| 6 | Design and development of a mitochondrial-targeted photosensitizer for two-photon fluorescence imaging and photodynamic therapy显示文摘Mitochondria are well-acknowledged as ideal targets for tumor therapy due to their important role in energy supply and cellular signal regulation.Mitochondria-specific photosensitizers have been reported to be critical for inducing cell apoptosis.Two-photon fluorescence imaging provides a new technique for delineating biological structures and activities in deep tissues.Herein,we developed a new aggregation-induced emission(AIE)active photosensitizer by attaching a pyridinium group for mitochondrial target-ing.The rationally designed photosensitizer(TTTP)exhibited excellent photophysical properties,good biocompatibility,reactive oxygen species(ROS)stimulation ability,anticancer efficacy,and two-photon imaging properties.TTTP was highly taken up by cells and accumulated specifically in mitochondria but was selectively cytotoxic to cancer cells.Under light irradiation,the generation of ROS was significantly boosted,leading to actively induced apoptosis.The in vivo tumor photodynamic therapeutic efficacy of TTTP showed significant inhibition of tumor growth.Furthermore,the underlying mechanism of TTTP tu-mor suppression revealed that the apoptosis agonist Bax was markedly up-regulated while the antagonist Bcl-xL was down-regulated.This research provides a potential mitochondrial-targeted phototherapeutic agent for effective therapy and two-photon fluorescence imaging. | Yu Chen Sheng Zhang Tianjiao Cheng Wei Lin Linlin Mao Zhonghui Chen Yang Yang Hanqing Huang Jinqiu Li Zhiyong Ke Zhong-Kai Cui | 2023 | Journal of Materials Science & Technology2023,,10: | 0 |
| 7 | PP2A interacts with KATANIN to promote microtubule organization and conical cell morphogenesis显示文摘The organization of the microtubule cytoskeleton is critical for cell and organ morphogenesis.The evolutionarily conserved microtubule-severing enzyme KATANIN plays critical roles in microtubule organization in the plant and animal kingdoms.We previously used conical cell of Arabidopsis thaliana petals as a model system to investigate cortical microtubule organization and cell morphogenesis and determined that KATANIN promotes the formation of circumferential cortical microtubule arrays in conical cells.Here,we demonstrate that the conserved protein phosphatase PP2A interacts with and dephosphorylates KATANIN to promote the formation of circumferential cortical microtubule arrays in conical cells.KATANIN undergoes cycles of phosphorylation and dephosphorylation.Using co-immunoprecipitation coupled with mass spectrometry,we identified PP2A subunits as KATANIN-interacting proteins.Further biochemical studies showed that PP2 A interacts with and dephosphorylates KATANIN to stabilize its cellular abundance.Similar to the katanin mutant,mutants for genes encoding PP2A subunits showed disordered cortical microtubule arrays and defective conical cell shape.Taken together,these findings identify PP2A as a regulator of conical cell shape and suggest that PP2A mediates KATANIN phospho-regulation during plant cell morphogenesis. | Huibo Ren Jinqiu Rao Min Tang Yaxing Li Xie Dang Deshu Lin | 2022 | Journal of Integrative Plant Biology2022,64,8: | 0 |
| 8 | Modulation of vagal activity to atria electrical remodeling resulted from rapid atrial pacing显示文摘Background Atrial electrical remodeling(AER) plays an important role in the pathogenesis and maintenance of atrial fibrillation.However,little is known about modulation of vagal activity to AER.This study aimed to investigate the relationship between vagal moduation and AER.Methods Twenty four adult mongrel dogs under general anesthesia were randomized into 3 groups.Sympathetic activity was blocked by administration of metoprolol in 3 groups.The changes in vagal modulation to atria after AER were observed in 10 dogs without vagal interruption in group A.The effects ofvagal intervention on AER were investigated in 8 dogs with administration of atropine in group B.The impact of aggressively vagal activity on AER was studied in 6 dogs with bilateral cervical vag sympathetic trunks stimulation during AER in group C.Bilateral cervical vagosympathetic trunks were decentralized. Multipolar catheters were placed into high right atria (RA),coronary sinus (CS) and right ventricle (RV).AER was induced by 600 bpm pacing through RA catheter for 30 minutes.Atrial effective refractory period (ERP) and vulnerability window (VW) of atrial fibrillation were measured with and without vagal stimulation before and after AER.Results In group A,ERP decreased significantly at baseline and during vagal stimulation after AER compared with that before AER (all P<0.05).In group B,ERP remained unchanged at baseline and vagal stimulation after AER compared with that before AER (all P>0.05).In group C,ERP shortened significantly at baseline and vagal stimulation after AER compared with that before AER (all P<0.05).ERP shortening after AER in Groups A and C increased significantly than that in group B (all P<0.05).Atrial fibrillation could not be induced at baseline (VW close to 0) before and after AER in three groups.VW became widen significantly during vagal stimulation after AER compared with that before AER in Groups A and C (all P<0.05),while VW remained unchanged in group B (VW close to 0).Conclusions Short-term AER results in the decrease in ERP.AER is accompanied by the increases in atrial vagal modulation.The increased vagal activity and vagal stimulation promote AER, thereby increase the susceptibility to atrial fibrillation.The interrupted vagal activity attenuates AER,thereby suppresses the atrial fibrillation mediated by vagal stimulation. | Shulong Zhang Yingxue Dong Lianjun Gao Donghui Yang Chunyue Zhao Hongwei Zhao Xiaomeng Yin Jinqiu Liu Zhihu Lin Yanzong Yang Department of Cardiology, The First Affiliated Hospital, Dalian Medical University, Dalian 116011, China | 2008 | Journal of Geriatric Cardiology2008,5,3: | 0 |
