| 1 | Molecular basis of CONSTANS oligomerization in FLOWERING LOCUS T activation显示文摘The transcription factor CONSTANS(CO)integrates day-length information to induce the expression of florigen FLOWERING LOCUS T(FT)in Arabidopsis.We recently reported that the C-terminal CCT domain of CO forms a complex with NUCLEAR FACTOR-YB/YC to recognize multiple cis-elements in the FT promoter,and the N-terminal tandem B-box domains form a homomultimeric assembly.However,the mechanism and biological function of CO multimerization remained unclear.Here,we report that CO takes on a head-to-tail oligomeric configuration via its B-boxes to mediate FT activation in long days.The crystal structure of B-boxesCOreveals a closely connected tandem B-box fold forming a continuous head-to-tail assembly through unique CDHH zinc fingers.Mutating the key residues involved in CO oligomerization resulted in a non-functional CO,as evidenced by the inability to rescue co mutants.By contrast,a transgene encoding a human p53-derived tetrameric peptide in place of the B-boxesCOrescued co mutant,emphasizing the essential role of BboxesCO-mediated oligomerization.Furthermore,we found that the four TGTG-bearing cis-elements in FT proximal promoter are required for FT activation in long days.Our results suggest that CO forms a multimer to bind to the four TGTG motifs in the FT promoter to mediate FT activation. | Xiaolin Zeng Xinchen Lv Rui Liu Hang He Shiqi Liang Lixian Chen Fan Zhang Liu Chen Yuehui He Jiamu Du | 2022 | Journal of Integrative Plant Biology2022,64,3: | 2 |
| 2 | Identification and bioinformatics analysis of lactate dehydrogenase genes from Echinococcus granulosus显示文摘Objective:To identify full length cDNA sequence of lactate dehydrogenase(LDH) from adult Echinococcus granulosus(E.granulosus) and to predict the structure and function of its encoding protein using bioinformatics methods.Methods:With the help of NCBI,EMBI, Expasy and other online sites,the open reading frame(ORF),conserved domain,physical and chemical parameters,signal peptide,epitope,topological structures of the protein sequences were predicted and a homology tertiary structure model was created:Vector NT1 software was used for sequence alignment,phylogenetic tree construction and tertiary structure prediction. Results:The target sequence was 1 233 bp length with a 996 bp biggest ORF encoding 331 amino acids protein with typical L-LDH conserved domain.It was confirmed as full length cDNA of LDH from E.granulosus and named as EgLDH(GenBank accession number:HM748917).The predicted molecular weight and isoelectric point of the deduced protein were 3 5516.2Da and 6.32 respectively.Compared with LDHs from Taenia solium,Taenia saginata asiatica,Spirometra erinaceieuropaei.Schistosoma japonicum,Clonorchis sinensis and human,it showed similarity of 86% ,85% ,55% ,58% ,58% and 53% ,respectively.EgLDH contained 3 putative transmembrane regions and 4 major epitopes(54aa-59aa.81aa-87aa,97aa-102aa,307aa-313aa),the latter were significant different from the corresponding regions of human LDH.In addition,some NAD and substrate binding sites located on epitopes 54aa-59aa and 97aa-102aa,respectively.Tertiary structure prediction showed that 3 key catalytic residues 105R,165D and 192H forming a catalytic center near the epitope 97aa-102aa,most NAD and substrate binding sites located around the center.Conclusions:The full length cDNA sequences of EgLDH were identified.It encoded a putative transmembrane protein which might be an ideal target molecule for vaccine and drugs. | Gang Lu Yajun Lu Lihua Li Lixian Wu Zhigang Fan Dazhong Shi Hu Wang Xiumin Han | 2010 | Asian Pacific Journal of Tropical Medicine2010,3,10: | 2 |

| 9 | Achieving High-Performance Molecular Rectification through Fast Screening Alkanethiol Carboxylate- Metal Complexes Electroactive Units显示文摘Achieving high performance in molecular scale diode devices remains a formidable challenge due to the complexity of the charge transport process and the difficulty in device structure modulation.Herein,we demonstrated a low-cost and fast screening strategy to search for proper electroactive units and construct high performance self-assembled monolayer(SAM)-based molecular diodes.The strategy is based on off-the-shelf carboxylate-terminated alkane thiols and simple carboxylate-metal ion complexes,structures denoted as Au-S-(CH2)n-1COO−Mm+(Cn+Mm+),where n=11,12,13,14,16,18,and Mm+=Ca^(2+),Mn^(2+),Fe_(2)+,Fe3+,Co^(2+),Ni^(2+),Cu^(2+),and Zn^(2+),as the library of functional SAM layers on gold substrate.Combing the fast screening measurements using a eutectic indium-gallium alloy top contact(EGaIn),C18+Ca^(2+)and C18+Zn^(2+)structures were found to afford record high rectification ratio(RR)>700 at±1.5 V.Theoretical analysis based on a single level tunneling model shows that the C18+Ca^(2+)and C18+Zn^(2+)devices possessed an optimized combination of asymmetric voltage division,energy offset,and coupling of carboxylate-metal complexes with the electrode.This newly developed method represents a general strategy for fast,inexpensive,and effective exploration of the functional metal complex chemical space,and can largely accelerate the development of practical high performance molecular diode devices. | Lixian Tian Aiqing Fan Xi Yu Wenping Hu | 2023 | CCS Chemistry2023,5,4: | 0 |
