| 1 | Establishing a human pancreatic stem cell line and transplanting induced pancreatic islets to reverse experimental diabetes in rats显示文摘The major obstacle in using pancreatic islet transplantation to cure type I and some type II diabetes is the shortage of the donors. One of ways to overcome such obstacle is to isolate and clone pancreatic stem cells as 'seed cells' and induce their differentiation into functional islets as an abundant trans-plantation source. In this study, a monoclonal human pancreatic stem cell (mhPSC) line was obtained from abortive fetal pancreatic tissues. Pancreatic tissues were taken from abortive fetus by sterile procedures, and digested into single cells and cell clusters with 0.1% type IV collagenase. Cultured in modified glucose-low DMEM with 10% fetal bovine serum (FBS), these single cells and cell clusters adhered to culture dishes, and then primary epidermal-like pancreatic stem cells started to clone. After digesting with 0.25% trypsin and 0.04% EDTA, fibroblasts and other cells were gradually eliminated and epithelioid pancreatic stem cells were gradually purified during generations. Using clone-ring selection, the mhPSCs were obtained. After addition of 10 ng/mL epidermal growth factor (EGF) in cell culture medium, the mhPSCs quickly grew and formed a gravelstone-like monolayer. Continuously proliferated, a mhPSC line, which was derived from a male abortive fetus of 4 months old, has been passed through 50 generations. More than 1×109 mhPSCs were cryo-preserved in liquid nitrogen. Karyotype analysis showed that the chromosome set of the mhPSC line was normal diploid. Immunocytochemistry results demonstrated that the mhPSC line was positive for the pdx1, glucagon, nestin and CK19, and negative for the insulin, CD34, CD44 and CD45 protein expression. RT-PCR revealed further that the mhPSCs expressed transcription factors of the pdx1, glucagon, nestin and CK19. Also, in vitro induced with β-mercaptoethanol, the mhPSCs differentiated into nerve cells that expressed the NF protein. Induced with nicotinamide, the mhPSCs differentiated into functional islet-like clusters, as identified by di-thizone staining, which expressed the transcription factor of the insulin and secreted the insulin and C-peptide. | 3 | Strategies for improving the lithium-storage performance of 2D nanomaterials显示文摘2D nanomaterials, including graphene, transition metal oxide(TMO) nanosheets, transition metal dichalcogenide(TMD) nanosheets, etc., have offered an appealing and unprecedented opportunity for the development of high-performance electrode materials for lithium-ion batteries(LIBs). Although significant progress has been made on 2D nanomaterials for LIB applications in the recent years, some major challenges still exist for the direct use of these sheet-like nanomaterials, such as their serious self-agglomerating tendency during electrode fabrication and low conductivity as well as the large volume changes over repeated charging–discharging cycles for most TMOs/TMDs, which have resulted in large irreversible capacity, low initial Coulombic efficiency and fast capacity fading. To address these issues,considerable progress has been made in the exploitation of 2D nanosheets for enhanced lithium storage. In this review, we intend to summarize the recent progress on the strategies for enhancing the lithium-storage performance of 2D nanomaterials, including hybridization with conductive materials, surface/edge functionalization and structural optimization. | 4 | Ultrasonic diagnosis of patients with clonorchiasis and preliminary study of pathogenic mechanism显示文摘Objective:To discuss the liver function damage mechanism of patients with clonorchiasis by analyzing the ultrasound characteristics,liver function,change of the serum inflammatory factors and cell apoptosis factors.Methods:Color Doppler ultrasound technique was adopted to detect the portal vein and blood flow change of patients with clonorchiasis;ELISA was used to determine the level of different serum inflammatory factors.The levels of serum total bilirubin,serum albumin and glutamic-pyruvic transaminase were detected by automatic biochemical analyzer.Western blot was used to determine the expression of proteins relevant to apoptosis.Results:Compared with the health control group,the trunk diameter of portal vein and the thickness of spleen,as well as the hepatic artery pulsation index of clonorchiasis patients increased obviously,the mean blood flow velocity of portal vein(P<0.05 or P<0.01) decreased.The content of total bilirubin and transaminase in plasma increased significantly,but albumin decreased(P<0.05).Levels of TNF-α,IL-6 and IFN-γ increased remarkably,and the level of every factor was significantly different among patients with Child-Pugh Ⅰ,Child-Pugh Ⅱ and Child-Pugh Ⅲ classification of liver function(P<0.05 or P<0.01).With the exacerbation of liver dysfunction,levels of TNF-α,IL-6 and IFN-γ gradually increased(P<0.05).Compared with the healthy control group,the expression quantity of apoptosis protein Fas,FasL,Bax and Caspase-3 increased significantly(P<0.05 or P<0.01),but Bcl-2 decreased(P<0.05).Conclusions:Changes of ultrasonic characteristics and liver dysfunction,caused by liver fluke infection,may be related to that both inflammatory response and apoptosis response have participated in the pathogenic process and liver damage course of clonorchiasis. | Mei Fan Lin Lu Chun Su Mei Xue Ji-Mei Dou Pei Li Han-Qi Feng Yan-Bing Fan | 2016 | Asian Pacific Journal of Tropical Medicine2016,9,7: | 3 |

| 6 | Research on Isolation and Clone of Embryonic Stem Cell-Like in Bovine显示文摘Bovine embryonic stem cell would be invaluable for researching the aspect of animal cloning, production transgenic animal and discussion of gene function in vitro. With the object of establishing an effective culture system for isolation and clone of bovine pluripotent stem cell, we cultured bovine embryos and mouse embryos including morula blastula and hatached blastula and obtained animal ICM on Primary marine embryonic fibroblast (Primary murine embryonic fibroblast, PMEF) feeder layer with tissue medium(DMEM supplemented with 15ml/100ml NBS ,0.1μmol/L Na2SeO3, 0. 1mmol/L β-mercaptoethanol, 1 000ng/ml LIF,10 ng/ml IGF, 1mmol/L necessary amino acid and 1mmol/L L-glutamine), then, we obtained mouse ICM and bovine ICM. Moreover, we isolated and cloned the 6 passage bovine ES like cells(12 cell lines) and 9 passage marine ES like cells (52 cell lines) deriving from bovine ICM and murine ICM respectively on the feeder layer of PMEF by disaggregating ICM and ES cell clones of bovine and murine into smaller clumps through digesting with 0. 125g/100ml trypsin and 0.02g/100ml EDTA and scattering with a glass needle. The pluripotency of both murine and bovine ES like cells was identified with morphological character, histochemistry identification, karyotype analysis and differentiation of ES cells in vitro or in vivo. | 14 | Sialylation of TLR2 initiates osteoclast fusion显示文摘The molecular control of osteoclast formation is still not clearly elucidated. Here, we show that a process of cell recognition mediated by Siglec15-TLR2 binding is indispensable and occurs prior to cell fusion in RANKL-mediated osteoclastogenesis. Siglec15 has been shown to regulate osteoclastic bone resorption. However, the receptor for Siglec15 has not been identified, and the signaling mechanism involving Siglec15 in osteoclast function remains unclear. We found that Siglec15 bound sialylated TLR2 as its receptor and that the binding of sialylated TLR2 to Siglec15 in macrophages committed to the osteoclast-lineage initiated cell fusion for osteoclast formation, in which sialic acid was transferred by the sialyltransferase ST3 Gal1. Interestingly, the expression of Siglec15 in macrophages was activated by M-CSF, whereas ST3 Gal1 expression was induced by RANKL. Both Siglec15-specific deletion in macrophages and intrafemoral injection of sialidase abrogated cell recognition and reduced subsequent cell fusion for the formation of osteoclasts, resulting in increased bone formation in mice. | 18 | The Massive Expansion and Spatial Transformation of Potentially Contaminated Land Across China in 1990–2020 Observed from Remote Sensing and Big-data显示文摘Identifying and monitoring the spatiotemporal patterns of potentially contaminated land(PCL) in China is a key concern of ecological governance. However, the dynamics of PCL’s expansion remain unclear nationwide. Integrating high-resolution remote sensing images, a land-use/cover change database, crawler data from websites, and other multisource data, we produced a new dataset of China’s PCL in 1990, 2000, 2010, and 2020 using data fusion technology. Then we analyzed the spatiotemporal patterns of China’s PCL from 1990 to 2020. Our study shows that the acquired vector dataset of China’s PCL is of high quality and reliability, with an overall accuracy of 93.21%. The area of China’s PCL has kept growing for the past 30 years, and the growth rate was especially rapid during2000–2010, 2.32 and 6.13 times as rapid as that during 1990–2000 and 2010–2020, respectively. PCL has also been trending toward higher aggregation over markedly enlarged areas and has transferred progressively from north and southeast of China to northwest and southwest of China and Qinghai-Tibet Plateau. | 20 | Research on Growth Behavior of Embryos for Bovine and Murine on Primary Murine Embryos Fibroblast Cell Feeder Layer显示文摘The difference in growth behavior between bovine embryos and murine embryos was studied on PMEF(primary murine embryos fibroblast)feeder layer. The results showed as follows: With embryos having attached, bovine embryonic trophoblast formed a transparent membranous structure covering on inner cell mass (ICM), however, murine embryonic trophoblast formed disc structure. Bovine embryos formed four kinds of ICM colonies with different morphology including the mass-like, the net-like, the stream-like and the mixture-like colonies. Compared with Murine ICM, the bovine ICM grew more fast. So, the bovine ICM was passaged at first after a culture of approximately 5 - 6 days in vitro, but murine ICM was passaged at first after an attachment of 3 - 4 days on PMEF feeder layer. The mixture colonies of bovine ICM differentiated very early, while the others differentiated very late. Most ICM-like mass of Bovine grew in a defined spot, but bovine ICMs like stream and ICMs like net proliferated fast and dispersed quickly.
