15Improved degradation of azo dyes by lignin peroxidase following mutagenesis at two sites near the catalytic pocket and the application of peroxidase-coated yeast cell walls显示文摘The enzymatic degradation of azo dyes is a promising alternative to ineffective chemical and physical remediation methods.Lignin peroxidase(LiP)from Phanerochaete chrysosporium is a hemecontaining lignin-degrading oxidoreductase that catalyzes the peroxide-dependent oxidation of diverse molecules,including industrial dyes.This enzyme is therefore ideal as a starting point for protein engineering.Accordingly,we subjected two positions(165 and 264)in the environment of the catalytic Trp171 residue to saturation mutagenesis,and the resulting library of 104 independent clones was expressed on the surface of yeast cells.This yeast display library was used for the selection of variants with the ability to break down structurally-distinct azo dyes more efficiently.We identified mutants with up to 10-fold greater affinity than wild-type LiP for three diverse azo dyes(Evans blue,amido black 10B and Guinea green)and up to 13-fold higher catalytic activity.Additionally,cell wall fragments displaying mutant LiP enzymes were prepared by toluene-induced cell lysis,achieving significant increases in both enzyme activity and stability compared to a whole-cell biocatalyst.LiPcoated cell wall fragments retained their initial dye degradation activity after 10 reaction cycles each lasting 8 h.The best-performing mutants removed up to 2.5-fold more of each dye than the wild-type LiP in multiple reaction cycles.Karla IlićĐurđić Raluca Ostafe Olivera Prodanović AleksandraĐurđevićĐelmaš Nikolina Popović Rainer Fischer Stefan Schillberg Radivoje Prodanović 2021Frontiers of Environmental Science & Engineering2021,15,2:1
16The Effects of Ground Cover Mowing Height on the Fauna of Ground-dwelling Arthropods in Olive Grove显示文摘Ground dwelling insects,spiders,and other arthropods play an important role in agroecosystems as predators and saprophytic organisms.Their presence on the soil surface helps the biological control of pests,enhances organic matter decomposition and promotes biodiversity.Soil disturbance,particularly tilling and mowing has greatly affected the assemblage and abundance of epigeic arthropods.This study aimed to determine the effects of herbaceous vegetation cover mowing height on arthropods abundance and structure in olive orchards.The experimental site was divided into three zones(Z1,Z2,and Z3)regarding mowing heights(0 cm,10 cm,and 15 cm).This research was done in 2019 from the beginning of May untill the end of September.During this research,1490 individuals were recorded belonging to 6 classes,11 orders and 13 families.The number of individuals was higher in the zones Z2 and Z3 comparing with Z1.The highest number of individuals showed woodlices(Isopoda)comprising 67,5%of all individuals collected.There were significant differences in the number of Carabidae,Bleteliidae and Forficulidae between the zones of research.Unlike Forficulidae,higher heights of mown positively affected the population of Carabidae and Blateliidae.Kristijan Franin Nikolina Kapić Šime Marcelić ZoranŠikić 2020Research in Ecology2020,2,2:0
17Flooding as a cause of ungulate mortality in floodplain forests in Croatia显示文摘Floodplain forests with regular flooding regimes are the largest natural retentions areas in Croatia and are important as natural habitats for ungulates.The aim of this study was to determine the scale of mortality caused by flooding within these forests.Over a 10-year period,data on ungulate mortality(red deer,roe deer and wild boar),flood duration and flooded surface area were recorded.The study was conducted in primary(Posavske Sume—RET I)and secondary(Opeke II—RET II)retention areas within Lonjsko Polje Nature Park(Sava River region,Croatia).The longest flood period and the largest flooded surface area were recorded in RET I.Total ungulate mortality was749 individuals,with 482 individuals in RET I and 267 individuals in RET II,predominantly wild boar.Flood mortality did not differ by gender.The highest mortality of wild boar was recorded for the juvenile and yearling ageclasses.Low mortality of red and roe deer can be attributed to their body size and ecological niches.Differences in mortality between the primary and secondary retention areas corresponded to differences in flood regimes,flood column heights and micro relief structures.In both retention areas,wild boar mortality and flood duration,i.e.flooded surface area,were positively correlated.Because the growth rate of the analysed ungulate populations was higher than the recorded mortality,no long-term effect of floods is expected on species abundance in these areas.Damir Ugarkovic Nikica Sprem Nikolina Kelava Ugarkovic Milan Orsanic 2020Journal of Forestry Research2020,31,3:0
