| 1 | Effect of Simulated Acid Rain on Potential Carbon and Nitrogen Mineralization in Forest Soils显示文摘Acid rain is a serious environmental problem worldwide. In this study, a pot experiment using forest soils planted with the seedlings of four woody species was performed with weekly treatments of pH 4.40, 4.00, 3.52, and 3.05 simulated acid rain (SAR) for 42 months compared to a control of pH 5.00 lake water. The cumulative amounts of C and N mineralization in the five treated soils were determined after incubation at 25 ℃ for 65 d to examine the effects of SAR treatments. For all five treatments, cumulative CO2-C production ranged from 20.24 to 27.81 mg kg-1 dry soil, net production of available N from 17.37 to 48.95 mg kg-1 dry soil, and net production of NO-3 -N from 9.09 to 46.23 mg kg-1 dry soil. SAR treatments generally enhanced the emission of CO2-C from the soils; however, SAR with pH 3.05 inhibited the emission. SAR treatments decreased the net production of available N and NO3-N. The cumulative CH4 and N2O productions from the soils increased with increasing amount of simulated acid rain. The cumulative CO2-C production and the net production of available N of the soil under Acmena acuminatissima were significantly higher (P ≤ 0.05) than those under Schima superba and Cryptocarya concinna. The mineralization of soil organic C was related to the contents of soil organic C and N, but was not related to soil pH. | 3 | Spatial Characteristics of Extreme Rainfall over China with Hourly through 24-Hour Accumulation Periods Based on National-Level Hourly Rain Gauge Data显示文摘Hourly rainfall measurements of 1919 national-level meteorological stations from 1981 through 2012 are used to document,for the first time,the climatology of extreme rainfall in hourly through 24-h accumulation periods in China. Rainfall amounts for 3-,6-,12- and 24-h periods at each station are constructed through running accumulation from hourly rainfall data that have been screened by proper quality control procedures. For each station and for each accumulation period,the historical maximum is found,and the corresponding 50-year return values are estimated using generalized extreme value theory. Based on the percentiles of the two types of extreme rainfall values among all the stations,standard thresholds separating Grade I,Grade II and Grade III extreme rainfall are established,which roughly correspond to the 70th and 90th percentiles for each of the accumulation periods. The spatial characteristics of the two types of extreme rainfall are then examined for different accumulation periods. The spatial distributions of extreme rainfall in hourly through 6-h periods are more similar than those of 12- and 24-h periods. Grade III rainfall is mostly found over South China,the western Sichuan Basin,along the southern and eastern coastlines,and in the large river basins and plains. There are similar numbers of stations with Grade III extreme hourly rainfall north and south of 30°N,but the percentage increases to about 70% south of 30°N as the accumulation period increases to 24 hours,reflecting richer moisture and more prolonged rain events in southern China. | 6 | Effect of N and P addition on soil organic C potential mineralization in forest soils in South China显示文摘Atmospheric nitrogen deposition is at a high level in some forests of South China. The effects of addition of exogenous N and P on soil organic carbon mineralization were studied to address: (1) if the atmospheric N deposition promotes soil C storage through decreasing mineralization; (2) if the soil available P is a limitation to organic carbon mineralization. Soils (0-10 cm) was sampled from monsoon evergreen broad-leaved forest (MEBF), coniferous and broad-leaved mixed forest (CBMF), and Pinus massoniana forest (PMF) in Dinghushan Biosphere Reserve (located in Guangdong Province, China). The soils were incubated at 25°C for 45 weeks, with addition of N (NH4NO3 solution) or P (KH2PO4 solution). CO2-C emission and the inorganic N (NH4+-N and NO3?-N) of the soils were determined during the incubation. The results showed that CO2-C emission decreased with the N addition. The addition of P led to a short-term sharp increase in CO2 emission after P application, and the responses of CO2-C evolution to P addition in the later period of incubation related to forest types. Strong P inhibition to CO2 emission occurred in both PMF and CBMF soils in the later incubation. The two-pool kinetic model was fitted well to the data for C turnover in this experiment. The model analysis demonstrated that the addition of N and P changed the distribution of soil organic C between the labile and recalcitrant pool, as well as their mineralization rates. In our experiment, soil pH can not completely explain the negative effect of N addition on CO2-C emission. The changes of soil inorganic N during incubation seemed to support the hypothesis that the polymerization of added nitrogen with soil organic compound by abiotic reactions during incubation made the added nitrogen retard the soil organic carbon mineralization. | 13 | Involvement of ayu NOD2 in NF-κB and MAPK signaling pathways: Insights into functional conservation of NOD2 in antibacterial innate immunity显示文摘Nucleotide oligomerization domain 2(NOD2) is a major cytoplasmic sensor for pathogens and is critical for the clearance of cytosolic bacteria in mammals.However, studies regarding NOD2, especially the initiated signaling pathways, are scarce in teleost species. In this study, we identified a NOD2 molecule(PaNOD2) from ayu(Plecoglossus altivelis).Bioinformatics analysis showed the structure of NOD2 to be highly conserved during vertebrate evolution. Dual-luciferase reporter assays examined the activation of NF-κB signaling and Western blotting analysis detected the phosphorylation of three MAP kinases(p-38, Erk1/2, and JNK1/2).Functional study revealed that, like its mammalian counterparts, PaNOD2 was the receptor of the bacterial cell wall component muramyl dipeptide(MDP), and the leucine-rich repeat motif was responsible for the recognition and binding of Pa NOD2 with the ligand. Overexpression of PaNOD2 activated the NF-κB signaling pathway, leading to the upregulation of inflammatory cytokines, including TNF-α and IL-1β in HEK293 T cells and ayu head kidney-derived monocytes/macrophages(MO/MΦ).Particularly, we found that PaNOD2 activated the MAPK signaling pathways, as indicated by the increased phosphorylation of p-38, Erk1/2, and JNK1/2, which have not been characterized in any teleost species previously. Our findings proved that the NOD2 molecule and initiated pathways are conserved between mammals and ayu. | 14 | Clinical risk score for invasive fungal diseases in patients with hematological malignancies undergoing chemotherapy:China Assessment of Antifungal Therapy in Hematological Diseases (CAESAR) study显示文摘Invasive fungal disease (IFD) is a major infectious complication in patients with hematological malignancies.In this study,we examined 4889 courses of chemotherapy in patients with hematological diseases to establish a training dataset (n=3500) by simple random sampling to develop a weighted risk score for proven or probable IFD through multivariate regression,which included the following variables: male patients,induction chemotherapy for newly diagnosed or relapsed disease,neutropenia,neutropenia longer than 10 days,hypoalbuminemia,central-venous catheter,and history of IFD.The patients were classified into three groups,which had low (0-10,~1.2%),intermediate (11-15,6.4%),and high risk (> 15,17.5%) of IFD.In the validation set (n=1389),the IFD incidences of the groups were ~1.4%,5.0%,and 21.4%.In addition,we demonstrated that antifungal prophylaxis offered no benefits in low-risk patients,whereas benefits were documented in intermediate (2.1% vs.6.6%,P=0.007) and high-risk patients (8.4% vs.23.3%,P=0.007).To make the risk score applicable for clinical settings,a pre-chemo risk score that deleted all unpredictable factors before chemotherapy was established,and it confirmed that anti-fungal prophylaxis was beneficial in patients with intermediate and high risk of IFD.In conclusion,an objective,weighted risk score for IFD was developed,and it may be useful in guiding antifungal prophylaxis. | Ling Wang Ying Wang Jiong Hu Yuqian Sun He Huang Jing Chen Jianyong Li Jun Ma Juan Li Yingmin Liang Jianmin Wang Yan Li Kang Yu Jianda Hu Jie Jin Chun Wang Depei Wu Yang Xiao Xiaojun Huang | 2019 | Frontiers of Medicine2019,13,3: | 10 |

| 16 | Molecular characterization of an IL-1β gene from the large yellow croaker(Larimichthys crocea) and its effect on fish defense against Vibrio alginolyticus infection显示文摘Interleukin 1β(IL-1β), the first interleukin to be characterized, plays a key role in regulating the immune response. In this study, we determined the c DNA and genomic DNA sequences of the IL-1β gene from the large yellow croaker, Larimichthys crocea. Phylogenetic analysis indicated that the IL-1β(Lc IL-1β) gene was most closely related to that of European seabass(Dicentrarchus labrax), sharing 67.8% amino acid identity. In healthy large yellow croaker, Lc IL-1β transcription was detected in all tested tissues, with the highest level found in the head kidney. Upon Vibrio alginolyticus infection, Lc IL-1β transcription in all tested tissues was significantly upregulated. Intraperitoneal injection of recombinant Lc IL-1β(r Lc IL-1β) improved the survival rate and reduced the tissue bacterial load after V. alginolyticus infection. In addition, r Lc IL-1β induced monocytes/macrophages(MO/MΦ) chemotaxis and increased phagocytosis and bactericidal activity in vitro. In this study, the soils at 0–3, 3–13, and 13–23 cm depths were sampled from six urban parks, differing in reconstruction intensity(mainly changes made during conversion of natural forests into parklands), in the Pearl River Delta, China to determine how reconstruction intensity influenced the extent of acidification and heavy metal levels in the soils of urban parks in a humid subtropical environment. High reconstruction intensity(HRI) was practiced in three parks and low reconstruction intensity(LRI) in three other parks. The LRI soils were strongly to extremely acidic(with low exchangeable Ca, Mg, and K concentrations) while the HRI soils were much less acidic. Both total and extractable concentrations of soil heavy metals were related to the specific management practices and age of the park, but did not differ significantly between LRI and HRI parks or among soil depths. Soil p H was significantly related to soil exchangeable cation concentrations and base saturation but was weakly related or unrelated to soil heavy metal levels. | 20 | Induction of cytotoxic T lymphocyte respones in vivo after immunotherapy with dendritic cells in patients with nasopharyngeal carcinoma显示文摘The aim of the present study was to determine the efficacy of immunotherapy with dendritic cells to elicit EBV-specific CTL-immunity in advanced cases of EBV-positive patients with nasopharyngeal carcinoma (NPC) and to determine the safety and toxicity of this preparation. Nine cases of histologically confirmed patients with NPC undergoing treatment with radiological therapy were enrolled in this study. Dendritic cells, generated in vitro from blood monocytes of patients were cultured and matured with cytokines and then infected with recombinant adenovirus vaccine containing EBV-latent membrane protein-2 (Ad-LMP2). On 9 days' cultivation of cells, the matured DCs were harvested, irradiated with Co and then injected intradermally to patients with NPC. The injections were performed 3 times totally. After immunization, the CTL responses were assayed by means of cytotoxicity and epitope-specific IFN-γproduction. The results of this trial showed that all patients could tolerate this kind of treatment without any side effect, during which marked increase of LMP2-specific CTL-responses could be demonstrated in 5 patients of this group. And the level of IgA/VCA antibody decreased in 8 of 9 patients, thus accounting for a better prognosis for these patients. All patients will be followed up for another one year. At least, the present work shows that intradermal vaccination with autologous DCs infected with recombinant Ad-LMP2 adenovirus is a safe procedure in NPC patients, in which this procedure can enhance the LMP2-specific CTL responses in patients.
