1Sonodynamic therapy(SDT): a novel strategy for cancer nanotheranostics显示文摘Sonodynamic therapy(SDT) is a promising non-invasive therapeutic modality. Compared to photo-inspired therapy, SDT provides many opportunities and benefits, including deeper tissue penetration, high precision, less side effects, and good patient compliance. Thanks to the facile engineerable nature of nanotechnology, nanoparticles-based sonosensitizers exhibit predominant advantages, such as increased SDT efficacy, binding avidity, and targeting specificity. This review aims to summarize the possible mechanisms of SDT, which can be expected to provide the theoretical basis for SDT development in the future. We also extensively discuss nanoparticle-assisted sonosensitizers to enhance the outcome of SDT. 3An EGLN1 mutation may regulate hypoxic response in cyanotic congenital heart disease through the PHD2/HIF-1A pathway显示文摘Cyanotic congenital heart disease(CCHD),a term describing the most severe congenital heart diseases are characterized by the anatomic malformation of a right to left shunt.Although the incidence of CCHD are far less than the that of congenital heart diseases(CHD),patients with CCHD always present severe clinical features such as hypoxia,dyspnea,and heart failure.Chronic hypoxia induces hypoxemia that significantly contributes to poor prognosis in CCHD.Current studies have demonstrated that the prolyl-4-hydroxylase2(PHD2,encoded by EGLN1)/hypoxia-inducible factor-1A(HIF-1A)pathway is a key regulator of hypoxic response.Thus,we aim to assess the associations of single polymorphisms(SNPs)of the EGLN1 gene and hypoxic response in CCHD.A missense variant of EGLN1 c.380G>C(rs1209790)was found in 46 patients(46/126),with lower hypoxia incidence and higher rate of collateral vessel formation,compared with the wild type(P<0.05).In vitro experiments,during hypoxia,EGLN1 mutation reduced EGLN1 expression compared with the wild type,with higher HIF-1A,VEGF and EPO expression levels in the mutant.No difference in HK1 expression was observed between the mutant and wild type.CCHD patients with c.380G>C showed improved response to hypoxia compared with the wild-type counterparts.The EGLN1 c.380G>C mutation improves hypoxic response through the PHD2/HIF-1A pathway,which may provide a molecular mechanism for hypoxic response in CCHD.The effects of the EGLN1 c.380G>C mutation on CCHD prognosis deserve further investigation.Yuanlin Zhou Na Ouyang Lingjuan Liu Jie Tian Xupei Huang Tiewei Lu 2019Genes & Diseases2019,6,1:7
4A high-fiber diet synergizes with Prevotella copri and exacerbates rheumatoid arthritis显示文摘Both preclinical and established rheumatoid arthritis(RA)patients display alterations in the gut microbiome.Prevotella spp.are preferentially enriched in a subset of RA patients.Here,we isolated a Prevotella strain,P.copri RA,from the feces of RA patients and showed that colonization of P.copri RA exacerbated arthritis in a collagen-induced arthritis(CIA)model.With the presence of P.copri RA colonization,a high-fiber diet exacerbated arthritis via microbial alterations and intestinal inflammation.Colonization of P.copri together with a high-fiber diet enabled the digestion of complex fiber,which led to the overproduction of organic acids,including fumarate,succinate and short-chain fatty acids.Succinate promoted proinflammatory responses in macrophages,and supplementation with succinate exacerbated arthritis in the CIA model.Our findings highlight the importance of dysbiosis when evaluating the effects of dietary interventions on RA pathogenesis and provide new insight into dietary interventions or microbiome modifications to improve RA management.Lingjuan Jiang Mengmeng Shang Shengnan Yu Yudong Liu Hui Zhang Yangzhong Zhou Min Wang Tingting Wang Hui Li Zhihua Liu Xuan Zhang 2022Cellular & Molecular Immunology2022,19,12:6

6Variation of Indo-Pacific upper ocean heat content during 2001–2012 revealed by Argo显示文摘Understanding of the temporal variation of oceanic heat content(OHC) is of fundamental importance to the prediction of climate change and associated global meteorological phenomena. However, OHC characteristics in the Pacific and Indian oceans are not well understood. Based on in situ ocean temperature and salinity profiles mainly from the Argo program, we estimated the upper layer(0–750 m) OHC in the Indo-Pacific Ocean(40°S–40°N, 30°E–80°W). Spatial and temporal variability of OHC and its likely physical mechanisms are also analyzed. Climatic distributions of upper-layer OHC in the Indian and Pacific oceans have a similar saddle pattern in the subtropics, and the highest OHC value was in the northern Arabian Sea. However, OHC variabilities in the two oceans were different. OHC in the Pacific has an east-west see-saw pattern, which does not appear in the Indian Ocean. In the Indian Ocean, the largest change was around 10°S. The most interesting phenomenon is that, there was a long-term shift of OHC in the Indo-Pacific Ocean during 2001–2012. Such variation coincided with modulation of subsurface temperature/salinity. During 2001–2007, there was subsurface cooling(freshening)nearly the entire upper 400 m layer in the western Pacific and warming(salting) in the eastern Pacific. During2008–2012, the thermocline deepened in the western Pacific but shoaled in the east. In the Indian Ocean, there was only cooling(upper 150 m only) and freshening(almost the entire upper 400 m) during 2001–2007. The thermocline deepened during 2008–2012 in the Indian Ocean. Such change appeared from the equator to off the equator and even to the subtropics(about 20°N/S) in the two oceans. 7Helicobacter pylori may participate in the development of inflammatory bowel disease by modulating the intestinal microbiota显示文摘Inflammatory bowel disease(IBD)is a non-specific inflammatory disease of the gastrointestinal(GI)tract that is generally accepted to be closely related to intestinal dysbiosis in the host.GI infections contribute a key role in the pathogenesis of IBD;however,although the results of recent clinical studies have revealed an inverse correlation between Helicobacter pylori(H.pylori)infection and IBD,the exact mechanism underlying the development of IBD remains unclear.H.pylori,as a star microorganism,has been a focus for decades,and recent preclinical and real-world studies have demonstrated that H.pylori not only affects the changes in the gastric microbiota and microenvironment but also influences the intestinal microbiota,indicating a potential correlation with IBD.Detailed analysis revealed that H.pylori infection increased the diversity of the intestinal microbiota,reduced the abundance of Bacteroidetes,augmented the abundance of Firmicutes,and produced short-chain fatty acid-producing bacteria such as Akkermansia.All these factors may decrease vulnerability to IBD.Further studies investigating the H.pylori-intestinal microbiota metabolite axis should be performed to understand the mechanism underlying the development of IBD.Xiaoyin Bai Lingjuan Jiang Gechong Ruan Tingting Liu Hong Yang 2022Chinese Medical Journal2022,,6:5
8Epigenetic regulation of phosphodiesterase 4d in restrictive cardiomyopathy mice with cTnI mutations显示文摘Epigenetic regulations play an important role in disease development.In this study,we have investigated epigenetic regulations in restrictive cardiomyopathy mice with cTnI 193 His mutation.Our results demonstrated that phosphodiesterase(PDEs)4d was down-regulated in the heart of these mice.Further studies showed that the epigenetic modifications were associated with enhanced acetylation of histone 3 lysine 4 and lysines 9,whereas tri-methylation of histone 3 lysine 4,were decreased in histones near the PDE4d gene promoter regions.The binding levels of histone transmethylase SMYD1 and histone deacetylase HDAC1 were increased in the gene promoter regions in cTnI193 His transgenic hearts.Using immune-fluorescent labeling we found an evidence of cTnI existence in the nucleus of cardiomyocytes and Western blotting further confirmed that both wild type and mutated cTnI could be detected in the cell nucleus of the hearts.Furthermore,an interaction between cTnI and SMYD1,or cTnI and HDAC1 was observed.Overexpression of the mutated cTnI in cultured cardiomyocytes reduced the expression of PDE4d.Our data suggest that the decrease of PDE4d expression in RCM mice caused by cTnI mutations may be related to epigenetic regulation,i.e.,histone acetylation and methylation,and cTnI might be involved in this procedure via an interaction with HDAC1 and SMAD1 in the hearts.Weian Zhao Xiaoqi Wu Zhiyuan Wang Bo Pan Lifei Liu Lingjuan Liu Xupei Huang Jie Tian 2020Science China(Life Sciences)2020,63,4:4

10Acetylation of H3K4,H3K9,and H3K27 mediated by p300 regulates the expression of GATA4 in cardiocytes显示文摘GATA4 is a particularly important cardiogenic transcription factor and serves as a potent driver of cardiogenesis.Recent progress in the field has made it clear that histone acetylation can influence gene expression through changing the structure of chromatin.Our previous research had revealed that hypo-acetylation could repress gata4 expression in cardiocytes,however the underlying mechanism by which this occurred was still unclear.To reveal the mechanism of histone acetylation involved in the regulation of gata4 transcription,we concentrated on P300,one of the important histone acetyltransferase associated with cardiogenesis.We found that P300 participated in gata4 expression through regulating histone acetylation in embryonic mouse hearts.RNAi-mediated downregulation of P300 modulated the global acetylation of H3 and the acetylation of H3K4,H3K9,and H3K27 in gata4 and Tbx5 promoters.Interestingly,there was an obvious inhibition of gata4 transcription,whereas Tbx5 was not influenced.Furthermore,SGC-CBP30,the selective inhibitor of the bromodomain in CBP/P300,downregulated gata4 transcription by repressing the acetylation of H3K4,H3K9,and H3K27 in the gata4 promoters.Taken together,our results identified that acetylation of H3K4,H3K9,and H3K27 mediated by P300 plays an important role in regulation of gata4 expression in cardiogenesis.Wei Zhou Dagui Jiang Jie Tian Lingjuan Liu Tiewei Lu Xupei Huang Huichao Sun 2019Genes & Diseases2019,6,3:3

13Argonaute protein as a linker to command center of physiological processes显示文摘MicroRNAs (miRNAs) post-transcriptionally regulate gene expression by binding to target mRNAs with perfect or imperfect complementarity, recruiting an Argonaute (AGO) protein complex that usually results in degradation or translational repression of the target mRNA. AGO proteins function as the Slicer enzyme in miRNA and small interfering RNA (siRNA) pathways involved in human physiological and pathophysiological processes, such as antiviral responses and disease formation. Although the past decade has witnessed rapid advancement in studies of AGO protein functions, to further elucidate the molecular mechanism of AGO proteins in cellular function and biochemical process is really a challenging area for researchers. In order to understand the molecular causes underlying the pathological processes, we mainly focus on five fundamental problems of AGO proteins, including evolution, functional domain, subcellular location, post-translational modification and protein-protein interactions. 14An effective amperometric biosensor based on graphene modified gold nanowire arrays for glucose detection显示文摘A novel glucose biosensor based on graphene nanosheets(GNs)modified gold nanowire arrays(AuNWAs)electrode was constructed.Highly ordered gold nanowire arrays were prepared by direct electrodeposition in anodic aluminum oxide templates.GNs were synthesized through a public route involving graphite oxidation,exfoliation,and chemical reduction.Field emission scanning electron microscope and high-resolution transmission electron microscope were employed to characterize the asprepared AuNWAs and GNs.Glucose oxidase was immobilized on the surface of GNs-AuNWAs modified electrode via a cross-linking method.The cyclic voltammetry results showed that the GNs-AuNWAs-based glucose biosensors have high catalysis activity to hydrogen peroxide(H2O2)than those modified with GNs or AuNWAs only.Furthermore,amperometric response was employed to detect glucose concentration owing to its simplicity,high selectivity,and relative low cost.Glucose biosensors based on GNs-AuNWAs showed excellent performance with high sensitivity of 40.25 lA cm-2(mmol/L)-1,low detection limit of 0.02 mmol/L,and a linear range from 0.02 to 3 mmol/L.Jianing Hui Jiewu Cui Lingjuan Liu Guangqing Xu Yucheng Wu 2014Chinese Science Bulletin2014,59,17:2

17Molecular mechanism of mureidomycin biosynthesis activated by introduction of an exogenous regulatory gene ssa A into Streptomyces roseosporus显示文摘Mureidomycins(MRDs), a group of unique uridyl-peptide antibiotics, exhibit antibacterial activity against the highly refractory pathogen Pseudomonas aeruginosa. Our previous study showed that the cryptic MRD biosynthetic gene cluster(BGC) mrd in Streptomyces roseosporus NRRL 15998 could not be activated by its endogenous regulator 02995 but activated by an exogenous activator Ssa A from sansanmycin’s BGC ssa of Streptomyces sp. strain SS. Here we report the molecular mechanism for this inexplicable regulation. EMSAs and footprinting experiments revealed that Ssa A could directly bind to a 14-nt palindrome sequence of 5′-CTGRCNNNNGTCAG-3′ within six promoter regions of mrd. Disruption of three representative target genes(SSGG-02981, SSGG-02987 and SSGG-02994) showed that the target genes directly controlled by Ssa Awere essential for MRD production. The regulatory function was further investigated by replacing six regions of SSGG-02995 with those of ssa A.Surprisingly, only the replacement of 343–450 nt fragment encoding the 115–150 amino acids(AA) of Ssa A could activate MRD biosynthesis.
