, qCPV5-1; qPKV7-1, qHPV7-1, qCPV7-1, qSBV7-1; and qPKV8-1 on chromosomes 2, 5, 7 and 8, respectively, with contributions ranging from-95.6% to 47.1%. | 7 | Risk stratification in symptomatic intracranial atherosclerotic disease with conventional vascular risk factors and cerebral haemodynamics显示文摘Background and purpose Symptomatic intracranial atherosclerotic stenosis(sICAS)is associated with a considerable risk of recurrent stroke despite contemporarily optimal medical treatment.Severity of luminal stenosis in sICAS and its haemodynamic significance quantified with computational fluid dynamics(CFD)models were associated with the risk of stroke recurrence.We aimed to develop and compare stroke risk prediction nomograms in sICAS,based on vascular risk factors and these metrics.Methods Patients with 50%-99%sICAS confirmed in CT angiography(CTA)were enrolled.Conventional vascular risk factors were collected.Severity of luminal stenosis in sICAS was dichotomised as moderate(50%-69%)and severe(70%-99%).Translesional pressure ratio(PR)and wall shear stress ratio(WSSR)were quantified via CTA-based CFD modelling;the haemodynamic status of sICAS was classified as normal(normal PR&WSSR),intermediate(otherwise)and abnormal(abnormal PR&WSSR).All patients received guideline-recommended medical treatment.We developed and compared performance of nomograms composed of these variables and independent predictors identified in multivariate logistic regression,in predicting the primary outcome,recurrent ischaemic stroke in the same territory(SIT)within 1 year.Results Among 245 sICAS patients,20(8.2%)had SIT.The D2H2A nomogram,incorporating diabetes,dyslipidaemia,haemodynamic status of sICAS,hypertension and age≥50 years,showed good calibration(P for Hosmer-Lemeshow test=0.560)and discrimination(C-statistic 0.73,95%CI 0.60 to 0.85).It also had better performance in risk reclassification and provided larger net benefits in decision curve analysis,compared with nomograms composed of conventional vascular risk factors only,and plus the severity of luminal stenosis in sICAS.Sensitivity analysis in patients with anterior-circulation sICAS showed similar results.Conclusions The D2H2A nomogram,incorporating conventional vascular risk factors and the haemodynamic significance of sICAS as assessed in CFD models,could be a useful tool to stratify sICAS patients for the risk of recurrent stroke under contemporarily optimal medical treatment. | Xuan Tian Hui Fang Linfang Lan Hing Lung Ip Jill Abrigo Haipeng Liu Lina Zheng Florence S Y Fan Sze Ho Ma Bonaventure Ip Bo Song Yuming Xu Jingwei Li Bing Zhang Yun Xu Yannie O Y Soo Vincent Mok Ka Sing Wong Thomas W Leung Xinyi Leng | 2023 | Stroke & Vascular Neurology2023,8,1: | 2 |
| 8 | Power scaling on tellurite glass Raman fibre lasers for mid-infrared applications显示文摘The power scaling on mid-infrared Raman fibre lasers(RFLs) is in demand for applications in health, environment and security. In this paper, we present the simulated laser behaviours of the tellurite glass RFLs pumped by 300-W Tm-doped fibre lasers(TDFLs) at 2 μm for the first time. By combining the advantages of the TDFLs and tellurite fibre, the output power at 2.35 μm has reached over hundreds of watts by first-order Raman shift. Moreover, the cascaded RFLs have been demonstrated with a wavelength extension greater than 3 μm and output power of tens of watts.
