1Effects of lipopolysaccharides stimulated Kupffer cells on activation of rat hepatic stellate cells显示文摘AIM:To study the effects of Kupffer cell-conditioned medium (KCCM) derived from lipopolysaccharide (LPS) treatment on proliferation of rat hepatic stellate cells (HSC).METHODS:HSC and Kupffer cells were isolated from the liver of Wistar rats by in situ perfusion with pronase and collagenase and density gradient centrifugation with Nycodenz and cultured. KCCM was prepared and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) colorimetric assay was used to detect HSC proliferation. The content of type Ⅳ collagen and laminin secreted by HSC in the HSC-conditioned medium was determined by radioimmunoassay.TGF-β1 production in the KCCM was detected by enzymelinked immunosorbent assay (ELISA).RESULTS:HSC and Kupffer cells isolated had high purity.One microgram per mililiter LPS-activated KCCM and unstimulated KCCM could significantly promote HSC proliferation [0.132±0.005 and 0.123±0.008 vscontrol group (0.100±0.003), P<0.01], and there was a difference between them (P<0.05). Ten microgram per mililiter LPS-activated KCCM (0.106±0.010) was unable to promote HSC proliferation (P>0.05). Adding anti-TGF-β1 antibodies could suppress the proliferation promoted by unstimulated KCCM and LPS(1μg/ml)-activated KCCM (0.109±0.009 vs 0.123±0.008,0.115±0.008 vs 0.132±0.005, P<0.01). LPS (1μg/ml or 10μg/ml) could not promote HSC proliferation immediately (0.096±0.003 and 0.101±0.004 vs 0.100±0.003, P>0.05). There was a parallel behavior between HSC proliferation and increased ECM level. One microgram per mililiter LPS-activated KCCM contained a larger amount of TGF-β1 than unstimulated KCCM.CONCLUSION:The technique for isolation of HSC and Kupffer cells described here is simple and reliable. 2The Study of Corneal Topography in Myopic and Hyperopic Children显示文摘Purpose: To compare the differences of corneal topographies in myopic and hyperopic children and study the effect of Atropin on their changes.Methods: The refractive components of 136 eyes with different refractive conditions were measured with A-Scan and their corneal topographies with and without cycloplegia were obtained respectively.Results: The mean corneal power of zones 3mm (MD3, P=0.031 ) and minor keratometer K2 (P=0.003) of myopia are greater than those of hyperopia without cycloplegia. MD3 (P=0.009) and Keratometer K1 (P = 0.025) increased in hyperopic eyes, while MD3(P=0.033), K1 (P = 0.035) and K2 (P = 0.002) decreased in myopic eyes significantly after cycloplegia. Similarly, the mean corneal power of zones 5mm (MD5) and 7mm (MD7) in myopic eyes decreased dramatically (P ≤ 0.001 ).Conclusions: The corneal power was found to be greater in myopia than that in hyperopia. 4The Synergistic Inhibitory Effect of STI571 in Combination with Arsenic Trioxide (As2O3) on Multidrug-Resistant Leukemic Cells显示文摘OBJECTIVE To study the synergistic effect of STI571, an inhibitor of tyrosine kinase, in combination with arsenic trioxide As2O3 on a multidrug-resistant leukemia cell line expressing bcr-abl. METNODS The cytotoxic effect of STI571 alone or in combination with different concentrations of As2O3 on the bcr-abl and mdrl -positive leukemia cell line, K562-n/VCR, was examined by the MTT method. RESULTS One μmol/L of STI571 alone had no significant cytotoxic effect on K562-n/VCR cells. However the cytotoxic effect increased markedly when combined with As2O3 at concentrations of 10^-5, 10^-6, 10^-7 and 10^-8 mol/L. The IC50 of K562-n/VCR cells in As2O3 group was 1.879 μmol/L, with. Upon addition of STI571, the IC50 decreased to 0.155 μmol/L resulting in a synergistic cytotoxic effect on K562-n/VCR cells that was increased 12.1 times.
