| 1 | A Novel Hydrolytic Product from Flesh of Mactra veneriformis and Its Bioactivities in Calcium Supplement显示文摘To prepare calcium-binding peptides, the flesh residue of Mactra Veneriformis was subjected to enzymatic hydrolysis. By comparing the capability of combining calcium of the hydrolyzates, pepsin was confirmed to be the most suitable enzyme for hydrolyzing the flesh residue to release calcium-binding peptides among the seven tested proteases. The pepsin hydrolyzate (PHM) was divided into three fractions according to the molecule weight of its composition, which ranged from 0.5 to 15 kDa. The low-molecule-weight fraction named PHM-3 had the highest capability in combining calcium. The peptides existing in the PHM-3 fraction consisted of higher contents of Glu, Ala and Leu, and could produce one type of calcium-peptide complex by powerfully chelating calcium ions. PHM-3 products could effectively increase calcium absorption and retention while they decreased the calcium excretion in animal tests. | 2 | Kinematics Analysis and Optimization of the Fast Shearing-extrusion Joining Mechanism for Solid-state Metal显示文摘Dynamical Joining of the solid-state metal is the key technology to realize endless hot rolling. The heating and laser welding method both require long joining time. Based on super deformation method, a 7-bar and 2-slider mechanism was developed in Japan, and the joining time is less than 0.5 s, however the length of each bar are not reported and this mechanism is complex. A relatively simple 6-bar and 1-slider mechanism is put forward, which can realize the shearing and extrusion motion of the top and bottom blades with a speed approximately equal to the speed of the metal plates. In order to study the kinematics property of the double blades, based on complex vector method, the multi-rigid-body model is built, and the displacement and speed functions of the double blades, the joining time and joining thickness are deduced, the kinematics analysis shows that the initial parameters can't satisfy the joining process. Hence, optimization of this mechanism is employed using genetic algorithm(GA) and the optimization parameters of this mechanism are obtained, the kinematics analysis show that the joining time is less than 0.1 s, the joining thickness is more than 80% of the thickness of the solid-state metal, and the horizontal speeds of the blades are improved. | 10 | Quantifying E2F1 protein dynamics in single cells显示文摘Background:E2F1 protein,a major effector of the Rb/E2F pathway plays a central role in regulating cell-fate decisions involved in proliferation,apoptosis,and differentiation.Its expression is highly dynamic and tightly modulated through a combination of transcriptional,translational and posttranslational controls.However,the mechanisms by which its expression and activity can promote different cellular outcomes remain to be fully elucidated.To better document E2F1 expression in live cells,we have engineered a series of fluorescent E2F1 protein reporters that quantitatively capture E2F1 protein dynamics.Methods:Reporter constructs,under the control of the mouse or human E2F1 proximal promoter,were designed to express an E2F1-Venus fusion protein incapable of binding DNA.In addition,constructs either included or excluded the y untranslated region(3fUTR)of the E2F1 gene.These constructs were introduced into fibroblasts and epithelial cells,and expression of the fusion reporter protein was validated and quantified in single cells using live imaging.Results:In all cases,expression of the reporter protein effectively recapitulated the behavior of E2F1 under various conditions,including cell cycle progression and genotoxic stress.No or little fluorescent signal of the reporter was detected in G〇,but as the cycle progressed,expression of the reporter protein steadily increased in the nucleus,peaking a few hours before cell division,but declining to baseline 2-3 h prior to the onset of mitosis.The absence of the E2F13 fUTR in the constructs led to considerably higher steady-state levels of the fusion protein,which although normally regulated,exhibited a slightly less complex dynamic profile during the cell cycle or genotoxic stress.Lastly,the presence or absence of Rb failed to impact the overall detection and levels of the reporter proteins.Conclusions:Our validated E2F1 protein reporters complement nicely other reporters of the Rb/E2F pathway and provide a unique tool to follow the complex dynamics of E2F1 expression in real time in single cells. | Bernard Mathey-Prevot Bao-Tran Parker Carolyn Im Cierra Hong Peng Dong Guang Yao Lingchong You | 2020 | Quantitative Biology2020,8,1: | 0 |

| 14 | Anti-hyperglycemic effect of the polysaccharide fraction isolated from mactra veneriformis显示文摘Total macromolecule extract was obtained from the soft body of Mactra veneriformis by the coupling techniques of decoction and alcohol precipitation.The extract was deproteinized with an ion exchange column,and resulted in the purifying of the crude polysaccharide fraction.It was found by chemical analysis that the crude polysaccharide part is composed of abundant polysacchar-ides(>95%)and few proteins(<1%).Furthermore,only one type of monosaccharide,glucose,was detected from its hydrolytes by thin-layer chromatography,indicating that the polysaccharides might be analogs of glucosan.The anti-hyperglycemia effects of the crude polysaccharide part were preliminarily investigated using several pharmacolo-gical methods in normal and diabetic mice.Animal experimental results showed that the crude polysaccharide fraction exhibited proper glycemia inhibition activity,and 300 mg/kg-weight dose has the optimal effect among all the studied doses.It is concluded that the crude polysaccharide fraction can be explored as a novel health product that possesses potential as an anti-hyperglycemic agent. | Lingchong WANG Hao WU Nian CHANG Kun ZHANG | 2011 | Frontiers of Chemical Science and Engineering2011,5,2: | 0 |
