| 1 | Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy. | Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen | 2021 | Cancer Communications2021，41，11： | 29 |

| 3 | Clinical profiles and outcomes of acute type A aortic dissection and intramural hematoma in the current era: lessons from the first registry of aortic dissection in China显示文摘Background::Acute type A aortic dissection(ATAAD)and acute type A intramural hematoma(ATAIMH)are life-threatening diseases with high mortality.To better understand their clinical features in the Chinese population,we analyzed the data from the first Registry of Aortic Dissection in China(Sino-RAD)to promote the understanding and management of the diseases.Methods::All patients with ATAAD and ATAIMH enrolled in Sino-RAD from January 1,2012 to December 31,2016 were involved.The data of patients’selection,history,symptoms,management,outcomes,and postoperation complications were analyzed in the study.The continuous variables were compared using the Student’s t test for normal distributions and the Mann-Whitney U test for non-normal distributions.Categorical variables were compared using the Chi-square test or Fisher exact test.Results::A total of 1582 patients with ATAAD and 130 patients with ATAIMH were included.The mean age of all patients was 48.4 years.Patients with ATAAD were significantly younger than patients with ATAIMH(48.9 years vs.55.6 years,P<0.001).For the total cohort,males were dominant,but the male ratio of patients with ATAAD was significantly higher compared to those with ATAIMH(P=0.01).The time range from the onset of symptom to hospitalization was 2.0 days.More patients of ATAIMH had hypertension than that of ATAAD(82.3%vs.67.6%,P<0.05).Chest and back pain were the most common clinical symptoms.Computerized tomography(CT)was the most common initial diagnostic imaging modality.84.7%received surgical treatment and in-hospital mortality was 5.3%.Patients with ATAAD mainly received surgical treatment(89.6%),while most patients with ATAIMH received medical treatment(39.2%)or endovascular repair(35.4%).Conclusions::Our study suggests that doctors should comprehensively use clinical examination and genetic background screening for patients with ATAAD and ATAIMH and further shorten the time range from symptoms onset to intervention,achieving early diagnosis and treatment,thereby reducing the mortality rate of patients with aortic dissection in China.We should standardize the procedures of aortic dissection treatment and improve people’s understanding.Meanwhile,the curing and transferring efficiency should also be improved. | Wei-Xun Duan Wei-Guang Wang Lin Xia Chao Xue Bo Yu Kai Ren Wei Yi Hong-Liang Liang Xiao-Chao Dong Jian Zuo Jin-Cheng Liu Shi-Qiang Yu Ding-Hua Yi | 2021 | Chinese Medical Journal2021，，8： | 15 |
| 4 | Inhibition of Rac1-dependent forgetting alleviates memory deficits in animal models of Alzheimer's disease显示文摘Accelerated forgetting has been identified as a feature of Alzheimer's disease(AD),but the therapeutic efficacy of the manipulation of biological mechanisms of forgetting has not been assessed in AD animal models.Ras-re-lated C3 botulinum toxin substrate 1(Rac1),a small GTPase,has been shown to regulate active forgetting in Drosophila and mice?Here,we showed that Rac1 activity is aberrantly elevated in the hippocampal tissues of AD patients and AD animal models.Moreover,amyloid-beta 42 could induce Rac1 activation in cultured cells.The elevation of Rac1 activity not only accelerated 6-hour spatial memory decay in 3-month-old APP/PS1 mice,but also significantly contributed to severe memory loss in aged APP/PS1 mice.A similar age-dependent Rac1 activity-based memory loss was also observed in an AD fly model.Moreover,inhibition of Rac1 activity could ameliorate cognitive defects and synaptic plasticity in AD animal models.Finally,two novel compounds,identified through behavioral screening of a randomly selected pool of brain permeable small molecules for their positive effect in rescuing memory loss in both fly and mouse models,were found to be capable of inhibiting Rac1 activity.Thus,multiple lines of evidence corroborate in supporting the idea that inhibition of Rac1 activity is effective for treating AD-related memory loss. | Wenjuan Wu Shuwen Du Wei Shi Yunlong Liu Ying Hu Zuolei Xie Xinsheng Yao Zhenyu Liu Weiwei Ma Lin Xu Chao Ma Yi Zhong | 2019 | Protein & Cell2019，10，10： | 10 |

| 8 | Adult metaplastic hutch diverticulum with robotic-assisted diverticulectomy and reconstruction:A case report显示文摘BACKGROUND Hutch diverticulum arises from the compromised muscular development at the ureteral orifice.It is a congenital disease and extremely rare in adult,only accounting for about 3%occurrence worldwide.It can be either symptomatic or asymptomatic,and relies on image tools for diagnosis and preoperative planning.Indications for surgery are dependent on the complications from the diverticulum.Metaplasia is about 10%among those with hutch diverticulum,and it still has chances turning into malignancy,especially urothelial cell carcinoma.CASE SUMMARY A 27-year-old man was presented with frequently recurrent urinary tract infection for one year,and had suffered from intermittent right flank pain for 3 mo.No past medical histories were recorded before.No obvious abnormalities on laboratory data and urine examination were found.Under ultrasound,right hydronephrosis was seen and an anatomical abnormality was observed on intravenous pyelography.Further computed tomography urogram showed one diverticulum seated at superolateral side of right ureteral orifice.Cystoscopy was done and biopsy results showed focal metaplasia.After discussing with him,roboticassisted diverticulectomy with reconstruction was performed.Right hydronephrosis was greatly improved after surgery.He has completed his 1.5-year follow-ups,and no malignancies were seen from urine cytology and image of intravenous pyelography.CONCLUSION Robotic-assisted diverticulectomy and reconstruction to hutch diverticulum is a safe and efficient operation,providing several advantages over open and laparoscopic ones. | Che H Yang Yi S Lin Yen C Ou Wei C Weng Li H Huang Chin H Lu Chao Y Hsu Min C Tung | 2020 | World Journal of Clinical Cases2020，8，20： | 2 |
| 9 | Response Characteristics and Adiabatic Heating during High Strain Rate for TRIP Steel and DP Steel显示文摘By using a static and high-speed material testing machine,tensile deformation behaviors of two kinds of SiMn TRIP(transformation induced plasticity)steels and DP(dual phase)steel were studied in a large range of strain rates(0.001-2 000s-1).Temperature variation during adiabatic heating and the amount of retained austenite at fracture were measured by an infrared thermometer and an X-ray stress analyser,respectively.The microstructure of steels was observed by optical microscopy(OM)and scanning electron microscopy(SEM)before and after tensile test.It was found from the experimental results that the tensile strength of these steels increased,and the fracture elongation firstly decreased and subsequently increased,as the strain rate increased in the range of 0.1-2 000s-1.The temperature raised during adiabatic heating of TRIP steel was in the range of 100-300℃,while that of the DP steel was in the range of 100-220 ℃.The temperature rise of these steels increased with increasing the strain rate,as well as the amount of the transformed retained austenite in TRIP steels.It was confirmed that austenite to martensite transformation is not suppressed by adiabatic heating. | Yi GAO Chao XU Zhong-ping HE Yan-lin HE Lin LI | 2015 | Journal of Iron and Steel Research(International)2015，22，1： | 2 |
| 10 | Recent Advances in Two-Dimensional Magnets:Physics and Devices towards Spintronic Applications显示文摘The emergence of low-dimensional nanomaterials has brought revolutionized development of magnetism,as the size effect can significantly influence the spin arrangement.Since the first demonstration of truly two-dimensional magnetic materials(2DMMs)in 2017,a wide variety of magnetic phases and associated properties have been exhibited in these 2DMMs,which offer a new opportunity to manipulate the spin-based devices efficiently in the future.Herein,we focus on the recent progress of 2DMMs and heterostructures in the aspects of their structural characteristics,physical properties,and spintronic applications.Firstly,the microscopy characterization of the spatial arrangement of spins in 2D lattices is reviewed.Afterwards,the optical probes in the light-matter-spin interactions at the 2D scale are discussed.Then,particularly,we systematically summarize the recent work on the electronic and spintronic devices of 2DMMs.In the section of electronic properties,we raise several exciting phenomena in 2DMMs,i.e.,long-distance magnon transport,field-effect transistors,varying magnetoresistance behavior,and(quantum)anomalous Hall effect.In the section of spintronic applications,we highlight spintronic devices based on 2DMMs,e.g.,spin valves,spin-orbit torque,spin field-effect transistors,spin tunneling field-effect transistors,and spin-filter magnetic tunnel junctions.At last,we also provide our perspectives on the current challenges and future expectations in this field,which may be a helpful guide for theorists and experimentalists who are exploring the optical,electronic,and spintronic properties of 2DMMs. | Vertikasari PNingrum Bowen Liu Wei Wang Yao Yin Yi Cao Chenyang Zha Hongguang Xie Xiaohong Jiang Yan Sun Sichen Qin Xiaolong Chen Tianshi Qin Chao Zhu Lin Wang Wei Huang | 2020 | Research2020，，1： | 2 |
| 11 | Eight-year follow-up of locally advanced lymphoepithelioma-like carcinoma at upper urinary tract: A case report显示文摘BACKGROUND Urinary tract lymphoepithelioma-like carcinoma is rarely seen.Although it is termed after lymphoepithelioma at the nasopharynx,it behaves more like high grade urothelial carcinoma by immunohistochemical features.Most published literatures focused on its rarity but few discussed results of long-term follow-ups.As no available guidelines are applicable,we postulated that principles should be similar to that of urothelial carcinoma at urinary tract.As of now,this work features the longest follow-up of this cancer at the upper urinary tract.CASE SUMMARY A 63-year-old female had a chief complaint of intermittent left flank pain for 2 mo,along with accompanying symptoms including vomiting and body weight loss,about 7 kg over 2 mo.Laboratory data showed normocytic anemia,mildly poor renal function,and hyperparathyroidism.Urine analysis showed mild hematuria.Computed tomography showed a 4.2-cm-width irregular mass over left renal pelvic and enlarged lymph node at the left renal hilum.Whole-body bone scan was negative of active bone lesions.Biopsy from ureteroscopy showed urothelial carcinoma.Specimen from laparoscopic nephroureterectomy with bladder cuff resection showed lymphoepithelioma-like carcinoma with muscular invasion(pT3).She took adjuvant chemotherapies of 2 cycles and full courses of radiation therapy.No recurrence was observed with designed investigative programs.CONCLUSION Locally advanced urinary tract lymphoepithelioma-like carcinoma could benefit from nephroureterectomy and bladder cuff excision in terms of recurrence-free survival. | Che H Yang Wei C Weng Yi S Lin Li H Huang Chin H Lu Chao Y Hsu Yen C Ou Min C Tung | 2020 | World Journal of Clinical Cases2020，8，19： | 1 |

| 17 | Exosomal miR-29b found in aqueous humour mediates calcium signaling in diabetic patients with cataract显示文摘AIM:To investigate the role of exosomal miR-29b and Ca^(2+)in regulating the function of human lens epithelial cells(HLECs).METHODS:Exosomes were isolated from human aqueous humour(AH)by ultracentrifugation,and visualized by nanopar ticle tracking and transmission electron microscopy.Exosomal miRNA sequencing was performed to identify differentially expressed miRNAs between diabetes with cataracts(DMC)group and age-related cataracts(ARC)group.TargetScan was used to predict potential target of certain miRNA.The expression of CACNA1C mRNA was determined by quantitative real-time polymerase chain reaction and CACNA1C protein was determined by Western blotting.Concentration of Ca^(2+)in human AH and the culture supernatant of cells were detected by the calcium assay kit.Cell counting kit-8 was used to determine cell viability.RESULTS:Exosomes were isolated from human AH,which had a typical cup-shaped phenotype and a particle size distribution in accordance with micro extracellular vesicles.Exosomal miRNA sequencing revealed that miR-29b was significantly downregulated in DMC group compared with ARC.Ca^(2+)concentration of human AH in DMC was higher than that in ARC.The culture supernatant of cells transfected with miR-29b inhibitors had a higher concentration of Ca^(2+)than that transfected with miR-29b mimics.miR-29b reduced the viability of HLECs by upregulating CACNA1C expression.CONCLUSION:Exosomes isolated from human AH contains abundant miRNAs.A significantly expressed miR NA,miR-29b,can affect the concentration of Ca^(2+)and regulate HLEC processes by upregulating CACNA1C. | Chao Gao Xin Liu Fan Fan Jia-Ning Yang Xi-Yue Zhou Heng-Jun Mei Xiao-Lei Lin Yi Luo | 2021 | International Journal of Ophthalmology(English edition)2021，14，10： | 1 |
