1Effect of extracorporeal bioartificial liver support system on fulminant hepatic failure rabbits显示文摘AIM To evaluate the possibility of usingcultured human hepatocytes as a bridge betweenbioartificial liver and liver transplantation.METHODS In this experiment,the efficacy ofextracorporeal bioartificial liver support system(EBLSS)consisting of spheriodal human livercells and cultured hepatocytes supernatant wasassessed in vivo using galactosamine inducedrabbit model of fulminant hepatic failure.RESULTS There was no difference of survivalbetween the two groups of rabbits,but in thesupported rabbits serum alanineaminotransferase,total bilirubin and creatininewere significantly lower and hepatocyte necrosiswas markedly milder than those in controlanimals.In addition,a good viability of humanliver cells was noted after the experiment.CONCLUSION EBLSS plays a biologic role inmaintaining and compensating the function ofthe liver.Wang YJ Li MD Wang YM Chen GZ Lu GD Tan ZX 2000World Journal of Gastroenterology2000,6,2:17
2Portal vein embolization by fine needle ethanol injection :experimental and clinicalstudies显示文摘AIM To improve the technique of intraportal embolization (PVE) therapy, a new embolic method, was devised and the safety, effectiveness and feasibility were evaluated. METHODS PVE with intraportal ethanol injection via a fine needle was performed in 28 normal dogs, 22 SD rats, and 24 cirrhotic SD rats. After PVE, portography, histological and functional alteration of the liver were evaluated in dogs and rats, and the changes in portal hemodynamics as well as hepatic anatomy were observed in rats. In the clinical study, PVE by ethanol injection was performed in 61 patients with hepatocellular carcinoma under the guidance of portoechography with intraportal injection of CO 2. The effect of PVE was evaluated by ultrasonography and laparotomy. RESULTS The effectiveness and toxicity were dependent on the dose of ethanol. In the dogs, 0 25*!mg/*!kg of ethanol caused incomplete embolization with least liver damage, while 1 0*!mg/*!kg induced complete embolization with a high mortality of 57 1% (4/*!7) due to respiratory arrest. The dose of 0 5*!mg/*!kg resulted in complete embolization with slight toxicity to the liver. In the rats, the survival rate was 100% in normal group but 40 9% in cirrhotic models after ethanol injection by dose of 0 05*!mg/*!100*!g . PVE for cirrhotic rats with 0 03*!mg/*!100*!g of ethanol induced satisfactory embolization with significant hypertrophy in nonembolized lobes, and only slight damage to the hepatic parenchyma, and transient alteration in liver function, portal pressure and portal flow. In the clinical study, 12 cases with reverse portal flow were excluded judged by portoechography. Satisfactory embolization was gained in 90 2% (55/*!61) of the remaining patients determined by ultrasonography and surgery. All cases ran an uneventful postembolization course with no aberrant embolization. CONCLUSION PVE with intraportal ethanol injection of appropriate dosage via a fine needle is safe and effective and has several advantages comparing with transcatheter method. 5Advances in the development of molecular genetic tools for Mycobacterium tuberculosis显示文摘Mycobacterium tuberculosis, a Gram-positive bacterium of great clinical relevance, is a lethal pathogen owing to its complex physiological characteristics and development of drug resistance. Several molecular genetic tools have been developed in the past few decades to study this microorganism. These tools have been instrumental in understanding how M. tuberculosis became a successful pathogen. Advanced molecular genetic tools have played a significant role in exploring the complex pathways involved in M. tuberculosis pathogenesis. Here, we review various molecular genetic tools used in the study of M. tuberculosis. Further, we discuss the applications of clustered regularly interspaced short palindromic repeat interference(CRISPRi), a novel technology recently applied in M. tuberculosis research to study target gene functions. 10Meeting report of the chief editorial board meeting for World Journal of Diabetes 2021显示文摘The 2021 online editorial board meeting of the World Journal of Diabetes(WJD)was held on November 9,2021.Jin-Lei Wang,General Manager on behalf of the Baishideng Publishing Group,and Professor Islam,one of the Editors-in-Chiefs(EiCs)of the WJD,organized the meeting.Three EiCs and 18 Baishideng Publishing Group staff attended the meeting.The meeting goal was to brief the EiCs on the journal’s performance,discuss the issues of concern of the EiCs,and gather ideas for the journal’s development in 2022.As of November 8,the WJD had received 287 manuscripts since the year’s start,among which 122 met the criteria for publication.These numbers represent an increase of 117.4%for submissions and 110.3%for publications compared to those in 2020.However,how to effectively control the academic quality of manuscripts and attract highquality original article submissions remain a challenge.The EiCs provided feedback and suggestions centered on three topics:(1)Who should and how to control the academic quality of the manuscripts;(2)How the EiCs perform their responsibilities;and(3)The distinctive and shared responsibilities of the publisher and the EiCs.Jin-Lei Wang Jia-Ping Yan Xiang Li Md Shahidul Islam Jian-Bo Xiao Lu Cai Na Ma Li Ma Lian-Sheng Ma 2021World Journal of Diabetes2021,12,12:1
