1Analysis of cytochrome P450 genes in silkworm genome (Bombyx mori)显示文摘We have searched the Bombyx mori genome for members of the major enzyme family, the Cytochrome P450s, which carry out multiple reactions to enable organisms to rid themselves of foreign compounds. As a result, 86 putative P450s were discovered in silkworm genome, which are thought to belong to 32 subfamilies. A comparative genomic analysis with Drosophila melanogaster reveals that the two insects have some similar P450 distribution pat-terns but still have some obvious differences. Especially, the diverse distribution exists in 7 p450 subfamilies, which are CYP4A, CYP4C, CYP4D, CYP6A, CYP6AE, CYP6B and CYP9A. 14Tissue-specific genome editing of laminA/C in the posterior silk glands of Bombyx mori显示文摘The RNA-guided CRISPR/Cas9 system has been shown to be a powerful tool for genome editing in various organisms. A comprehensive toolbox for multiplex genome editing has been developed for the silkworm,Bombyx mori, a lepidopteran model insect of economic importance. However, as previous methods mainly relied on delivery of transient Cas9/guide RNA(gRNA), they could not be used in loss-of-function studies of essential genes. Here, we report a simple and versatile tissue-specific genome editing strategy.We perform a proof-of-principle demonstration by establishing and crossing two transgenic B. mori lines,one expressing Cas9 protein in the posterior silk glands(PSGs) and the other constitutively expressing BmlaminA/C(BmLMN) gRNA. All BmLMN alleles in the PSG cells were edited precisely at the target genome region, resulting in diverse mutations. mRNA expression of BmLMN was reduced by up to 75%,and only very low levels of BmLaminA/C protein were detected. Knockout of BmLMN produced obvious defects in gland cell development and cocoon production. 15Facile and Low-Cost Fabrication of a Thread/Paper-Based Wearable System for Simultaneous Detection of Lactate and pH in Human Sweat显示文摘Wearable devices have received tremendous interests in human sweat analysis in the past few years.However,the widely used polymeric substrates and the layer-by-layer stacking structures greatly influence the cost-efficiency,conformability and breathability of the devices,further hindering their practical applications.Herein,we report a facile and low-cost strategy for the fabrication of a skin-friendly thread/paper-based wearable system consisting of a sweat reservoir and a multi-sensing component for simultaneous in situ analysis of sweat pH and lactate.In the system,hydrophilic silk thread serves as the micro-channel to guide the liquid flow.Filter papers were functionalized to prepare colorimetric sensors for lactate and pH.The smartphone-based quantitative analysis shows that the sensors are sensitive and reliable.Although pH may interfere the lactate detection,the pH detected simultaneously could be employed to correct the measured data for the achievement of a precise lactate level.After being integrated with a hydrophobic arm guard,the system was successfully used for the on-body measurement of pH and lactate in the sweats secreted from the volunteers.This low-cost,easy-to-fabricate,light-weight and flexible thread/paper-based microfluidic sensing device may hold great potentials as a wearable system in human sweat analysis and point-of-care diagnostics.Gang Xiao Jing He Yan Qiao Feng Wang Qingyou Xia Xin Wang Ling Yu Zhisong Lu Chang-Ming Li 2020Advanced Fiber Materials2020,2,5:0
16Estimation of Enthalpy of Formation Using Benson’s Group Addition and Functional Group Correction显示文摘The enthalpies of formation of solid organic compounds containing carbon,nitrogen,oxygen,and hydrogen were estimated using two suggested descriptor sets,separately,by machine learning methods.The two descriptor sets are both composed of descriptors of Benson’s groups and corrected groups.The main differences between them are that one is generated based on atoms and the other is based on bonds.An in-house program was specially written in Java to extract all the descriptors with a function to ensure that each atom(or bond)of a molecule is represented by Benson’s groups once for an atom-based(or bond-based)descriptor set.Multiple linear regression and partial least squares were used,separately,to build models to predict the enthalpy of formation for two descriptor sets.The combination of the models constructed by two descriptor sets based on the atoms and the bonds achieved the best-predicted results in this paper,and the corresponding results of the test set are better than that in the literature,from which the original data were retrieved.Further,a small data set of fluorinated molecules was collected,and satisfactory results were also obtained for these molecules containing fluorine with the assistance of the former data set.LI Xianlan LUAN Yue LU Yanhua LI Wei MA Lihong ZHANG Qingyou PANG Aimin 2023Chemical Research in Chinese Universities2023,39,2:0

19Individual variation in buffalo somatic cell cloning efficiency is related to glycolytic metabolism显示文摘Mammalian individuals differ in their somatic cell cloning efficiency,but the mechanisms leading to this variation is poorly understood.Here we found that high cloning efficiency buffalo fetal fibroblasts(BFFs)displayed robust energy metabolism,looser chromatin structure,high H3 K9 acetylation and low heterochromatin protein 1α(HP1α)expression.High cloning efficiency BFFs had more H3 K9 ac regions near to the upstream of glycolysis genes by Ch IP-seq,and involved more openness loci related to glycolysis genes through ATAC-seq.The expression of these glycolysis genes was also found to be higher in high cloning efficiency BFFs by q RT-PCR.Two key enzymes of glycolysis,PDKs and LDH,were confirmed to be associated with histone acetylation and chromatin openness of BFFs.Treatment of low cloning efficiency BFFs with PS48(activator of PDK1)resulted in an increase in the intracellular lactate production and H3 K9 acetylation,decrease in histone deacetylase activity and HP1αexpression,less condensed chromatin structure and more cloning embryos developing to blastocysts.These results indicate that the cloning efficiency of buffalo somatic cells is associated with their glycolytic metabolism and chromatin structure,and can be improved by increasing glycolytic metabolism.Chan Luo Zhiqiang Wang Jinling Wang Feng Yun Fenghua Lu Jiayuan Fu Qingyou Liu Deshun Shi 2022Science China(Life Sciences)2022,65,10:0
