| 1 | Cycloastragenol upregulates SIRT1 expression,attenuates apoptosis and suppresses neuroinflammation after brain ischemia显示文摘Cycloastragenol(CAG)is the active form of astragaloside V isolated from Astragalus Radix,which displays multiple pharmacological effects.Silent information regulator 1(SIRT1),a classll histone deacetylase,has been shown to play an important role in neuroprotection against cerebral ischemia.In this study,we investigated whether CAG protected against ischemic brain injury and,if so,whether the beneficial effects were associated with the regulation of SIRT1 in the ischemic brain.Mice were subjected to 45 min of middle cerebral artery occlusion(MCAO)followed by reperfusion.CAG(5,10,20 mg/kg)was injected intraperitoneally at the onset of reperfusion,12h later and then twice daily for up to three days.CAG dose-dependently reduced brain infarct volume,significantly ameliorated functional deficits,and prevented neuronal cell oss in MCAO mice.Meanwhile,CAG signicantly reduced matrix metalloproteinase-9 activity,prevented tight junction degradation and subsequently ameliorated blood-brain barier disruption.Moreover,CAG significantly upregulated SIRT1 expression in the ischemic brain but did not directly activate its enzymatic activity.Concomitant with SIRT1 upregulation,CAG reduced p53 acetylation and the ratio of Bax to Bcl-2 in the ischemic brain.CAG also inhibited NF-κB p65 nuclear translocation.As a result,CAG suppressed the mRNA expression of pro-inflammatory cytokines,including TNF-α and IL-1β,and inhibited the activation of microglia and astrocytes in the ischemic brain.Our findings suggest that CAG is neuroprotective against ischemic brain injury in mice and that its beneficial effect may involve SIRT1 upregulation and the inhibition of apoptosis and neuroinflammation in the ischemic brain. | Man Li Shichun Li Bao-kai Dou Yving-xiang Zou Hao-zhen Han Dong-xiang Liu Zunji Ke Zhi-fei Wang | 2020 | Acta Pharmacologica Sinica2020,41,8: | 22 |
| 2 | Investigation of quality of life in patients with lung cancer by the EORTC QLQ-C30(V3.0) Chinese version显示文摘Objective To explore quality of life(QOL) and its influencing factors in patients with lung cancer.Methods A QOL questionnaire(European Organization for Research and Treatment of Cancer [EORTC] Quality of Life Questionnaire-Core 30 [QLQ-C30] Chinese version) was used with 138 patients with lung cancer participating in the study. A statistical description of the general characteristics of the patients with lung cancer was performed. The patients' QLQ-C30 scores were compared with the reference value for each dimension. To analyze the influence of QOL in different genders, age groups, and cancer stages, ttests and rank sum tests were used to compare the differences in QOL using a 5% significance level. Results The QLQ-C30 function scores in PF(physical functioning), EF(emotional functioning), SF(social functioning), and GH(global functioning), and symptom scales in NV(nausea and vomiting), DY(dyspnea), SL(insomnia), and FI(financial difficulties) were significantly different(P < 0.05) in comparison to the reference values. Female patients were worse than males(P < 0.05) in EF, NV, and DI(diarrhea). The later the stage of lung cancer, the worse the quality of life became; the functional scales in RF(role functioning), EF, CF, SF, and GH, and symptom scales in PA(pain), AP(appetite), and SL differences were statistically significant(P < 0.05). | 3 | The FGOALS climate system model as a modeling tool for supporting climate sciences:An overview显示文摘Climate system models are useful tools for understanding the interactions among the components of the climate system and predicting/projecting future climate change. The development of climate models has been a central focus of the State Key Laboratory of Numerical Modeling for Atmospheric Sciences and Geophysical Fluid Dynamics, Institute of Atmospheric Physics, Chinese Academy of Sciences(LASG/IAP) since the establishment of the laboratory in 1985. In China, many pioneering component models and fully coupled models of the climate system have been developed by LASG/IAP. The fully coupled climate system developed in the recent decade is named FGOALS(Flexible Global Ocean-Atmosphere-Land System Model). In this paper, an application-oriented review of the LASG/IAP FGOALS model is presented. The improved model performances are demonstrated in the context of cloud-radiation processes, Asian monsoon, ENSO phenomena, Atlantic Meridional Overturning Circulation(AMOC) and sea ice. The FGOALS model has contributed to both CMIP5(Coupled Model Intercomparison Project-phase 5) and IPCC(Intergovernmental Panel on Climate Change) AR5(the Fifth Assessment Report). The release of FGOALS data has supported the publication of nearly 500 papers around the world. The results of FGOALS are cited ~106 times in the IPCC WG1(Working Group 1) AR5. In addition to the traditional long-term simulations and projections, near-term decadal climate prediction is a new set of CMIP experiment, progress of LAGS/IAP in the development of nearterm decadal prediction system is reviewed. The FGOALS model has supported many Chinese national-level research projects and contributed to the national climate change assessment report. | 6 | Effects of elevated atmospheric CO_2 concentration and temperature on the soil profile methane distribution and diffusion in rice–wheat rotation system显示文摘The aim of this experiment was to determine the impacts of climate change on soil profile concentrations and diffusion effluxes of methane in a rice–wheat annual rotation ecosystem in Southeastern China. We initiated a field experiment with four treatments:ambient conditions(CKs), CO_2 concentration elevated to ~ 500 μmol/mol(FACE),temperature elevated by ca. 2°C(T) and combined elevation of CO_2 concentration and temperature(FACE + T). A multilevel sampling probe was designed to collect the soil gas at four different depths, namely, 7 cm, 15 cm, 30 cm and 50 cm. Methane concentrations were higher during the rice season and decreased with depth, while lower during the wheat season and increased with depth. Compared to CK, mean methane concentration was increased by 42%, 57% and 71% under the FACE, FACE + T and T treatments, respectively, at the 7 cm depth during the rice season(p < 0.05). Mean methane diffusion effluxes to the 7 cm depth were positive in the rice season and negative in the wheat season, resulting in the paddy field being a source and weak sink, respectively. Moreover, mean methane diffusion effluxes in the rice season were 0.94, 1.19 and 1.42 mg C/(m^2·hr) in the FACE,FACE + T and T treatments, respectively, being clearly higher than that in the CK. | 8 | Creatine Pyruvate Enhances Lipolysis and Protein Synthesis in Broiler Chicken显示文摘To assess the effects of creatine pyruvate (Cr-Pyr) on lipid and protein metabolism in broiler chickens, a total of 400 1-day-old male birds (Aconred) were randomly allocated to four groups, with each group replicating four?times and each replicate involving 25?birds. The broilers were provided with a commercial diet supplemented with Cr-Pyr at 0, 1, 5, or 10% of the diet, respectively, for a period of 3 wk ad libitum (from 22 to 42 d). In the present study, body weight (BW) and average daily gain (ADG) of broilers decreased in 10% Cr-Pyr group (P<0.01), whereas the relative leg and pectoral muscle weights were significantly higher than they were in the control group (P<0.05). 5 or 10% Cr-Pyr of diets decreased the abdominal fat rate (AFR, abdominal fat/live weight) of the broilers. The serum or hepatic triglyceride (TG) concentrations were significantly lower in the 5 and 10% groups (P<0.01). In contrast, Cr-Pyr caused a marked increase in the serum non-esterified fatty acid (NEFA), high-density lipoprotein cholesterol (HDL-C) and creatine kinase (CK) concentrations (P<0.01). Supplementation with Cr-Pyr (5 and 10%) in the diet also increased glucagons (GLU), insulin (INS) or leptin (LEP) contents (P<0.01). The expression of hepatic peroxisomal proliferators-activated receptor a (PPAR-a) and carnitine palmitoyl transferase-I (CPT-I), muscle insulin-like growth factor I (IGF-I) were significantly elevated and myostatin mRNA level was reduced in the 5 and 10% groups (P<0.05). | 9 | In Situ Exfoliation and Pt Deposition of Antimonene for Formic Acid Oxidation via a Predominant Dehydrogenation Pathway显示文摘Direct formic acid fuel cell(DFAFC)has been considered as a promising energy conversion device for stationary and mobile applications.Advanced platinum(Pt)electrocatalysts for formic acid oxidation reaction(FAOR)are critical for DFAFC.However,the oxidation of formic acid on Pt catalysts often occurs via a dual pathway mechanism,which hinders the catalytic activity owing to the CO poisoning.Herein,we directly exfoliate bulk antimony to 2D antimonene(Sb)and in situ load Pt nanoparticles onto antimonene sheets with the assistance of ethylenediamine.According to the Bader charge analysis,the charge transfer from antimonene to Pt occurs,confirming the electronic interaction between Pt and Sb.Interestingly,antimonene,as a cocatalyst,alters the oxidation pathway for FAOR over Pt catalyst and makes FAOR follow the more efficient dehydrogenation pathway.The density functional theory(DFT)calculation demonstrates that antimonene can activate Pt to be a lower oxidative state and facilitate the oxidation of HCOOH into CO_(2) via a direct pathway,resulting in a weakened intermediate binding strength and better CO tolerance for FAOR.The specific activity of FAOR on Pt/Sb is 4.5 times,and the mass activity is 2.6 times higher than the conventional Pt/C. | Yiqiong Zhang Man Qiao Yucheng Huang Yuqin Zou Zhijuan Liu Li Tao Yafei Li Chung-Li Dong Shuangyin Wang | 2020 | Research2020,,1: | 3 |
| 10 | Metabolic regulation of T cell development by Sinl-mTORC2 is mediated by pyruvate kinase M2显示文摘Glucose metabolism plays a key role in thymocyte development. The mammalian target of rapamycin complex 2 (mT0RC2) is a critical regulator of cell growth and metabolism, but its role in early thymocyte development and metabolism has not been fully studied. We show here that genetic ablation of Sinl, an essential component of mTORC2, in T lineage cells results in severely impaired thymocyte development at the CD4^- CD8^- double negative (DN) stages but not at the CD4^+ CD8^+ double positive (DP) or later stages. Notably, Sinl-deficient DN thymocytes show markedly reduced proliferation and glycolysis.Importantly, we discover that the M2 isoform of pyruvate kinase (PKM2) is a novel and crucial Sinl effector in promoting DN thymocyte development and metabolism. At the molecular level, we show that Sinl-mTORC2 controls PKM2 expression through an AKT-dependent PPAR-y nuclear translocation. | 11 | Cholesterol homeostasis regulated by ABCA1 is critical for retinal ganglion cell survival显示文摘Genome-wide association studies have suggested a link between primary open-angle glaucoma and the function of ABCA1.ABCA1 is a key regulator of cholesterol efflux and the biogenesis of high-density lipoprotein(HDL) particles. Here, we showed that the POAG risk allele near ABCA1 attenuated ABCA1 expression in cultured cells. Consistently, POAG patients exhibited lower ABCA1 expression, reduced HDL, and higher cholesterol in white blood cells. Ablation of Abca1 in mice failed to form HDL, leading to elevated cholesterol levels in the retina. Counting retinal ganglion cells(RGCs) by using an artificial intelligence(AI) program revealed that Abca1-deficient mice progressively lost RGCs with age. Single-cell RNA sequencing(scRNA-seq) revealed aberrant oxidative phosphorylation in the Abca1-/-retina, as well as activation of the mTORC1 signaling pathway and suppression of autophagy. Treatment of Abca1-/-mice using atorvastatin reduced the cholesterol level in the retina,thereby improving metabolism and protecting RGCs from death. Collectively, we show that lower ABCA1 expression and lower HDL are risk factors for POAG. Accumulated cholesterol in the Abca1-/-retina causes profound aberrant metabolism, leading to a POAG-like phenotype that can be prevented by atorvastatin. | 12 | Allogeneic stem cell transplantation in the treatment of acute myeloid leukemia: An overview of obstacles and opportunities显示文摘As an important treatment for acute myeloid leukemia, allogeneic hematopoietic stem cell transplantation(allo-HSCT) plays an important role in reducing relapse and improving long-term survival. With rapid advancements in basic research in molecular biology and immunology and with deepening understanding of the biological characteristics of hematopoietic stem cells, allo-HSCT has been widely applied in clinical practice. During allo-HSCT, preconditioning, the donor, and the source of stem cells can be tailored to the patient’s conditions, greatly broadening the indications for HSCT, with clear survival benefits. However, the risks associated with allo-HSCT remain high, i.e. hematopoietic reconstitution failure, delayed immune reconstitution, graft-versus-host disease, and posttransplant relapse, which are bottlenecks for further improvements in allo-HSCT efficacy and have become hot topics in the field of HSCT. Other bottlenecks recognized in the current treatment of individuals diagnosed with acute myeloid leukemia and subjected to allo-HSCT include the selection of the most appropriate conditioning regimen and post-transplantation management. | 18 | Efficacy and adverse effects of olanzapine in the treatment of moderate to severe refractory neuropathic pain显示文摘Objective The aim of the study was to investigate the efficacy and adverse effects of olanzapine in the treatment of moderate to severe refractory neuropathic pain.Methods Forty patients with digestive system cancer were enrolled,who had moderate to severe refractory neuropathic pain;the patients were treated with olanzapine for 2 weeks at a daily dosage of 5 mg to 10 mg per night according to patients’response and tolerability,combined with conventional analgesic therapy.Pain intensity was evaluated by using a Numeral Rating Scale(NRS)at baseline,3 days,and 2 weeks after therapy.The Pittsburg Sleep Quality Index(PSQI)was evaluated at baseline and 2 weeks after therapy.Data on adverse events were recorded.The dosage of conventional analgesics was adjusted over time based on the severity of pain.Results The mean pain score decreased by 2.575±1.318(P<0.000)at 3 days and by 3.400±1.614(P<0.000)at 2 weeks;30%of the patients experienced significant pain relief at 3 days and 50%at 2 weeks.The PSQI decreased by 4.725±2.828(P<0.000)at 2 weeks.The adverse events induced by olanzapine included sleepiness,weight gain,dizziness,fatigue,dry mouth,and constipation;all the side effects were mild.Conclusion When combined with conventional analgesic therapy,olanzapine was effective in relieving pain and sleep disturbance,and was well-tolerated among patients with refractory neuropathic pain. | Yuhong Dai Man Zou Tingting Huang Hong Qiu | 2020 | Oncology and Translational Medicine2020,6,2: | 0 |
| 19 | Dose escalation guided by^(18)F-FDG PET/CT for esophageal cancer显示文摘Objective:To assess the feasibility of dose escalation guided by ^(18)F-deoxyglucose positron emission tomography/computer tomography(^(18)F-FDG PET/CT)for esophageal cancer(EC).Methods and materials:Ten random patients treated with definitive chemoradiotherapy and pre-therapeutic ^(18)FFDG PET/CT were included in this study.Retrospectively,a threshold of 50%of SUVmax was used to define the high FDG uptake region of the GTV(GTVPET).Three intensity-modulated radiation therapy(IMRT)plans were generated,delivering three dose levels to three different planning target volumes(PTVs).50.4 Gy delivered to PTV50.4 was defined on computed tomography(CT)as Plan50.4,63 Gy was delivered to PTV63 was defined as GTV plus a uniform margin of 0.5 cm as Plan63,and 70 Gy delivered to PTV70 was defined as GTVPET plus a 0.5 cm margin as Plan70.A dosimetric comparison was performed based on normal tissue complication probability(NTCP)for the lung and heart.Results:Clinically acceptable dose escalation failed for 2 of 10 patients in Plan63 due to heart dose constraints.One patient failed heart dose constraint for Plan70.Two patients failed spinal cord constraint for Plan63.Three patients failed lung dose constraints for both Plan63 and Plan70,two of which were even not suitable for Plan50.4 for the same reason.NTCP modeling for lung showed increased risk for Plan63 and Plan70 compared to Plan50.4.The difference between Plan63 and Plan70 was insignificant.NTCP modeling for heart showed an increased risk from 6.38%of Plan50.4 to 8.88%of Plan70(P=0.009)or 9.79%of Plan63(P=0.007).The risk of heart mortality was significantly higher for Plan63 than Plan70(P=0.047).Conclusions:Selective boosting of sub-volumes based on ^(18)F-FDG PET/CT is feasible way with a modest increase in the risk of cardiac and lung toxicities. | Bingjie Fan Chengqiang Li Fengchun Mu Wenru Qin Linlin Wang Xindong Sun Chunni Wang Bing Zou Shijiang Wang Wanlong Li Man Hu | 2021 | Radiation Medicine and Protection2021,2,2: | 0 |
