1Prediction of blasting mean fragment size using support vector regression combined with five optimization algorithms显示文摘The main purpose of blasting operation is to produce desired and optimum mean size rock fragments.Smaller or fine fragments cause the loss of ore during loading and transportation,whereas large or coarser fragments need to be further processed,which enhances production cost.Therefore,accurate prediction of rock fragmentation is crucial in blasting operations.Mean fragment size(MFS) is a crucial index that measures the goodness of blasting designs.Over the past decades,various models have been proposed to evaluate and predict blasting fragmentation.Among these models,artificial intelligence(AI)-based models are becoming more popular due to their outstanding prediction results for multiinfluential factors.In this study,support vector regression(SVR) techniques are adopted as the basic prediction tools,and five types of optimization algorithms,i.e.grid search(GS),grey wolf optimization(GWO),particle swarm optimization(PSO),genetic algorithm(GA) and salp swarm algorithm(SSA),are implemented to improve the prediction performance and optimize the hyper-parameters.The prediction model involves 19 influential factors that constitute a comprehensive blasting MFS evaluation system based on AI techniques.Among all the models,the GWO-v-SVR-based model shows the best comprehensive performance in predicting MFS in blasting operation.Three types of mathematical indices,i.e.mean square error(MSE),coefficient of determination(R^(2)) and variance accounted for(VAF),are utilized for evaluating the performance of different prediction models.The R^(2),MSE and VAF values for the training set are 0.8355,0.00138 and 80.98,respectively,whereas 0.8353,0.00348 and 82.41,respectively for the testing set.Finally,sensitivity analysis is performed to understand the influence of input parameters on MFS.It shows that the most sensitive factor in blasting MFS is the uniaxial compressive strength.Enming Li Fenghao Yang Meiheng Ren Xiliang Zhang Jian Zhou Manoj Khandelwal 2021Journal of Rock Mechanics and Geotechnical Engineering2021,13,6:3
2Rational design of a visible-light photochromic diarylethene:a simple strategy by extending conjugation with electron donating groups显示文摘Photochromic diarylethenes have been widely used in many fields. However, their cyclization process must be induced by UV light. In this article, a simple strategy is developed by extending π-conjugation with electron donating groups. The modified dirylethene derivative can photocyclolize under 405-nm light with a good photochromic efficiency. Meanwhile, its absorption and moderate fluorescence can be switched effectively in both directions by visible lights(405 and 520 nm, respectively) in different solutions and in living cells. 12Drug discovery of sclerostin inhibitors显示文摘Sclerostin, a protein secreted from osteocytes, negatively regulates the WNT signaling pathway by binding to the LRP5/6 co-receptors and further inhibits bone formation and promotes bone resorption. Sclerostin contributes to musculoskeletal system-related diseases, making it a promising therapeutic target for the treatment of WNT-related bone diseases. Additionally, emerging evidence indicates that sclerostin contributes to the development of cancers, obesity, and diabetes, suggesting that it may be a promising therapeutic target for these diseases. Notably, cardiovascular diseases are related to the protective role of sclerostin. In this review, we summarize three distinct types of inhibitors targeting sclerostin, monoclonal antibodies, aptamers, and small-molecule inhibitors, from which monoclonal antibodies have been developed. As the first-in-class sclerostin inhibitor approved by the U.S. FDA,the monoclonal antibody romosozumab has demonstrated excellent effectiveness in the treatment of postmenopausal osteoporosis;however, it conferred high cardiovascular risk in clinical trials. Furthermore,romosozumab could only be administered by injection, which may cause compliance issues for patients who prefer oral therapy. 14Structural determinants specific for retromer protein sorting nexin 5 in regulating subcellular retrograde membrane trafficking显示文摘The endosomal trafficking of signaling membrane proteins, such as receptors, transporters and channels, is mediated by the retromer-mediated sorting machinery, composed of a cargo-selective vacuolar protein sorting trimer and a membrane-deforming subunit of sorting nexin proteins. Recent studies have shown that the isoforms, sorting nexin 5 (SNX5) and SNX6, have played distinctive regulatory roles in retrograde membrane trafficking. However, the molecular insight determined functional differences within the proteins remains unclear. We reported that SNX5 and SNX6 had distinct binding affinity to the cargo protein vesicular monoamine transporter 2 (VMAT2). SNX5, but not SNX6, specifically interacted with VMAT2 through the Phox domain, which contains an alpha-helix binding motif. Using chimeric mutagenesis, we identified that several key residues within this domain were unique in SNX5, but not SNX6, and played an auxiliary role in its binding to VMAT2. Importantly, we generated a set of mutant SNX6, in which the corresponding key residues were mutated to those in SNX5. In addition to the gain in binding affinity to VMAT2, their overexpression functionally rescued the altered retrograde trafficking of VMAT2 induced by siRNA-mediated depletion of SNX5. These data strongly suggest that SNX5 and SNX6 have different functions in retrograde membrane trafficking, which is determined by the different structural elements within the Phox domain of two proteins.
