1Seafloor geodetic network establishment and key technologies显示文摘Seafloor geodetic network construction involves the development of geodetic station shelter, network configuration design, location selection and layout, surveying strategy, observation model establishment and optimization, data processing strategy and so on. This paper tries to present main technological problems involved in the seafloor geodetic network construction, and seek the technically feasible solutions. Basic conceptions of developing seafloor geodetic station shelters for shallow sea and deep-sea are described respectively. The overall criteria of seafloor geodetic network construction for submarine navigation and those of network design for crustal motion monitoring are both proposed. In order to enhance application performances of the seafloor geodetic network, the seafloor network configuration should prefer a symmetrical network structure. The sea surface tracking line measurements for determining the seafloor geodetic station position should also adopt an approximately symmetrical configuration, and we recommend circle tracking line observations combined with cross-shaped line(or double cross-shape line) observations for the seafloor positioning mode. As to the offset correction between the Global Navigation Satellite System antenna phase center and the acoustic transducer, it is recommended to combine the calibration through external measurements and model parameter estimation. Besides, it is suggested to correct the sound speed error with a combination of observation value correction and parameterized model correction, and to mainly use the model correction to reduce the influence of acoustic ray error on the seafloor positioning. Following the proposed basic designs, experiments are performed in shallow sea area and deep-sea area respectively. 2IL-17 sustains the plasma cell response via p38-mediated Bcl-xL RNA stability in lupus pathogenesis显示文摘Recent studies have demonstrated a central role for plasma cells in the development of autoimmune diseases,such as systemic lupus erythematosus(SLE).Currently,both the phenotypic features and functional regulation of autoreactive plasma cells during SLE pathogenesis remain largely unclear.In this study,we first found that a major subset of IL-17 receptor-expressing plasma cells potently produced anti-dsDNA IgG upon IL-17A(IL-17)stimulation in SLE patients and lupus mice.Using a humanized lupus mouse model,we showed that the transfer of Th17 cell-depleted PBMCs from lupus patients resulted in a significantly reduced plasma cell response and attenuated renal damage in recipient mice compared to the transfer of total SLE PBMCs.Moreover,long-term BrdU incorporation in lupus mice detected highly enriched long-lived BrdU+subsets among IL-17 receptor-expressing plasma cells.Lupus mice deficient in IL-17 or IL-17 receptor C(IL-17RC)exhibited a diminished plasma cell response and reduced autoantibody production with attenuated renal damage,while the adoptive transfer of Th17 cells triggered the plasma cell response and renal damage in IL-17-deficient lupus mice.In reconstituted chimeric mice,IL-17RC deficiency resulted in severely impaired plasma cell generation but showed no obvious effect on germinal center B cells.Further mechanistic studies revealed that IL-17 significantly promoted plasma cell survival via p38-mediated Bcl-xL transcript stabilization.Together,our findings identified a novel function of IL-17 in enhancing plasma cell survival for autoantibody production in lupus pathogenesis,which may provide new therapeutic strategies for the treatment of SLE.Kongyang Ma Wenhan Du Fan Xiao Man Han Enyu Huang Na Peng Yuan Tang Chong Deng Lixiong Liu Yulan Chen Jingjing Li Shiwen Yuan Qin Huang Xiaoping Hong Dajun Hu Xiaoyan Cai Quan Jiang Dongzhou Liu Liwei Lu 2021Cellular & Molecular Immunology2021,18,7:6
3Parthenolide increases the sensitivity of gastric cancer cells to chemotherapy显示文摘OBJECTIVE:To investigate the effect of parthenolide(PTL),a sesquiterpene lactone medicinal compound,on the sensitivity of the gastric cancer cell line SGC7901 and the DPP-and ADR-resistant sublines SGC7901/DDP and SGC7901/ADR to cisplatin[diamminedichloroplatinum(Ⅱ),DDP]and adriamycin(ADR)in vitro.METHODS:SGC7901,SGC7901/DDP,and SGC7901/ADR were treated with various concentrations of PTL alone or in combination with DDP or ADR.The effects on cell proliferation,apoptosis,and expression/activity of several proliferation/apoptosis-related proteins[B-cell lymphoma-2(Bcl-2),cyclin D1,nuclear factor-kappa B(NF-κB),and Caspase-8]and drug transporters(P-glycoprotein and multidrug resistance protein-1)were measured using flow cytometry,Western blotting,and in vitro activity assays.RESULTS:Treatment of SGC7901 cells with PTL inhibited cell growth,increased apoptosis,and sensitized the cells to DPP.Mechanistically,PTL treatment resulted in downregulation of NF-κB activity and Bcl-2 expression,and upregulation of Caspase-8 activity.Similarly,PTL co-treatment of SGC7901/DDP and SGC7901/ADR overcame their resistance to DDP and ADR,respectively,with concomitant inhibition of NF-κB,Bcl-2,Cyclin D1,P-glycoprotein,and multidrug resistance protein-1 expression and/or activity.CONCLUSION:PTL treatment decreases drug resistance in SGC7901,SGC7901/DDP,and SGC7901/ADR cells,as reflected by induction of apoptosis,inhibition of proliferation,downregulation of pro-survival and drug resistance pathways,and upregulation of pro-apoptotic pathways.Our results suggest that co-treatment with PTL may thus complement existing therapies for gastric cancer.Liu Minghua Yang Ying Liu Dajun Cao Ye Li Yan 2020Journal of Traditional Chinese Medicine2020,40,6:5
4Characterization of a PDR type ABC transporter gene from wheat (Triticum aestivum L.)显示文摘DON, as a virulence factor, plays an important role in the infection of Fusarium graminearum in wheat. The infection ability of F. graminearum depends on its capacity of producing DON. The production of DON by F. graminearum is significantly decreased in the wheat varieties with scab resistance. In this study, GeneChip analysis indicated that an EST encoding an ATP-binding cassette (ABC) transporter was up-regulated by 45 times in a wheat landrace Wangshuibai, which is resistant to DON accumulation. A pair of EST-derived primers were designed based on the EST sequence, and a clone was then isolated from a wheat genomic DNA TAC library. The TAC clone was sequenced using chromosome walking and gene prediction was conducted using Softberry. A cDNA clone of this gene was subsequently isolated from Wangshuibai induced by DON using gene-specific primers designed according to the untranslated sequence of the gene. The genome size of the gene is 7377 bp, consisting of 19 exons with coding sequences of 4308 bp. It encodes a protein with 1435 amino acid residues and the calculated molecular weight is about 161 kD. BLAST analysis indicated that the gene may belong to pleiotropic drug resistance (PDR) sub-family, and hence designated as TaPDR1 (Triticum aestivum pleiotropic drug resistance). TaPDR1 was located on chromosome 5A of wheat using nullisomic-tetrasomic lines of Chinese Spring. TaPDR1 was up-regulated by induction of both DON and F. graminearum. Expression patterns of TaPDR1 were different in wild-type Wangshuibai and the fast-neutron induced Wangshuibai mutant lacking FHB1, a major QTL of FHB resistance and DON resistance in chromosome arm 3BS. These results suggested that TaPDR1 might be a candidate gene responsible for DON ac-cumulation resistance. The expression profile showed that TaPDR1 expression was neither induced by hormones typically involved in biotic stress, such as JA and SA, nor by abiotic stresses, such as heat, cold, wounding and NaCl. 5Coordinated transcription of ANRIL and P16 genes is silenced by P16 DNA methylation显示文摘Objective: To investigate the relationship between the transcription of ANRIL, P15, P14 and P16 at the same locus and the regulation mechanism of ANRIL.Methods: Publicly available database of Cancer Cell Line Encyclopedia(CCLE) was used in bioinformatic analyses. Methylation of Cp G islands was detected by denaturing high performance liquid chromatography(DHPLC). Gene transcript levels were determined using quantitative real-time polymerase chain reaction(q RTPCR) assays. An engineered P16-specific transcription factor and DNA methyltransferase were used to induce P16-specific DNA demethylation and methylation.Results: The expression level of ANRIL was positively and significantly correlated with that of P16 but not with that of P15 in the CCLE database. This was confirmed in human cell lines and patient colon tissue samples. In addition, ANRIL was significantly upregulated in colon cancer tissues. Transcription of ANRIL and P16 was observed only in cell lines in which the P16 alleles were unmethylated and not in cell lines with fully methylated P16 alleles.Notably, P16-specific methylation significantly decreased transcription of P16 and ANRIL in BGC823 and GES1 cells. 14Effects of CeO2 preparation methods on the catalytic performance of MoO3/CeO2 toward sulfur-resistant methanation显示文摘CeO_2 supports were prepared by calcination or precipitation method and 5% MoO_3/CeO_2 catalysts were prepared by incipient-wetness impregnation method. The catalytic performance of the 5% MoO_3/CeO_2 catalysts toward sulfur-resistant methanation was investigated. The results showed that the Mo/Ce-1 catalysts with CeO_2 support prepared by calcination method exhibited the best sulfur-resistant methanation activity and stability with CO conversion as high as 75% while the Mo/Ce-3 catalysts the poorest. The supports and catalysts were characterized by N_2-adsorption–desorption, temperature-programmed reduction(TPR), X-ray diffraction(XRD), Raman spectroscopy(RS) and scanning electron microscope(SEM). The results indicated that the saturated monolayer loading MoO_3 on Ce-3 support was lower than 5% and there were some crystalline MoO_3 particles on the surface of the Mo/Ce-3. 15A Block Parallel l_0-Norm Penalized Shrinkage and Widely Linear Affine Projection Algorithm for Adaptive Filter显示文摘To improve the identification capability of AP algorithm in time-varying sparse system, we propose a block parallel l_0-SWL-DCD-AP algorithm in this paper. In the proposed algorithm, we first introduce the l_0-norm constraint to promote its application for sparse system. Second, we use the shrinkage denoising method to improve its track ability. Third, we adopt the widely linear processing to take advantage of the non-circular properties of communication signals. Last, to reduce the high computational complexity and make it easy to implemented, we utilize the dichotomous coordinate descent(DCD) iterations and the parallel processing to deal with the tapweight update in the proposed algorithm. To verify the convergence condition of the proposed algorithm, we also analyze its steadystate behavior. Several simulation are done and results show that the proposed algorithm can achieve a faster convergence speed and a lower steady-state misalignment than similar APA-type algorithm. 19Review of stopping power and Coulomb explosion for molecular ion in plasmas显示文摘We summarize our theoretical studies for stopping power of energetic heavy ion,diatomic molecular ions and small clusters penetrating through plasmas.As a relevant research field for the heavy ion inertial confinement fusion(HICF),we lay the emphasis on the dynamic po-larization and correlation effects of the constituent ion within the molecular ion and cluster for stopping power in order to disclose the role of the vicinage effect on the Coulomb explosion and energy deposition of molecules and clusters in plasma.On the other hand,as a promising scheme for ICF,both a strong laser field and an intense ion beam are used to irradiate a plasma target.So the influence of a strong laser field on stopping power is significant.We discussed a large range of laser and plasma parameters on the coulomb explosion and stopping power for correlated-ion cluster and C 60 cluster.Furthermore,in order to indicate the effects of different cluster types and sizes on the stopping power,a comparison is made for hydrogen and carbon clusters.In addition,the deflection of molecular axis for diatomic molecules during the Coulomb explosion is also given for the cases both in the presence of a laser field and laser free.Finally,a future experimental scheme is put forward to measure molecular ion stopping power in plasmas in Xi’an Jiaotong University of China.Guiqiu Wang He Yi Yujiao Li Yaochuan Wang Dajun Liu Fei Gao Wei Liu Jieru Ren Xing Wang Yongtao Zhao Younian Wang 2018Matter and Radiation at Extremes2018,3,2:1
