| 3 | Isolation and characteristics of autoreactive T cells specific to aggrecan G1 domain from rheumatoid arthritis patients显示文摘Our previous work showed that the cartilage proteoglycan aggrecan could induce an erosive polyarthritis and spondylitis in BALB/c mice and the GI globular domain of the aggrecan (GI) contained the arthritogenic region. To elucidate whether autoreactive T cells to G1 are expressed in rheumatoid arthritis patients, we analyzed the frequency of human G1-specific T cells in the peripheral blood of five rheumatoid arthritis patients and tried to establish G1-reactive T cell lines from these rheumatoid arthritis patients. The results showed that the G1-specific T cells in PBL were detectable at the range of 4.97 ±0.5 ×10-6 in peripheral blood lymphocytes. We have also generated 15 G1-specific T lymphocyte lines from these pateints with a standard split-well method. All these cells expressed fine specificity to human recombinant G1, but not to unrelated antigen. All the 15 lines expressed a panT cell marker and 13 of them selectively used the αβ T cell receptor. Two of them used rye T cell receptor. The 13 of these T cell lines was CD4 positive. One line expressed CD8. One line expressed both CD4 and CD8. Moreover, 14 out of 15 lines expressed the Th-1 cytokine profile, characterized by interferon-γpositivity and IL-4 negativity. No Th-2 type cell line was generated. These data provide strong evidence in favor of the presence of autoreactive T cells in the rheumatoid arthritis pateints. On the basis of analysis, the author put forward recommendations, including increasing scientific research input for agricultural machinery, supporting agricultural machinery enterprises, promoting scientific and technical innovation, taking characteristic industries (such as flowers and vegetables) as pillar industry, speeding up development of modern facility agriculture, and upgrading primary processing of agricultural products. | 7 | Face and object visual working memory deficits in first-episode schizophrenia correlate with multiple neurocognitive performances显示文摘Background Working memory(WM)deficit is considered a core feature and cognitive biomarker in patients with schizophrenia.Several studies have reported prominent object WM deficits in patients with schizophrenia,suggesting that visual WM in these patients extends to non-spatial domains.However,whether non-spatial WM is similarly affected remains unclear.Aim This study primarily aimed to identify the processing of visual object WM in patients with first-episode schizophrenia.Methods The study included 36 patients with first-episode schizophrenia and 35 healthy controls.Visual object WM capacity,including face and house WM capacity,was assessed by means of delayed matching-to sample visual WM tasks,in which participants must distribute memory so that they can discriminate a target sample.We specifically examined their anhedonia experience by the Temporal Experience of Pleasure Scale and the Snaith-Hamilton Pleasure Scale.Cognitive performance was measured by the Repeatable Battery for the Assessment of Neuropsychological Status(RBANS).Results Both face and house WM capacity was significantly impaired in patients with schizophrenia.For both tasks,the performance of all the subjects was worse under the high-load condition than under the low-load condition.We found that WM capacity was highly positively correlated with the performance on RBANS total scores(r=−0.528,p=0.005),RBANS delayed memory scores(r=−0.470,p=0.013),RBANS attention scores(r=−0.584,p=0.001),RBANS language scores(r=−0.448,p=0.019),Trail-Making Test:Part A raw scores(r=0.465,p=0.015)and simple IQ total scores(r=−0.538,p=0.005),and correlated with scores of the vocabulary test(r=−0.490,p=0.011)and scores of the Block Diagram Test(r=−0.426,p=0.027)in schizophrenia.No significant correlations were observed between WM capacity and Positive and Negative Syndrome Scale symptoms.Conclusions Our research found that visual object WM capacity is dramatically impaired in patients with schizophrenia and is strongly correlated with other measures of cognition,suggesting a mechanism that is critical in explaining a portion of the broad cognitive deficits observed in schizophrenia. | Yi Liu Taiyong Bi Bei Zhang Qijie Kuang Haijing Li Kunlun Zong Jingping Zhao Yuping Ning Shenglin She Yingjun Zheng | 2021 | General Psychiatry2021,34,1: | 2 |

| 14 | Resistance potential of soil bacterial communities along a biodiversity gradient in forest ecosystems显示文摘Higher biodiversity is often assumed to be a more desirable scenario for maintaining the functioning of ecosystems,but whether species-richer communities are also more disturbance-tolerant remains controversial.In this study,we investigated the bacterial communities based on 472 soil samples from 28 forests across China with associated edaphic and climatic properties.We developed two indexes(i.e.,community mean tolerance breadth[CMTB]and community mean response asynchrony[CMRA])to explore the relationship between diversity and community resistance potential.Moreover,we examined this resistance potential along the climatic and latitudinal gradients.We revealed that CMTB was significantly and negatively related to species richness,resulting from the changes in balance between relative abundances of putative specialists and generalists.In comparison,we found a unimodal relationship between CMRA and richness,suggesting that higher biodiversity might not always lead to higher community resistance.Moreover,our results showed differential local patterns along latitude.In particular,local patterns in the northern region mainly followed general relationships rather than those for the southern forests,which may be attributed to the differences in annual means and annual variations of climate conditions.Our findings highlight that the community resistance potential depends on the composition of diverse species with differential environmental tolerance and responses.This study provides a new,testable evaluation by considering tolerance breadth and response asynchrony at the community level,which will be helpful in assessing the influence of disturbance under rapid shifts in biodiversity and species composition as a result of global environmental change. | Jialiang Kuang Dongmei Deng Shun Han Colin T.Bates Daliang Ning Wensheng Shu Jizhong Zhou | 2022 | mLife2022,1,4: | 0 |

| 18 | Integrated analysis of micro RNA and m RNA expression profiles in HBx-expressing hepatic cells显示文摘AIM To identify the mi RNA-m RNA regulatory network in hepatitis B virus X(HBx)-expressing hepatic cells. METHODS A stable HBx-expressing human liver cell line L02 was established. The mR NA and miR NA expression profiles of L02/HBx and L02/pc DNA liver cells were identified by RNA-sequencing analysis. Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis was performed to investigate the function of candidate biomarkers, and the relationship between mi RNA and mR NA was studied by network analysis.RESULTS Compared with L02/pc DNA cells, 742 unregulated genes and 501 downregulated genes were determined as differentially expressed in L02/HBx cells. Gene ontology analysis suggested that the differentially expressed genes were relevant to different biologicalprocesses. Concurrently, 22 differential mi RNAs were also determined in L02/HBx cells. Furthermore, integrated analysis of mi RNA and m RNA expression profiles identified a core mi RNA-m RNA regulatory network that is correlated with the carcinogenic role of HBx.
