| 4 | Mechanisms of antinociceptive effects of ouabain in combination with neostigmine in the rat显示文摘BACKGROUND: It has been previously shown that intrathecal administration of either ouabain or neostigmine can produce antinociceptive effects. Moreover, ouabain and neostigmine are differently associated with acetylcholine. OBJECTIVE: It has been hypothesized that intrathecal administration of ouabain, in combination with neostigmine, can produce antinociceptive synergistic effects. Atropine, as a competitive antagonist, was pre-injected to verify the mechanisms of action. DESIGN, TIME AND SETTING: This study was a randomized, controlled, animal experiment, performed at the State Key Laboratory of Oncology in Southern China between May 2006 and February 2007. MATERIALS: A total of 102 healthy, adult, Sprague Dawley rats were included. Ouabain and neostigmine (Sigma, USA), as well as atropine (Tanabe Seiyaku, Japan), were also used. METHODS: Varied doses of ouabain, neostigmine, and a combination of the two were intrathecally injected into rats. Six rats were allotted for each dose group. Intrathecal pretreatment with atropine was tested 10 minutes prior to intrathecal administration of neostigmine or the combination of ouabain and neostigmine. MAIN OUTCOME MEASURES: Tail-flick tests were performed to measure tail-flick latency (seconds) prior to and after administration. The response in the tail-flick test was expressed as the percentage of maximum possible effect (% MPE), where % MPE = [tail-flick latency after administration (seconds) -mean baseline value for tail-flick latency]/[10 seconds - the mean baseline value for tail-flick latency (seconds)] × 100%. RESULTS: Rat spinal intrathecal administration of either ouabain or neostigmine alone produced antinociceptive effects in a dose-dependent manner. Intrathecally administration of neostigmine (0.05, 0.1, 0.3 μg ) in combination with ouabain (1 μg ) produced enhanced antinociceptive effects, with a % MPE of 29%, 78%, and 95%, respectively (P < 0.05). Intrathecally administration of 0.3 μg neostigmine (% MPE: 45%), in combination with 1 μg ouabain (% MPE: 27%) produced potent antinociceptive effects (% MPE: 95%). Intrathecally pre-injected atropine antagonized the antinociceptive effects of neostigmine (3 μg), or a combination of ouabain (1 μg) and neostigmine (0.3 μg) (P < 0.01). CONCLUSION: Rat spinal intrathecal administration of either ouabain or neostigmine alone produced dose-dependent antinociceptive effects. Ouabain enhanced the antinociceptive effects of neostigmine. Atropine antagonized the antinociceptive effects of neostigmine or the combination of ouabain and neostigmine. | 10 | Increased dopamine and its receptor dopamine receptor D1 promote tumor growth in human hepatocellular carcinoma显示文摘Background:Dopamine and dopamine receptor D1(DRD1),a member of the dopamine receptor family,have been indicated to play important roles in cancer progression,but dopamine secretion in hepatocellular carcinoma(HCC)and the effects of DRD1 on HCC remain unclear.This study was designed to explore the contribution of the dopaminergic system to HCC and determine the relationship between DRD1 and prognosis in HCC patients.Methods:The dopamine metabolic system was monitored using enzyme-linked immunosorbent assays(ELISAs).The expression of DRD1 was detected by microarray analysis,immunohistochemistry(IHC),and quantitative real-time PCR(qRT-PCR).Stable DRD1 knockout and overexpression cell lines were established for investigation.Transwell,colony formation,and Cell Counting Kit 8(CCK8)assays were performed to assess the malignant behaviors of cancer cells.The cAMP/PI3K/AKT/cAMP response element-binding(CREB)signaling pathway was evaluated by Western blot.This pathway,which is agitated by DRD1 in striatal neurons,had been proven to participate in tumor progression.Xenograft HCC tumors were generated for in vivo experiments.Results:Dopamine secretion increased locally in HCC due to an imbalance in dopamine metabolism,including the upregulation of dopa decarboxylase(DDC)and the downregulation of monoamine oxidase A(MAOA).Dopamine promoted the proliferation and metastasis of HCC.DRD1 was highly expressed in HCC tissues and positive DRD1 expression was related to a poor prognosis in HCC patients.The upregulation of DRD1 agitated malignant activities,including proliferation and metastasis in HCC by regulating the cAMP/PI3K/AKT/CREB pathway,and the downregulation of DRD1 had opposing effects.The effects of dopamine on HCC was reversed by depleting DRD1.SCH23390,a selective DRD1 antagonist,inhibited the proliferation and metastasis of HCC cells both in vitro and in vivo.Conclusion:Dopamine secretion was locally increased in HCC and promoted HCC cell proliferation and metastasis.DRD1 was found to exert positive effects on HCC progression and play a vital role in the dopamine system,and could be a potential therapeutic target and prognostic biomarker for HCC. | Yan Yan Jiahao Pan Yonghua Chen Wei Xing Qiang Li Dongyin Wang Xiaoshuang Zhou Jingdun Xie Changhong Miao Yunfei Yuan Weian Zeng Dongtai Chen | 2020 | Cancer Communications2020,40,12: | 1 |

| 13 | Intranuclear cardiac troponin I plays a functional role in regulating Atp2a2 expression in cardiomyocytes显示文摘In the past studies,it is shown that cardiac troponin I(cTnI,encoded by TNNI3),as a cytoplasmic protein,is an inhibitory subunit in troponin complex,and involves in cardiomyocyte diastolic regulation.Here,we assessed a novel role of cTnI as a nucleoprotein.Firstly,the nuclear translocation of cTnI was found in mouse,human fetuses and rat heart tissues.In addition,there were differences in percentage of intranuclear cTnI in different conditions.Based on weighted gene co-expression network analyses(WGCNA)and verification in cell experiments,a strong expression correlation was found between TNNI3 and Atp2a2,which encodes sarco-endoplasmic reticulum Ca2t ATPase isoform 2a(SERCA2a),and involves in ATP hydrolysis and Ca2t transient.TNNI3 gain and loss caused Atpa2a2 increase/decrease in a dosedependent manner both in mRNA and protein levels,in vivo and in vitro.By using ChIP-sequence we demonstrated specific binding DNA sequences of cTnI were enriched in ATP2a2 promoter239we889 region and the specific binding sequence motif of cTnI was analyzed by software as'CCAT',which has been reported to be required for YY1 binding to the promoter region of YY1-related genes.Moreover,it was further verified that pcDNA3.1()-TNNI3 could express cTnI proteins and increase the promoter activity of Atp2a2 through luciferase report assay.In the end,we evaluated beat frequencies,total ATP contents,Ca2t transients in TNNI3-siRNA myocardial cells.These findings indicated,for the first time,cTnI may regulate Atp2a2 in cardiomyocytes as a co-regulatory factor and participate in the regulation of intracellular Ca ions. | Qian Lu Bo Pan Haobo Bai Weian Zhao Lingjuan Liu Gu Li Ruimin Liu Tiewei Lv Xupei Huang Xi Li Jie Tian | 2022 | Genes & Diseases2022,9,6: | 0 |

| 15 | Face anti-spoofing based on multi-modal and multi-scale features fusion显示文摘Face anti-spoofing is used to assist face recognition system to judge whether the detected face is real face or fake face. In the traditional face anti-spoofing methods, features extracted by hand are used to describe the difference between living face and fraudulent face. But these handmade features do not apply to different variations in an unconstrained environment. The convolutional neural network(CNN) for face deceptions achieves considerable results. However, most existing neural network-based methods simply use neural networks to extract single-scale features from single-modal data, while ignoring multi-scale and multi-modal information. To address this problem, a novel face anti-spoofing method based on multi-modal and multi-scale features fusion(MMFF) is proposed. Specifically, first residual network(Resnet)-34 is adopted to extract features of different scales from each modality, then these features of different scales are fused by feature pyramid network(FPN), finally squeeze-and-excitation fusion(SEF) module and self-attention network(SAN) are combined to fuse features from different modalities for classification. | 16 | A Near-Infrared Organoplatinum(Ⅱ) Metallacycle Conjugated with Heptamethine Cyanine for Trimodal Cancer Therapy显示文摘Discrete Pt(Ⅱ)metallacycles have attracted particular attention for the chemotherapeutic treatment of cancer.However,a single chemotherapy cannot simultaneously balance efficiency and safety because the continuous administration throughout the entire therapy period will lead to inefficient therapy and potentially long-term systemic toxicity.Therefore,the development of a novel organoplatinum(Ⅱ)metallacycle with multimodal treatment capabilities is urgently needed to overcome these issues.Herein,a discrete Pt(II)metallacycle(SCY)bearing the near-infrared(NIR)photosensitizer heptamethine cyanine was fabricated and further encapsulated by amphiphilic 1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-methoxy(polyethyleneglycol)(DSPEmPEG)to form P@SCY nanoparticles.Heptamethine cyanine,which has excellent photoconversion efficiency,can generate reactive oxygen species(ROS)and heat simultaneously,and the cyanine moiety can target mitochondria in cancer cells due to their quaternary ammonium salt cations,which improve the effect of phototherapy.Due to its excellent phototherapy and chemotherapy properties,P@SCY exhibited remarkable trimodal therapeutic effects[chemo-/photodynamic therapy(PDT)/photothermal therapy(PTT)]against cancer cells(HepG2 cells,MCF-7 cells,and 4T1 cells)in vitro.Furthermore,in vivo results also confirmed thatP@SCY had superior antitumor properties with minimal side effects in the 4T1 tumor model.Thiswork presents a practicable approach to develop a multifunctional organoplatinum(Ⅱ)metallacycle for multimodal tumor therapy. | Baoxuan Huang Xi Liu Guoliang Yang Jia Tian Zhiyong Liu Yucheng Zhu Xiaopeng Li Guangqiang Yin Wei Zheng Lin Xu Weian Zhang | 2022 | CCS Chemistry2022,4,6: | 0 |
