2Ozone sensitivity analysis with the MM5-CMAQ modeling system for Shanghai显示文摘Ozone has become one of the most important air pollution issues around the world. This article applied both O 3 /(NOy-NOx) and H 2 O 2 /HNO 3 indicators to analyze the ozone sensitivity in urban and rural areas of Shanghai, with implementation of the MM5-CMAQ modeling system in July, 2007. The meteorological parameters were obtained by using the MM5 model. A regional emission inventory with spatial and temporal allocation based on the statistical data has been developed to provide input emission data to the MM5-CMAQ modeling system. Results showed that the ozone concentrations in Shanghai show clear regional differences. The ozone concentration in rural areas was much higher than that in the urban area. 3Kinetics of the accumulation of group 2 innate lymphoid cells in IL-33-induced and IL-25-induced murine models of asthma:a potential role for the chemokine CXCL16显示文摘ILC2s are implicated in asthma pathogenesis, but little is known about the mechanisms underlying their accumulation in airways.We investigated the time course of ILC2 accumulation in different tissues in murine models of asthma induced by a serial per-nasalchallenge with ovalbumin (OVA), house dust mice (HDM), IL-25 and IL-33 and explored the potential roles of ILC2-attractingchemokines in this phenomenon. Flow cytometry was used to enumerate ILC2s at various time points. The effects of cytokines andchemokines on ILC2 migration were measured in vitro using a chemotaxis assay and in vivo using small animal imaging. Comparedwith saline and OVA challenge, both IL-25 and IL-33 challenge alone induced significant accumulation of ILC2s in the mediastinallymph nodes, lung tissue and bronchoalveolar lavage fluid of challenged animals, but with a distinct potency and kinetics. In vitro,IL-33 and CXCL16, but not IL-25 or CCL25, directly induced ILC2 migration. Small animal in vivo imaging further confirmed that asingle intranasal provocation with IL-33 or CXCL16 was sufficient to induce the accumulation of ILC2s in the lungs followinginjection via the tail vein. Moreover, IL-33-induced ILC2 migration involved the activation of ERK1/2, p38, Akt, JNK and NF-κB, whileCXCL16-induced ILC2 migration involved the activation of ERK1/2, p38 and Akt. These data support the hypothesis that epitheliumderived IL-25 and IL-33 induce lung accumulation of ILC2s, while IL-33 exerts a direct chemotactic effect in this process. 4Anlotinib has good efficacy and low toxicity: a phase Ⅱ study of anlotinib in pre-treated HER-2 negative metastatic breast cancer显示文摘Objective:Anlotinib is a novel tyrosine kinase inhibitor blocking angiogenesis.This study was performed to assess the efficacy and safety of anlotinib in patients with metastatic breast cancer.Methods:Patients with HER2-negative breast cancer,who were pre-treated with anthracycline or taxanes in a neoadjuvant,adjuvant,or metastatic setting,and had treatment failure after at least one prior chemotherapy regimen in the metastatic setting were enrolled.Anlotinib was administered at 12 mg daily for 14 days in a 21-day cycle until disease progression or unacceptable toxicity occurred.Simultaneously,5–10 m L of venous blood was collected to perform circulating tumor DNA(ct DNA)testing every 2 treatment cycles.The primary endpoint was the objective response rate(ORR).Secondary endpoints included the disease control rate(DCR),progression-free survival(PFS),overall survival,safety,and biomarkers.Results:Twenty-six eligible patients were enrolled,with a median age of 56(30–75)years.The median follow-up time was 10.5 months.The ORR was 15.4%,the DCR was 80.8%,and the median PFS was 5.22 months(95%confidence interval 2.86–6.24).Fourteen(53.8%)patients survived for more than 10 months.The changes in the detectable ct DNA variant allele frequency were consistent with the tumor response.The most common treatment-related adverse events were hypertension(57.7%),thyroidstimulating hormone elevation(34.6%),and hand-foot syndrome(23.1%).Conclusion:Anlotinib showed objective efficacy with tolerable toxicity in heavily pre-treated,metastatic HER2-negative breast cancer.The dynamic changes in the ct DNA variant allele fraction may be predictive of the tumor response.Nanlin Hu Yiran Si Jian Yue Tingting Sun Xue Wang Zhuqing Jia Songlin Gao Qiao Li Yang Shao Jiayu Wang Yang Luo Fei Ma Binghe Xu Peng Yuan 2021Cancer Biology & Medicine2021,18,3:11
5Role of apoptosis-inducing factor in perinatal hypoxic-ischemic brain injury显示文摘Perinatal complications,such as asphyxia,can cause brain injuries that are often associated with subsequent neurological deficits,such as cerebral palsy or mental retardation.The mechanisms of perinatal brain injury are not fully understood,but mitochondria play a prominent role not only due to their central function in metabolism but also because many proteins with apoptosis-related functions are located in the mitochondrion.Among these proteins,apoptosis-inducing factor has already been shown to be an important factor involved in neuronal cell death upon hypoxia-ischemia,but a better understanding of the mechanisms behind these processes is required for the development of more effective treatments during the early stages of perinatal brain injury.In this review,we focus on the molecular mechanisms of hypoxic-ischemic encephalopathy,specifically on the importance of apoptosis-inducing factor.The relevance of apoptosis-inducing factor is based not only because it participates in the caspase-independent apoptotic pathway but also because it plays a crucial role in mitochondrial energetic functionality,especially with regard to the maintenance of electron transport during oxidative phosphorylation and in oxidative stress,acting as a free radical scavenger.We also discuss all the different apoptosis-inducing factor isoforms discovered,focusing especially on apoptosis-inducing factor 2,which is only expressed in the brain and the functions of which are starting now to be clarified.Finally,we summarized the interaction of apoptosis-inducing factor with several proteins that are crucial for both apoptosis-inducing factor functions(prosurvival and pro-apoptotic)and that are highly important in order to develop promising therapeutic targets for improving outcomes after perinatal brain injury.Juan Rodriguez Tao Li Yiran Xu Yanyan Sun Changlian Zhu 2021Neural Regeneration Research2021,16,2:11
63D Geometry-Based UAV-MIMO Channel Modeling and Simulation显示文摘A more general narrowband regular-shaped geometry-based statistical model(RS-GBSM) combined with the line of sight(LoS) and single bounce(SB) rays for unmanned aerial vehicle(UAV) multiple-input multiple-output(MIMO) channel is proposed in this paper. The channel characteristics, including space-time correlation function(STCF), Doppler power spectral density(DPSD), level crossing rate(LCR) and average fade duration(AFD), are derived based on the single sphere reference model for a non-isotropic environment. The corresponding sum-of-sinusoids(SoS) simulation models including both the deterministic model and statistical model with finite scatterers are also proposed for practicable implementation. The simulation results illustrate that the simulation models well reproduce the channel characteristics of the single sphere reference model with sufficient simulation scatterers. And the statistical model has a better approximation of the reference model in comparison with the deterministic one when the simulation trials of the stochastic model are sufficient. 7Biomass-derived porous carbon highly efficient for removal of Pb(Ⅱ) and Cd(Ⅱ)显示文摘The utilization of abundant and renewable biomass to fabricate advanced functional materials is considered a promising route for environmental applications.Herein,Lignin-based porous carbon with layered graphene-like structure(LPC)is successfully synthesized and applied to efficiently remove Pb(Ⅱ)and Cd(Ⅱ).The as-synthesized LPC materials are systematically characterized and these results show that LPC has a porous graphene-like structure,facilitating the diffusion and immobilization of heavy metal ions.The influence of different reaction parameters(solution pH,initial concentration of metal ions,contact time and adsorbent amount)on the adsorption performance is investigated in details.The results demonstrate that LPC can achieve superior adsorption capacities of 250.5 mg·g^-1 for Pb(Ⅱ)and 126.4 mg·g^-1 for Cd(Ⅱ),which are far superior to the previously reported adsorbents.Pseudo-second order kinetics model and Freundlich isotherm model describe the adsorption process well.Furthermore,the exhausted LPC can be regenerated easily and exhibits the removal efficiency of 96%and 92%for Pb(Ⅱ)and Cd(Ⅱ)after five continuous runs,respectively.This study shows a sustainable strategy for the design of porous carbon material from na?ve biomass and highlights the great potential in wastewater treatment.Anqi Wang Zhikeng Zheng Ruiqi Li Di Hu Yiran Lu Huixia Luo Kai Yan 2019Green Energy & Environment2019,4,4:10
8Percent free prostate-specific antigen for prostate cancer diagnosis in Chinese men with a PSA of 4.0-10.0 ng/mL:Results from the Chinese Prostate Cancer Consortium显示文摘Objective:To test the diagnostic performance of percent free prostate-specific antigen(%fPSA)in predicting any prostate cancer(PCa)and high-grade prostate cancer(HGPCa)in a retrospective multi-center biopsy cohort with a PSA level of 4.0e10.0 ng/mL in China.Methods:Consecutive patients with a PSA of 4.0-10.0 ng/mL who underwent transrectal ultrasound-guided biopsy were enrolled at 16 Chinese medical centers from January 1st,2010 to December 31st,2013.Total and free serum PSA determinations were performed using three types of electro-chemiluminescence immunoassays recalibrated to the World Health Organization(WHO)standard.The diagnostic accuracy of PSA,%fPSA,and %fPSA in combination with PSA(%fPSA t PSA)was determined using the area under the receiver operating characteristic(ROC)curve(AUC).Results:A total of 2310 consecutive men with PSA levels between 4.0 and 10.0 ng/mL were included,and the detection rate of PCa was 25.1%.The AUC of%fPSA and %fPSA t PSA in predicting any PCa was superior to PSA alone in men aged≥60 years(0.623 vs.0.534,p<0.0001)but not in men aged 40e59 years(0.517 vs.0.518,p=0.939).Similar result was yield in predicting HGPCa.Conclusion:In a clinical setting of Chinese men with 4.0e10.0 ng/mL PSA undergoing initial prostate biopsy,adding %fPSA to PSA can moderately improve the diagnostic accuracy for any PCa and HGPCa compared with PSA alone in patients≥60 but not in patients aged 40-59 years.Rui Chen Liping Xie Xiaobing Cai Yiran Huang Liqun Zhou Lulin Ma Xu Gao Chuanliang Xu Shancheng Ren Pengfei Shao Danfeng Xu Kexin Xu Zhangqun Ye Chunxiao Liu Dingwei Ye Li Lu Qiang Fu Jianquan Hou Jianlin Yuan Dalin He Tie Zhou Fubo Wang Biming He Yinghao Sun 2015Asian Journal of Urology2015,2,2:8
9Preparation of cationic polyacrylamide microsphere emulsion and its performance for permeability reduction显示文摘In this paper, cationic polyacrylamide microspheres(CPAM) were synthesized using acrylamide(AM) and methacryloyloxyethyl trimethyl ammonium chloride(TMAEMC) as monomers, ammonium sulfate as dispersant, poly(acryloyloxyethyl trimethyl ammonium chloride)(PAETAC) as dispersion stabilizer, and ammonium persulfate as initiator. The synthetic method was dispersion polymerization. The effects of monomer ratio(AM/TMAEMC), dispersant concentration, and dispersion stabilizer dosage on dispersion polymerization were systematically studied to determine the optimal preparation conditions. The structure and viscosity of the synthesized polymer were characterized by FTIR and capillary viscometry, respectively, and the particle sizes and distribution of the polymer microspheres were characterized by microscopy and dynamic light scattering, respectively. Finally, flow tests were conducted to measure the permeability reduction performance of the microspheres at various concentrations in sand packs with different permeability. 10Injectable peptide hydrogel as intraperitoneal triptolide depot for the treatment of orthotopic hepatocellular carcinoma显示文摘Chemotherapy is among the limited choices approved for the treatment of hepatocellular carcinoma(HCC) at intermediate and advanced stages. Preferential and prolonged drug exposure in diseased sites is required to maximize the therapeutic index of the drug. Here, we report an injectable supramolecular peptide hydrogel as an intraperitoneal depot for localized and sustained release of triptolide for the treatment of orthotopic HCC. We chose peptide amphiphile C16-GNNQQNYKD-OH-based nanofibers as gelators and carriers for triptolide. Sustained triptolide release from the hydrogel was achieved over 14 days in vitro, with higher accumulation in and cytotoxicity against human HCC Bel-7402 in comparison with L-02 fetal hepatocytes. After intraperitoneal injection, the hydrogel showed prolonged retention over 13 days and preferential accumulation in the liver, realizing HCC growth inhibition by99.7 ± 0.1% and animal median survival extension from 19 to 43 days, without causing noticeable pathological changes in the major organs. 12Metastatic status of sentinel lymph nodes in breast cancer determined with photoacoustic microscopy via dual-targeting nanoparticles显示文摘Detection of sentinel lymph nodes(SLNs)is critical to guide the treatment of breast cancer.However,distinguishing metastatic SLNs from normal and inflamed lymph nodes(LNs)during surgical resection remains a challenge.Here,we report a CD44 and scavenger receptor class B1 dual-targeting hyaluronic acid nanoparticle(5K-HA-HPPS)loaded with the near-infra-red fluorescent dye DiR-BOA for SLN imaging in breast cancer.The small sized(~40 nm)self-assembled 5K-HA-HPPSs accumulated rapidly in the SLNs after intradermal injection.Compared with normal popliteal LNs(N-LN),there were ~3.2-fold and ~2.4-fold increases in fluorescence intensity in tumour metastatic SLNs(T-MLN)and inflamed LNs(Inf-LN),respectively,6 h after nanoparticle inoculation.More importantly,photoacoustic microscopy(PAM)of 5KHA-HPPS showed a significantly distinct distribution in T-MLN compared with N-LN and Inf-LN.Signals were mainly distributed at the centre of T-MLN but at the periphery of N-LN and Inf-LN.The ratio of PA intensity(R)at the centre of the LNs compared with that at the periphery was 5.93±0.75 for T-MLNs of the 5K-HA-HPPS group,which was much higher than that for the Inf-LNs(R=0.2±0.07)and N-LNs(R=0.45±0.09).These results suggest that 5K-HA-HPPS injection combined with PAM provides a powerful tool for distinguishing metastatic SLNs from pLNs and inflamed LNs,thus guiding the removal of SLNs during breast cancer surgery.Yanfeng Dai Xiang Yu Jianshuang Wei Fanxin Zeng Yiran Li Xiaoquan Yang Qingming Luo Zhihong Zhang 2020Light(Science & Applications)2020,9,1:6
13Deterministic and Stochastic Simulators for Non-Isotropic V2V-MIMO Wideband Channels显示文摘In this paper, we consider a novel two-dimensional(2D) geometry-based stochastic model(GBSM) for multiple-input multiple-output(MIMO) vehicle-to-vehicle(V2V) wideband fading channels. The proposed model employs the combination of a two-ring model and a multiple confocal ellipses model, where the signal is sum of the line-of-sight(Lo S) component, single-bounced(SB) rays, and double-bounced(DB) rays. Based on the reference model, we derive some expressions of channel statistical properties, including space-time correlation function(STCF), Doppler spectral power density(DPSD), envelope level crossing rate(LCR) and average fade duration(AFD). In addition, corresponding deterministic and stochastic simulation models are developed based on the reference model. Moreover, we compare the statistical properties of the reference model and the two simulation models in different scenarios and investigate the impact of different vehicular traffic densities(VTDs) on the channel statistical properties of the proposed model. 14Tumor microenvironment-responsive docetaxel-loaded micelle combats metastatic breast cancer显示文摘Efficient tumor-targeting drug delivery systems are urgently needed for treating metastatic breast cancer.In this work, a docetaxel(DTX)-loaded micelle(pDM) as the tumor-microenvironment-responsive delivery platform is developed. The micelle is composed of a pH-sensitive amphiphilic copolymer,poly((1,4-butanediol)-diacrylate-b-N,N-diisopropylethylenediamine)-polyethyleneimine(BD-PEI), and a matrix metalloproteinase(MMP)-responsive polymer, poly((1,4-butanediol)-diacrylate-b-N,N-diisopropy lethylenediamine)-peptide-polyethylene glycol(PEG)(BD-peptide-PEG). The PEG block of BD-peptidePEG will be split by MMPs at the tumor microenvironment, which leads to the change of the surface charge and particle size of the micelle to more positive and smaller one. Owing to this transformation and enhanced permeability and retention(EPR) effect, pDM delivers more DTX into tumor tissues and is internalized more efficiently by tumor cells than the non-MMP-sensitive micelles in the 4 T1 tumorbearing mice model. In addition, DTX is released in acidic endo/lysosomes due to the dissociation of the micelle, triggered by the protonation of the hydrophobic block of BD-PEI. As a result, the DTX-loaded micelle inhibits primary tumor growth and pulmonary metastasis effectively. 15Unraveling a genetic roadmap for improved taste in the domesticated apple显示文摘Although taste is an important aspect of fruit quality, an understanding of its genetic control remains elusive in apple and other fruit crops. In this study, we conducted genomic sequence analysis of 497 Malus accessions and revealed erosion of genetic diversity caused by apple breeding and possible independent domestication events of dessert and cider apples. Signatures of selection for fruit acidity and size, but not for fruit sugar content, were detected during the processes of both domestication and improvement. Furthermore, we found that single mutations in major genes affecting fruit taste, including Ma1, MdTDT, and MdSOT2, dramatically decrease malate, citrate, and sorbitol accumulation, respectively, and correspond to important domestication events. Interestingly, Ma1 was identified to have pleiotropic effects on both organic acid content and sugar:acid ratio, suggesting that it plays a vital role in determining fruit taste. Fruit taste is unlikely to have been negatively affected by linkage drag associated with selection for larger fruit that resulted from the pyramiding of multiple genes with minor effects on fruit size. 17Characterization of VOC emissions from construction machinery and river ships in the Pearl River Delta of China显示文摘Speciated characterization of Volatile Organic Compounds (VOCs),including oxygenated VOCs (OVOCs),from construction machinery and river ships in China is currently lacking.In this regard,we conducted field measurement on speciated VOC (including OVOC) emissions from six construction machinery and five river ships in the Pearl River Delta (PRD) region to identify VOC emission characteristics.We noticed that OVOC emissions from construction machinery and ships accounted for more than 50%of the total VOC emissions,followed by alkenes,aromatics and alkanes.Formaldehyde and acetaldehyde were the most emission species,accounting for 61.8%-83.2%of OVOCs.For construction machinery,the fuel-based emission factors of roller,grader and pile driver were 3.12,3.12 and 7.36 g/kg,respectively.With the rigorous restraint by the national emission standards,VOC emissions of construction machinery had decreased considerably,especially during stageⅢ.Ozone formation potential was also significantly reduced due to the significant decrease in emissions of OVOCs and alkenes with higher reactivity.For river ships,the fuel-based emission factors of cargo ships and speedboat were 1.46 and 0.44 g/kg,respectively.VOC emissions from construction machinery and river ships in Guangdong Province in 2017 were 8851.0 and 4361.0 ton,respectively.This study filled the knowledge gaps of reactive gas emissions from different kinds of non-road mobile sources over the PRD,and more importantly,highlighted the necessity in adding OVOC measurement to give a complete and accurate depiction of reactive gas emissions from non-road mobile sources.Richao Wang Zibing Yuan Junyu Zheng Cheng Li Zhijiong Huang Wenshi Li Yan Xie Yiran Wang Kaiyang Yu Lejun Duan 2020Journal of Environmental Sciences2020,32,10:5

19PCL2 regulates p53 stability and functions as a tumor suppressor in breast cancer显示文摘Polycomblike2(PCL2) is a well-known component of polycomb repressive complex 2(PRC2) and plays important roles in H3 K27 methylation and homeotic gene silencing.However,the involvement of PCL2 in breast cancer development remains unclear.Here,we established PCL2 as a tumor suppressor gene in breast cancer.Expression level of PCL2 was significantly downregulated in breast cancer tissue samples observed at different TNM stages.Ectopic expression of PCL2 could significantly inhibit cell proliferation and promoted apoptosis.PCL2 also remarkably elevated levels of p53 and its targets by increasing p53 stability.Mechanistically,PCL2 protected p53 proteins from MDM2-mediated ubiquitination and degradation by sequestering MDM2 into the nucleolus.Overexpression of PCL2 also suppressed migration and invasion by inhibiting epithelial-mesenchymal transition.PCL2 expression was correlated with Ecadherin expression and was inversely correlated with vimentin expression.Furthermore,PCL2 knockdown could attenuate anti-tumor effect of MLN4924.Taken together,our findings indicated that PCL2 played a tumor suppressor role in development and progression of breast cancer and may be a prognostic and predictive marker for breast cancer.Yiran Liang Yang Yang Renbo Guo Shuang Gao Xinghong Guo Danyang Li Meng Wang Haruhiko Koseki Xiangzhi Li 2018Science Bulletin2018,63,10:4
20All VN-graphene architecture derived self-powered wearable sensors for ultrasensitive health monitoring显示文摘The booming of wearable electronics has nourished the progress on developing multifunctional energy storage systems with versatile flexibility, which enable the continuous and steady power supply even under various deformed states. In this sense, the synergy of flexible energy and electronic devices to construct integrative wearable microsystems is meaningful but remains quite challenging by far. Herein, we devise an innovative supercapacitor/sensor integrative wearable device that is based upon our designed vanadium nitride-graphene (VN-G) architectures. Flexible quasi-solid-state VN-G supercapacitor with ultralight and binder-free features deliver a specific capacitance of^53 F·g^-1 with good cycle stability. On the other hand, VN-G derived pressure sensors fabricated throughout a spray-printing process also manifest favorably high sensitivity (40 kPa^-1 at the range of 2-10 kPa), fast response time (~130 ms), perfect skin conformability, and outstanding stability under static and dynamic pressure conditions.
