| 1 | Fusing T5 exonuclease with Cas9 and Cas12a increases the frequency and size of deletion at target sites显示文摘CRISPR/Cas systems, especially CRISPR/Cas9, generally result in small insertions/deletions, which are unlikely to eliminate the functions of regulatory and other non-coding sequences. To generate larger genomic deletions usually requires the use of pairs of guide RNAs. Here we show that it is possible to create such deletions with a single guide RNA by fusing Cas9 or Cas12a with T5 exonuclease(T5exo). These fusion constructs were found to increase both the frequency and size of deletions at target loci in rice protoplasts and seedlings. Moreover, the genome editing efficiencies of Cas9 and Cas12a were also enhanced by fusion with T5 exonuclease. These T5exo-Cas fusions expand the CRISPR toolbox, and facilitate knockout of regulatory and non-coding DNA sequences. | 2 | Effect of Hydrological Connectivity on Soil Carbon Storage in the Yellow River Delta Wetlands of China显示文摘Hydrological connectivity has significant effects on the functions of estuarine wetland ecosystem.This study aimed to examine the dynamics of hydrological connectivity and its impact on soil carbon pool in the Yellow River Delta,China.We calculated the hydrological connectivity based on the hydraulic resistance and graph theory,and measured soil total carbon and organic carbon under four different hydrological connectivity gradients(Ⅰ0‒0.03,Ⅱ0.03‒0.06,Ⅲ0.06‒0.12,Ⅳ0.12‒0.39).The results showed that hydrological connectivity increased in the north shore of the Yellow River and the south tidal flat from 2007 to 2018,which concentrated in the mainstream of the Yellow River and the tidal creek.High hydrological connectivity was maintained in the wetland restoration area.The soil total carbon storage and organic carbon storage significantly increased with increasing hydrological connectivity fromⅠtoⅢgradient and decreased inⅣgradient.The highest soil total carbon storage of 0‒30 cm depth was 5172.34 g/m^(2),and organic carbon storage 2764.31 g/m^(2)inⅢgradient.The hydrological connectivity changed with temporal and spatial change during 2007‒2018 and had a noticeable impact on soil carbon storage in the Yellow River Delta.The results indicated that appropriate hydrological connectivity,i.e.0.08,could effectively promote soil carbon storage. | FENG Jiuge LIANG Jinfeng LI Qianwei ZHANG Xiaoya YUE Yi GAO Junqin | 2021 | Chinese Geographical Science2021,31,2: | 4 |
| 3 | Relationship between COX-2 and cell cycle-regulatory proteins in patients with esophageal squamous cell carcinoma显示文摘AIM: To investigate the correlation between cyclooxygenase-2 (COX-2) and cell cycle-regulatory proteins in patients with esophageal squamous cell carcinoma (ESCC). METHODS: One hundred and two surgically obtained specimens of ESCC were randomly collected. All specimens were obtained from patients who had not received chemoor radiotherapy prior to surgical resection.Twenty-eight specimens of normal squamous epithelium served as controls. The expression of COX-2, Ki-67, cyclin A and p27 was examined by immunohistochemistry. The Pearson test was used to analyze the relationship between groups. RESULTS: The protein level of COX-2, Ki-67 and cyclin A was significantly higher in ESCC than in normal squamous epithelium (74.7±61.2 vs 30.2 ± 43.4, 64.0 ± 51.6 vs 11.6 ± 2.3, 44.2 ± 32.2 vs 11.7 ± 5.0, respectively, all P<0.01). In contrast, the protein level of p27 was signifi cantly lower in ESCC than in normal squamous epithelium (182.0 ±69.0 vs 266.4±28.0, P<0.01). In ESCC, COX-2 expression was correlated with T stage, the score of T1-T2 stage was lower than that of T3-T4 stage (55.0±42.3 vs 83.0 ± 66.5, P<0.05), and Ki-67, cyclin A and p27 expressions were correlated with the tumor differentiation (43.8±31.7 vs 98.4± 84.8, 32.0 ± 19.0 vs 54.1 ±53.7,206.2±61.5 vs 123.5±68.3, respectively, all P<0.01). | 8 | In-situ coupling construction of interface bridge to enhance electrochemical stability of all solid-state lithium metal batteries显示文摘Polymer-based composite electrolytes composed of three-dimensional Li_(6.4)La_(3)Zr_(2)Al_(0.2)O_(12)(3D-LLZAO)have attracted increasing attention due to their continuous ion conduction and satisfactory mechanical properties.However,the organic/inorganic interface is incompatible,resulting in slow lithium-ion transport at the interface.Therefore,the compatibility of organic/inorganic interface is an urgent problem to be solved.Inspired by the concept of“gecko eaves”,polymer-based composite solid electrolytes with dense interface structures were designed.The bridging of organic/inorganic interfaces was established by introducing silane coupling agent(3-chloropropyl)trimethoxysilane(CTMS)into the PEO-3D-LLZAO(PL)electrolyte.The in-situ coupling reaction improves the interface affinity,strengthens the organic/inorganic interaction,reduces the interface resistance,and thus achieves an efficient interface ion transport network.The prepared PEO-3D-LLZAO-CTMS(PLC)electrolyte exhibits enhanced ionic conductivity of 6.04×10^(-4)S cm^(-1)and high ion migration number(0.61)at 60℃and broadens the electrochemical window(5.1 V).At the same time,the PLC electrolyte has good thermal stability and high mechanical properties.Moreover,the Li Fe PO_(4)|PLC|Li battery has excellent rate performance and cycling stability with a capacity decay rate of 2.2%after 100 cycles at 60℃and 0.1 C.These advantages of PLC membranes indicate that this design approach is indeed practical,and the in-situ coupling method provides a new approach to address interface compatibility issues. | Qianwei Zhang Rong Yang Chao Li Lei Mao Bohai Wang Meng Luo Yinglin Yan Yiming Zou Lisheng Zhong Yunhua Xu | 2024 | Journal of Energy Chemistry2024,89,2: | 0 |

| 12 | Biomineralized nanoparticles for the immobilization and degradation of crude oil-contaminated soil显示文摘Accidental oil leaks and spills often cause server soil pollution,and in situ remediation is a powerful and economical treatment technology.While during in situ remediation process,unpredicted migration of petroleum hydrocarbon in heterogeneous soil will lead to a long-term source of persistent aquifer contamination.To reduce the migration of petroleum hydrocarbon and effectively improve the in situ remediation efficiency,herein,fungal biomineralization strategy was proposed for the immobilization of petroleum contaminants.A ureolytic fungi strain with crude oil-degradation ability was screened and identified as Chaetomium globosum.When incubated in medium containing Ca2+and crude oil,a mineral corona with spiny nanoparticles was formed at the edge of oil and the interface characters were analyzed using fluorescent pH and dissolved oxygen(DO)sensing films,respectively.Results indicated that biominerals preferred to aggregate around the edge of crude oil,providing favorable microenvironment for fungal growth and then leading to the increase of pH in the microenvironment,eventually accompanied by the formation of mineral corona.The mineral corona with numerous nanoparticles may act as a solid and stable shell,limiting or reducing the mobility of crude oil,and providing enough time for fungal biodegradation.After 28 days incubation,oilcontaminated soil treated with fungal biomineralization showed better immobilization ability for total petroleum hydrocarbon(TPH)under simulated acid-rain condition and higher TPH removal efficiency.This is the first demonstration for the immobilization of oil through fungal biomineralized nanoparticles,thus providing a novel strategy for the in situ remediation of oilcontaminated sites. | Daoqing Liu Qianwei Li Enhui Liu Miao Zhang Jicheng Liu Chunmao Chen | 2023 | Nano Research2023,16,10: | 0 |
| 13 | Calcification of planktonic foraminifer Neogloboquadrina pachyderma(sinistral) controlled by seawater temperature rather than ocean acidification in the Antarctic Zone of modern Sothern Ocean显示文摘Neogloboquadrina pachyderma(sinistral), the dominant planktonic foraminiferal species in the mid-to-high latitude oceans, represents a major component of local calcium carbonate(CaCO) production. However, the predominant factors,governing the calcification of this species and its potential response to the future marine environmental changes, are poorly understood. The present study utilized an improved cleaning method for the size-normalized weight(SNW) measurement to estimate the SNW of N. pachyderma(sin.) in surface sediments from the Amundsen Sea, the Ross Sea, and the Prydz Bay in the Antarctic Zone of the Southern Ocean. It was found that SNW of N. pachyderma(sin.) is not controlled by deep-water carbonate dissolution post-mortem, and can be therefore, used to reflect the degree of calcification. The comparison between N. pachyderma(sin.) SNW and environmental parameters(temperature, salinity, nutrient concentration, and carbonate system) in the calcification depth revealed that N. pachyderma(sin.) SNWs in the size ranges of 200–250, 250–300, and 300–355 μm are significantly and positively correlated with seawater temperature. Moreover, SNW would increase by ~30% per degree increase in temperature, thereby suggesting that the calcification of N. pachyderma(sin.) in the modern Antarctic Zone of the Southern Ocean is mainly controlled by temperature, rather than by other environmental parameters such as ocean acidification. Importantly, a potential increase in calcification of N. pachyderma(sin.) in the Antarctic Zone to produce CaCOwill release COinto the atmosphere. | 15 | Effect of pressure cooking on phenolic compounds of quinoa显示文摘Due to good nutritional properties and potential health benefits,quinoa has gained an increasing attention.The study aimed to analyze the effect of pressure cooking on the composition,antioxidant activity,antibacterial activity and bioavailability of phenolic compounds in four types of quinoa,and to evaluate the correlation between phenolics and its biological activities by correlation analysis.The results showed that different varieties of quinoa contained different phenolic components and their biological activities were different.Pressure cooking could significantly increase(P<0.05)the phenolic contents of quinoa,and decrease in vitro digestibility of protein.The antioxidant activity,antibacterial activity and bioavailability of quinoa were also enhanced which were positively related with phenolic contents.HPLC analysis indicated that at least twelve phenolic compounds were found in quinoa,and hyperoside,sinapic acid,rutin and ferulic acid occupied a majority of them.Correlation analysis suggested that hyperoside,quercetin,sinapic acid,ferulic acid and gallic acid made the key contribution to antioxidant and antibacterial activities of phenolic compounds of quinoa.The results provided valuable information for quinoa processing with phenolics as functional ingredient. | Qianwei Ma Zhixiang Li Thanutchaporn Kumrungsee Wenda Huang Ruge Cao | 2023 | Grain & Oil Science and Technology2023,6,3: | 0 |
