1The Blocking Effect of Phenolic Acid on N-Nitrosomorpholine Formation in vitro显示文摘Phenolic acids (PAs) are widely found in many daily consumed vegetables and fruits.The inhibitory effects of PAs on Nnitrosomorpholine (NMOR) formation in vitro undersimulated gastric juice condition were studied. The results showed that the inhibjtoryIX)tency of thirteen varieties of PAs differed greatly, which may be related to theirchemical structures; the blocking rate (BR) of different killds of PAs were as follows:caf feic acid, 92.5%; tannic acid, 90.0%; gallic acid, 86.8%; sinaPinic acid, 86.2%; ferulicacid, 8l.l%; chlorogenic acid, 69.4%; gentisic acid, 69.2%; syringic acid, 62.1%; protocatechuic acid, 56.0%; p-coumaric acid, 52.5%; vannilic acid, 35.4%; moreover,p-hydroxybenzoic acid and m-coumaric acid had the least blocking effect or even slightcatalyzing effect. 6Physical Activity,Sedentary Behavior,and the Risk of Cardiovascular Disease in Type 2 Diabetes Mellitus Patients:The MIDiab Study显示文摘The aim of this study was to explore the associations of moderate-to-vigorous-intensity physical activity(MVPA)time and sedentary(SED)time with a history of cardiovascular disease(CVD)and multifactorial(i.e.,blood pressure(BP),body mass index(BMI),low-density lipoprotein cholesterol(LDL-C),and glycated hemoglobin A1c(HbA1c))control status among type 2 diabetes mellitus(T2DM)patients in China.A cross-sectional analysis of 9152 people with type 2 diabetes from the Multifactorial Intervention on Type 2 Diabetes(MIDiab)study was performed.Patients were grouped according to their self-reported MVPA time(low,<150 min·week−1;moderate,150 to<450 min·week−1;high,≥450 min·week−1)and SED time(low,<4 h·d–1;moderate,4 to<8 h·d–1;high,≥8 h·d–1).Participants who self-reported a history of CVD were identified as having a CVD risk.Odds ratios(ORs)and 95%confidence intervals(CIs)of CVD risk and multifactorial control status associated with MVPA time and SED time were estimated using mixed-effect logistic regression models,adjusting for China’s geographical region characteristics.The participants had a mean±standard deviation(SD)age of(60.87±8.44)years,44.5%were women,and 25.1%had CVD.After adjustment for potential confounding factors,an inverse association between high MVPA time and CVD risk that was independent of SED time was found,whereas this association was not observed in the moderate-MVPA group.A higher MVPA time was more likely to have a positive effect on the control of BMI.Compared with the reference group(i.e.,those with MVPA time≥450 min·week−1 and SED time<4 h·d–1),CVD risk was higher in the low-MVPA group:The OR associated with an SED time<4 h·d–1 was 1.270(95%CI,1.040–1.553)and that associated with an SED time≥8 h·d–1 was 1.499(95%CI,1.149–1.955).We found that a high MVPA time(i.e.,≥450 min·week−1)was associated with lower odds of CVD risk regardless of SED time among patients with T2DM.Yafei Wu Guijun Qin Guixia Wang Libin Liu Bing Chen Qingbo Guan Zhongshang Yuan Xu Hou Ling Gao Chao Xu Haiqing Zhang Xu Zhang Qiu Li Yongfeng Song Fei Jing Shizhan Ma Shanshan Shao Meng Zhao Qingling Guo Nanwei Tong Hongyan Zhao Xiaomin Xie Chao Liu Zhongyan Shan Zhifeng Cheng Xuefeng Yu Shulin Chen Tao Yang Yangang Wang Dongmei Li Zhaoli Yan Lixin Guo Qiuhe Ji Wenjuan Wang Jiajun Zhao 2023Engineering2023,,1:1
7Mutations in epigenetic regulator KMT2C detected by liquid biopsy are associated with worse survival in prostate cancer patients显示文摘Background:KMT2(lysine methyltransferase)family enzymes are epigenetic regulators that activate gene transcription.KMT2C is mainly involved in enhancer-associated H3K4me1,and is also one of the top mutated genes in cancer(6.6%in pan-cancer).Currently,the clinical significance of KMT2C mutations in prostate cancer is understudied.Methods:We included 221 prostate cancer patients diagnosed between 2014 and 2021 in West China Hospital of Sichuan University with cell-free DNA-based liquid biopsy test results in this study.We investigated the association between KMT2C mutations,other mutations,and pathways.Furthermore,we evaluated the prognostic value of KMT2C mutations,measured by overall survival(OS)and castration resistance-free survival(CRFS).Also,we explored the prognostic value of KMT2C mutations in different patient subgroups.Lastly,we investigated the predictive value of KMT2C mutations in individuals receiving conventional combined anti-androgen blockade(CAB)and abiraterone(ABI)as measured by PSA progression-free survival(PSA-PFS).Results:The KMT2C mutation rate in this cohort is 7.24%(16/221).KMT2C-mutated patients showed worse survival than KMT2C-wild type(WT)patients regarding both CRFS and OS(CRFS:mutated:9.9 vs.WT:22.0 months,p=0.015;OS:mutated:71.9 vs.WT 137.4 months,p=0.012).KMT2C mutations were also an independent risk factor in OS[hazard ratio:3.815(1.461,9.96),p=0.006]in multivariate analyses.Additionally,we explored the association of KMT2C mutations with other genes.This showed that KMT2C mutations were associated with Serine/Threonine-Protein Kinase 11(STK11,p=0.004)and Catenin Beta 1(CTNNB1,p=0.008)mutations.In the CAB treatment,KMT2C-mutated patients had a significantly shorter PSA-PFS compared to KMT2C-WT patients.(PSA-PFS:mutated:9.9 vs.WT:17.6 months,p=0.014).Moreover,KMT2C mutations could effectively predict shorter PSA-PFS in 10 out of 23 subgroups and exhibited a strong trend in the remaining subgroups.Conclusions:KMT2C-mutated patients showed worse survival compared to KMT2C-WT patients in terms of both CRFS and OS,and KMT2C mutations were associated with STK11 and CTNNB1 mutations.Furthermore,KMT2C mutations indicated rapid progression during CAB therapy and could serve as a potential biomarker to predict therapeutic response in prostate cancer.SHA ZHU NANWEI XU JIAYU LIANG FENGNIAN ZHAO ZILIN WANG YUCHAO NI JINDONG DAI JINGE ZHAO XINGMING ZHANG JUNRU CHEN GUANGXI SUN PENGFEI SHEN HAO ZENG 2023Oncology Research2023,31,4:0
8High Performance Liquid Chromatographic Determination of Phenolic Acids in Fruits and Vegetables显示文摘A simple isocratic HPLC technique has been developed for the quantitative analysis of phenolic acids(PAs)in fruits and vegetables.Nine benzoic and cinnamic acid derivatives were separated in less than 30min,and the resolution was all more than 1.23.The ranges of linearity for PAs standards were 0.2-100ng,even up to 600 ng(r=0.983-1.000)and the detection limits were 0.02-0.24mg/kg,Samples of fresh vegetables and fruits were extracted with 80% methanol and ethyl acetate,then purified with C18 Sep-Pak cartridge and determined by HPLC.This method was applied to the determination of PAs in 7 kinds of fruits and vegetables.i.e.,apple,pear,Chinese cabbage,cauliflower,turnip,soybean sprout and white grape wine,The content of the 9 PAs varied widely in the 7 kinds of fodds studied.The average concentrations of chorogenic acid in apple(100.2mg/kg)and pear(30.8mg/kg)were quite high,and sinapinic acid was remarkable(42.5mg/kg)in chinese cabbage,and protocatechuic acid had the highest concentration of all the PAs in white wine.LI PING, WANG Xu-QlNG, WANG HlJAI-ZHOU, AND WU YONG-NlNGInstitute of Nutrition and Food Hygiene, Chinese Academy of Preventive Medicine, 29 Nanwei Road, Beijing 100050, China 1993Biomedical and Environmental Sciences1993,6,4:0
