| 1 | Regulation of extracellular signal-regulated kinase 1/2 influences hippocampal neuronal survival in a rat model of diabetic cerebral ischemia显示文摘Activation of extracellular signal-regulated kinase 1/2 has been demonstrated in acute brain ischemia. We hypothesized that activated extracellular signal-regulated kinase 1/2 can protect hippocampal neurons from injury in a diabetic model after cerebral ischemia/reperfusion. In this study, transient whole-brain ischemia was induced by four-vessel occlusion in normal and diabetic rats, and extracellular signal-regulated kinase 1/2 inhibitor(U0126) was administered into diabetic rats 30 minutes before ischemia as a pretreatment. Results showed that the number of surviving neurons in the hippocampal CA1 region was reduced, extracellular signal-regulated kinase 1/2 phosphorylation and Ku70 activity were decreased, and pro-apoptotic Bax expression was upregulated after intervention using U0126. These findings demonstrate that inhibition of extracellular signal-regulated kinase 1/2 activity aggravated neuronal loss in the hippocampus in a diabetic rat after cerebral ischemia/reperfusion, further decreased DNA repairing ability and accelerated apoptosis in hippocampal neurons. | 2 | Simulation and experimental investigation of inner-jet electrochemical grinding of GH4169 alloy显示文摘GH4169 alloy is one of the most commonly used materials in aero engine turbine blades,but its machinability is poor because of its excellent strength at high temperatures. Electrochemical machining(ECM) has become a common method for machining this alloy and other difficult-tomachine materials. Electrochemical grinding(ECG) is a hybrid process combining ECM and conventional grinding. In this paper, investigations conducted on inner-jet ECG of GH4169 alloy are described. Two types of inner-jet ECG grinding wheels were used to machine a flat bottom surface.The machining process was simulated using COMSOL software, and machining gaps under different machining parameters were obtained. In addition, maximum feed rates and maximum material removal rates under different machining parameters were studied experimentally. The maximum sizes and the uniformity of the distributions of the gaps machined by the two grinding wheels were compared. | 3 | Protective effects of dl-3n-butylphthalide against diffuse brain injury显示文摘Dl-3n-butylphthalide can effectively treat cerebral ischemia;however,the mechanisms underlying the effects of dl-3n-butylphthalide on microcirculation disorders following diffuse brain injury remain unclear.In this study,models of diffuse brain injury were established in Sprague-Dawley rats with the vertical impact method.Dl-3n-butylphthalide at 80 and 160 mg/kg was given via intraperitoneal injection immediately after diffuse brain injury.Ultrastructural changes in the cerebral cortex were observed using electron microscopy.Cerebral blood flow was measured by laser Doppler flowmetry,vascular density was marked by tannic acid-ferric chloride staining,vascular permeability was estimated by the Evans blue method,brain water content was measured using the dry-wet method,and rat behavior was measured by motor function and sensory function tests.At 6,24,48,and 72hours after administration of dl-3n-butylphthalide,reduced cerebral ultrastructure damage,increased vascular density and cerebral blood flow,and improved motor and sensory functions were observed.Our findings demonstrate that dl-3n-butylphthalide may have protective effects against diffuse brain injury by ameliorating microcirculation disorder and reducing blood-brain barrier damage and cerebral edema. | Yaning Zhao Jianmin Li Pan Zhang Changxiang Chen Shuxing Li | 2013 | Neural Regeneration Research2013,8,28: | 7 |
| 4 | Gene characterization and molecular pathway analysis of reverse thermosensitive genic male sterility in eggplant(Solanum melongena L.)显示文摘The naturally occurring mutant eggplant line 05ms was identified with reverse thermosensitive genic male sterility(rTGMS),but its temperature-responsive fertility mechanisms remain largely unknown.Here,we studied the flower morphology,anther cellular structure,and genome-wide gene expression of this rTGMS line.Candidate genes for thermosensitive male sterility during the microspore development of 05ms and the temperature-insensitive line S63 under low-temperature(LT)and high-temperature(HT)conditions were identified.Under LT,tapetum cells were vacuolated and had delayed disintegration in 05ms.RNA-seq analysis indicated that DEGs were enriched in the KEGG pathways‘plant hormone signal transduction’,‘starch and sucrose metabolism’,and‘phenylpropanoid biosynthesis’.We identified two genes,4CLL1(Sme2.5_00368.1_g00010.1)and CKI1(Sme2.5_10056.1_g00002.1),which could potentially regulate eggplant anther development and may be candidate genes for rTGMS.Finally,we propose a working model of anther abortion for rTGMS in eggplant.CKI1 responds to LT stress and causes expression changes in genes related to anther development,such as 4CLL1,and the cellular structure of the tapetum becomes abnormal,causing male sterility.The findings of this study explain the underlying molecular mechanisms of male sterility in eggplant rTGMS lines. | Bing Li Xueping Chen Yanrong Wu Aixia Gu Jingjing Zhang Shuangxia Luo Xiurui Gao Jianjun Zhao Xiuqing Pan Shuxing Shen | 2019 | Horticulture Research2019,6,1: | 5 |
| 5 | Controlling the size of fragrance microcapsules using designed agitator paddles: Experiment and CFD simulation显示文摘Controlling the size of fragranee microcapsules using designed agitator paddles was investigated and studied by CFD simulation. First, different fluid flows were established by varying stirring speeds, reactor scales, and agitator paddle design, and the effects of each on particle size and distribution of prepared microcapsules were determined. The experimental results showed that the pattern design of orifices in the plate paddles control the flow field well. Narrow particle-size distributio ns of the microcapsules were obtained. The fluid flow characteristics including fluid velocity field, turbulent kinetic energy field, and shear stress distribution for thedifferent agitator paddle types in different reaction kettles were simulated using CFD tech no logy. The correlations between simulated data and experime ntal results were an alyzed. | 6 | Quantification and assessment of fault uncertainty and risk using stochastic conditional simulations显示文摘The effect of geological uncertainty on the development and mining of underground coal deposits is a key issue for longwall mining, as the presence of faults generates substantial monetary losses. This paper develops a method for the conditional simulation of fault systems and uses the method to quantify and assess fault uncertainty. The method is based on the statistical modelling of fault attributes and the simulation of the locations of the centres of the fault traces. Fault locations are generated from the thinning of a Poisson process using a spatially correlated probability field. The proposed algorithm for simulating fault traces takes into account soft data such as geological interpretations and geomechanical data. | 7 | Exogenous melatonin improves the chilling tolerance and preharvest fruit shelf life in eggplant by affecting ROS-and senescence-related processes显示文摘Low temperature is the most common abiotic stress factors during the eggplant cultivation in solar greenhouses.Melatonin plays important roles in plant resistance to low temperature.However,the role of melatonin in regulating chilling tolerance and extending the preharvest shelf life of eggplant fruits is still unknown.In this study,we investigated the effects of exogenous melatonin on eggplant plants and fruits in response to low temperature.Under simulated low-temperature conditions,exogenous melatonin significantly relieved the chilling symptoms of seedlings by reducing reactive oxygen species (ROS) and malondialdehyde (MDA) levels and relative leakage rates.These reductions were caused by higher superoxide dismutase (SOD) and catalase (CAT) activities and increased endogenous polyamine and melatonin levels compared with those in untreated seedlings.Notably,the expression levels of SOD,CAT1/2,and polyamine synthesis genes (ADC and ODC) were also increased by 100μmol·L~(-1)melatonin,as well as those of genes involved in melatonin synthesis (TDC,T5H,SNAT,ASMT,and COMT) and cold regulation (COR1,CBFa/b,and ZAT2/6/12).To further investigate the effects of melatonin on eggplant leaves and fruits under natural low temperature conditions,100μmol·L~(-1)melatonin was sprayed on the functional leaves at three days before commodity maturation.Melatonin significantly alleviated chilling injury in the leaves and pericarp and extended the preharvest shelf life of the fruit by increasing the expression of COR1,CBF,ZAT2/6/12,and API5 and decreasing the expression of senescence-related genes (NCED1/2 and SAG12).Therefore,100μmol·L~(-1)melatonin improved chilling tolerance and fruit shelf life by upregulating ZAT2/6/12 to affect ROS-and senescence-related processes,which provides a reference for alleviating cold stress and extending the preharvest fruit shelf life in eggplant. | Lijun Song Zhuo Tan Weiwei Zhang Qiang Li Zhaoxiang Jiang Shuxing Shen Shuangxia Luo Xueping Chen | 2023 | Horticultural Plant Journal2023,9,3: | 2 |

| 10 | Apolipoprotein E mimetic peptide protects against diffuse brain injury显示文摘Apolipoprotein E plays a crucial role in inhibiting chronic neurodegenerative processes. However, its impact on neurological function following diffuse brain injury is still unclear. This study was designed to evaluate the therapeutic effects and mechanisms of action of apolipoprotein E mimetic peptide on diffuse brain injury. Apolipoprotein E mimetic peptide was administered into the caudal vein of rats with diffuse brain injury before and after injury. We found that apolipoprotein E mimetic peptide significantly decreased the number of apoptotic neurons, reduced extracellular signal-regulated kinase1/2 phosphorylation, down-regulated Bax and cytochrome c expression, decreased malondialdehyde content, and increased superoxide dismutase activity in a dose-dependent manner. | 16 | Construction of a high-density mutant population of Chinese cabbage facilitates the genetic dissection of agronomic traits显示文摘Chinese cabbage(Brassica rapa ssp.pekinensis)is an economically important vegetable crop throughout the world,especially in Asia.High-quality genome sequences are available for Chinese cabbage,but gene functional studies remain challenging.To promote functional genomic studies of Chinese cabbage,we generated an ethyl methane sulfonate(EMS)mutant population of~8000 M_(2) plants using the double haploid inbred line A03 as the parent.The genome of A03 was sequenced and used as a reference for high-throughput functional characterization of gene mutations at the whole-genome level.A total of 300 M_(2) to M_(5) EMS mutants were phenotypically screened and then sequenced,revealing 750629 SNPs and 46272 InDel mutations that cover 98.27%of all predicted genes in the A03 genome.A forward-genetics approach was successfully used to identify two genes with chloroplast-related functions that are responsible for the yellow leaf mutant trait.A reverse-genetics approach was also used to identify associations between mutations in five genes of the glucosinolate biosynthetic pathway and variations in glucosinolate content of the mutant plants.In addition,we built the Chinese cabbage EMS mutation database(CCEMD,www.bioinformaticslab.cn/EMSmutation/home)to increase the usability of this mutant population resource.In summary,we performed large-scale screening of a heading Chinese cabbage EMS mutant collection at the phenotypic and genotypic levels,which will facilitate gene mining of Chinese cabbage and might also be useful for the study of other Brassica crops. | Xiaoxue Sun Xing Li Yin Lu Shan Wang Xiaomeng Zhang Kang Zhang Xiangjie Su Mengyang Liu Daling Feng Shuangxia Luo Aixia Gu Yu Fu Xueping Chen Shuxin Xuan Yanhua Wang Donghui Xu Shumin Chen Wei Ma Shuxing Shen Feng Cheng Jianjun Zhao | 2022 | Molecular Plant2022,15,5: | 1 |
