1Leonurine protects ischemia-induced brain injury via modulating SOD, MDA and GABA levels显示文摘The present study was designed to investigate the protective effects of leonurine, a compound purified from Herba Leonuri that is active on ischemic rat behavior and cortical neurons, and explore the underlying mechanism. The general rat activity, cortical neuron morphology, superoxide dismutase (SOD), malondialdehyde (MDA),γ- aminobutyric acid (GABA) and glutamate decarboxylase 67 (GAD67) levels were measured. We found leonurine significantly improve the general activity of rats in an opefield test, which was associated with attenuated neuronal damage induced by ischemia. Moreover, serum SOD activity was significantly greater, MDA level lower in the leonurine group as compared with ischemia group. In addition, GABA content in the cerebral cortex was significantly greater in high-dose leonurine group. Correspondingly, GAD67 protein level coincided with the GABA level. 2Characteristics of secondary inorganic aerosol and sulfate species in size-fractionated aerosol particles in Shanghai显示文摘Sulfate, nitrate and ammonium(SNA) are the dominant species in secondary inorganic aerosol, and are considered an important factor in regional haze formation. Size-fractionated aerosol particles for a whole year were collected to study the size distribution of SNA as well as their chemical species in Shanghai. SNA mainly accumulated in fine particles and the highest average ratio of SNA to particulate matter(PM) was observed to be 47% in the fine size fraction(0.49–0.95 μm). Higher sulfur oxidation ratio and nitrogen oxidation ratio values were observed in PM of fine size less than 0.95 μm. Ion balance calculations indicated that more secondary sulfate and nitrate would be generated in PM of fine size(0.49–0.95 μm). Sulfur K-edge X-ray absorption near-edge structure(XANES) spectra of typical samples were analyzed. Results revealed that sulfur mainly existed as sulfate with a proportion(atomic basis) more than 73% in all size of PM and even higher at 90% in fine particles. Sulfate mainly existed as(NH4)2SO4 and gypsum in PM of Shanghai. Compared to non-haze days, a dramatic increase of(NH4)2SO4 content was found in fine particles on haze days only, which suggested the promoting impact of(NH4)2SO4 on haze formation. According to the result of air mass backward trajectory analysis, more(NH4)2SO4 would be generated during the periods of air mass stagnation. 3Sulfur speciation and bioaccumulation in camphor tree leaves as atmospheric sulfur indicator analyzed by synchrotron radiation XRF and XANES显示文摘Analyzing and understanding the effects of ambient pollution on plants is getting more and more attention as a topic of environmental biology.A method based on synchrotron radiation X-ray fluorescence and X-ray absorption near edge structure spectroscopy was established to analyze the sulfur concentration and speciation in mature camphor tree leaves (CTLs),which were sampled from 5 local fields in Shanghai,China.Annual SO2 concentration,SO42-concentration in atmospheric particulate,SO42-and sulfur concentration in soil were also analyzed to explore the relationship between ambient sulfur sources and the sulfur nutrient cycling in CTLs.Total sulfur concentration in mature camphor tree leaves was 766-1704 mg/kg.The mainly detected sulfur states and their corresponding compounds were +6 (sulfate,include inorganic sulfate and organic sulfate),+5.2 (sulfonate),+2.2 (suloxides),+0.6 (thiols and thiothers),+0.2 (organic sulfides).Total sulfur concentration was strongly correlated with sulfate proportion with a linear correlation coefficient up to 0.977,which suggested that sulfur accumulated in CTLs as sulfate form.Reduced sulfur compounds (organic sulfides,thiols,thioethers,sulfoxide and sulfonate) assimilation was sufficed to meet the nutrient requirement for growth at a balanced level around 526 mg/kg.The sulfate accumulation mainly caused by atmospheric sulfur pollution such as SO2 and airborne sulfate particulate instead of soil contamination.From urban to suburb place,sulfate in mature CTLs decreased as the atmospheric sulfur pollution reduced,but a dramatic increase presented near the seashore,where the marine sulfate emission and maritime activity pollution were significant.The sulfur concentration and speciation in mature CTLs effectively represented the long-term biological accumulation of atmospheric sulfur pollution in local environment.Jianrong Zeng Guilin Zhang Liangman Bao Shilei Long Mingguang Tan Yan Li Chenyan Ma Yidong Zhao 2013Journal of Environmental Sciences2013,25,3:9
4Geological cycling of potassium and the K isotopic response:insights from loess and shales显示文摘Shales are a major sink for K into seawater delivered from continental weathering,and are potential recorders of K cycling.High precision K isotope analyses reveal a[0.6%variation in δ^41K values(41K/39K relative to NIST SRM 3141a)from a set of well characterized postArchean Australian shale(PAAS)samples.By contrast,loess samples have relatively homogenous δ^41K values(-0.5±0.1%),which may represent the average K composition of upper continental crust.Most of the shales analyzed in this study have experienced K enrichment relative to average continental crust,and the majority of them define a trend of decreasing δ^41K value(from-0.5to-0.7%)with increasing K content and K/Na ratio,indicating cation exchange in clays minerals is accompanied by K isotope fractionation.Several shale samples do not follow the trend and have elevated δ^41K values up to-0.1%,and these samples are characterized by variable Fe isotope compositions,which reflect post-depositional processes.The K isotope variability observed in shales,in combination with recent findings about K isotope fractionation during continental weathering,indicates that K isotopes fractionate during cycling of K between different reservoirs,and K isotopes in sediments may be used to trace geological cycling of K.Weiqiang Li Shilei Li Brian L.Beard 2019Acta Geochimica2019,38,4:8
5Sediment pollution characteristics and in situ control in a deep drinking water reservoir显示文摘Sediment pollution characteristics, in situ sediment release potential, and in situ inhibition of sediment release were investigated in a drinking water reservoir. Results showed that organic carbon(OC), total nitrogen(TN), and total phosphorus(TP) in sediments increased from the reservoir mouth to the main reservoir. Fraction analysis indicated that nitrogen in ion exchangeable form and Na OH-extractable P(Fe/Al-P) accounted for 43% and 26% of TN and TP in sediments of the main reservoir. The Risk Assessment Code for metal elements showed that Fe and Mn posed high to very high risk. The results of the in situ reactor experiment in the main reservoir showed the same trends as those observed in the natural state of the reservoir in 2011 and 2012; the maximum concentrations of total OC, TN, TP, Fe,and Mn reached 4.42 mg/L, 3.33 mg/L, 0.22 mg/L, 2.56 mg/L, and 0.61 mg/L, respectively. An in situ sediment release inhibition technology, the water-lifting aerator, was utilized in the reservoir. 6Cromakalin pretreatment affects mitochondrial structure and function in a rat model of ischemia/reperfusion injury显示文摘BACKGROUND:Mitochondrial structural changes and energy dysmetabolism frequently occur subsequent to cerebral ischemia.Adenosine triphosphate(ATP)-sensitive potassium channel openers exhibit protective effects on cerebral ischemia/reperfusion injury.OBJECTIVE:To validate the effects of cromakalin on mitochondrial structure and function in ischemic penumbra brain tissue in a rat model of middle cerebral artery occlusion(MCAO).DESIGN,TIME AND SETTING:The present single-factor analysis of variance,randomized,controlled,animal experiment was performed at the Institute of Brain Science,Affiliated Hospital of Qingdao University Medical College between October 2007 and March 2008.MATERIALS:Forty male,Wistar rats were randomly divided into four groups,with 10 rats per group:sham-operated,MCAO,MCAO+ATP-sensitive potassium channel opener(cromakalin),and MCAO+cromakalin+ATP-sensitive potassium channel blocking agent(glibenclamide).METHODS:Focal cerebral ischemia/reperfusion injury was induced by MCAO in all groups except the sham-operated group.The MCAO cromakalin group was administered 10 mg/kg cromakalin(i.p.) prior to MCAO induction.The MCAO+cromakalin+glibenclamide group received an injection of 10 mg/kg cromakalin(i.v.),and subsequently an injection of 10 mg/kg cromakalin(i.p.) prior to MCAO induction.MAIN OUTCOME MEASURES:At 24 hours after cerebral ischemia/reperfusion injury,cellular apoptosis was detected by terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate(dUTP) nick-end labeling technique.Cytochrome C expression was measured by immunohistochemistry.In addition,mitochondrial swelling,membrane fluidity,membrane phospholipid and malonaldehyde(MDA) contents,as well as Na+-K+-ATPase,Ca2 +-ATPase,and superoxide dismutase(SOD) activities were determined.RESULTS:Compared with the sham-operated group,the three ischemia groups exhibited significantly elevated mitochondrial MDA content,reduced membrane phospholipid and ATP contents,down-regulated membrane fluidity,and reduced Na+-K+-ATPase,Ca2+-ATPase,and SOD activities(P < 0.05-0.01).In the MCAO+cromakalin group,the number of apoptotic cells decreased,and cytochrome C expression,as well as MDA content,were reduced.However,ATP content and Na+-K+-ATPase,Ca2+-ATPase,and SOD activities significantly increased compared with the MCAO group(P < 0.05-0.01).Glibenclamide noticeably antagonized cromakalin protection of mitochondria.CONCLUSION:Pretreatment with the ATP-sensitive potassium channel opener cromakalin increased mitochondrial Na+-K+-ATPase,Ca2+-ATPase,and SOD activities,decreased neuronal apoptosis,and inhibited cytochrome C expression following MCAO.Shilei Wang Peng Wang Qingxian Chang Yu Li Yan Jiang Shiduan Wang 2008Neural Regeneration Research2008,3,9:6
7Deformation-induced martensitic transformation kinetics and correlative micromechanical behavior of medium-Mn transformation-induced plasticity steel显示文摘An in situ high-energy X-ray diffraction(HE-XRD) technique was mainly used to investigate the micromechanical behavior of medium-Mn Fe-0.12 C-10.16 Mn-1.87 Al(in wt%) transformation-induced plasticit(TRIP) steel subjected to intercritical annealing at 625℃, 650℃, 675℃ and 700℃ for 1 h. As the intercritical annealing temperature increased, the volume fraction of retained austenite(RA) and ultimate tensilstress(UTS) increased, while the Lüders strain and yield stress(YS) decreased. The incremental workhardening exponent of experimental steel increased with increasing intercritical annealing temperatureThe overall trend of the transformation kinetics of the RA with respect to the true strain followed thsigmoidal shape predicted by the Olson and Cohen(OC) model. Load partitioning occurred among the ferrite, austenite and martensite immediately after entering the yielding stage. Because the stability of thRA decreased with increasing intercritical annealing temperature, the load undertaken by the martensitincreased. 8Effects of the mitochondrial calcium uniporter on cerebral edema in a rat model of cerebral ischemia reperfusion injury显示文摘The present study investigated the effects of the mitochondrial calcium uniporter inhibitor ruthenium red and the agonist spermine on cerebral edema in rats with cerebral ischemia reperfusion injury. Left middle cerebral artery occlusion (MCAO) was induced in rats using the suture method. Following 24 hours of ischemic reperfusion, neurological function scores of rats with MCAO, and rats pretreated with ruthenium red and spermine were significantly lower, however, water content of brain tissue, aquaporin 4 expression and immunoglobulin G (IgG) exudation were significantly higher than those of sham-operated rats. Compared with MCAO rats and spermine-treated rats, neurological function scores were considerably higher, and brain tissue water content, aquaporin 4 expression and IgG exudation decreased in ruthenium red-treated rats. These findings suggest that preventive application of the mitochondrial calcium uniporter inhibitor ruthenium red can significantly decrease aquaporin 4 and IgG expression, influence the permeability of the blood brain barrier, and thereby decrease the extent of cerebral edema.Linlin Li Shilei Wang Haihong Luan 2011Neural Regeneration Research2011,6,22:4
9Fingerprinting hydrothermal fluids in porphyry Cu deposits using K and Mg isotopes显示文摘In this study, we performed an integrated investigation of K and Mg isotopes in hydrothermally altered rocks from the giant Dexing porphyry Cu deposit in China. Both the altered porphyry intrusion and the surrounding wall rocks exhibit large variations in K and Mg isotope compositions, with δ41K values ranging between-1.02‰ and 0.38‰, and δ26Mg values ranging between-0.49‰ and 0.32‰. The δ41K and δ26Mg values of the majority of altered samples are higher than the isotopic baseline values for upper continental crust. We attribute the general increase in δ41 K and δ26Mg in altered rocks to hydrothermal alteration,which caused preferential incorporation of heavy K and Mg isotopes in alteration products, particularly phyllosilicates. However,a few altered samples show anomalously low δ41K and δ26Mg values. The δ41K and δ26Mg values do not correlate with K and Mg concentrations, or mineralogy of altered samples. The variable K-Mg isotope data likely reflect fluids of different physicalchemical properties, or different isotopic compositions. Based on the combined K-Mg isotope data, at least three groups of hydrothermal fluids are distinguished from the Dexing porphyry deposit. 10Perfection of Perovskite Grain Boundary Passivation by Rhodium Incorporation for Efficient and Stable Solar Cells显示文摘Organic cation and halide anion defects are omnipresent in the perovskite films,which will destroy perovskite electronic structure and downgrade the properties of devices.Defect passivation in halide perovskites is crucial to the application of solar cells.Herein,tiny amounts of trivalent rhodium ion incorporation can help the nucleation of perovskite grain and passivate the defects in the grain boundaries,which can improve efficiency and stability of perovskite solar cells.Through first-principle calculations,rhodium ion incorporation into the perovskite structure can induce ordered arrangement and tune bandgap.In experiment,rhodium ion incorporation with perovskite can contribute to preparing larger crystalline and uniform film,reducing trap-state density and enlarging charge carrier lifetime.After optimizing the content of 1% rhodium,the devices achieved an efficiency up to 20.71% without obvious hysteresis,from 19.09% of that pristine perovskite.In addition,the unencapsulated solar cells maintain 92% of its initial efficiency after 500 h in dry air.This work highlights the advantages of trivalent rhodium ion incorporation in the characteristics of perovskite solar cells,which will promote the future industrial application.Wei Liu Nanjing Liu Shilei Ji Hongfeng Hua Yuhui Ma Ruiyuan Hu Jian Zhang Liang Chu Xing’ao Li Wei Huang 2020Nano-Micro Letters2020,12,9:4
11The calcium uniporter regulates the permeability transition pore in isolated cortical mitochondria显示文摘To investigate the influence of the mitochondrial calcium uniporter on the mitochondrial permeability transition pore, the present study observed mitochondrial morphology in cortical neurons isolated from adult rats using transmission electron microscopy, and confirmed the morphology and activity of isolated mitochondria by detecting succinic dehydrogenase and monoamine oxidase, two mitochondrial enzymes. Isolated mitochondria were treated with either ruthenium red, an inhibitor of the uniporter, spermine, an activator of the uniporter, or in combination with cyclosporin A, an inhibitor of the mitochondrial permeability transition pore. Results showed that ruthenium red inhibited CaCl2-induced mitochondrial permeability transition pore opening, spermine enhanced opening, and cyclosporin A attenuated the effects of spermine. 13Porcine skeletal muscle tissue fabrication for cultured meat production using three-dimensional bioprinting technology显示文摘Cultured meat produced through in vitro cell culture technology is regarded as a technical revolution.In this study,three-dimensional(3D)bioprinting technology was used to mimic the growth environment in vivo and construct a 3D culture system in vilro.Hydroge1 bioinks,namely,sodium alginate-gelatin and gelatin-methacrylate(GelMA)-silk fbroin,produced using two different curing processes were blended,and their rheological properties,mechanical properties,and biocompatibilities were compared.The 4%GelMA-20%silk fibroin hydroge1(GS2)demonstrated good performance and was hybridized with porcine skeleta1 muscle satellite cells for 3D printing to construct network structures of size 15 mm×15 mm and porosity 1000 um in 4-,6-,and 8-1ayer struchures.After 16 days of culture,4-and 6-1ayer grid structures formed compact muscle fbers organized by multinucleated myotubes.These results suggested that 3D bioprinting and GeIMA-silk fbroin hydrogels have great potential in fabricating porcine skeleta1 muscle tissue for use as cultured meat.Yingying Li Wenting Liu Shilei Li Mingyue Zhang Feng Yang Shouwei Wang 2021Journal of Future Foods2021,1,1:3
14Cholecystokinin octapeptide antagonizes apoptosis in human retinal pigment epithelial cells显示文摘Although cholecystokinin octapeptide-8 is important for neurological function, its neuroprotective properties remain unclear. We speculated that cholecystokinin octapeptide-8 can protect human retinal pigment epithelial cells against oxidative injury. In this study, retinal pigment epithelial cells were treated with peroxynitrite to induce oxidative stress. Peroxynitrite triggered apoptosis in these cells, and increased the expression of Fas-associated death domain, Bax, caspase-8 and Bcl-2. These changes were suppressed by treatment with cholecystokinin octapeptide-8. 15Neuroprotective effects of cromakalim on cerebral ischemia-reperfusion injury in rats Correlation with hippocampal metabotropic glutamate receptor 1 alpha and glutamate transporter 1显示文摘BACKGROUND:Studies have reported that potassium channel openers exhibit a protective effect on cerebral ischemia-reperfusion injury and inhibit glutamate excitotoxicity in rats.However,the effects of the glutamate receptor 1α and glutamate transporter 1 remain poorly understood.OBJECTIVE:To investigate the prophylactic use of the adenosine triphosphate-sensitive potassium channel opener cromakalim on neurological function and cerebral infarct size,as well as glutamate receptor 1α and glutamate transporter 1 expression,in rats with cerebral ischemia-reperfusion injury,and to explore action mechanisms underlying reduced glutamate excitotoxicity and neuroprotection in rats.DESIGN,TIME AND SETTING:Randomized,controlled,animal experiment was performed at the Brain Institute,Qingdao University Medical College,Between July 2008 and April 2009.MATERIALS:Cromakalim was purchased from Sigma,USA;rabbit anti-glutamate receptor 1α polyclonal antibody was offered by Wuhan Boster,China;rabbit anti-glutamate transporter 1 polyclonal antibody was offered by Santa Cruz Biotechnology,USA.METHODS:Sixty male,Wistar rats,aged 6 months,were randomly assigned to three groups (n= 20):sham-surgery,model,and cromakalim.Intraluminal thread methods were used to establish middle cerebral artery occlusion in rats from the model and cromakalim groups.Rats from the sham-surgery group were subjected to exposed common carotid artery,external carotid artery,and internal carotid artery,without occlusion.Cromakalim (10 mg/kg) was administered 30 minutes prior to middle cerebral artery occlusion,but there was no intervention in the model and sham-surgery groups.MAIN OUTCOME MEASURES:At 24 hours post-surgery,neurological behavioral functions were evaluated using Bederson's test,cerebral infarction volume was determined following tetrazolium chloride staining,and glutamate receptor 1α and glutamate transporter 1 expressions were detected using immunohistochemistry.RESULTS:Following cerebral ischemia-reperfusion injury,neurological behavioral malfunctions were obvious in all mice.Focal cerebral infarction was detected in ischemic hemispheres,glutamate receptor 1α expression increased,and glutamate transporter 1 expression decreased in the ischemic hemisphere (P < 0.05).Compared with the model group,neurological behavioral functions significantly improved,cerebral infarction volume was significantly reduced (P < 0.05),glutamate receptor 1α expression was significantly decreased,and glutamate transporter 1 expression was increased in the cromakalim group (P < 0.05).CONCLUSION:Improved neurological function and reduced cerebral infarction volume in rats through the preventive use of cromakalim could be related to decreased glutamate receptor 1α expression and enhanced glutamate transporter 1 expression.Shilei Wang Junchao Liu Qingxian Chang Yu Li, Yan Jiang Shiduan Wang 2010Neural Regeneration Research2010,5,9:2

17Population Genetics of SARS-CoV-2: Disentangling Effects of Sampling Bias and Infection Clusters显示文摘A novel RNA virus,the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),is responsible for the ongoing outbreak of coronavirus disease 2019(COVID-19).Population genetic analysis could be useful for investigating the origin and evolutionary dynamics of COVID-19.However,due to extensive sampling bias and existence of infection clusters during the epidemic spread,direct applications of existing approaches can lead to biased parameter estimations and data misinterpretation.In this study,we first present robust estimator for the time to the most recent common ancestor(TMRCA)and the mutation rate,and then apply the approach to analyze 12,909 genomic sequences of SARS-CoV-2.The mutation rate is inferred to be 8.69×10^(−4) per site per year with a 95%confidence interval(CI)of[8.61×10^(−4),8.77×10^(−4)],and the TMRCA of the samples inferred to be Nov 28,2019 with a 95%CI of[Oct 20,2019,Dec 9,2019].The results indicate that COVID-19 might originate earlier than and outside of Wuhan Seafood Market.We further demonstrate that genetic polymorphism patterns,including the enrichment of specific haplotypes and the temporal allele frequency trajectories generated from infection clusters,are similar to those caused by evolutionary forces such as natural selection.Our results show that population genetic methods need to be developed to efficiently detangle the effects of sampling bias and infection clusters to gain insights into the evolutionary mechanism of SARS-CoV-2.Software for implementing VirusMuT can be downloaded at http://gffzzdbc7b6aaae734bdds5cpnxb0qbc5v6xpx.ffgz.tsg.suse.edu.cn/biocode/tools/BT007081.Qi Liu Shilei Zhao Cheng-Min Shi Shuhui Song Sihui Zhu Yankai Su Wenming Zhao Mingkun Li Yiming Bao Yongbiao Xue Hua Chen 2020Genomics, Proteomics & Bioinformatics2020,18,6:1

19Petal Development in Lotus japonicus显示文摘Previous studies have demonstrated that petal shape and size in legume flowers are determined by two separate mechanisms,dorsoventral (DV) and organ internal (IN) asymmetric mechanisms,respectively.However,little is known about the molecular mechanisms controlling petal development in legumes.To address this question,we investigated petal development along the floral DV axis in Lotus japonicus with respect to cell and developmental biology by comparing wild-type legumes to mutants.Based on morphological markers,the entire course of petal development,from initiation to maturity,was grouped to define 3 phases or 13 stages.In terms of epidermal micromorphology from adaxial surface,mature petals were divided into several distinct domains,and characteristic epidermal cells of each petal differentiated at stage 9,while epidermal cells of all domains were observed until stage 12.TCP and MIXTA-like genes were found to be differentially expressed in various domains of petals at stages 9 and 12.Our results suggest that DV and IN mechanisms interplay at different stages of petal development,and their interaction at the cellular and molecular level guides the elaboration of domains within petals to achieve their ideal shape,and further suggest that TCP genes determine petal identity along the DV axis by regulating MIXTA-like gene expression.Lin Weng Zhaoxia Tian Xianzhong Feng Xin Li Shilei Xu Xiaohe Hu Da Luo Jun Yang 2011Journal of Integrative Plant Biology2011,53,10:1
