2Interactions Between TLR7 and TLR9 Agonists and Receptors Regulate Innate Immune Responses by Astrocytes and Microglia显示文摘Toll样受体7(TLR7)和Toll样受体9(TLR9)是固有免疫反应中的重要介质。它们均定位于内涵体间隔,识别核酸和经过髓样分化因子88(MyD88)传导来的信号。本文分析TLR7和TLR9导致的在中枢神经系统固有免疫反应中起作用的星形胶质细胞和小胶质细胞的活化。TLR7和TLR9激动剂引起星形胶质细胞和小胶质细胞相似的细胞因子反应,但二者仍有区别,小胶质细胞产生抗炎因子IL-10、抗凋亡因子G-CSF及IL-9,但星形胶质细胞不产生。对2种细胞的共刺激研究均显示,TLR7激动剂——咪喹莫特,能浓度依赖性地抑制TLR9激动剂引起的固有免疫反应。令人惊讶的是,这种抑制并非由TLR7介导,因为在TLR7的缺乏的情况下,TLR9激动剂导致的固有免疫反应仍然被抑制。固有免疫反应的抑制也并非由于抑制TLR9激动剂摄取所致。这提示咪喹莫特的抑制效应可能是直接作用,也许是通过阻滞CpG-ODN结合和/或TLR9介导的信号转导所致,从而限制细胞活化。这种拮抗关系也存在于小胶质细胞中的这2类受体之间,TLR7的缺乏导致CpG-ODN刺激的细胞因子反应增强。因此,在胶质细胞中,TLR7及其激动剂对TLR9介导的细胞因子反应有抑制作用。NIRANJAN B. 3Use of a Bioaugmented Organic Soil Amendment in Combination with Gypsum for Withania somnifera Growth on Sodic Soil显示文摘Limited availability of organic matter is a problem to sustain crop growth on sodic soil. Organic soil amendments are a costeffective source of nutrients to enhance crop growth. A field study was conducted to evaluate the effect of an organic soil amendment bioaugmented with plant growth-promoting fungi(SF_(OA) ) in combination with gypsum on soil properties and growth and yield attributes of Withania somnifera, one of the most valuable crops of the traditional medicinal system in the world, on a sodic soil at the Aurawan Research Farm of CSIR-National Botanical Research Institute, Lucknow, India. The SF_(OA) used was prepared by pre-enriching farm waste vermicompost with plant growth-promoting fungi before mixing with pressmud and Azadirachta indica seed cake. The application of SF_(OA) at 10 Mg ha^(-1)after gypsum(25.0 Mg ha^(-1)) treatment significantly(P < 0.05) increased root length(by 96%) and biomass(by 125%) of Withania plants compared to the control without SF_(OA) and gypsum. Similarly, the highest withanolide contents were observed in leaves and roots of Withania plants under 10 Mg ha^(-1)SF_(OA) and gypsum. Combined application of SF_(OA) and gypsum also improved physical, chemical and enzymatic properties of the soil, with the soil bulk density decreasing by 25%, water-holding capacity increasing by 121%, total organic C increasing by 90%, p H decreasing by 17% and alkaline phosphatase, β-glucosidase, dehydrogenase and cellulase activities increasing by 54%, 128%, 81% and 96%, respectively, compared to those of the control. 4Computational Identification of Sweet Wormwood (Artemisia annua) microRNA and Their mRNA Targets显示文摘Despite its efficacy against malaria, the relatively low yield (0.01%-0.8%) of artemisinin in Artemisia annua is a serious limitation to the commercialization of the drug. A better understanding of the biosynthetic pathway of artemisinin and its regulation by both exogenous and endogenous factors is essential to improve artemisinin yield. Increasing evidence has shown that microRNAs (miRNAs) play multiple roles in various biological processes. In this study, we used previously known miRNAs from Arabidopsis and rice against expressed sequence tag (EST) database of A. annua to search for potential miRNAs and their targets in A. annua. A total of six potential miRNAs were predicted, which belong to the miR414 and miR1310 families. Furthermore, eight potential target genes were identified in this species. Among them, seven genes encode proteins that play important roles in ar- temisinin biosynthesis, including HMG-CoA reductase (HMGR), amorpha-4,11-diene synthase (ADS), farnesyl pyrophosphate synthase (FPS) and cytochrome P450. In addition, a gene coding for putative AINTEGUMENTA, which is involved in signal transduction and development, was also predicted as one of the targets. 5Hidden Markov Models Incorporating Fuzzy Measures and Integrals for Protein Sequence Identification and Alignment显示文摘Profile hidden Markov models (HMMs) based on classical HMMs have been widely applied for protein sequence identification. The formulation of the forward and backward variables in profile HMMs is made under statistical independence as-sumption of the probability theory. We propose a fuzzy profile HMM to overcome the limitations of that assumption and to achieve an improved alignment for protein sequences belonging to a given family. The proposed model fuzzifies the forward and backward variables by incorporating Sugeno fuzzy measures and Choquet inte-grals, thus further extends the generalized HMM. Based on the fuzzified forward and backward variables, we propose a fuzzy Baum-Welch parameter estimation al-gorithm for profiles. The strong correlations and the sequence preference involved in the protein structures make this fuzzy architecture based model as a suitable candidate for building profiles of a given family, since the fuzzy set can handle uncertainties better than classical methods.Niranjan P. 6Autologous bone marrow derived stem cell therapy in patients with type 2 diabetes mellitus-defining adequate administration methods显示文摘AIM To carry out randomized trial for evaluating effects of autologous bone marrow derived stem cell therapy(ABMSCT) through different routes.METHODS Bone marrow aspirate was taken from the iliac crest of patients. Bone marrow mononuclear cells were separatedand purified using centrifugation. These cells were then infused in a total of 21 patients comprising three groups of 7 patients each. Cells were infused into the superior pancreaticoduodenal artery(Group Ⅰ), splenic artery(Group Ⅱ) and through the peripheral intravenous route(Group Ⅲ). Another group of 7 patients acted as controls and a sham procedure was carried out on them(Group Ⅳ). The cells were labelled with the PET tracer F18-FDG to see their homing and in vivo distribution. Data for clinical outcome was expressed as mean ± SE. All other data was expressed as mean ± SD. Baseline and post treatment data was compared at the end of six months, using paired t-test. Cases and controls data were analyzed using independent t-test. A probability(P) value of < 0.05 was regarded as statistically significant. Measures of clinical outcome were taken as the change or improvement in the following parameters:(1) C-peptide assay;(2) HOMA-IR and HOMA-B;(3) reduction in Insulin dose; subjects who showed reduction of insulin requirement of more than 50% from baseline requirement were regarded as responders; and(4) reduction in HbA 1c. RESULTS All the patients, after being advised for healthy lifestyle changes, were evaluated at periodical intervals and at the end of 6 mo. The changes in body weight, body mass index, waist circumference and percentage of body fat in all groups were not significantly different at the end of this period. The results of intra-group comparison before and after ABMSCT at the end of six months duration was as follows:(1) the area under C-peptide response curve was increased at the end of 6 mo however the difference remained statistically non-significant(P values for fasting C-peptide were 0.973, 0.103, 0.263 and 0.287 respectively and the P values for stimulated C-peptide were 0.989, 0.395, 0.325 and 0.408 respectively for groups Ⅰ?to Ⅳ);(2) the Insulin sensitivity indices of HOMA IR and HOMA B also did not show any significant differences(P values for HOMA IR were 0.368, 0.223, 0.918 and 0.895 respectively and P values for HOMA B were 0.183, 0.664, 0.206 and 0.618 respectively for groups Ⅰto Ⅳ);(3) Group Ⅰshowed a significant reduction in Insulin dose requirement(P < 0.01). Group Ⅱ patients also achieved a significant reduction in Insulin dosages(P = 0.01). The Group Ⅰand Group Ⅱ patients together constituted the targeted group wherein the feeding arteries to pancreas were used for infusing stem cells. Group Ⅲ, which was the intravenous group, showed a non-significant reduction in Insulin dose requirement(P = 0.137). Group Ⅳ patients which comprised the control arm also showed a significant reduction in Insulin dosages at the end of six months(P < 0.05); and(4) there was a non-significant change in the Hb A1 c levels at the end of 6 mo across all groups(P = 0.355, P = 0.351, P = 0.999 and P = 0.408 respectively for groups Ⅰto Ⅳ). 7Locoregional treatment for hepatocellular carcinoma:The best is yet to come显示文摘Hepatocellular carcinoma(HCC) is the sixth-most common type of cancer worldwide. The only definitive treatment modalities capable of achieving a cure are hepatic resection and hepatic transplantation. However, most patients are not candidates for these therapies. Overall, treatment options are driven by the stage of HCC. Early-stage disease is treated with ablative therapies, with radiofrequency ablation the ablative therapy of choice. Microwave ablation and irreversible electroporation are the other upcoming alternatives. 8Imaging in pulmonary hydatid cysts显示文摘Hydatid disease is a zoonosis that can involve almost any organ in the human body. After the liver, the lungs are the most common site for hydatid disease in adults. Imaging plays a pivotal role in the diagnosis of the disease, as clinical features are often nonspecific. Classical radiological signs of pulmonary hydatid cysts have been described in the literature, aiding in the diagnosis of the disease. However, complicated hydatid cysts can prove to be a diagnostic challenge at times due to their atypical imaging features. Radiography is the initial imaging modality. Computed tomography can provide a specific diagnosis in complicated cases. Ultrasound is particularly useful in peripheral lung lesions. 9Extracellular synthesis of silver nanoparticles using dried leaves of pongamia pinnata(L) pierre显示文摘Extract of oven dried leaves of Pongamia pinnata(L) Pierre was used for the synthesis of silver nanoparticles. Stable and crystalline silver nanoparticles were formed by the treatment of aqueous solution of AgNO_3(1m M) with dried leaf extract of Pongamia pinnata(L) Pierre. UV-visible spectroscopy studies were carried out to quantify the formation of silver nanoparticles. Transmission electron microscopy, X-ray diffraction and Fourier transform infrared spectroscopy were used to characterize the silver nanoparticles. TEM image divulges that silver nanoparticles are quite polydispersed, the size ranging from 20 nm to 50 nm with an average of 38 nm. Water soluble heterocyclic compounds such as flavones were mainly responsible for the reduction and stabilization of the nanoparticles. Silver nanoparticles were effective against Escherichia coli(ATCC 8739), Staphylococcus aureus(ATCC 6538p), Pseudomonas aeruginosa(ATCC 9027) and Klebsiella pneumoniae(clinical isolate). 10Differences in atheroma between Caucasian and Asian subjects with anterior stroke:A vessel wall MRI study显示文摘Background and purpose While extracranial carotid artery stenosis is more common among Caucasians and intracranial artery stenosis is more common among Asians,the differences in atherosclerotic plaque characteristics have not yet been extensively examined.We sought to investigate plaque location and characteristics within extracranial carotid and intracranial arteries in symptomatic Caucasians and Chinese using vessel wall MRI.Methods Subjects with recent anterior circulation ischaemic stroke were recruited and imaged at two sites in the USA and China using similar protocols.Both extracranial carotid and intracranial arteries were reviewed to determine plaque location and characteristics.Results The prevalence of extracranial carotid plaque in Caucasians and Chinese was 73.1%and 49.1%,respectively(p=0.055).Prevalence of intracranial plaque was 38.5%and 69.1%in Caucasians and Chinese,respectively(p=0.02).Furthermore,42% of Caucasians and 16%of Chinese had high-risk plaque(HRP)features(intraplaque haemorrhage,luminal surface disruption)in the extracranial carotid artery(p=0.03).The prevalence of HRP features in intracranial arteries was not significantly different between the two cohorts(4%vs 11%;p=0.42).Conclusions Differences in the location and characteristics of cerebrovascular atherosclerosis were identified by vessel wall MRI in US Caucasian and Chinese subjects with recent anterior circulation ischaemic stroke.Extracranial carotid plaques with HRP features were more common in Caucasians.Intracranial plaques were more common in Chinese subjects,but no significant difference between the two cohorts in intracranial HRP prevalence was found.Larger studies using vessel wall imaging to investigate racial differences in cerebrovascular disease may inform underlying mechanisms of HRP development and may ultimately help guide appropriate therapy.Hiroko Watase Mi Shen Binbin Sui Peiyi Gao Dong Zhang Jie Sun Niranjan Balu Daniel S Hippe Gail P Jarvik Xihai Zhao Rui Li Shuo Chen Chun Yuan Thomas S Hatsukami 2021Stroke & Vascular Neurology2021,6,1:4

12Solar-Driven Hydrogen Peroxide Production Using Polymer-Supported Carbon Dots as Heterogeneous Catalyst显示文摘Safe, sustainable, and green production of hydro gen peroxide is an exciting proposition due to the role of hydrogen peroxide as a green oxidant and energy carrier for fuel cells. The current work reports the development of carbon dot-impregnated waterborne hyperbranched polyurethane as a heterogeneous photo-catalyst for solar-driven production of hydrogen peroxide. The results reveal that the carbon dots possess a suitable band-gap of 2.98 eV,which facilitates effective splitting of both water and ethanol under solar irradiation. Inclusion of the carbon dots within the eco-friendly polymeric material ensures their catalytic activity and also provides a facile route for easy catalyst separation, especially from a solubilizing medium.The overall process was performed in accordance with the principles of green chemistry using bio-based precursorsand aqueous medium. 14髋关节置换术后短期内患者生活质量调查分析显示文摘目的:观察全髋关节置换和半髋置换术后3个月内患者生活质量果。方法:对2013年3月到12月期间在荆州市中心医院骨科进行髋关节置换术的31例患者进行回访调查,并进行健康生活质量(HRQoL)、生理功能(PF)调查与评估以及体能健康的局限(RP)、躯体疼痛(BP)、一般健康(GH)的评分和36项健康调查问卷(SF-36)的填写。调查方式是以病人的医疗记录为调查内容。SF-36评分的平均变化的临床意义在于术后和术前评分之间的平均差异有统计学意义且术后平均SF-36评分能与健康的65岁或以上的老年人一致。结果:3个月随访后,术前患者SF-36问卷评分后PF、RP、BP和GH值分别为39.4、18.5、30.6和47.3。术后患者的PF、RP、BP和GH值分别为45.2、30.6、46.9和52.6。术后患者以上4个值的平均分数均高于术前得分,平均BP得分在统计学上有显著改善(P<0.01),PF、RP和GH的变化无统计学差异(P>0.05)。结论:全髋关节置换3个月后,虽然患者疼痛显著减轻,但患者总体生活质量无明显改善。Niranjan Aryal 许永涛 汪勇刚 2015长江大学学报(自科版)(下旬)2015,12,6:2

17Improvement in the dissolution rate and tableting properties of cefuroxime axetil by melt-granulated dispersion and surface adsorption显示文摘A combination of melt-granulated dispersion and surface adsorption techniques was used to enhance the dissolution and tableting properties of cefuroxime axetil(CA).Gelucire 50/13 was used as the melt-dispersion carrier and Sylysia 350 was used to adsorb the melt dispersion.Solubility studies showed an 8-fold increase in solubility at a ratio of 1:1.5 for CA:Gelucire 50/13.The minimum quantity of Sylysia 350 required to achieve the desired flowability and compressibility was 0.5 parts of Sylysia 350 per unit of Gelucire 50/13.Phase solubility studies showed negative ΔG_(tr)^(0) values for Gelucire 50/13 at various concentrations(2-10%,vv/v),indicating the spontaneous nature of solubilization.FT-IR and DSC spectra exhibited drug-excipient compatibility.Molecular modeling by a computational method employing energy minimization revealed entrapment of CA in Gelucire 50/13.The total potential energy of CA(70.562 keal/mol)was reduced to 33.578 keal/mol after solid dispersion with Gelucire 50/13.P-XRD studies indicated that the presence of Sylysia 350 is less likely to promote the reversion of the amorphous CA to a crystalline state.In vitro dissolution studies demonstrated an improved dissolution rate,and drug release at 15 min(Q_(15min))exhibited a 15-fold improvement.The rapidly dissolving CA tablets showed improved dissolution with improved tableting properties.Jammula Sruti Chinam Niranjan Patra Suryakanta Swain Kanhu Charan Panigrahi Allupati Premananda Patro Sarwar Beg Subas Chandra Dinda Muddana Eswara Bhanoji Rao 2013Acta Pharmaceutica Sinica B2013,3,2:2
