| 3 | Pathologically triggered in situ aggregation of nanoparticles for inflammation-targeting amplification and therapeutic potentiation显示文摘Uncontrolled and persistent inflammation is closely related to numerous acute and chronic diseases.However,effective targeting delivery systems remain to be developed for precision therapy of inflammatory diseases.Herein we report a novel strategy for engineering inflammationaccumulation nanoparticles via phenolic functionalization.Different phenol-functionalized nanoparticles were first developed,which can undergo in situ aggregation upon triggering by the inflammatory/oxidative microenvironment.Phenolic compound-decorated poly(lactide-co-glycolide)nanoparticles,in particular tyramine(Tyr)-coated nanoparticles,showed significantly enhanced accumulation at inflammatory sites in mouse models of colitis,acute liver injury,and acute lung injury,mainly resulting from in situ cross-linking and tissue anchoring of nanoparticles triggered by local myeloperoxidase and reactive oxygen species.By combining a cyclodextrin-derived bioactive material with Tyr decoration,a multifunctional nanotherapy(TTN)was further developed,which displayed enhanced cellular uptake,antiinflammatory activities,and inflammatory tissue accumulation,thereby affording amplified therapeutic effects in mice with colitis or acute liver injury.Moreover,TTN can serve as a bioactive and inflammation-targeting nanoplatform for site-specifically delivering a therapeutic peptide to the inflamed colon post oral administration,leading to considerably potentiated in vivo efficacies.Preliminary studies also revealed good safety of orally delivered TTN.Consequently,Tyr-based functionalization is promising for inflammation targeting amplification and therapeutic potentiation of nanotherapies. | Qiang Nie Chenwen Li Yu Wang Yi Hu Wendan Pu Qixiong Zhang Jiajun Cai Yongyao Lin Gang Li Chenping Wang Lanlan Li Yin Dou Jianxiang Zhang | 2023 | Acta Pharmaceutica Sinica B2023,13,1: | 1 |
| 4 | Identification of two pIgR-like molecules in teleost fish with opposite roles in mucosal immunity against bacterial infection显示文摘Polymeric immunoglobulin(Ig)receptor-like(pIgRL)molecules have been identified in teleost fish.However,compared to functional studies of their related genes(e.g.,mammalian CD300 family)in eliminating pathogen invasion while preserving homeostasis,the roles of pIgRL in teleost fish remain unclear.In this study,we demonstrated that a pair of pIgRL molecules in zebrafish,pIgRL3.5 and pIgRL4.2,were highly expressed in the intestine and immune cells.Moreover,we constructed an Edwardsiella piscicida infection model,which induced strong inflammatory responses in the zebrafish intestine.Interestingly,pIgRL3.5 and pIgRL4.2 exhibited opposite inducible expression patterns in response to bacterial infection,suggesting that they perform different roles.More importantly,by conducting overexpression and knockdown experiments,our findings demonstrated that zebrafish pIgRL3.5 played a protective role in host defense against E.piscicida infection by inhibiting excessive inflammatory responses.In contrast,pIgRL4.2 facilitated pathogen growth and dissemination in zebrafish intestine.Collectively,our findings are the first to demonstrate that a pair of pIgRL molecules in teleost fish play opposite roles in mucosal immune response to bacterial infection.Therefore,our results provide crucial insights into the conserved role of pIgRL molecules in immune regulatory functions throughout vertebrate evolution. | Haoyue Xu Zixuan Wang Zhenyu Huang Xiaoyun Chen Ruiqi Lin Yongyao Yu Zhen Xu | 2023 | Water Biology and Security2023,2,4: | 0 |

| 6 | Machine learning molecular dynamics simulation identifying weakly negative effect of polyanion rotation on Li-ion migration显示文摘Understanding the physical picture of Li ion transport in the current ionic conductors is quite essential to further develop lithium superionic conductors for solid-state batteries.The traditional practice of directly extrapolating room temperature ion diffusion properties from the high-temperature(>600 K)ab initio molecular dynamics simulations(AIMD)simulations by the Arrhenius assumption unavoidably cause some deviations.Fortunately,the ultralong-time molecular dynamics simulation based on the machine-learning interatomic potentials(MLMD)is a more suitable tool to probe into ion diffusion events at low temperatures and simultaneously keeps the accuracy at the density functional theory level.Herein,by the low-temperature MLMD simulations,the non-linear Arrhenius behavior of Li ion was found for Li3ErCl6,which is the main reason for the traditional AIMD simulation overestimating its ionic conductivity.The 1μs MLMD simulations capture polyanion rotation events in Li_(7)P_(3)S_(11) at room temperature,in which four[PS_(4)]^(3−)tetrahedra belonging to a part of the longer-chain[P_(2)S_(7)]4−group are noticed with remarkable rotational motions,while the isolated group[PS_(4)]^(3−)does not rotate.However,no polyanion rotation is observed in Li10GeP_(2)S12,β-Li3PS_(4),Li3ErCl6,and Li3YBr6 at 300 K during 1μs simulation time.Additionally,the ultralong-time MLMD simulations demonstrate that not only there is no paddle-wheel effect in the crystalline Li_(7)P_(3)S_(11) at room temperature,but also the rotational[PS_(4)]^(3−)polyanion groups have weakly negative impacts on the overall Li ion diffusion.The ultralong-time MLMD simulations deepen our understanding of the relationship between the polyanion rotation and cation diffusion in ionic conductors at room environments. | Zhenming Xu Huiyu Duan Zhi Dou Mingbo Zheng Yixi Lin Yinghui Xia Haitao Zhao Yongyao Xia | 2023 | npj Computational Materials2023,,1: | 0 |
