| 1 | Knockdown of OsHox33, a member of the class III homeodomain-leucine zipper gene family, accelerates leaf senescence in rice显示文摘The class III homeodomain-leucine zipper(HD-Zip III)gene family plays important roles in plant growth and development,including regulation of apical embryo patterning,embryonic shoot meristem formation,leaf polarity,vascular development,and meristem function,with a particularly crucial function in leaf development.Although HD-Zip III members are highly conserved in land plants,previous studies,such as genetic analyses based on multiple mutants in Arabidopsis and other plants,suggest that various HD-Zip III family genes have evolved with distinct functions and pleiotropic effects on plant growth and development.In this study,we analyzed a HD-Zip III member,OsHox33,and demonstrated that it plays an important role in age-dependent leaf senescence in rice.We constructed two specific RNAi vectors derived from the 5′-end region and 3′-UTR of OsHox33 to knockdown its expression.Transgenic plants harboring either RNAi construct displayed similar phenotypes of precocious leaf senescence symptoms,suggesting that knockdown of OsHox33 accelerates leaf senescence in rice.pOsHox33::GUS fusion expression and RT-PCR revealed that OsHox33 is highly expressed in young organs,especially in young meristems such as shoot apical meristems,intercalary meristems,and young callus.In addition,real-time PCR indicated that OsHox33 was more highly expressed in young leaves than in old leaves.To further investigate OsHox33 function,we analyzed chloroplast ultrastructure in different-aged leaves of RNAi plants,and found that OsHox33 knockdown accelerated chloroplast degradation,which is consistent with RNAi phenotypes.Finally,real-time PCR studies showed that OsHox33 can regulate the expression of GS1 and GS2,two senescence-associated genes.Taken together,the work presented here provides new insights into the function of HD-Zip III members in plants. | LUAN WeiJiang SHEN Ao JIN ZhiPing SONG SuSheng LI ZhengLong SHA AiHua | 2013 | Science China(Life Sciences)2013,56,12: | 13 |
| 2 | Analysis of DNA methylation variation in wheat genetic background after alien chromatin introduction based on methylation-sensitive amplification polymorphism显示文摘During the process of alien germplasm introduced into wheat genome by chromosome engineering, extensive genetic variations of genome structure and gene expression in recipient could be induced. In this study, we performed GISH (genome in situ hybridization) and AFLP (amplified fragment length po-lymorphism) on wheat-rye chromosome translocation lines and their parents to detect the identity in genomic structure of different translocation lines. The results showed that the genome primary struc-ture variations were not obviously detected in different translocation lines except the same 1RS chromosome translocation. Methylation sensitive amplification polymorphism (MSAP) analyses on genomic DNA showed that the ratios of fully-methylated sites were significantly increased in translo-cation lines (CN12, 20.15%; CN17, 20.91%; CN18, 22.42%), but the ratios of hemimethylated sites were significantly lowered (CN12, 21.41%; CN17, 23.43%; CN18, 22.42%), whereas 16.37% were fully-me-thylated and 25.44% were hemimethylated in case of their wheat parent. Twenty-nine classes of me-thylation patterns were identified in a comparative assay of cytosine methylation patterns between wheat-rye translocation lines and their wheat parent, including 13 hypermethylation patterns (33.74%), 9 demethylation patterns (22.76%) and 7 uncertain patterns (4.07%). | 3 | Remotely excited Raman optical activity using chiral plasmon propagation in Ag nanowires显示文摘We experimentally investigated remotely excited Raman optical activity(ROA)using propagating surface plasmons in chiral Ag nanowires.Using chiral fmoc-glycyl-glycine-OH(FGGO)molecules,we first studied the local surface plasmon-enhanced ROA.We found that the Raman intensity can be excited by left-and right-circularly polarized lights and that the circular intensity difference(CID)can be significantly enhanced.Second,by selecting vibrational modes with large Raman and ROA intensities that are not influenced by chemical enhancements,we studied remotely excited ROA imaging and the CID of FGGO molecules by propagating a plasmonic waveguide using Ag chiral nanostructures.When laser light was radiated on one of the Ag terminals,the measured CID of the FGG at the other terminal showed little change compared to the local excited CID.Meanwhile,when the laser light was radiated on the Ag nanowires(not on the terminals)and was coupled to the nearby nanoantenna,the CID of the ROA could be manipulated by altering the coupling angle between the Ag nanowires.To directly demonstrate the propagation of ROA along the nanowire and its remote detection,we also measured the remotely excited ROA spectra.Our experimental method has the potential to remotely determine the chirality of molecular structures and the absolute configuration or conformation of a chiral live cell. | Mengtao Sun Zhenglong Zhang Peijie Wang Qiang Li Fengcai Ma Hongxing Xu | 2013 | Light(Science & Applications)2013,2,1: | 7 |
| 4 | Effects of Yishengukang decoction on expression of bone-specific alkaline phosphatase,carboxyterminal propeptide of typeⅠprocollagen,and carboxyterminal cross-linked telepeptide of typeⅠcollagen in malignant tumor patients with bone metastasis显示文摘OBJECTIVE: To investigate the effect of Yishengukang decoction on the expression of the metabolic bone markers, bone-specific alkaline phosphatase(BAP), carboxyterminal propeptide of type Ⅰ procollagen(PICP), and carboxyterminal cross-linked telepeptide of type Ⅰ collagen(ICTP), in cancer patients with bone metastasis.METHODS: Patients(n = 180) were divided into three groups:(a) bone metastasis patients treated with Yishengukang and pamidronate disodium injection(treatment group, n = 60);(b) bone metastasis patients treated with pamidronate disodium injection alone(control group, n = 60);(c) cancer patients without metastatic bone lesion(non-bone metastasis group, n = 60). Serum levels of the metabolic markers BAP, PICP, and ICTP were detected by enzyme-linked immunosorbent assay pre- and post-therapy.RESULTS: A significant decrease in serum BAP level was observed in the treatment group compared with the control group. However there were no significant differences in serum levels of PICP and ICTP before or after treatment compared with the control group.CONCLUSION: Yishengukang decoction combined with pamidronate disodium injection reduced serum BAP level to a greater extent that pamidronate disodium injection alone. | 7 | Value-added Impact of Geostationary Hyperspectral Infrared Sounders on Local Severe Storm Forecasts——via a Quick Regional OSSE显示文摘Accurate atmospheric temperature and moisture information with high temporal/spatial resolutions are two of the key parameters needed in regional numerical weather prediction(NWP) models to reliably predict high-impact weather events such as local severe storms(LSSs). High spectral resolution or hyperspectral infrared(HIR) sounders from geostationary orbit(GEO) provide an unprecedented source of near time-continuous, three-dimensional information on the dynamic and thermodynamic atmospheric fields—an important benefit for nowcasting and NWP-based forecasting. In order to demonstrate the value of GEO HIR sounder radiances on LSS forecasts, a quick regional OSSE(Observing System Simulation Experiment)framework has been developed, including high-resolution nature run generation, synthetic observation simulation and validation, and impact study on LSS forecasts. Results show that, on top of the existing LEO(low earth orbit) sounders, a GEO HIR sounder may provide value-added impact [a reduction of 3.56% in normalized root-mean-square difference(RMSD)] on LSS forecasts due to large spatial coverage and high temporal resolution, even though the data are assimilated every 6 h with a thinning of 60 km. Additionally, more frequent assimilations and smaller thinning distances allow more observations to be assimilated, and may further increase the positive impact from a GEO HIR sounder. On the other hand, with denser and more frequent observations assimilated, it becomes more difficult to handle the spatial error correlation in observations and gravity waves due to the limitations of current assimilation and forecast systems(such as a static background error covariance). | 8 | Application of computational fluid dynamics simulation for submarine oil spill显示文摘Computational fluid dynamics(CFD) codes are being increasingly used in the simulation of submarine oil spills.This study focuses on the process of oil spills, from damaged submarine pipes, to the sea surface, using numerical models. The underwater oil spill model is developed, and a description of the governing equations is proposed,along with modifications required for the particalization of the control volume. Available experimental data were introduced to evaluate the validity of the CFD predictions, the results of which proved to be in good agreement with the experimental data. The effects of oil leak rate, leak diameter, current velocity, and oil density are investigated, by the validated CFD model, to estimate the undersea leakage time, the lateral migration distance,and surface diffusion range when the oil reaches the sea surface. Results indicate that the leakage time and lateral migration distance increase with decreasing leak rates and leak diameter, and increase with increasing current velocity and oil density. On the other hand, a large leak diameter, high density, high leak rate, or fast currents result in a greater surface diffusion range. | 9 | Ensemble Retrieval of Atmospheric Temperature Profiles from AIRS显示文摘Satellite-based observations provide great opportunities for improving weather forecasting. Physical retrieval of atmospheric profiles from satellite observations is sensitive to the uncertainty of the first guess and other factors. In order to improve the accuracy of the physical retrieval, an ensemble methodology was developed with an emphasis on perturbing the first guess. In the methodology, a normal probability density function(PDF) is used to select the optimal profile from the ensemble retrievals. The ensemble retrieval algorithm contains four steps:(1) regression retrieval for original first guess;(2) perturbation of the original first guess to generate new first guesses(ensemble first guesses);(3) using the ensemble first guesses and nonlinear iterative physical retrieval to generate ensemble physical results; and(4) the final optimal profile is selected from the ensemble physical results by using PDF. Temperature eigenvectors(EVs) were used to generate the perturbation and generate the ensemble first guess. Compared with the regular temperature profile retrievals from the Atmospheric InfraRed Sounder(AIRS), the ensemble retrievals RMSE of temperature profiles selected by the PDF was reduced between150 and 320 hPa and below 400 hPa, with a maximum improvement of 0.3 K at 400 hPa. The bias was also reduced in many layers, with a maximum improvement of 0.69 K at 460 hPa. The combined optimal(CombOpt) profile and a mean optimal(MeanOpt) profile of all ensemble physical results were improved below 150 hPa. The MeanOpt profile was better than the CombOpt profile, and was regarded as the final optimal(FinOpt) profile. As a genetically amenable model organism, Caenorhabditis elegans has often been used to explore the molecular mechanisms of fat storage regulation. Dye staining of fixed animals and stimulated Raman scattering(SRS) microscopy methods have been used successfully to study fat storage, but a genetic screening system that takes full advantage of C. elegans transparency to perform live imaging of fluorescent protein reporters has not yet been reported.Here, we investigated the tissue-specific expression of the GFP fusion of Perilipin 1(PLIN1), a Drosophila lipid droplet-associated protein, in C. elegans. Our results indicate that PLIN1::GFP labels lipid droplets and can be used as a fat storage indicator in live worms. | 12 | Added-value of GEO-hyperspectral Infrared Radiances for Local Severe Storm Forecasts Using the Hybrid OSSE Method显示文摘High spectral resolution(or hyperspectral)infrared(IR)sounders onboard low earth orbiting satellites provide high vertical resolution atmospheric information for numerical weather prediction(NWP)models.In contrast,imagers on geostationary(GEO)satellites provide high temporal and spatial resolution which are important for monitoring the moisture associated with severe weather systems,such as rapidly developing local severe storms(LSS).A hyperspectral IR sounder onboard a geostationary satellite would provide four-dimensional atmospheric temperature,moisture,and wind profiles that have both high vertical resolution and high temporal/spatial resolutions.In this work,the added-value from a GEO-hyperspectral IR sounder is studied and discussed using a hybrid Observing System Simulation Experiment(OSSE)method.A hybrid OSSE is distinctively different from the traditional OSSE in that,(a)only future sensors are simulated from the nature run and(b)the forecasts can be evaluated using real observations.This avoids simulating the complicated observation characteristics of the current systems(but not the new proposed system)and allows the impact to be assessed against real observations.The Cross-track Infrared Sounder(CrIS)full spectral resolution(FSR)is assumed to be onboard a GEO for the impact studies,and the GEO CrIS radiances are simulated from the ECMWF Reanalysis v5(ERA5)with the hyperspectral IR all-sky radiative transfer model(HIRTM).The simulated GEO CrIS radiances are validated and the hybrid OSSE system is verified before the impact assessment.Two LSS cases from 2018 and 2019 are selected to evaluate the value-added impacts from the GEO CrIS-FSR data.The impact studies show improved atmospheric temperature,moisture,and precipitation forecasts,along with some improvements in the wind forecasts.An added-value,consisting of an overall 5%Root Mean Square Error(RMSE)reduction,was found when a GEO CrIS-FSR is used in replacement of LEO ones indicat-ing the potential for applications of data from a GEO hyperspectral IR sounder to improve local severe storm forecasts. | Pei WANG Zhenglong LI Jun LI Timothy JSCHMIT | 2021 | Advances in Atmospheric Sciences2021,38,8: | 1 |
