1Secalonic acid D induces cell apoptosis in both sensitive and ABCG2-overexpressing multidrug resistant cancer cells through upregulating c-Jun expression显示文摘Secalonic acid D(SAD) could inhibit cell growth in not only sensitive cells but also multidrug resistant(MDR) cells. However, the molecular mechanisms need to be elucidated. Here, we identified that SAD possessed potent cytotoxicity in 3 pairs of MDR and their parental sensitive cells including S1-MI-80 and S1,H460/MX20 and H460, MCF-7/ADR and MCF-7 cells. Furthermore, SAD induced cell G2/M phase arrest via the downregulation of cyclin B1 and the increase of CDC2 phosphorylation. Importantly, JNK pathway upregulated the expression of c-Jun in protein level and increased c-Jun phosphorylation induced by SAD, which was linked to cell apoptosis via c-Jun/Src/STAT3 pathway. To investigate the mechanisms of upregulation of c-Jun protein by SAD, the mR NA expression level and degradation of c-Jun were examined. We found that SAD did not alter the mR NA level of c-Jun but inhibited its proteasome-dependent degradation. Taken together, these results implicate that SAD induces cancer cell death through c-Jun/Src/STAT3 signaling axis by inhibiting the proteasome-dependent degradation of c-Jun in both sensitive cells and ATP-binding cassette transporter sub-family G member 2(ABCG2)-mediated MDR cells.Hong Zhang Liyan Huang Liyang Tao Jianye Zhang Fang Wang Xu Zhang Liwu Fu 2019Acta Pharmaceutica Sinica B2019,9,3:5
2Growth Inhibitory Effects of Garlic Polysaccharide on Human HepG2 Cells显示文摘[Objective] The growth inhibitory effects of garlic polysaccharide(GPS) on human Hep G2 cells were evaluated in this paper. [Method] Hep G2 cells were treated with GPS for 48 h for morphology assay by transition electron microscope. Anti-proliferative effects with the same treatment for 24 hand 48 h were assayed by MTT method.Cell cycle distribution and apoptosis assay of treated cells were performed in flow cytometry. [Result] The results showed that GPS enhanced growth inhibitory effect on Hep G2 cells in a time-and dose-dependent manner. PI(Propidium iodide)/Annexin V staining analyzed by FCM(flow cytometry) demonstrated that GPS has a cytotoxic effect on tumor cells. Cell cycle arrest of Hep G2 treated with GPS occurred in G2 phase. 3Effects of Ligustrazine Combined with Cisdichlorodiamine Platinum on Anti-proliferation,Cell Cycle and Apoptosis of Human Gastric Carcinoma SGC-7901 Cell Lines显示文摘[Objective] This study aimed to investigate the combination effects of ligustrazine and cis-dichlorodiamine platinum(DDP) on anti-proliferation, cell cycle and apoptosis of SGC-7901 cell lines in vitro. [Method] SGC-7901 cells were treated with ligustrazine and DDP alone or combined for 48 h for morphology assay.Anti-proliferative effects with the same treatment for 24, 48 and 72 h were assayed by MTT method, respectively. Cell cycle distribution and apoptosis assay of treated cells were performed in flow cytometry. Zhengjun Jin's protocol was used to assay the effect of drug combination. [Result] Ligustrazine significantly increased the proliferation inhibition rate of DDP on SGC-7901 cells in combination with DDP, comparing with the effects of ligustrazine or DDP alone, and exhibited synergistic antitumor effect. The combination drug treatment induced cell cycle arrest occurred in S and G2 phase of the cell cycle and increased the apoptosis rate significantly.[Conclusion] Our results indicate that ligustrazine, as a low-toxic and natural herbal component, can significantly increase the anti-proliferative effect and apoptosis rate of antitumor drug DDP on human gastric carcinoma SGC-7901 cells.Liyan LI Tao HUANG An JIA Huili HOU Sen PANG Yuzhen WANG 2014Agricultural Science & Technology2014,15,9:2
4Changes of NF-κB, Bax and Caspase 3 in Apoptosis Induced by Ligustrazine Combined with Cis-dichlorodiamine Platinum in Human Gastric Carcinoma SGC-7901 Cell Lines显示文摘[Objective] This study aimed to investigate the mechanism of apoptosis induced by ligustrazine(TMP) and cis-dichlorodiamine platinum(DDP) in SGC-7901 cell lines in vitro. [Methods] SGC-7901 cell lines were treated with ligustrazine and DDP alone or combined for 48 h for Western blot analysis, respectively. Western blot analysis was used to determine the expression of proteins involved in apoptosis including NF-κB p65, bax and caspase-3. [Results] The viability of SGC-7901 cells was inhibited after treated with ligustrazine and/or combined with DDP. The expression of NF-κB P65 protein decreased after treated with drugs, in which the protein decreased significantly in 1.2 mg/ml of TMP combined with 2 μg/ml of DDP group.Meanwhile, we investigated the protein expression of bax and caspase-3. The results showed that the expression of the two proteins increased following with the increasing concentration of TMP. 6Anti-inflammatory Activity and Mechanism of Total Flavonoids from the Phloem of Paulownia elongate S.Y. Hu in LPS-stimulated RAW264.7 Macrophages显示文摘The juice leaked from broken phloem of Paulownia elongate S.Y.Hu has been used to cure acute inflammation resulted from stung injury by poisonous insects for a long time in China,but its potential mechanism remains unclear.The present study was designed to evaluate anti-inflammatory activity and mechanism of total flavonoids from the phloem of P.elongate S.Y.Hu in RAW264.7 cells.Lipopolysaccharide(LPS)-induced nitric oxide(NO)was measured by Griess and mRNA of pro-inflammatory mediators was analyzed by q-PCR.Cell viability was measured using cell counting kit(CCK)-8 assay.The protein level was analyzed by Western blot.The results showed that the total flavonoids of P.elongate significantly inhibited the production of the pro-inflammatory mediators such as NO,interleukin(IL)-1β and interleukin(IL)-6 in LPS-stimulated RAW264.7 cells.Total flavonoids of P.elongate exerted potential anti-inflammatory activity through the regulation of several signaling pathways.Total flavonoids of P.elongate inhibited JAK/STAT by blocking JAK2 and STAT3 phosphorylation levels.This is the first report on anti-inflammatory activity of total flavonoids from the phloem of P.elongate,which suggests that the total flavonoids of P.elongate may have great potential for the development of anti-inflammatory drug to treat inflammatory disorders.Liyan LI Tao HUANG Chong LAN 2021Agricultural Biotechnology2021,10,3:1
7Recovery of Ni(Ⅱ)from real electroplating wastewater using fixed-bed resin adsorption and subsequent electrodeposition显示文摘Effective recovery of high-value heavy metals from electroplating wastewater is of great significance,but recovering nickel ions from real electroplating wastewater as nickel sheet has not been reported.In this study,the pilot-scale fixed-bed resin adsorption was conducted to recover Ni(Ⅱ)ions from real nickel plating wastewater,and then the concentrated Ni(Ⅱ)ions in the regenerated solution were reduced to nickel sheet via electrodeposition.A commercial cation-exchange resin was selected and the optimal resin adsorption and regeneration conditions were investigated.The resin exhibited an adsorption capacity of 63 mg/g for Ni(Ⅱ)ions,and the average amount of treated water was 84.6 bed volumes(BV)in the pilot-scale experiments.After the adsorption by two ion-exchange resin columns in series and one chelating resin column,the concentrations of Ni(Ⅱ)in the treated wastewater were below 0.1 mg/L.After the regeneration of the spent resin using 3 BVof 4%(w/w)HC1 solution,1.5 BV of concentrated neutral nickel solution(>30 g/L)was obtained and used in the subsequent electrodeposition process.Using the aeration method,alkali and water required in resin activation process were greatly reduced to 2 BV and 3 BV,respectively.Under the optimal electrodeposition conditions,95.6%of Ni(Ⅱ)in desorption eluent could be recovered as the elemental nickel on the cathode,llie total treatment cost for the resin adsorption and regeneration as well as the electrodeposition was calculated.Tong Li Ke Xiao Bo Yang Guilong Peng Fenglei Liu Liyan Tao Siyuan Chen Haoran Wei Gang Yu Shubo Deng 2019Frontiers of Environmental Science & Engineering2019,13,6:1
8Erythrocytic α-Synuclein as a potential biomarker for Parkinson’s disease显示文摘Background:Erythrocytes are a major source of peripheralα-synuclein(α-Syn).The goal of the current investigation is to evaluate erythrocytic total,oligomeric/aggregated,and phosphorylatedα-Syn species as biomarkers of Parkinson’s disease(PD).PD and healthy control blood samples were collected along with extensive clinical history to determine whether total,phosphorylated,or aggregatedα-Syn derived from erythrocytes(the major source of bloodα-Syn)are more promising and consistent biomarkers for PD than are freeα-Syn species in serum or plasma.Methods:Using newly developed electrochemiluminescence assays,concentrations of erythrocytic total,aggregated and phosphorylated at Ser129(pS129)α-Syn,separated into membrane and cytosolic components,were measured in 225 PD patients and 133 healthy controls and analyzed with extensive clinical measures.Results:The total and aggregatedα-Syn levels were significantly higher in the membrane fraction of PD patients compared to healthy controls,but without alterations in the cytosolic component.The pS129 level was remarkably higher in PD subjects than in controls in the cytosolic fraction,and to a lesser extent,higher in the membrane fraction.Combining age,erythrocytic membrane aggregatedα-Syn,and cytosolic pS129 levels,a model generated by using logistic regression analysis was able to discriminate patients with PD from neurologically normal controls,with a sensitivity and a specificity of 72 and 68%,respectively.Conclusions:These results suggest that total,aggregated and phosphorylatedα-Syn levels are altered in PD erythrocytes and peripheral erythrocyticα-Syn is a potential PD biomarker that needs further validation.Chen Tian Genliang Liu Liyan Gao David Soltys Catherine Pan Tessandra Stewart Min Shi Zhiying Xie Na Liu Tao Feng Jing Zhang 2019Translational Neurodegeneration2019,8,1:1
9Primary Research on UVB-irradiation Protective Effect of Total Flavonoids from Large-leaved Kuding Tea (Ilex latifolia Thunb.)显示文摘Total flavonoids extracted from large-leaved Kuding tea(Ilex latifolia Thunb.) were investigated to develop anti-UV sun-screening agents. The results showed that total flavonoids of I. latifolia Thunb.(TFILT) showed maximum absorb peak at 325 nm between UVB and UVA by wavelength scanning spectrum. TFILT could inhibit the decrease of cell viability induced by UVB irradiation by CCK-8 method, and 0.25 mg/ml of TFILT showed the most significant protective activity on cells. The dermal thickness and collagen fiber density as skin structural characteristic were analyzed by Histological staining. The results showed that TFILT protected mice skin by significantly attenuating the dermal thickness and inhibiting collagen fiber degradation. 10Natural products and extracts from plants as natural UV filters for sunscreens:A review显示文摘Although solar exposure is necessary for human health,phototoxicology induced by excessive UVB and UVA radiation,which involves sunburns,skin aging and even tu-morigenesis,has been widely researched.Sunscreen is one of the most important ways to protect skin from UV phototoxic damage.As well as inorganic and organic UV filters,some natural products or plant extracts with aromatic rings in their structures,such as flavonoids or polyphenols,can absorb UV to reduce sunburn,acting as a natu-ral UV filter;they also show antioxidant or/and anti-inflammatory activity.This could explain why,although there are no officially approval natural commercial sun-filters,more and more commercial sunscreen products containing plant extracts are avail-able on the market.Here we summarize articles focusing on natural UV filters from plant published in the last 6 years,selecting the most significant data in order to better understand the photoprotective activity of natural products and extracts from plants,including their major constituents and main biological effects,methods for evaluating UV radiation resistance,anti-UV radiation experimental models and anti-UV radiation mechanisms.Liyan Li Lan Chong Tao Huang Yunge Ma Yingyan Li Hui Ding 2023Animal Models and Experimental Medicine2023,6,3:0
11Diversity and bioactivity of cultured aquatic fungi from the High Arctic region显示文摘This study assessed the diversity and α-glycosidase inhibitory activity of cultured fungi isolated from four aquatic environments(stream, pond, glacial ice, and estuary) in the Ny-?lesund region(Svalbard, Norway, High Arctic). A total of 134 fungal isolates were obtained from 13 water samples. Based on morphological characteristics and sequence analyses of the nuclear ribosomal DNA internal transcribed spacer region, these fungal isolates were identified as belonging to 47 species, with 26 belonging to the Ascomycota, 20 to the Basidiomycota, and one to the Zygomycota. The most frequently detected fungal species were Vishniacozyma sp. 2, Cadophora sp. 2, Phenoliferia sp. 1, Dioszegia sp. 2, and Mortierella sp.; these species occurred in 10, eight, seven, six, and five of the samples, respectively. Among the 134 fungal isolates, 17 isolates of 15 species displayed high α-glycosidase inhibitory activity in culture. 12Late-stage cascade of oxidation reactions during the biosynthesis of oxalicine B in Penicillium oxalicum显示文摘Oxalicine B(1)is an a-pyrone meroterpenoid with a unique bispirocyclic ring system derived from Penicillium oxalicum.The biosynthetic pathway of 15-deoxyoxalicine B(4)was preliminarily reported in Penicillium canescens,however,the genetic base and biochemical characterization of tailoring reactions for oxalicine B(1)has remained enigmatic.In this study,we characterized three oxygenases from the metabolic pathway of oxalicine B(1),including a cytochrome P450 hydroxylase OxaL,a hydroxylating Fe(II)/a-KG-dependent dioxygenase OxaK,and a multifunctional cytochrome P450 OxaB.Intriguingly,OxaK can catalyze various multicyclic intermediates or shunt products of oxalicines with impressive substrate promiscuity.OxaB was further proven via biochemical assays to have the ability to convert 15-hydroxdecaturin A(3)to 1 with a spiro-lactone core skeleton through oxidative rearrangement.We also solved the mystery of OxaL that controls C-15 hydroxylation.Chemical investigation of the wild-type strain and deletants enabled us to identify 10 metabolites including three new compounds,and the isolated compounds displayed potent anti-influenza A virus bioactivities exhibiting IC50values in the range of 4.0-19.9μmol/L.Our studies have allowed us to propose a late-stage biosynthetic pathway for oxalicine B(1)and create downstream derivatizations of oxalicines by employing enzymatic strategies.Tao Zhang Guowei Gu Guodong Liu Jinhua Su Zhilai Zhan Jianyuan Zhao Jinxiu Qian Guowei Cai Shan Cen Dewu Zhang Liyan Yu 2023Acta Pharmaceutica Sinica B2023,13,1:0
