| 4 | Lung epithelial cell-derived IL-25 negatively regulates LPS-induced exosome release from macrophages显示文摘Background: Acute lung injury(ALI) is a major component of multiple organ dysfunction syndrome(MODS) following pulmonary and systemic infection. Alveolar macrophages(AMφ) are at the center of ALI pathogenesis. Emerging evidence has shown that cell-cell interactions in the lungs play an important regulatory role in the development of acute lung inflammation. However, the underneath mechanisms remain poorly addressed. In this study, we explore a novel function of lung epithelial cells(LEPCs) in regulating the release of exosomes from AMφ following LPS stimulation.Methods: For the in vivo experiments, C57 BL/6 wildtype(WT) mice were treated with lipopolysaccharide(LPS)(2 mg/kg) in 0.2 ml of saline via intratracheal aerosol administration. Bronchoalveolar lavage fluid was collected at 0–24 h after LPS treatment, and exosomes derived from AMφ were measured. For the in vitro studies, LEPCs and bone marrowderived Mφ(BMDM) were isolated from WT or TLR4-/-mice and were then cocultured in the Transwell? system. After coculture for 0–24 h, the BMDM and supernatant were harvested for the measurement of exosomes and cytokines.Results: We demonstrate that LPS induces macrophages(Mφ) to release exosomes, which are then internalized by neighboring Mφ to promote TNF-α expression. The secreted interleukin(IL)-25 from LEPCs downregulates Rab27 a and Rab27 b expression in Mφ, resulting in suppressed exosome release and thereby attenuating exosome-induced TNF-α expression and secretion.Conclusion: These findings reveal a previously unidentified crosstalk pathway between LEPCs and Mφ that negatively regulates the inflammatory responses of Mφ to LPS. | 6 | Dark,Beyond Deep:A Paradigm Shift to Cognitive AI with Humanlike Common Sense显示文摘Recent progress in deep learning is essentially based on a“big data for small tasks”paradigm,under which massive amounts of data are used to train a classifier for a single narrow task.In this paper,we call for a shift that flips this paradigm upside down.Specifically,we propose a“small data for big tasks”paradigm,wherein a single artificial intelligence(AI)system is challenged to develop“common sense,”enabling it to solve a wide range of tasks with little training data.We illustrate the potential power of this new paradigm by reviewing models of common sense that synthesize recent breakthroughs in both machine and human vision.We identify functionality,physics,intent,causality,and utility(FPICU)as the five core domains of cognitive AI with humanlike common sense.When taken as a unified concept,FPICU is concerned with the questions of“why”and“how,”beyond the dominant“what”and“where”framework for understanding vision.They are invisible in terms of pixels but nevertheless drive the creation,maintenance,and development of visual scenes.We therefore coin them the“dark matter”of vision.Just as our universe cannot be understood by merely studying observable matter,we argue that vision cannot be understood without studying FPICU.We demonstrate the power of this perspective to develop cognitive AI systems with humanlike common sense by showing how to observe and apply FPICU with little training data to solve a wide range of challenging tasks,including tool use,planning,utility inference,and social learning.In summary,we argue that the next generation of AI must embrace“dark”humanlike common sense for solving novel tasks. | Yixin Zhu Tao Gao Lifeng Fan Siyuan Huang Mark Edmonds Hangxin Liu Feng Gao Chi Zhang Siyuan Qi Ying Nian Wu Joshua B.Tenenbaum Song-Chun Zhu | 2020 | Engineering2020,6,3: | 3 |
| 7 | Including predator presence in a refined model for assessing resistance of alfalfa cultivar to aphids显示文摘The aphid quantity ratio(AQR) is defined as the number of aphids on each cultivar divided by the number of aphids on all cultivars. AQR is based on the correlation between aphid populations and their host plants and is an important tool that has been utilized in evaluating Medicago sativa(alfalfa) cultivar resistance to aphids. However, assessment of alfalfa resistance to aphids can be confused by the presence of aphid predators, causing the assessment of plant resistance to aphids to be based on incorrect aphid population data. To refine the AQR and account for the effect of predators on aphid population assessments, we introduced a parameter ‘α', corresponding to the predator quantity ratio, and used αAQR as the ratio to quantify aphid populations. Populations of both aphids(4 species) and their predators(12 species) occurring in 28 M. sativa cultivars were sampled over two years at a research station near Cangzhou, Hebei Province, China. Results showed that the most suitable evaluation period was from May to June, as the aphid population was stable during this period. Compared with the AQR method, the predator population numbers based on the αAQR had a significant inverse relationship with aphid population numbers and the 28 cultivars were clustered into three classes: the resistant class, tolerant class, and susceptible class. In addition, 17 cultivars were reassigned when evaluated using αAQR. All numerical values calculated by αAQR were displayed as a Gaussian distribution, which showed that the 28 cultivars could be clustered into nine groups using a median value(±SE) of 1±0.1. | 10 | Cambro-Silurian magmatisms at the northern Gondwana margin(Penninic basement of the Ligurian Alps)显示文摘The Early Paleozoic evolution of the northern margin of Gondwana is characterized by several episodes of bimodal magmatism intruded or outpoured within thick sedimentary basins. These processes are well recorded in the Variscan blocks incorporated in the Ligurian Alps because they experienced low temperature Alpine metamorphism. During the Paleozoic, these blocks, together with the other Alpine basements, were placed between the Corsica-Sardinia and the Bohemian Massif along the northern margin of Gondwana. In this framework, they host several a variegated lithostratigraphy forming two main complexes(Complexs I and II) that can be distinguished by both the protoliths and their crosscutting relationships, which indicate that the acidic and mafic intrusives of Complex II cut an already folded sequence made of sediments, basalts and granitoids of Complex I. Both complexes were involved in the Variscan orogenic phases as highlighted by the pervasive eclogite-amphibolite facies schistosity(foliation II). However, rare relicts of a metamorphic foliation at amphibolite facies conditions(foliation I)is locally preserved only in the rocks of Complex I. It is debatable if this schistosity was produced during the early folding event e occurred between the emplacement of Complex I and II e rather than during an early stage of the Variscan metamorphic cycle.New SHRIMP and LA ICP-MS Ue Pb zircon dating integrated with literature data, provide emplacement ages of the several volcanic or intrusive bodies of both complexes. The igneous activity of Complex I is dated between 507 ± 15 Ma and 494 ± 5 Ma, while Complex II between 467 ± 12 Ma and 445.5 ± 12 Ma.The folding event recorded only by the Complex I should therefore have occurred between 494 ± 5 Ma and 467 ± 12 Ma. The Variscan eclogite-amphibolite facies metamorphism is instead constrained between ~420 Ma and ~300 Ma. These ages and the geochemical signature of these rocks allow constraining the Early Paleozoic tectono-magmatic evolution of the Ligurian blocks, from a middleeupper Cambrian rifting stage, through the formation of an Early Ordovician volcanic arc during the Rheic Ocean subduction, until a Late Ordovician extension related to the arc collapse and subsequent rifting of the PaleoThetys.
