1The roles of surfactant protein D during Aspergillus fumigatus infection in human corneal epithelial cells显示文摘AIM: To investigate roles of surfactant protein D (SP-D) and relative cytokines in human corneal epithelial(HCE) cells exposed to aspergillus fumigatus (AF) antigens.METHODS: HCE cells cultured in vitro with AF antigens and sampled at 0, 0.5, 1 hour, 2, 4, 6 and 8 hours. The expression of SP-D mRNA was evaluated by semiquantitative reverse transcription-polymerase chain reaction (RT-PCR).The expression of SP-D protein was shown by ELISA and immunocytochemistry SP methods. The expression of NF-κB and relative downstream cytokines such as TNF-α, IL-1β, IL-8 and IL-10 in supernatant fluid were measured by ELISA.RESULTS: SP-D mRNA and protein were detected in untreated HCE cells. The expression of SP-D and the relative downstream cytokines rose after being stimulated with AF antigens. SP-D mRNA began to rise at 0.5 hour and the most significantly peak was in 2 hours. The protein of SP-D in supernatant fluid had the same trend with mRNA. Immunocytochemistry of SP-D showed positive expression and gradually increased to 6 hours, and then the expression began to decline. NF-κB was activated after treated by AF antigens and the changes had correlation with SP-D. TNF-α, IL-1β, IL-8 and IL-10 began to rise after given AF antigens 1 hour and were 1.82, 1.43, 1.12 and 1.28 times higher than the untreated HCE cells separately. The expression of TNF-α and IL-1β reached the peak at 2 hours, separately 2.80 and 2.86 times than the untreated. The expression of IL-8 and IL-10 gradually increased with a time-dependent manner.CONCLUSION: HCE cells exists SP-D and it may play a significant role in pathogenesis of keratomycosis. AF may induce human corneal epithelial cells to express inflammatory cytokines via SP-D and NF-κB pathway. 2Mapping the permafrost stability on the Tibetan Plateau for 2005–2015显示文摘Data scarcity is a major obstacle for high-resolution mapping of permafrost on the Tibetan Plateau(TP).This study produces a new permafrost stability distribution map for the 2010 s(2005–2015)derived from the predicted mean annual ground temperature(MAGT)at a depth of zero annual amplitude(10–25 m)by integrating remotely sensed freezing degree-days and thawing degree-days,snow cover days,leaf area index,soil bulk density,high-accuracy soil moisture data,and in situ MAGT measurements from 237 boreholes on the TP by using an ensemble learning method that employs a support vector regression model based on distance-blocked resampled training data with 200 repetitions.Validation of the new permafrost map indicates that it is probably the most accurate of all currently available maps.This map shows that the total area of permafrost on the TP,excluding glaciers and lakes,is approximately 115.02(105.47–129.59)×10^4 km^2.The areas corresponding to the very stable,stable,semi-stable,transitional,and unstable types are 0.86×10^4,9.62×10^4,38.45×10^4,42.29×10^4,and 23.80×10^4 km^2,respectively.This new map is of fundamental importance for engineering planning and design,ecosystem management,and evaluation of the permafrost change in the future on the TP as a baseline.Youhua RAN Xin LI Guodong CHENG Zhuotong NAN Jinxing CHE Yu SHENG Qingbai WU Huijun JIN Dongliang LUO Zhiguang TANG Xiaobo WU 2021Science China Earth Sciences2021,64,1:11

4Comparison of Histological, Microbiological, and Molecular Methods in Diagnosis of Patients with TBLN Having Different Anti-TB Treatment Background显示文摘Objective The influence of anti-tuberculosis(TB) treatment history on tuberculous lymphadenitis(TBLN) diagnosis is unclear. Therefore, this study aims to evaluate the diagnostic methods, including histology, microbiology, and molecular tests, used for TBLN. Methods In this study, suspected patients with TBLN and having different anti-TB treatment background were enrolled. All the samples were tested simultaneously by histology, Ziehl-Neelsen(ZN) staining, mycobacterial culture(culture), Xpert MTB/RIF(xpert), real-time PCR, and high-resolution melting curve PCR(HRM). Thereafter, the performance of these methods on samples with different anti-TB treatment background was assessed. Results In our study, 89 patients were prospectively included 82 patients with TBLN and 7 with other diseases. The overall sensitivities of Xpert, real-time PCR, histology, ZN staining, and culture were 86.6%, 69.5%, 58.5%, 43.9%, and 22.0%, respectively. The anti-TB treatment history revealed dramatic influences on the sensitivity of culture(P < 0.0001). In fact, the treatment that lasted over 3 months also influenced the sensitivity of Xpert(P < 0.05). However, the treatment history did not affect the performance of remaining tests(P > 0.05). For rifampicin drug susceptibility test(DST), the anti-TB treatment showed only significant influence on the success rate of culture DST(P = 0.001), but not on those of Xpert and HRM tests(P > 0.05). 5Effect of corneal graft diameter on therapeutic penetrating keratoplasty for fungal keratitis显示文摘AIM: To evaluate the effect of corneal graft diameter on therapeutic penetrating keratoplasty(PKP) for fungal keratitis. METHODS: A total of 116 patients (116 eyes) suffered from fungal keratitis underwent PKP at the Affiliated Hospital of Medical College Qingdao University from May 2006 to May 2010. They were divided into two groups according to the corneal graft diameter. 64 eyes' corneal graft diameter was 8.00mm or larger and 52 eyes' graft diameter was smaller than 8.00mm. The follow-up time was 2 years. The postoperative visual acuity and complications were documented and compared. RESULTS: Sixty-two (96.88%) eyes and fifty (96.15%) eyes preserved eyeballs respectively in two groups. There was no statistical difference in postoperative visual acuity (P =0.961), corneal graft clear rate (P =0.132) or the incidence of recurred fungal infection (P =0.770) between two groups. But there was a higher incidence of graft rejection (P =0.020) and secondary glaucoma (P =0.039) in group with corneal graft diameter 8.00mm or larger. CONCLUSION: PKP is an effective treatment approach for fungal keratitis. 6Microwave ablation of primary breast cancer inhibits metastatic progression in model mice via activation of natural killer cells显示文摘Surgery is essential for controlling the symptoms and complications of stage IV breast cancer.However,locoregional treatment of primary tumors often results in distant progression,including lung metastasis,the most common type of visceral metastasis.As a minimally invasive thermal therapy,microwave ablation(MWA)has been attempted in the treatment of breast cancer,but the innate immune response after MWA has not yet been reported.Using two murine models of stage IV breast cancer,we found that MWA of primary breast cancer inhibited the progression of lung metastasis and improved survival.NK cells were activated after MWA of the primary tumor and exhibited enhanced cytotoxic functions,and the cytotoxic pathways of NK cells were activated.Depletion experiments showed that NK cells but not CD4+or CD8+T cells played a pivotal role in prolonging survival.Then,we found that compared with surgery or control treatment,MWA of the primary tumor induced completely different NK-cell-related cytokine profiles.Macrophages were activated after MWA of the primary tumor and produced IL-15 that activated NK cells to inhibit the progression of metastasis.In addition,MWA of human breast cancer stimulated an autologous NK-cell response.These results demonstrate that MWA of the primary tumor in metastatic breast cancer inhibits metastatic progression via the macrophage/IL-15/NK-cell axis.MWA of the primary tumor may be a promising treatment strategy for de novo stage IV breast cancer,although further substantiation is essential for clinical testing.Muxin Yu Hong Pan Nan Che Li Li Cong Wang Yue Wang Ge Ma Mengjia Qian Jiawei Liu Mingjie Zheng Hui Xie Lijun Ling Yi Zhao Xiaoxiang Guan Qiang Ding Wenbin Zhou Shui Wang 2021Cellular & Molecular Immunology2021,18,9:5
7Expression of S100B during the innate immune of corneal epithelium against fungi invasion显示文摘AIM: To explore the expression of S100 B in corneal epithelial cells under Aspergillus stimulation both in vivo and in vitro.·METHODS: Immortalized human corneal epithelial cells(HCECs) were exposed to inactive Aspergillus fumigatus(A. fumigatus) conidia at 0, 4, 8, 12, 16, and 24 h respectively. The corneas of Wistar rats were exposed to active A. fumigatus at 0, 12, 24, 48 h and the normal rat corneas were used for normal control. The m RNA level of S100 B was evaluated by real time quantitative reverse transcription-polymerase chain reaction(q RT-PCR).S100 B protein expression in cornea epithelium was detected by immunohistochemical/immunocytochemical staining(IHC/ICC).· RESULTS: Histopathology revealed a significant inflammatory cell infiltration in fungal keratitis human and rat cornea. Corneal epithelial cells didn't express or rarely express S100 B at baseline. A. fumigatus significantly induced S100 B m RNA expression in cultured corneal epithelial cells in a time depended manner in vitro, the m RNA began to rise significantly at8 h in vitro(P <0.05) and continue to rise as time prolonged(P <0.01). In vivo, S100 B m RNA level was low in the normal corneas. However, it was increased in keratitis corneas from 12 h after infection(P <0.05) and reached to a peak at 24h(P <0.001). 9Early expression of surfactant proteins D in Fusarium solani infected rat cornea显示文摘AIM:To investigate the early expression of surfactant proteins D(SP-D) in Fusarium solani infected rat cornea.· METHODS:Wistar rats were divided into group A,B and C randomly.The right eyes were chosen as the experiment one.Group A was control group.Group B was not inoculated with Fusarium solani.Group C was taken as fusarium solani keratitis model.Five rats in group B and C were executed randomly at 6,12,24,48 and 96 hours respectively after the experimental model being established.The expression of SP-D was assessed through immunohistochemistry and reverse transcription polymerase chain reaction(RT-PCR).· RESULTS:RT-PCR detected that the SP-D mRNA expression was low in the corneal of normal rats and group B.The expression of fungal infected cornea increased gradually and reached the peak at 24 hours in group C.The synchronous expression of group B and C were in significant difference(P <0.01).Immunohistochemisty discovered the protein of SP-D expression was increased gradually from 12 hours and reached the peak at 48 hours in group C.The synchronous expression of group B and C were also in significant difference(P <0.01).· CONCLUSION:There exists SP-D in rat corneal tissue and the expression is significantly increased at the early period of fusarium solani infected cornea.SP-D may play a role in the early innate immunity response of the corneal resistance to Fusarium solani infection.Cheng-Ye Che, Gui-Qiu Zhao 2012International Journal of Ophthalmology(English edition)2012,5,3:4
10Expression of indoleamine 2,3-dioxygenase in a murine model of Aspergillus fumigatus keratitis显示文摘AIM: To observe the presence and expression of indoleamine 2,3-dioxygenase(IDO) during the corneal immunity to Aspergillus fumigatus(A. fumigatus) in the murine models.·METHODS: The murine model of fungal keratitis was established by smearing with colonies of A. fumigatus after scraping central epithelium of cornea and covering with contact lenses in C57BL/6 mice. The mice were randomly divided into control group, sham group and A.fumigatus keratitis group. The cornea was monitored daily using a slit lamp and recorded disease score after infection. Corneal lesion was detected by immunofluorescence staining. IDO m RNA and protein were also detected by quantitative reverse transcription-polymerase chain reaction(q RT-PCR) and Western blot.· RESULTS: The disease score and slit lamp photography indicated that disease severity was consistent with corneal inflammation in the murine models, and the disease scores in A. fumigatus keratitis group were obviously higher than those in the sham group. By immunofluorescence staining, IDO was mainly localized in corneal epithelium and stroma in the murine corneal tissues with A. fumigatus keratitis. Compared with the sham group, IDO m RNA expression was significantly enhanced in corneal epithelium infected by A. fumigatus. Furthermore, IDO protein expression detected by Western blot was in accord with transcript levels of IDO m RNA measured by q RT-PCR. 11Prognostic value of dynamic electroencephalogram in comatose patients with different diseased regions显示文摘BACKGROUND: It has proved that dynamic electroencephalogram (EEG) is definite in judging the outcome of ischemic hypoxic comatose patients, EEG is more sensitive to the cortical affection, but not sensitive to the subcortical and brainstem affections, thus it is necessary to clarify the indications of this technique in the clinical application. OBJECTIVE: To observe and compare the prognostic value of dynamic EEG and Glasgow coma score in comatose patients with different diseased region. DESIGN: A clinical case-controlled observation. SETTING: Union Hospital of Fujian Medical University. PARTICIPANTS: Sixty-eight comatose patients were selected from the Union Hospital affiliated to Fujian Medical University from June 1998 to January 2005. The diseased regions were identified using cranial CT (n =43) or MR (n =25). According to different primarily diseased regions, the comatose patients were divided into two groups: ① brainstem affection group (n =23): 13 males and 10 females, 14–62 years of age; ② diffuse cortical affection group (n =45): 28 males and 17 females, 23–75 years of age. METHODS: The dynamic EEG and Glasgow coma score were examined in the 45 comatose patients with primarily cortical affection and 22 comatose patients with primarily brainstem affection at acute phase. The patients were followed-up for 3 months to observe the outcome, The termination of outcome judgment was 3 months after attack or the death. The clinical outcome was classified as complete rehabilitation, survived with disability, death or vegetative state. Correlations of dynamic EEG and Glasgow coma score with the outcome of patients were analyzed. The correlations of dynamic EEG grades and Glasgow coma scores with the outcome were analyzed, and the prognostic value of dynamic EEG grades was compared between the two groups. MAIN OUTCOME MEASURES: ① Correlations of dynamic EEG and Glasgow coma score with the outcome of patients; ② Comparison of the prognostic value of dynamic EEG grades between the two groups. RESULTS: All the 68 patients were involved in the analysis of results. ① Correlations of dynamic EEG grades and Glasgow scores and their correlation analysis: EEG grades had significant negative correlation with Glasgow coma scores in both the cortical affection group and brainstem affection group (r =–0.743, –0.564, P < 0.01, 0.05). In the cortical affection group, the Glasgow coma scores and dynamic EEG grades in the patients with the outcome of death or vegetative state were significantly different from those with the outcome of rehabilitation (P < 0.05–0.01). In the brainstem affection group, the Glasgow coma scores were only significantly different between the patients with outcome of rehabilitation and death (P < 0.05), and there was no significant difference in dynamic EEG grades among the three prognostic states (P > 0.05). ② Comparison of the prognostic value of dynamic EEG grades between comatose patients with cortical affection and brainstem affection: The sensitivity, specificity and accuracy were all higher (P < 0.05), while the error rate was lower (P < 0.05) in the cortical affection group than in the brainstem affection group. 12Nucleotide oligomerization domain 2 contributes to the innate immune response in THCE cells stimulated by Aspergillus fumigatus conidia显示文摘·AIM:To investigate the expression of nucleotide oligomerization domain 2 (NOD2) in the immortalized human corneal epithelial cell line (THCE), and its role in the innate immune response triggered by inactive Aspergillus fumigatus (Af ) conidia.·METHODS:The normal THCE cells were investigated as controls. After incubation with inactive Af conidia for 0.5, 2, 4, 6, and 8 hours, THCE cells were harvested, mRNA expression of NOD2 and receptor interacting protein 2 (RIP2) was detected by RT-PCR. Intracellular proteins including NOD2, NF-κB and proinflammatory cytokines such as TNF-α, IL-8, IL-6 in the cell supernatant were analyzed by ELISA.·RESULTS:Our data indicate that NOD2 expressed in the normal THCE cells. After triggered by the inactive Af conidia, the expression of NOD2, RIP2 mRNA and the secretion of NOD2, NF-κB, TNF-α, IL-8, IL-6 both increased in a time-depended manner, and reached the peak point at 4, 6, 6, 4, 6, 6, 4 hours, respectively. 14Diagnostic Value of Cerebrospinal Fluid T-SPOT.TB for Tuberculousis Meningitis in China显示文摘The aim of this study was to evaluate the diagnostic value of the cerebrospinal fluid(CSF) T‐SPOT.TB test for the diagnosis of TB meningitis(TBM). A retrospective analysis of 96 patients with manifested meningitis was conducted; T‐SPOT.TB test was performed for diagnosing TBM to determine the diagnostic sensitivity, specificity, positive predictive value(PPV), and negative predictive value(NPV). A receiver operating characteristic(ROC) curve was also drawn to assess the diagnostic accuracy. The sensitivity, specificity, PPV, and NPV of CSF T‐SPOT.TB test were 97.8%, 78.0%, 80.3%, and 97.5%, respectively, for 52 patients(54.2%) of the 96 enrolled patients. The area under the curve(AUC) was 0.910, and the sensitivities of CSF T‐SPOT.TB for patients with stages I, II, and III of TBM were 96.7%, 97.2%, and 98.9%, respectively. 16Regulation of interleukin 33/ST2 signaling of human corneal epithelium in allergic diseases显示文摘AIM:To identify the function of ST2 and explore the role of IL-33/ST2 signaling in regulating the pro-allergic cytokine production in human corneal epithelial cells (HCECs). METHODS:Human corneal tissues and cultured primary HCECs were treated with IL-33 in different concentrations without or with different inhibitors to evaluate the expression, location and signaling pathways of ST2 in regulating production of pro-allergic cytokine and chemokine. The expression of mRNA was determined by reverse transcription and real time PCR, and protein production was measured by enzyme-linked immunosorbent assay (ELISA), immunohistochemical and immunofluorescent staining. ST2 protein was detected in donor corneal epithelium, and ST2 signal was enhanced by exposure to IL-33. ·RESULTS:IL-33 significantly stimulated production of pro-allergic cytokines thymic stromal lymphopoietin (TSLP) and chemokine (CCL2, CCL20, CCL22) in HCECs at both mRNA and protein levels. These stimulated productions of pro-allergic mediators by IL-33 were blocked by ST2 antibody or soluble ST2 protein(P <0.05). Interestingly, the IκB-α inhibitor BAY11-7082 or NF-κB activation inhibitor quinazoline blocked NF-κB p65 protein nuclear translocation, and also suppressed the productions of these pro-allergic cytokines and chemokine induced by IL-33. CONCLUSION:These findings demonstrate that IL-33/ ST2 signaling plays an important role in regulating IL-33 induced pro-allergic responses.
