| 1 | An acorn squash (Cucurbita pepo ssp. ovifera) fruit and seed transcriptome as a resource for the study of fruit traits in Cucurbita显示文摘Acorn squash(Cucurbita pepo)is an iconic fall vegetable in the United States,known for its unique fruit shape and also prized for its culinary properties.Little is known about the metabolism that underlies the development of fruit quality attributes such as color,sweetness,texture and nutritional qualities in acorn squash,or any other winter squash grown worldwide.To provide insight into winter squash fruit and seed development and add to the genomic resources in the Cucurbita genus,RNA sequencing was used to generate an acorn squash fruit and seed transcriptome from the cultivar Sweet REBA at critical points throughout fruit development.141838600 high-quality paired-end Illumina reads were assembled into 55949 unigenes.85%of unigenes with predicted open reading frames had homology with previously identified genes and over 62%could be functionally annotated.Comparison with the watermelon and cucumber genomes provided confirmation that the unigenes are full-length and comprehensive,covering an average of 90%of the coding sequence of their homologs and 72%of the cucumber and watermelon exomes.Key candidate genes associated with carotenoid and carbohydrate metabolism were identified toward a resource for winter squash fruit quality trait dissection.This transcriptome represents a major advance in C.pepo genomics,providing significant new sequence information and revealing the repertoire of genes expressed throughout winter squash fruit and seed development.Future studies on the genetic basis of fruit quality and future breeding efforts will be enhanced by tools and insights developed from this resource. | Lindsay E Wyatt Susan R Strickler Lukas A Mueller Michael Mazourek | 2015 | Horticulture Research2015，2，1： | 11 |
| 2 | Comparative analysis of Cucurbita pepo metabolism throughout fruit development in acorn squash and oilseed pumpkin显示文摘Both the fruit mesocarp and the seeds of winter squash can be used for consumption,although the focus of breeding efforts varies by cultivar.Cultivars bred for fruit consumption are selected for fruit mesocarp quality traits such as carotenoid content,percent dry matter,and percent soluble solids,while these traits are essentially ignored in oilseed pumpkins.To compare fruit development in these two types of squash,we sequenced the fruit transcriptome of two cultivars bred for different purposes:an acorn squash,‘Sweet REBA’,and an oilseed pumpkin,‘Lady Godiva’.Putative metabolic pathways were developed for carotenoid,starch,and sucrose synthesis in winter squash fruit and squash homologs were identified for each of the structural genes in the pathways.Gene expression,especially of known rate-limiting and branch point genes,corresponded with metabolite accumulation both across development and between the two cultivars.Thus,developmental regulation of metabolite genes is an important factor in winter squash fruit quality. | Lindsay E Wyatt Susan R Strickler Lukas A Mueller Michael Mazourek | 2016 | Horticulture Research2016，3，1： | 7 |
| 3 | Acute pancreatitis in aging animals:Loss of pancreatitis-associated protein protection?显示文摘AIM:To investigate the effect of age on severity of acute pancreatitis(AP) using biochemical markers,histology and expression of the protective pancreatitisassociated proteins(PAPs).METHODS:AP was induced via intraductal injection of 4% sodium taurocholate in young and old rats.Sera and pancreata were assayed at 24 h for the parameters listed above;we also employed a novel molecular technique to assess bacterial infiltration using polymerase chain reaction to measure bacterial genomic ribosomal RNA.RESULTS:At 24 h after induction of AP,the pancreata of older animals had less edema(mean ± SE histologic score of young vs old:3.11 ± 0.16 vs 2.50 ±-0.11,P < 0.05),decreased local inflammatory response(histologic score of stromal infiltrate:3.11 ± 0.27 vs 2.00 ± 0.17,P < 0.05) and increased bacterial infiltration(174% ± 52% increase from sham vs 377% ± 4%,P < 0.05).A decreased expression of PAP1 and PAP2 was demonstrated by Western blotting analysis and immunohistochemical staining.There were no differences in serum amylase and lipase activity,or tissue myeloperoxidase or monocyte chemotactic protein-1 levels.However,in the most-aged group,serum C-reactive protein levels were higher(young vs old:0.249 ± 0.04 mg/dL vs 2.45 ± 0.68 mg/dL,P < 0.05).CONCLUSION:In older animals,there is depressed PAP expression related to a blunted inflammatory response in AP which is associated with worsened bacterial infiltration and higher C-reactive protein level;this may explain the more aggressive clinical course. | Sophia Fu Albert Stanek Cathy M Mueller Nefertti A Brown Chongmin Huan Martin H Bluth Michael E Zenilman | 2012 | World Journal of Gastroenterology2012，18，26： | 5 |
| 4 | 子宫内膜异位症患者自然周期卵泡液中细胞因子研究显示文摘目的观察子宫内膜异位症(EMs)患者自然周期卵泡液中细胞因子的表达情况,探讨其与EMs的相关性。方法收集2013~2016年间在瑞士伯尔尼大学妇女医院生殖中心因EMs行自然周期IVF-ET或卵胞浆内单精子注射(ICSI)助孕治疗的17例患者为研究对象(EMs组),其中轻度EMs患者7例,中重度EMs患者10例。同时收集同期17例年龄匹配,因输卵管因素或男方因素在该中心行自然周期IVF/ICSI-ET助孕的患者做为对照组。在自然周期采卵日收集卵泡液,通过Bio-Plex技术检测卵泡液中白介素(IL)-1β、IL-6、IL-8、IL-15、IL-18、调节活化正常T细胞表达与分泌的趋化因子(RANTES)、肿瘤坏死因子(TNF)-α、血管内皮生长因子(VEGF)、白血病抑制因子(LIF)等9种细胞因子的浓度;ELISA法检测单核细胞趋化因子-1(MCP-1)和瘦素(Leptin)的浓度,分析两组间细胞因子浓度的差异及其与EMs的相关性。结果IL-1β和IL-6在EMs患者卵泡液中的浓度显著高于对照组(中位数比较分别为0.8pg/mlvs.0pg/ml和9.1pg/mlvs.6.1pg/ml,P均<0.05)。中重度EMs患者卵泡液中IL-1β浓度(1.44pg/ml)显著高于轻度EMs及对照组(分别为0.26pg/ml、0pg/ml),VEGF浓度(2464pg/ml)亦显著高于轻度EMs及对照组(分别为1268pg/ml、1560pg/ml,P均<0.05)。通过Spearman相关分析发现卵泡液中只有IL-1β和IL-6的浓度与EMs相关,大部分细胞因子相互之间存在较强的相关性。结论EMs患者自然周期卵泡液中细胞因子微环境发生了变化,IL-1β和IL-6的浓度均异常增高,且大部分细胞因子相互之间存在较强的相关性,可能与EMs患者生育力降低有关。 | 吴庚香 Nick Bersinger Michael Mueller Michael von Wolff | 2019 | 生殖医学杂志2019，28，9： | 4 |
