| 2 | Profiling of hepatocellular carcinoma neoantigens reveals immune microenvironment and clonal evolution related patterns显示文摘Objective: Neoantigens derived from tumor-specific genomic alterations have demonstrated great potential for immunotherapeutic interventions in cancers. However, the comprehensive profile of hepatocellular carcinoma(HCC) neoantigens and their complex interplay with immune microenvironment and tumor evolution have not been fully addressed.Methods: Here we integrated whole exome sequencing data, transcriptome sequencing data and clinical information of 72 primary HCC patients to characterize the HCC neoantigen profile, and systematically explored its interactions with tumor clonal evolution, driver mutations and immune microenvironments.Results: We observed that higher somatic mutation/neoantigen load was associated with better clinical outcomes and HCC patients could be further divided into two subgroups with distinct prognosis based on their neoantigen expression patterns. HCC subgroup with neoantigen expression probability high(NEP-H) showed more aggressive pathologic features including increased incidence of tumor thrombus(P=0.038), higher recurrence rate(P=0.029),more inclined to lack tumor capsule(P=0.026) and with more microsatellite instability sites(P=0.006). In addition,NEP-H subgroup was also characterized by higher chance to be involved in tumor clonal evolution [odds ratio(OR)=46.7, P<0.001]. Gene set enrichment analysis revealed that upregulation of MYC and its targets could suppress immune responses, leading to elevated neoantigen expression proportion in tumor cells. Furthermore, we discovered an immune escape mechanism that tumors could become more inconspicuous by evolving subclones with less immunogenicity. We observed that smaller clonal mutation clusters with higher immunogenicity in tumor were more likely to involve in clonal evolution. | 3 | A novel low pressure-difference fluctuation electro-hydraulic large flowrate control valve for fuel flowrate control of aeroengine afterburner system显示文摘To address the control accuracy of large fuel flowrate during pressure fluctuation,a novel electro-hydraulic fuel metering unit(FMU)is constructed for afterburner fuel system of military aeroengine.Different from the previous FMU,the proposed FMU can achieve the higher precision opening control by a new metering valve with double control chambers(MVDCC),and realize the lower pressure difference fluctuation regulating by a novel two-stage constant pressure difference compensated valve(CPDCV)with dynamic damping orifice and damping piston.The experimental and AMESim simulation results verify the validity and superiority of the novel FMU.Since the temperature-induced variation in fuel properties and device capabilities may degrade or even impair the properties of novel FMU,the discharge flowrate is analyzed by global sensitivity analysis to research the effect proportion of each factor,the temperature effect is explored to ensure the working reliability in long-span temperature variation.Finally,the optimization of structure parameters for novel CPDCV can further reduce pressure difference fluctuation during pressure regulation,and the overshoot,adjust time and the integral of time multiplied by absolute value of error(ITAE)can be reduced by 24%,30%and 26%,respectively.This paper provides a reference for improving the stability of large flowrate during pressure fluctuation. | Jie HANG Yunhua LI Liman YANG | 2022 | Chinese Journal of Aeronautics2022,35,11: | 1 |

| 6 | Tunable light emission from carbon dots by controlling surface defects显示文摘Carbon dots(CDs)are metal-free fluorescent materials that can be used in optical and electronic devices,but few studies have focused on one-step synthesis routes for CDs with tunable color and high photoluminescence quantum yield(PLQY).Herein,CDs with tunable light emission were synthesized using a novel amide-assisted solvothermal approach.The as-prepared CDs were well dispersed and homogeneous,with average diameters of approximately 2.0–4.0 nm,depending on the dopants.Owing to the surface states with different ratios of nitrogen-and oxygen-related species,different CDs can exhibit blue,green,red,or white emission with relatively high PLQYs of 61.6%,41.3%,29.1%and 19.7%,respectively.XPS measurements,in conjunction with DFT calculations,indicate that nitrogen substitution(pyridinic/pyrrolic nitrogen)dominates the blue emission,while introducing oxygen functional groups lowered the LUMO energy level,which resulted in redder emission.In addition,the CDs are demonstrated as a bioimaging probe in both in vitro and in vivo assays,and the white light CDs have been demonstrated to be potential fluorescent materials for white-light-emitting diode(WLED). | Huijun Li Sancan Han Bowen Lyu Ting Hong Shibo Zhi Ling Xu Fengfeng Xue Liman Sai Junhe Yang Xianying Wang Bin He | 2021 | Chinese Chemical Letters2021,32,9: | 1 |

| 12 | Vote-Based Feature Selection Method for Stratigraphic Recognition in Tunnelling Process of Shield Machine显示文摘Shield machines are currently the main tool for underground tunnel construction. Due to the complexity and variability of the underground construction environment, it is necessary to accurately identify the ground in real-time during the tunnel construction process to match and adjust the tunnel parameters according to the geological conditions to ensure construction safety. Compared with the traditional method of stratum identifcation based on staged drilling sampling, the real-time stratum identifcation method based on construction data has the advantages of low cost and high precision. Due to the huge amount of sensor data of the ultra-large diameter mud-water balance shield machine, in order to balance the identifcation time and recognition accuracy of the formation, it is necessary to screen the multivariate data features collected by hundreds of sensors. In response to this problem, this paper proposes a voting-based feature extraction method (VFS), which integrates multiple feature extraction algorithms FSM, and the frequency of each feature in all feature extraction algorithms is the basis for voting. At the same time, in order to verify the wide applicability of the method, several commonly used classifcation models are used to train and test the obtained efective feature data, and the model accuracy and recognition time are used as evaluation indicators, and the classifcation with the best combination with VFS is obtained. The experimental results of shield machine data of 6 diferent geological structures show that the average accuracy of 13 features obtained by VFS combined with diferent classifcation algorithms is 91%;among them, the random forest model takes less time and has the highest recognition accuracy, reaching 93%, showing best compatibility with VFS. Therefore, the VFS algorithm proposed in this paper has high reliability and wide applicability for stratum identifcation in the process of tunnel construction, and can be matched with a variety of classifer algorithms. | 13 | Molecular Cloning, Expression Pattern, and 3D Structural Prediction of the Cold Inducible RNA- Binding Protein(CIRP) in Japanese Flounder(Paralichthys olivaceus)显示文摘Cold-inducible RNA-binding protein(CIRP) is a kind of RNA binding proteins that plays important roles in many physiological processes. The CIRP has been widely studied in mammals and amphibians since it was first cloned from mammals. On the contrary, there are little reports in teleosts. In this study, the Po CIRP gene of the Japanese flounder was cloned and sequenced. The genomic sequence consists of seven exons and six introns. The putative Po CIRP protein of flounder was 198 amino acid residues long containing the RNA recognition motif(RRM). Phylogenetic analysis showed that the flounder Po CIRP is highly conserved with other teleost CIRPs. The 5' flanking sequence was cloned by genome walking and many transcription factor binding sites were identified. There is a Cp Gs region located in promoter and exon I region and the methylation state is low. Quantitative real-time PCR analysis uncovered that Po CIRP gene was widely expressed in adult tissues with the highest expression level in the ovary. The m RNA of the Po CIRP was maternally deposited and the expression level of the gene was regulated up during the gastrula and neurula stages. In order to gain the information how the protein interacts with m RNA, we performed the modeling of the 3D structure of the flounder Po CIRP. The results showed a cleft existing the surface of the molecular.
