Fuzzy Comprehensive Evaluation Model for Water Resources Carrying Capacity in Tarim River Basin,Xinjiang,China显示文摘This paper explores the method of comprehensive evaluation of water resources carrying capacity and sets up an evaluation model applying the fuzzy comprehensive evaluation method. Based on the data of nature, society, economics and water resources of the Tarim River Basin in 2002,we evaluated the water resources carrying capacity of the basin by means of the model. The results show that the comprehensive grades are 0.438 and 0.454 for Aksu and Kashi prefectures respectively, where the current water resources exploitation and utilization has reached a relative high degree and there is only a very limited water carrying capacity,0.620 for Kizilsu Kirgiz Autonomous Prefecture, where water resources carrying capacity is much higher, and in between for Hotan Prefecture and Bayingolin Mongo-lian Autonomous Prefecture. As a whole, the comprehensive grade of the Tarim River Basin is 0.508 and the current water resources exploitation and utilization has reached a relative high degree. Thus,we suggest that the integrated management of the water resources in the basin should be strengthened in order to utilize water resources scientifically and sustainably.MENG Lihong CHEN Yaning LI Weihong ZHAO Ruifeng 2009Chinese Geographical Science2009,19,1:55
Regulation of extracellular signal-regulated kinase 1/2 influences hippocampal neuronal survival in a rat model of diabetic cerebral ischemia显示文摘Activation of extracellular signal-regulated kinase 1/2 has been demonstrated in acute brain ischemia. We hypothesized that activated extracellular signal-regulated kinase 1/2 can protect hippocampal neurons from injury in a diabetic model after cerebral ischemia/reperfusion. In this study, transient whole-brain ischemia was induced by four-vessel occlusion in normal and diabetic rats, and extracellular signal-regulated kinase 1/2 inhibitor(U0126) was administered into diabetic rats 30 minutes before ischemia as a pretreatment. Results showed that the number of surviving neurons in the hippocampal CA1 region was reduced, extracellular signal-regulated kinase 1/2 phosphorylation and Ku70 activity were decreased, and pro-apoptotic Bax expression was upregulated after intervention using U0126. These findings demonstrate that inhibition of extracellular signal-regulated kinase 1/2 activity aggravated neuronal loss in the hippocampus in a diabetic rat after cerebral ischemia/reperfusion, further decreased DNA repairing ability and accelerated apoptosis in hippocampal neurons. Extracellular signal-regulated kinase 1/2 activation plays a neuroprotective role in hippocampal neurons in a diabetic rat after cerebral ischemia/ reperfusion.Yaning Zhao Jianmin Li Qiqun Tang Pan Zhang Liwei Jing Changxiang Chen Shuxing Li 2014Neural Regeneration Research2014,9,7:10
Protective effects of dl-3n-butylphthalide against diffuse brain injury显示文摘Dl-3n-butylphthalide can effectively treat cerebral ischemia;however, the mechanisms underlying the effects of dl-3n-butylphthalide on microcirculation disorders following diffuse brain injury remain unclear. In this study, models of diffuse brain injury were established in Sprague-Dawley rats with the vertical impact method.Dl-3n-butylphthalide at 80 and 160 mg/kg was given via intraperitoneal injection immediately after diffuse brain injury.Ultrastructural changes in the cerebral cortex were observed using electron microscopy.Cerebral blood flow was measured by laser Doppler flowmetry, vascular density was marked by tannic acid-ferric chloride staining, vascular permeability was estimated by the Evans blue method, brain water content was measured using the dry-wet method, and rat behavior was measured by motor function and sensory function tests. At 6,24,48,and 72hours after administration of dl-3n-butylphthalide,reduced cerebral ultrastructure damage, increased vascular density and cerebral blood flow, and improved motor and sensory functions were observed. Our findings demonstrate that dl-3n-butylphthalide may have protective effects against diffuse brain injury by ameliorating microcirculation disorder and reducing blood-brain barrier damage and cerebral edema.Yaning Zhao Jianmin Li Pan Zhang Changxiang Chen Shuxing Li 2013Neural Regeneration Research2013,8,28:7
Engineered trimeric ACE2 binds viral spike protein and locks it in'Thfee-up'conformation to potently inhibit SARS-CoV-2 infection显示文摘Dear Editor,Coronavirus disease 2019(COVID-19)caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has resulted in a severe global pandemic Following SARS-CoV,SARS-CoV-2 is yet another emerge nt beta-coronavirus threate ning human health.1 However, seventeen years after the SARS pandemic,no targeted vaccines or therapeutics have been approved for SARS,while some of them might have held promise for treating COVID-19.Many neutralizing antibodies against SARS-CoV-2 are currently being developed. However,RNA viruses are known to have high mutation rates.Liang Guo Wenwen Bi Xinling Wang Wei Xu Renhong Yan Yuanyuan Zhang Kai Zhao Yaning Li Mingfeng Zhang Xia Cai Shibo Jiang Youhua Xie Qiang Zhou Lu Lu Bobo Dang 2021Cell Research2021,31,1:5
Structural basis for bivalent binding and inhibition of SARS-CoV-2 infection by human potent neutralizing antibodies显示文摘Neutralizing monoclonal antibodies(nAbs)to severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)represent promising candidates for clinical intervention against coronavirus disease 2019(COVID-19).We isolated a large number of nAbs from SARS-CoV-2-infected individuals capable of disrupting proper interaction between the receptor binding domain(RBD)of the viral spike(S)protein and the receptor angiotensin converting enzyme 2(ACE2).However,the structural basis for their potent neutralizing activity remains unclear. Here,we report cryo-EM structures of the ten most potent nAbs in their native full-length IgG-form or in both IgG-form and Fab-form bound to the trimeric S protein of SARS-CoV-2.The bivalent binding of the full-length IgG is found to associate with more RBDs in the“up”conformation than the monovalent binding of Fab,perhaps contributing to the enhanced neutralizing activity of IgG and triggering more shedding of the S1 subunit from the S protein.Comparison of a large number of nAbs identified common and unique structural features associated with their potent neutralizing activities. This work provides a structural basis for further understanding the mechanism of nAbs,especially through revealing the bivalent binding and its correlation with more potent neutralization and the shedding of S1 subunit.Renhong Yan Ruoke Wang Bin Ju Jinfang Yu Yuanyuan Zhang Nan Liu Jia Wang Qi Zhang Peng Chen Bing Zhou Yaning Li Yaping Shen Shuyuan Zhang Long Tian Yingying Guo Lu Xia Xinyue Zhong Lin Cheng Xiangyang Ge Juanjuan Zhao Hong-Wei Wang Xinquan Wang Zheng Zhang Linqi Zhang Qiang Zhou 2021Cell Research2021,31,5:4
Dental plaque-inspired versatile nanosystem for caries prevention and tooth restoration显示文摘Dental caries is one of the most prevalent human diseases resulting from tooth demineralization caused by acid production of bacteria plaque. It remains challenges for current practice to specifically identify, intervene and interrupt the development of caries while restoring defects. In this study, inspired by natural dental plaque,a stimuli-responsive multidrug delivery system(PMs@NaF-SAP)has been developed to prevent tooth decay and promote enamel restoration.Classic spherical core-shell structures of micelles dual-loaded with antibacterial and restorative agents are self-assembled into bacteria-responsive multidrug delivery system based on the pH-cleavable boronate ester bond, followed by conjugation with salivary-acquired peptide(SAP)to endow the nanoparticle with strong adhesion to tooth enamel. The constructed PMs@NaF-SAP specifically adheres to tooth, identifies cariogenic conditions and intelligently releases drugs at acidic pH,thereby providing antibacterial adhesion and cariogenic biofilm resistance, and restoring the microarchitecture and mechanical properties of demineralized teeth.Topical treatment with PMs@NaF-SAP effectively diminishes the onset and severity of caries without impacting oral microbiota diversity or surrounding mucosal tissues. These findings demonstrate this novel nanotherapy has potential as a promising biomedical application for caries prevention and tooth defect restoration while resisting biofilm-associated diseases in a controlled manner activated by pathological bacteria.Yue Xu Yuan You Luyao Yi Xiaoyi Wu Yaning Zhao Jian Yu He Liu Ya Shen Jingmei Guo Cui Huang 2023Bioactive Materials2023,,2:3
Analysis of spatiotemporal variations in the characteristics of soil microbial communities in Castanopsis fargesii forests显示文摘Castanopsis fargesii is a good afforestation plants and various microorganisms play important roles in mediating the growth and ecological functions of this species. In this study,we evaluated changes in microbial communities in soil samples from C.fargesii forests. The phospholipid fatty acid(PLFA)biomarker method was used to obtain bacteria,fungi,actinomycetes,gram-positive bacteria(G?),gram-negative bacteria(G-),aerobic bacteria, and anaerobic bacteria to investigate spatiotemporal changes in microbial communities during the growing season. The results show that soil microorganisms were mainly concentrated in the upper 20-cm layer, demonstrating an obvious surface aggregation(P<0.05).Large amounts of litter and heavy rainfall during the early growing season resulted in the highest PLFA contents for various microorganisms, whereas relatively low and stable levels were observed during other times. The dominant species during each period were bacteria.G+ or aerobic bacteria were the main bacterial populations, providing insights into the overall trends of soil bacterial PLFA contents.Due to the relative accumulation of refractory substances during the later stages of litter decomposition, the effects of fungi increased significantly.Overall, our findings demonstrate that the main factors influencing microbial communities were litter, rainfall, and soil field capacity.Hongyong Qiao Yaning Luan Bing Wang Wei Dai Mengsai Zhao 2020Journal of Forestry Research2020,31,5:2
Biomarkers of aging显示文摘Aging biomarkers are a combination of biological parameters to(i)assess age-related changes,(ii)track the physiological aging process, and(iii)predict the transition into a pathological status.Although a broad spectrum of aging biomarkers has been developed, their potential uses and limitations remain poorly characterized. An immediate goal of biomarkers is to help us answer the following three fundamental questions in aging research:How old are we?Why do we get old?And how can we age slower? This review aims to address this need. Here,we summarize our current knowledge of biomarkers developed for cellular, organ, and organismal levels of aging, comprising six pillars:physiological characteristics, medical imaging, histological features, cellular alterations, molecular changes, and secretory factors. To fulfill all these requisites,we propose that aging biomarkers should qualify for being specific, systemic, and clinically relevant.Aging Biomarker Consortium Hainan Bao Jiani Cao Mengting Chen Min Chen Wei Chen Xiao Chen Yanhao Chen Yu Chen Yutian Chen Zhiyang Chen Jagadish K Chhetri Yingjie Ding Junlin Feng Jun Guo Mengmeng Guo Chuting He Yujuan Jia Haiping Jiang Ying Jing Dingfeng Li Jiaming Li Jingyi Li Qinhao Liang Rui Liang Feng Liu Xiaoqian Liu Zuojun Liu Oscar Junhong Luo Jianwei Lv Jingyi Ma Kehang Mao Jiawei Nie Xinhua Qiao Xinpei Sun Xiaoqiang Tang Jianfang Wang Qiaoran Wang Siyuan Wang Xuan Wang Yaning Wang Yuhan Wang Rimo Wu Kai Xia Fu-Hui Xiao Lingyan Xu Yingying Xu Haoteng Yan Liang Yang Ruici Yang Yuanxin Yang Yilin Ying Le Zhang Weiwei Zhang Wenwan Zhang Xing Zhang Zhuo Zhang Min Zhou Rui Zhou Qingchen Zhu Zhengmao Zhu Feng Cao Zhongwei Cao Piu Chan Chang Chen Guobing Chen Hou-Zao Chen Jun Chen Weimin Ci Bi-Sen Ding Qiurong Ding Feng Gao Jing-Dong JHan Kai Huang Zhenyu Ju Qing-Peng Kong Ji Li Jian Li Xin Li Baohua Liu Feng Liu Lin Liu Qiang Liu Qiang Liu Xingguo Liu Yong Liu Xianghang Luo Shuai Ma Xinran Ma Zhiyong Mao Jing Nie Yaojin Peng Jing Qu Jie Ren Ruibao Ren Moshi Song Zhou Songyang Yi Eve Sun Yu Sun Mei Tian Shusen Wang Si Wang Xia Wang Xiaoning Wang Yan-Jiang Wang Yunfang Wang Catherine CL Wong Andy Peng Xiang Yichuan Xiao Zhengwei Xie Daichao Xu Jing Ye Rui Yue Cuntai Zhang Hongbo Zhang Liang Zhang Weiqi Zhang Yong Zhang Yun-Wu Zhang Zhuohua Zhang Tongbiao Zhao Yuzheng Zhao Dahai Zhu Weiguo Zou Gang Pei Guang-Hui Liu 2023Science China(Life Sciences)2023,66,5:2

Inhibitory effect of oridonin on proliferation of RPMI8226 cells and the possible underlying mechanism显示文摘OBJECTIVE:To observe the effects of oridonin on proliferation and apoptosis of myeloma RPMI8226cells and to investigate the potential underlying mechanisms.METHODS:RPMI8226 cells were treated with various concentrations of oridonin.Cell proliferation was analyzed using the thiazolyl blue tetrazolium bromide method.Ultramicrostructure was observed by transmission electron microscopy.Annexin-V/PI staining and flow cytometry was performed to determine cell apoptosis.Expression of apoptosis-related proteins was evaluated by western blot analysis.RESULTS:Oridonin suppressed the proliferation of RPMI8226 cells and induced apoptosis in a timeand dose-dependent manner.Transmission electron microscopy confirmed apoptotic morphologyupon treatment with 20μmol/L oridonin and western blot revealed decreased expressions of the apoptosis suppressors survivin,Bcl-2 and pro-caspase-3 proteins, and the increased expression of the apoptosis inducer Bax.CONCLUSION:Our results show that oridonin exhibits an inhibitory effect on the proliferation of RPMI8226 cells and induces apoptosis. This is associated with altering the balance between Bcl-2 and Bax protein expressions and decreased survivin and pro-caspase-3 expressions.Hao Yaning Zhao Fei Luo Yuanyuan Zhang Mei Li Shasha 2016Journal of Traditional Chinese Medicine2016,36,2:1
Apolipoprotein E mimetic peptide protects against diffuse brain injury显示文摘Apolipoprotein E plays a crucial role in inhibiting chronic neurodegenerative processes. However, its impact on neurological function following diffuse brain injury is still unclear. This study was designed to evaluate the therapeutic effects and mechanisms of action of apolipoprotein E mimetic peptide on diffuse brain injury. Apolipoprotein E mimetic peptide was administered into the caudal vein of rats with diffuse brain injury before and after injury. We found that apolipoprotein E mimetic peptide significantly decreased the number of apoptotic neurons, reduced extracellular signal-regulated kinase1/2 phosphorylation, down-regulated Bax and cytochrome c expression, decreased malondialdehyde content, and increased superoxide dismutase activity in a dose-dependent manner. These experimental findings demonstrate that apolipoprotein E mimetic peptide improves learning and memory function and protects against diffuse brain injury-induced apoptosis by inhibiting the extracellular signal-regulated kinase1/2-Bax mitochondrial apoptotic pathway.Yaning Zhao Jianmin Li Qiqun Tang Junling Gao Changxiang Chen Liwei Jing Pan Zhang Shuxing Li 2014Neural Regeneration Research2014,9,5:1
