1Potentiating CD8^(+) T cell antitumor activity by inhibiting PCSK9 to promote LDLR-mediated TCR recycling and signaling显示文摘Metabolic regulation has been proven to play a critical role in T cell antitumor immunity.However,cholesterol metabolism as a key component of this regulation remains largely unexplored.Herein,we found that the low-density lipoprotein receptor(LDLR),which has been previously identified as a transporter for cholesterol,plays a pivotal role in regulating CD8+T cell antitumor activity.Besides the involvement of cholesterol uptake which is mediated by LDLR in T cell priming and clonal expansion,we also found a non-canonical function of LDLR in CD8+T cells:LDLR interacts with the T-cell receptor(TCR)complex and regulates TCR recycling and signaling,thus facilitating the effector function of cytotoxic T-lymphocytes(CTLs).Furthermore,we found that the tumor microenvironment(TME)downregulates CD8+T cell LDLR level and TCR signaling via tumor cell-derived proprotein convertase subtilisin/kexin type 9(PCSK9)which binds to LDLR and prevents the recycling of LDLR and TCR to the plasma membrane thus inhibits the effector function of CTLs.Moreover,genetic deletion or pharmacological inhibition of PCSK9 in tumor cells can enhance the antitumor activity of CD8+T cells by alleviating the suppressive effect on CD8+T cells and consequently inhibit tumor progression.While previously established as a hypercholesterolemia target,this study highlights PCSK9/LDLR as a potential target for cancer immunotherapy as well.Juanjuan Yuan Ting Cai Xiaojun Zheng Yangzi Ren Jingwen Qi Xiaofei Lu Huihui Chen Huizhen Lin Zijie Chen Mengnan Liu Shangwen He Qijun Chen Siyang Feng Yingjun Wu Zhenhai Zhang Yanqing Ding Wei Yang 2021Protein & Cell2021,12,4:12

8A novel humanized MUC1 antibody–drug conjugate for the treatment of trastuzumab-resistant breast cancer显示文摘Mucin 1(MUC1)has been regarded as an ideal target for cancer treatment,since it is overexpressed in a variety of different cancers including the majority of breast cancer.However,there are still no approved monoclonal antibody drugs targeting MUC1.In this study,we generated a humanized MUC1(HzMUC1)antibody from our previously developed MUC1 mouse monoclonal antibody that only recognizes MUC1 on the surface of tumor cells.Furthermore,an antibody–drug conjugate(ADC)was generated by conjugating HzMUC1 with monomethyl auristatin(MMAE),&the efficacy of HzMUC1-MMAE on the MUC1-positive HER2+breast cancer in vitro&in‘Xenograft’model was tested.Results from western blot analysis&immunoprecipitation revealed that the HzMUC1 antibody did not recognize cell-free MUC1-N in sera from breast cancer patients.Confocal microscopy analysis showed that HzMUC1 antibody bound to MUC1 on the surface of breast cancer cells.Results from mapping experiments suggested that HzMUC1 may recognize an epitope present in the interaction region between MUC1-N and MUC1-C.Results from colony formation assay&flow cytometry demonstrated that HzMUC1-MMAE significantly inhibited cell growth by inducing G2/M cell cycle arrest&apoptosis in trastuzumab-resistant HER2-positive breast cancer cells.Meanwhile,HzMUC1-MMAE significantly reduced the growth of HCC1954 xenograft tumors by inhibiting cell proliferation&enhancing cell death.In conclusion,our results indicate that HzMUC1-ADC is a novel therapeutic drug that can overcome trastuzumab resistance of breast cancer.HzMUC1-ADC should also be an effective therapeutic drug for the treatment of different MUC1-positive cancers in clinic.Guang Wu Lan Li Yuxin Qiu Wei Sun Tianhao Ren Yingshuai Lv Mengnan Liu Xiaoxia Wang Hongqun Tao Lingjie Zhao Jiawei Cao Licai He Hongzhi Li Haihua Gu 2021Acta Biochimica et Biophysica Sinica2021,53,12:2
9Reorganization of 3D genome architecture across wild boar and Bama pig adipose tissues显示文摘Background:A growing body of evidence has revealed that the mammalian genome is organized into hierarchical layers that are closely correlated with and may even be causally linked with variations in gene expression.Recent studies have characterized chromatin organization in various porcine tissues and cell types and compared them among species and during the early development of pigs.However,how chromatin organization differs among pig breeds is poorly understood.Results:In this study,we investigated the 3D genome organization and performed transcriptome characterization of two adipose depots(upper layer of backfat[ULB]and greater omentum[GOM])in wild boars and Bama pigs;the latter is a typical indigenous pig in China.We found that over 95%of the A/B compartments and topologically associating domains(TADs)are stable between wild boars and Bama pigs.In contrast,more than 70%of promoterenhancer interactions(PEIs)are dynamic and widespread,involving over a thousand genes.Alterations in chromatin structure are associated with changes in the expression of genes that are involved in widespread biological functions such as basic cellular functions,endocrine function,energy metabolism and the immune response.Approximately 95%and 97%of the genes associated with reorganized A/B compartments and PEIs in the two pig breeds differed between GOM and ULB,respectively.Conclusions:We reported 3D genome organization in adipose depots from different pig breeds.In a comparison of Bama pigs and wild boar,large-scale compartments and TADs were mostly conserved,while fine-scale PEIs were extensively reorganized.The chromatin architecture in these two pig breeds was reorganized in an adipose depotspecific manner.These results contribute to determining the regulatory mechanism of phenotypic differences between Bama pigs and wild boar.Jiaman Zhang Pengliang Liu Mengnan He Yujie Wang Hua Kui Long Jin Diyan Li Mingzhou Li 2022Journal of Animal Science and Biotechnology2022,13,4:2

11Numerical simulation study on shear resistance of anchorage joints considering tensile-shear fracture criterion of 2G-NPR bolt显示文摘2G-NPR bolt (the 2nd generation Negative Poisson’s Ratio bolt) is a new type of bolt with high strength, high toughness and no yield platform. It has signifcant efects on improving the shear strength of jointed rock mass and controlling the stability of surrounding rock. To achieve an accurate simulation of bolted joint shear tests, we have studied a numerical simulation method that takes into account the 2G-NPR bolt's tensile–shear fracture criterion. Firstly, the indoor experimental study on the tensile–shear mechanical properties of 2G-NPR bolt is carried out to explore its mechanical properties under diferent tensile–shear angles, and the fracture criterion of 2G-NPR bolt considering the tensile–shear angle is established. Then, a three-dimensional numerical simulation method considering the tensile–shear mechanical constitutive and fracture criterion of 2G-NPR bolt, the elastoplastic mechanical behavior of surrounding rock and the damage and deterioration of grouting body is proposed. The feasibility and accuracy of the method are verifed by comparing with the indoor shear test results of 2G-NPR bolt anchorage joints. Finally, based on the numerical simulation results, the deformation and stress of the bolt, the distribution of the plastic zone of the rock mass, the stress distribution and the damage of the grouting body are analyzed in detail.
