1Combined peripheral natural killer cell and circulating tumor cell enumeration enhance prognostic efficiency in patients with metastatic triple-negative breast cancer显示文摘Objective:Triple-negative breast cancer(TNBC)is a heterogeneous disease with poor prognosis.Circulating tumor cells(CTCs)are a promising predictor for breast cancer prognoses but their reliability regarding progression-free survival(PFS)is controversial.We aim to verify their predictive value in TNBC.Methods:In present prospective cohort study,we used the Pep@MNPs method to enumerate CTCs in baseline blood samples from 75 patients with TNBC(taken at inclusion in this study)and analyzed correlations between CTC numbers and outcomes and other clinical parameters.Results:Median PFS was 6.0(range:1.0–25.0)months for the entire cohort,in whom we found no correlations between baseline CTC status and initial tumor stage(P=0.167),tumor grade(P=0.783)or histological type(P=0.084).However,among those getting first-line treatment,baseline CTC status was positively correlated with ratio of peripheral natural killer(NK)cells(P=0.032),presence of lung metastasis(P=0.034)and number of visceral metastatic site(P=0.037).Baseline CTC status was predictive for PFS in first-line TNBC(P=0.033),but not for the cohort as a whole(P=0.118).This prognostic limitation of CTC could be ameliorated by combining CTC and NK cell enumeration(P=0.049).Conclusions:Baseline CTC status was predictive of lung metastasis,peripheral NK cell ratio and PFS in TNBC patients undergoing first-line treatment.We have developed a combined CTC-NK enumeration strategy that allows us to predict PFS in TNBC without any preconditions.Xiaoran Liu Ran Ran Bin Shao Hope S.Rugo Yanlian Yang Zhiyuan Hu Zewen Wei Fengling Wan Weiyao Kong Guohong Song Hanfang Jiang Xu Liang Ruyan Zhang Ying Yan Guobing Xu Huiping Li 2018Chinese Journal of Cancer Research2018,30,3:4
2A turn-on fluorescent probe for Au^(3+) based on rodamine derivative and its bioimaging application显示文摘A new fluorescent probe RY was synthesized for the detection of Au3+ions based on a rhodamine B derivative.The fluorescent probe showed good selectivity and sensitivity to Au3+ions.Obvious color and fluorescence changes could be observed with the naked eye while the fluorescent probe reacted with the Au3+ions.The detection limit of the probe was determined to be 36 ppb by the fluorescence titration.The excellent linear relationship suggests that the probe is potentially useful for quantitative detection of Au3+in vitro.We also demonstrated its bioimaging application in both living cells and mice.This was the first time that a fluorescent probe was successfully applied to imaging Au3+in living animals.SONG FengLing NING HouFu SHE HaiYing WANG JingYun PENG XiaoJun 2014Science China Chemistry2014,57,7:3
3Thermally activated delayed fluorescence molecules and their new applications aside from OLEDs显示文摘Thermally activated delayed fluorescence(TADF) organic molecules feature with long-lived delayed fluorescence, because they can undergo not only efficient intersystem crossing(ISC), but also efficient reverse intersystem crossing(RISC) at room temperature. As a new type of luminescent molecules, they have exhibited successful applications in organic light emitting diodes(OLEDs). Aside from OLEDs, they are also found to have potential applications in time-resolved luminescence imaging based on long-lived fluorescence property. Meanwhile, due to their excited triplet characteristic originated from efficient ISC,they were found to be applied in triplet-triplet annihilation upconversion(TTA-UC), photodynamic therapy(PDT) and organic photocatalytic synthesis. 4Rational design of a visible-light photochromic diarylethene:a simple strategy by extending conjugation with electron donating groups显示文摘Photochromic diarylethenes have been widely used in many fields. However, their cyclization process must be induced by UV light. In this article, a simple strategy is developed by extending π-conjugation with electron donating groups. The modified dirylethene derivative can photocyclolize under 405-nm light with a good photochromic efficiency. Meanwhile, its absorption and moderate fluorescence can be switched effectively in both directions by visible lights(405 and 520 nm, respectively) in different solutions and in living cells. 6Self-assembly of amphiphilic peptides to construct activatable nanophotosensitizers for theranostic photodynamic therapy显示文摘A variety of nano-engineered photosensitizers have been developed for photodynamic therapy(PDT)of cancer diseases. However, traditional nano-engineering methods usually cannot avoid drug leakage and premature release, and have disadvantages such as low drug load and inaccurate release.The self-assembly strategy based on amphiphilic peptides has been considered to be more attractive nano-engineering method. Here we developed novel acid-activatable self-assembled nanophotosensitizers based on an amphiphilic peptide derivative. The peptide derivative was synthesized from a fluorescein molecule with thermally activated delayed fluorescence(TADF). The self-assembled nanophotosensitizers can specifically enter the tumor cells and disassemble inside lysosomes companied with “turn-on” fluorescence and photodynamic therapy effect. 8Fine-tailoring the linker of near-infrared fluorescence probes for nitroreductase imaging in hypoxic tumor cells显示文摘Imaging hypoxia using fluorescence probes for nitroreductase(NTR) have attracted much attention in last decade. At least three different linkers have been commonly used to connect the recognition unit and reporting unit in reported probes for NTR. Meanwhile, the linker is known to be a key factor for achieving best sensing performance. In this work, three near-infrared fluorescence probes CyNP-1, CyNP-2 and CyNP-3 were designed and synthesized from an aminocyanine dye CyNP. The three probes have the same recognition unit and same fluorescence reporting unit, but different linkers. CyNP-1 was found to have the best sensing performance for NTR with 40-fold of fluorescence enhancement. It is well investigated how the difference of the linkers brings out the different sensing performance by HPLC, MS and docking calculations.
