2Construction of multiform scFv antibodies using linker peptide显示文摘Multiform single chain variable fragments (scFvs) including different length linker scFvs and bispecific scFv were constructed. The linker lengths of 0, 3, 5, 8, 12, and 15 amino acids between VH and VL of antideoxynivalenol (anti-DON) scFv were used to analyze the affinities of scFvs. The affinity constants of these scFvs increased when the linker was lower than 12 amino acids. The affinity constant would not change when the linker was longer than 12 amino acids. Fusion gene of anti-DON scFv and antizearalenone (anti-ZEN) scFv was also constructed through connection by a short peptide linker DNA to express a bispecific scFv. The affinity constants assay showed that the two scFvs of fusion bispecific scFv remained their own affinity compared to their parental scFvs. Competitive direct enzyme linked immunosorbent assay was used to detect DON and ZEN in contaminated wheat (Triticum aestivum L.) samples, and the results indicated that this bispecific scFv was applicable in DON and ZEN detection. 3Comparative QTL analysis of maize seed artificial aging between an immortalized F_2 population and its corresponding RILs显示文摘Seed aging decreases the quality and vigor of crop seeds,thereby causing substantial agricultural and economic losses in crops.To identify genetic differences in seed aging between homozygotes and heterozygotes in maize,the seeds of a set of recombinant inbred lines(RILs) and an immortalized F_2(IF_2) population were subjected to artificial aging treatments for 0,2,3,and 4 days under 45℃ and 85%relative humidity and seed vigor was then evaluated in a field experiment.Seed vigor of all entries tested decreased sharply with longer aging treatment and seed vigor decreased more slowly in heterozygotes than in homozygotes.Forty-nine QTL were detected for four measured seed vigor traits in the RIL(28QTL) and IF_2(21 QTL) populations.Only one QTL,qGP5,was detected in both populations,indicating that the genes involved in anti-aging mechanisms differed between inbred lines and hybrids.Several QTL were identified to be responsible for multiple seed vigor traits simultaneously in the RIL and IF_2 populations under artificial aging conditions.These QTL may include major genes for seed vigor or seed aging.QTL qVI4 b and qGE3 a detected in the RIL population coincided with genes ZmLOX1 and ZmPLD1 in the same respective chromosomal regions.These QTL would be useful for screening for anti-aging genes in maize breeding.Bin Wang Zhanhui Zhang Zhiyuan Fu Zonghua Liu Yanmin Hu Jihua Tang 2016The Crop Journal2016,4,1:5

Recently, signal response amplification has been shown to be implemented through networks exhibiting scale-free topology, which has potential applications in artificial information processing systems and devices. 5Spatial multi-scaled chimera states of cerebral cortex network and its inherent structure-dynamics relationship in human brain显示文摘Human cerebral cortex displays various dynamics patterns under different states,however the mechanism how such diverse patterns can be supported by the underlying brain network is still not well understood.Human brain has a unique network structure with different regions of interesting to perform cognitive tasks.Using coupled neural mass oscillators on human cortical network and paying attention to both global and local regions,we observe a new feature of chimera states with multiple spatial scales and a positive correlation between the synchronization preference of local region and the degree of symmetry of the connectivity of the region in the network.Further,we use the concept of effective symmetry in the network to build structural and dynamical hierarchical trees and find close matching between them.These results help to explain the multiple brain rhythms observed in experiments and suggest a generic principle for complex brain network as a structure substrate to support diverse functional patterns.Siyu Huo Changhai Tian Muhua Zheng Shuguang Guan Changsong Zhou Zonghua Liu 2021National Science Review2021,8,1:1

7Systematic dissection of disease resistance to southern corn rust by bulked-segregant and transcriptome analysis显示文摘Southern corn rust(SCR) is a destructive maize disease caused by Puccinia polysora Underw. To investigate the mechanism of SCR resistance in maize, a highly resistant inbred line, L119 A, and a highly susceptible line, Lx9801, were subjected to gene mapping and transcriptome analysis. Bulked-segregant analysis coupled with whole-genome sequencing revealed several quantitative trait loci(QTL) on chromosomes 1, 6, 8, and 10. A set of 25 genes, including two coiled-coil nucleotide-binding site leucine-rich repeat(CC-NBS-LRR) genes, were identified as candidate genes for a major-effect QTL on chromosome 10. To investigate the mechanism of SCR resistance in L119 A, RNA-seq of P. polysorainoculated and non-inoculated plants of L119 A and Lx9801 was performed. Unexpectedly, the number of differentially expressed genes in inoculated versus non-inoculated L119 A plants was about 10 times that of Lx9801, with only 29 common genes identified in both lines, suggesting extensive gene expression changes in the highly resistant but not in the susceptible line. Based on the transcriptome analysis, one of the CC-NBS-LRR candidate genes was confirmed to be upregulated in L119 A relative to Lx9801 independently of P. polysora inoculation. Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analyses indicated that transcription factors, as well as genes involved in defense responses and metabolic processes, were dominantly enriched, with the phenylpropanoid biosynthesis pathway most specifically activated. Consistently, accumulation of phenylpropanoid-derived lignin, especially S lignin, was drastically increased in L119 A after P. polysora inoculation, but remained unchanged in Lx9801, suggesting a critical role of lignin in SCR resistance. 18Nitrogen and atomic Fe dual-doped porous carbon nanocubes as superior electrocatalysts for acidic H2-O_(2) PEMFC and alkaline Zn-air battery显示文摘Air cathodes with high electrocatalytic activity are vital for developing H2/O_(2) proton exchange membrane fuel cells(PEMFC)and Zn-air batteries.However,the state-of-the-art air cathodes suffer from either limited catalytic activity or high cost,which thus hinder their applications.Herein,we designed ZIF-8 derived nitrogen and atomic iron dual-doped porous carbon nanocubes as high-quality catalysts for ORR,through a novel gas-doping approach.The porous carbon nanocubic architecture and abundant Fe-Nxactive species endow ZIF-8 derived single atomic iron catalyst(PCN-A@Fe SA)with superior catalytic activity,and surpass Pt/C and a majority of the reported catalysts.Both XAS and DFT calculations suggest that Fe2+N4 moieties are the main active centers that are favorable for oxygen affinity and OH*intermediate desorption,which can result in promising catalytic performance.Most importantly,PCNA@Fe SA can achieve power density of 514 m W cm^(-2) as cathodic catalyst in a PEMFC and discharge peak power density of 185 m W cm^(-2) in an alkaline Zn-air battery.The outstanding performance is derived from both the high specific surface area and high-density of iron single atom in nitrogen doped nanocubic carbon matrix.Xiudong Shi Zonghua Pu Bin Chi Mingrui Liu Siyan Yu Long Zheng Lijun Yang Ting Shu Shijun Liao 2021Journal of Energy Chemistry2021,30,8:1
19Thermal-siphon phenomenon and thermal/electric conduction in complex networks显示文摘In past decades,a lot of studies have been carried out on complex networks and heat conduction in regular lattices.However,very little attention has been paid to the heat conduction in complex networks.In this work,we study(both thermal and electric)energy transport in physical networks rewired from 2D regular lattices.It is found that the network can be transferred from a good conductor to a poor conductor,depending on the rewired network structure and coupling scheme.Two interesting phenomena were discovered:(i)the thermal-siphon effect——namely the heat flux can go from a low-temperature node to a higher-temperature node and(ii)there exits an optimal network structure that displays small thermal conductance and large electrical conductance.These discoveries reveal that network-structured materials have great potential in applications in thermal-energy management and thermal-electric-energy conversion.Kezhao Xiong Zonghua Liu Chunhua Zeng Baowen Li 2020National Science Review2020,7,2:1
