6Targeted core-shell nanoparticles for precise CTCF gene insert in treatment of metastatic breast cancer显示文摘Clustered regularly interspaced short palindromic repeats(CRISPR)technology emerges a remarkable potential for cure of refractory cancer like metastatic breast cancer.However,how to efficiently deliver the CRISPR system with non-viral carrier remains a major issue to be solved.Here,we report a kind of targeted core-shell nanoparticles(NPs)carrying dual plasmids(pHR-pCas9)for precise CCCTC-binding factor(CTCF)gene insert to circumvent metastatic breast cancer.The targeted core-shell NPs carrying pHR-pCas9 can accomplishγGTP-mediated cellular uptake and endosomal escape,facilitate the precise insert and stable expression of CTCF gene,inhibit the migration,metastasis,and colonization of metastatic breast cancer cells.Besides,the finding further reveals that the inhibitory mechanism of metastasis could be associated with up-regulating CTCF protein,followed by down-regulating stomatin(STOM)protein.The study offers a universal nanostrategy enabling the robust non-viral delivery of gene-editing system for treatment of severe illness.Jialun Duan Chunjie Bao Ying Xie Haitao Guo Yixuan Liu Jianwei Li Rui Liu Peishan Li Jing Bai Yan Yan Limin Mu Xueqi Li Guiling Wang Wanliang Lu 2022Bioactive Materials2022,7,5:1

13Substrate Effects on the High-Temperature Oxidation Behavior of Thermal Barrier Coatings显示文摘The high-temperature oxidation behaviors of the NiCrAlYSi/P-YSZ thermal barrier coatings (TBCs) produced by electron beam-physical vapor deposition (EB-PVD) on directionally solidified (DS) and single crystalline (SC) Ni-based superalloy substrates were investigated. The cross-sectional microstructure investigation, isothermal and cyclic oxidation tests were conducted for the comparison of oxidation behaviors of TBCs on different substrates. Although TBC on DS substrate has a relatively higher oxidation rate, it has a longer thermal cycling lifetime than that on SC substrate. The primary factor for TBC spallation is the mismatch of thermal expansion coefficient (TEC) of the bond coat and substrate. The morphological feature of thermally grown oxide (TGO) has a strong influence on the TBC performance. 14Amphiphilic small molecular mates match hydrophobic drugs to form nanoassemblies based on drug-mate strategy显示文摘Nanomedicine has made great progress in the targeted therapy of cancer. Here, we established a novel drug-mate strategy by studying the formulation of nanodrugs at the molecular level. In the drug-mate combination, the drug is a hydrophobic drug that is poorly soluble in water, and the mate is an amphiphilic small molecule (SMA) that has both hydrophilic and lipophilic properties. We proposed that the hydrophobic drug could co-assemble with a suitable SMA on a nanoscale without additive agents. The proof-ofconcept methodology and results were presented to support our hypothesis. We selected five hydrophobic drugs and more than ten amphiphilic small molecules to construct a library. Through molecular dynamic simulation and quantum chemistry computation,we speculated that the formation of nanoassemblies was related to the binding energy of the drug-mate, and the drug-mate interaction must overcome drug-drug interaction.Furthermore, the obtained SF/VECOONa nanoassemblieswas selected as a model, which had an ultra-high drug loading content (46%), improved pharmacokinetics, increased bioavailability, and enhanced therapeutic efficacy. 15Postmortem Metabolite Profile Changes of Mud Crab(Scylla paramamosain)Under Different Storage Conditions显示文摘Mud crab(Scylla paramamosain)is one of the most consumed crabs in China.Generally,it is believed that dead mud crabs are not edible.Herein,we investigated the postmortem metabolite profile changes of the muscle and hepatopancreas of mud crab associated with storage method using nuclear magnetic resonance-based metabolomics coupled with multivariate data analysis.Our results showed that postmortem changes of trimethylamine,organic acids(lactate,formate,and succinate),as well as ATP and its breakdown products are closely affected by the storage method.These metabolites can be used as indicators for monitoring loss of crab freshness.Furthermore,dead mud crabs preserved an edible quality at least for 4 h at room temperature,12h on ice,and 16 d at−20℃,strongly suggesting that dead crab consumption quality should be determined by freshness.They shouldn’t be discarded without considering the effects of different storage conditions on their freshness.ZHANG Limin GUO Liping MU Changkao YE Yangfang WANG Chunlin 2021Journal of Ocean University of China2021,20,3:0
16Roles of light transmission aggregometry and CYP2C19 genotype in predicting ischaemic complications during interventional therapy for intracranial aneurysms显示文摘Background and purpose Light transmission aggregometry(LTA)and CYP2C19 genotype analysis are commonly used to evaluate the antiplatelet effects of clopidogrel during the interventional treatment of intracranial aneurysms.The aim of this study was to determine which test can predict ischaemic events during these treatments.Methods Patient demographic information,imaging data,laboratory data and ischaemic complications were recorded.LTA and CYP2C19 genotype results were compared,and multiple linear regression was performed to examine factors related to platelet reactivity.Multivariate regression analysis was performed to determine whether LTA and CYP2C19 could predict ischaemic complications and to identify other clinical risk factors.Receiver operating characteristic curve analysis was conducted to calculate the cut-off value for predicting ischaemic complications.A subgroup analysis was also performed for different CYP2C19 genotype metabolisers,as well as for patients with flow diverters and traditional stents.Results A total of 379 patients were included,of which 22 developed ischaemic events.Maximum platelet aggregation induced by ADP(ADP-MPA)could predict ischaemic events(p<0.001;area under the curve,0.752(95%CI 0.663 to 0.842)),and its cut-off value was 41.5%.ADP-MPA(p=0.001)and hypertension duration>10 years(p=0.022)were independent risk factors for ischaemic events,while the CYP2C19 genotype was not associated with ischaemic events.In the subgroup analysis,ADP-MPA could predict ischaemic events in fast metabolisers(p=0.004)and intermediate metabolisers(p=0.003).The cut-off value for ischaemic events was lower in patients with flow diverters(ADP-MPA=36.4%)than in patients with traditional stents(ADP-MPA=42.9%).Conclusions ADP-MPA can predict ischaemic complications during endovascular treatment of intracranial aneurysms.Patients with flow diverters require stronger antiplatelet medication than patients with traditional stents.Yangyang Zhou Wenqiang Li Chao Wang Ruhang Xie Yongnan Zhu Qichen Peng Limin Zhang Hongqi Zhang Yuxiang Gu Shiqing Mu Jian Liu Xinjian Yang 2023Stroke & Vascular Neurology2023,8,4:0
17A Novel Multi-Points Laser Monitoring of Building Settlement and Its Risk Evaluation显示文摘Aiming at the defects of routine settlement measurement methods, such as complicated procedures, time-consuming and labor-intensive, high cost and low measurement accuracy, based on the analysis of existing engineering measurement technical requirements and specifications, a multi-point high precision and high efficiency based on laser reference is proposed. The automatic building settlement real-time monitoring system program gives the principle and system model of single-point settlement observation. The model of multi-point scanning settlement monitoring system and the model of multi-point network settlement monitoring system are designed, and their advantages and disadvantages are analyzed. We focus on the networked multi-point settlement monitoring system for network cumulative error analysis, and propose related evaluation and correction methods. The hardware schematic and software block diagram of the laser reference measurement and measurement system of the single point settlement acquisition system are given. Finally, the risk of subsidence state is quantitatively evaluated based on multi-point settlement monitoring data.
