1Apoptosis and non-alcoholic fatty liver diseases显示文摘The number of patients with nonalcoholic fatty liver diseases(NAFLD) including nonalcoholic steatohepatitis(NASH), has been increasing. NASH causes cirrhosis and hepatocellular carcinoma(HCC) and is one of the most serious health problems in the world. The mechanism through which NASH progresses is still largely unknown. Activation of caspases, Bcl-2 family proteins, and c-Jun N-terminal kinase-induced hepatocyte apoptosis plays a role in the activation of NAFLD/NASH. Apoptotic hepatocytes stimulate immune cells and hepatic stellate cells toward the progression of fibrosis in the liver through the production of inflammasomes and cytokines. Abnormalities in glucose and lipid metabolism as well as microbiota accelerate these processes. The production of reactive oxygen species, oxidative stress, and endoplasmic reticulum stress is also involved. Cell death, including apoptosis, seems very important in the progression of NAFLD and NASH. Recently, inhibitors of apoptosis have been developed as drugs for the treatment of NASH and may prevent cirrhosis and HCC. Increased hepatocyte apoptosis may distinguish NASH from NAFLD, and the improvement of apoptosis could play a role in controlling the development of NASH. In this review, the association between apoptosis and NAFLD/NASH are discussed. 2Reduced EGFR signaling enhances cartilage destruction in a mouse osteoarthritis model显示文摘Osteoarthritis(OA) is a degenerative joint disease and a major cause of pain and disability in older adults. We have previously identified epidermal growth factor receptor(EGFR) signaling as an important regulator of cartilage matrix degradation during epiphyseal cartilage development. To study its function in OA progression, we performed surgical destabilization of the medial meniscus(DMM) to induce OA in two mouse models with reduced EGFR activity, one with genetic modification(EgfrWa5/1mice) and the other one with pharmacological inhibition(gefitinib treatment). Histological analyses and scoring at 3 months post-surgery revealed increased cartilage destruction and accelerated OA progression in both mouse models. TUNEL staining demonstrated that EGFR signaling protects chondrocytes from OA-induced apoptosis, which was further confirmed in primary chondrocyte culture. Immunohistochemistry showed increased aggrecan degradation in these mouse models, which coincides with elevated amounts of ADAMTS5 and matrix metalloproteinase 13(MMP13), the principle proteinases responsible for aggrecan degradation, in the articular cartilage after DMM surgery. Furthermore, hypoxia-inducible factor 2a(HIF2a), a critical catabolic transcription factor stimulating MMP13 expression during OA, was also upregulated in mice with reduced EGFR signaling. 3Intervertebral disc development and disease-related genetic polymorphisms显示文摘The intervertebral disc(IVD)comprises a gelatinous inner core(nucleus pulposus;NP)and concentric rings(annulus fibrosus;AF).The NP,an important structure for shock absorption in the vertebrate spinal motion segment,can be traced back to the notochord in ontogenetic lineage.In vertebrates,the notochord undergoes mucinoid changes,and had been considered vestigial until recently.However,observed correlations between IVD degeneration and back pain in humans have renewed interest in the IVD in biomedical fields.Beyond its mechanical contribution to development,the notochord is also an essential signaling center,which coordinates formation of the neural tube and somites.The pertinent signaling molecules,particularly TGF-b and bone morphogenetic proteins(BMPs),continue to play roles in the adult tissues and have been utilized for tissue regeneration.Genetic factors are major determinants of who will develop IVD degeneration and related back pain,and seem to correlate better with disc degeneration and back pain than do external forces on the spine.In summary,the spinal column is a landmark development in evolution.Genes directing the development of the IVD may also contribute to its maintenance,degeneration,and regeneration.Likewise,structural genes as well as genes responsible for maintenance of the structure are related to IVD degeneration.Finally,genes responsible for inflammation may play a dual role in exacerbating degeneration or facilitating repair responses depending on the context.Jason W.Ashley Motomi Enomoto-Iwamoto Lachlan J.Smith Robert L.Mauck Danny Chan Joseph Lee Martin F.Heyworth Howard An Yejia Zhang 2016Genes & Diseases2016,3,3:3
