1NLRP3-dependent pyroptosis is required for HIV-1 gp120- induced neuropathology显示文摘The human immunodeficiency virus-1(HIV-1)envelope protein gp120 is the major contributor to the pathogenesis of HIVassociated neurocognitive disorder(HAND).Neuroinflammation plays a pivotal role in gp120-induced neuropathology,but how gp120 triggers neuroinflammatory processes and subsequent neuronal death remains unknown.Here,we provide evidence that NLRP3 is required for gp120-induced neuroinflammation and neuropathy.Our results showed that gp120-induced NLRP3-dependent pyroptosis and IL-1βproduction in microglia.Inhibition of microglial NLRP3 inflammasome activation alleviated gp120-mediated neuroinflammatory factor release and neuronal injury.Importantly,we showed that chronic administration of MCC950,a novel selective NLRP3 inhibitor,to gp120 transgenic mice not only attenuated neuroinflammation and neuronal death but also promoted neuronal regeneration and restored the impaired neurocognitive function.In conclusion,our data revealed that the NLRP3 inflammasome is important for gp120-induced neuroinflammation and neuropathology and suggest that NLRP3 is a potential novel target for the treatment of HAND.Xiaolong He Weijun Yang Zhijie Zeng Yi Wei Jie Gao Bao Zhang Li Li Liqun Liu Yu Wan Qing Zeng Zelong Gong Liting Liu Hanyun Zhang Yubin Li Shaojie Yang Tongtong Hu Lixian Wu Eliezer Masliah Shenghe Huang Hong Cao 2020Cellular & Molecular Immunology2020,17,3:11
2Accuracy of predicting metabolizable energy from in vitro digestible energy determined with a computer-controlled simulated digestion system in feed ingredients for ducks显示文摘Two experiments were conducted to study the accuracy of predicting true metabolizable energy(TME)of ingredients for ducks from in vitro digestible energy(IVDE)determined with a computer-controlled simulated digestion system.Experiment 1 was to establish TME prediction models from the IVDE of 9 energy feed ingredients and 12 protein feed ingredients using regression analysis.Experiment 2 was to validate the accuracy of the predicted ME of 10 ingredients randomly selected from Exp.1.Ten diets were formulated with 2 to 6 of 10 ingredients.Dietary in vivo TME values were compared with calculated values based on the TME predicted in Exp.1.In Exp.1,the correlation coefficients between TME and IVDE were 0.9339(P<0.05)in 9 energy feed ingredients and 0.8332(P<0.05)in 12 protein feed ingredients.No significant difference was observed on the slope and intercept of TME regression models between 9 energy feed ingredients and 12 protein feed ingredients.Therefore,the regression model of TME on IVDE for 21 feed ingredients was TME=0.7169×IVDE+1,224(R^(2)=0.7542,P<0.01).Determined and predicted TME differed by less than 100 kcal/kg of DM in 11 ingredients,and the difference ranged from 100 to 200 kcal/kg of DM in 5 ingredients.However,the difference between determined and predicted TME varied from 410 to 625 kcal/kg of DM in rice bran,rapeseed meal,corn gluten meal,and citric acid meal.In Exp.2,the determined and calculated TME were comparable(3,631 vs.3,639 kcal/kg of DM)and highly correlated(r=0.9014;P<0.05)in 10 diets.Determined and calculated TME differed by less than 100 kcal/kg of DM in 7 diets and by 106 to 133 kcal/kg of DM in 3 diets.These results have demonstrated that TME can be accurately predicted from IVDE in most feed ingredients,but it is less accurate for rice bran,rapeseed meal,corn gluten and citric acid meal.Yuming Wang Liting Yin Hu Zhang Ke Li Dailin Li Feng Zhao 2022Animal Nutrition2022,,1:2

4Effect of Roucongrong(Herba Cistanches Deserticolae) decoction on the substantia nigra through Wnt/β-catenin signaling pathway in rats with Parkinson's disease induced by 6-hydroxydopamine hydrochloride显示文摘OBJECTIVE:To investigate the effect of Roucongrong(Herba Cistanches Deserticolae)decoction on the substantia nigra in rats with Parkinson’s disease(PD)induced by 6-hydroxydopamine hydrochloride(6-OHDA).To further determine whether the Wnt/β-catenin signaling pathway is involved in the action.METHODS:A rat model of PD was established by intracranial injection of 6-OHDA.Subsequently,three concentrations of Roucongrong(Herba Cistanches Deserticolae)decoction were prepared and administered to rats by gavage therapy for 14 d.Behavioral changes were measured in PD rats.In vivo tyrosine hydroxylase(TH)levels in the substantia nigra were examined by immunohistochemistry.Additionally,gene and protein expression levels of members of the Wnt/β-catenin signaling pathway were examined by Western blotting and polymerase chain reaction.Lastly,a Wnt/β-catenin inhibitor was used to investigate the mechanism of action in 1-methyl-4-phenylpyridinium(MPP+)-treated MES23.5 cells in vitro.RESULTS:Roucongrong(Herba Cistanches Deserticolae)decoction improved performance in the stride and gait adjustment tests in PD rats.It also increased TH in the substantia nigra and altered the expression of genes and proteins in the Wnt/β-catenin signaling pathway.Wnt/β-catenin inhibitor reduced the effect of Roucongrong(Herba Cistanches Deserticolae)decoction in MPP+-treated MES23.5 cells.CONCLUSION:Roucongrong(Herba Cistanches Deserticolae)decoction may promote neuronal survival in PD in vivo and in vitro by increasing TH content in the substantia nigra and by activating the Wnt/β-catenin signaling pathway.XU Qian QIN Wei WU Fangzhen LIN Yao HONG Liting CHEN Dan HU Xuefeng CAI Jing 2021Journal of Traditional Chinese Medicine2021,41,5:1

10Constructing metal-free heterophotocatalyst using two-dimensional carbon nitride sheets and violet phosphorene for highly efficient visible-light photocatalysis显示文摘Two-dimensional(2D)carbon nitride sheets(CNs)with nanoscale thickness have great potential in solar energy conversion owing to their intrinsic bandgap.However,their photocatalytic efficiency falls far below the practical requirements because the generated electron-hole pairs rapidly recombine.We construct a metal-free 2D p–n junction heterophotocatalyst from CNs and violet phosphorene(VP)using a simple electrostatic self-assembly method.The experimental results and density functional theory calculations show that the built-in electric field of the p–n junction promotes photogenerated carrier transport at the heterojunction interface,thereby promoting the separation of photogenerated electron-hole pairs.Under visible-light illumination,the visible-light photocatalytic hydrogen and CO production rates of the CNs/VP heterojunction were increased by 5.85 and 1.51 times,respectively,when compared to that of CNs.This study provides new insights into the design of metal-free heterogeneous photocatalysts with high catalytic activity。Yong Wang Chengxin Zeng Liting Wu Yalin Dong Yu Zhang Dingyi Yang Wen Hu Jian Hao Hongzhe Pan Rusen Yang 2023Journal of Materials Science & Technology2023,,15:0
11SARS-CoV-2 spike-specific TFH cells exhibit unique responses in infected and vaccinated individuals显示文摘Long-term humoral immunity to SARS-CoV-2 is essential for preventing reinfection. The production of neutralizing antibody (nAb)and B cell differentiation are tightly regulated by T follicular help (T_(FH)) cells. However, the longevity and functional role of T_(FH) cellsubsets in COVID-19 convalescents and vaccine recipients remain poorly defined. Here, we show that SARS-CoV-2 infection andinactivated vaccine elicited both spike-specific CXCR3^(+) T_(FH) cell and CXCR3^(-) T_(FH) cell responses, which showed distinct responsepatterns. Spike- specific CXCR3^(+) T_(FH) cells exhibit a dominant and more durable response than CXCR3^(-) T_(FH) cells that positivelycorrelated with antibody responses. A third booster dose preferentially expands the spike-specific CXCR3^(+) T_(FH) cell subset inducedby two doses of inactivated vaccine, contributing to antibody maturation and potency. Functionally, spike-specific CXCR3^(+) T_(FH) cellshave a greater ability to induce spike-specific antibody secreting cells (ASCs) differentiation compared to spike-specific CXCR3^(-) T_(FH)cells. In conclusion, the persistent and functional role of spike-specific CXCR3^(+) T_(FH) cells following SARS-CoV-2 infection andvaccination may play an important role in antibody maintenance and recall response, thereby conferring long-term protection. 12Global methyl halide emissions from biomass burning during 2003-2021显示文摘Methyl halides(CH3Cl,CH3Br,and CH3I)are ozone-depleting substances.Biomass burning(BB)is an important source of methyl halides.The temporal variations and global spatial distribution of BB methyl halide emissions are unclear.Thus,global methyl halide emissions from BB during 2003e2021 were estimated based on satellite data.A significant decreasing trend(p<0.01)in global methyl halide emissions from BB was found between 2003 and 2021,with CH3Cl emissions decreasing from 302 to 220 Gg yr^(-1),CH3Br emissions decreasing from 16.5 to 11.7 Gg yr^(-1),and CH3I emissions decreasing from 8.9 to 6.1 Gg yr^(-1).From a latitudinal perspective,the northern high-latitude region(60e90N)was the only latitude zone with significant increases in BB methyl halide emissions(p<0.01).Based on an analysis of the drivers of BB methyl halide emissions,emissions from cropland,grassland,and shrubland fires were more correlated with the burned area,while BB emissions from forest fires were more correlated with the emissions per unit burned area.The non-BB emissions of CH3Cl increased from 4749 Gg yr^(-1)in 2003 to 4882 Gg yr^(-1)in 2020,while those of CH3Br decreased from 136 Gg yr^(-1)in 2003 to 118 Gg yr^(-1)in 2020(global total CH3I emissions are not available).The finding indicates that global CH3Cl and CH3Br emissions from sources besides BB increased and decreased during 2003e2020.Based on our findings,not only searching for unknown sources is important,but also re-evaluating known sources is necessary for addressing methyl halide emissions.Xiaoyi Hu Di Chen Liting Hu Bowei Li Xinhe Li Xuekun Fang 2023Environmental Science and Ecotechnology2023,,2:0
