1Caspase-3 activation as a bifurcation point between plasticity and cell death显示文摘Death-mediating proteases such as caspases and caspase-3 in particular,have been implicated in neurodegenerative processes,aging and Alzheimer's disease.However,emerging evidence suggests that in addition to their classical role in cell death,caspases play a key role in modulating synaptic function.It is remarkable that active caspases-3,which can trigger widespread damage and degeneration,aggregates in structures as delicate as synapses and persists in neurons without causing acute cell death.Here,we evaluate this dichotomy,and discuss the hypothesis that caspase-3 may be a bifurcation point in cellular signaling,able to orient the neuronal response to stress down either pathological/apoptotic pathways or towards physiological cellular remodeling.We propose that temporal,spatial and other regulators of caspase activity are key determinants of the ultimate effect of caspase-3 activation in neurons.This concept has implications for differential roles of caspase-3 activation across the lifespan.Specifically,we propose that limited caspase-3 activation is critical for synaptic function in the healthy adult brain while chronic activation is involved in degenerative processes in the aging brain.Shikha Snigdha Erica D Smith G Aleph Prieto Carl W Cotman 2012Neuroscience Bulletin2012,28,1:91
2Targeting the PI3K/Akt signaling pathway in gastric carcinoma: A reality for personalized medicine?显示文摘Frequent activation of phosphatidylinositol-3 kinases(PI3K)/Akt/m TOR signaling pathway in gastric cancer(GC) is gaining immense popularity with identification of mutations and/or amplifications of PIK3 CA gene or loss of function of PTEN,a tumor suppressor protein,to name a few; both playing a crucial role in regulating this pathway. These aberrations result in dysregulation of this pathway eventually leading to gastric oncogenesis,hence,there is a need for targeted therapy for more effective anticancer treatment. Several inhibitors are currently in either preclinical or clinical stages for treatment of solid tumors like GC. With so many inhibitors under development,further studies on predictive biomarkers are needed to measure the specificity of any therapeutic intervention. Herein,we review the common dysregulation of PI3K/Akt/m TOR pathway in GC and the various types of single or dual pathway inhibitors under development that might have a superior role in GC treatment. 5Use of a Bioaugmented Organic Soil Amendment in Combination with Gypsum for Withania somnifera Growth on Sodic Soil显示文摘Limited availability of organic matter is a problem to sustain crop growth on sodic soil. Organic soil amendments are a costeffective source of nutrients to enhance crop growth. A field study was conducted to evaluate the effect of an organic soil amendment bioaugmented with plant growth-promoting fungi(SF_(OA) ) in combination with gypsum on soil properties and growth and yield attributes of Withania somnifera, one of the most valuable crops of the traditional medicinal system in the world, on a sodic soil at the Aurawan Research Farm of CSIR-National Botanical Research Institute, Lucknow, India. The SF_(OA) used was prepared by pre-enriching farm waste vermicompost with plant growth-promoting fungi before mixing with pressmud and Azadirachta indica seed cake. The application of SF_(OA) at 10 Mg ha^(-1)after gypsum(25.0 Mg ha^(-1)) treatment significantly(P < 0.05) increased root length(by 96%) and biomass(by 125%) of Withania plants compared to the control without SF_(OA) and gypsum. Similarly, the highest withanolide contents were observed in leaves and roots of Withania plants under 10 Mg ha^(-1)SF_(OA) and gypsum. Combined application of SF_(OA) and gypsum also improved physical, chemical and enzymatic properties of the soil, with the soil bulk density decreasing by 25%, water-holding capacity increasing by 121%, total organic C increasing by 90%, p H decreasing by 17% and alkaline phosphatase, β-glucosidase, dehydrogenase and cellulase activities increasing by 54%, 128%, 81% and 96%, respectively, compared to those of the control. 6Attenuation of Collagen Induced Arthritis by Centella asiatica Methanol Fraction via Modulation of Cytokines and Oxidative Stress显示文摘Objective To investigate the anti-inflammatory, antioxidant and anti-arthritic effects of Centella asiatica methanolfraction(CaM E) on collagen-induced arthritis(CIA), an animal model of rheumatoid arthritis. Methods Arthritis was induced in female wistar rats by immunization with porcine type II collagen. The CIA rats were treated orally with CaM E(50, 150, and 250 mg/kg/day) for 15 d(beginning on day 21 of the experimental period). The clinical, histological, biochemical, and immunological parameters were assessed. Results CaM E treatment(150 and 250 mg/kg) significantly attenuated the severity of CIA and reduced the synovial inflammation, cartilage erosion, and bone erosion as evident from both histological and radiographic data. The escalated plasma levels of pro-inflammatory cytokines TNF-α, IL-1β, IL-6, and IL-12 alongwith nitric oxide in CIA rats decreased significantly on CaM E treatment. The serum levels of type-II collagen antibody were significantly lower in rats of CaM E(150 and 250 mg/kg) treated group than those in the arthritic group. Furthermore, by inhibiting the above mediators, CaM E also contributed towards the reversal of the disturbed antioxidant levels and peroxidative damage. 7Soil Carbon Dynamics Under Changing Climate A Research Transition from Absolute to Relative Roles of Inorganic Nitrogen Pools and Associated Microbial Processes:A Review显示文摘It is globally accepted that soil carbon(C) dynamics are at the core of interlinked environmental problems, deteriorating soil quality and changing climate. Its management remains a complex enigma for the scientific community due to its intricate relationship with soil nitrogen(N) availability and moisture-temperature interactions. This article reviews the management aspects of soil C dynamics in light of recent advances, particularly in relation to the availability of inorganic N pools and associated microbial processes under changing climate. Globally, drastic alterations in soil C dynamics under changing land use and management practices have been primarily attributed to the variation in soil N availability, resulting in a higher decomposition rate and a considerable decline in soil organic C(SOC) levels due to increased soil CO_2 emissions, degraded soil quality, and increased atmospheric CO_2 concentrations,leading to climate warming. Predicted climate warming is proposed to enhance SOC decomposition, which may further increase soil N availability, leading to higher soil CO_2 efflux. However, a literature survey revealed that soil may also act as a potential C sink, if we could manage soil inorganic N pools and link microbial processes properly. Studies also indicated that the relative, rather than the absolute, availability of inorganic N pools might be of key importance under changing climate, as these N pools are variably affected by moisture-temperature interactions, and they have variable impacts on SOC turnover. 8Critical role of OX40 signaling in the TCR-independent phase of human and murine thymic Treg generation显示文摘Regulatory T cells(Tregs)play a pivotal role in immune-tolerance,and loss of Treg function can lead to the development of autoimmunity.Natural Tregs generated in the thymus substantially contribute to the Treg pool in the periphery,where they suppress self-reactive effector T cells(Teff)responses.Recently,we showed that OX40L(TNFSF4)is able to drive selective proliferation of peripheral Tregs independent of canonical antigen presentation(CAP-independent)in the presence of low-dose IL-2.Therefore,we hypothesized that OX40 signaling might be integral to the TCR-independent phase of murine and human thymic Treg(tTreg)development.Development of tTregs is a two-step process:Strong T-cell receptor(TCR)signals in combination with co-signals from the TNFRSF members facilitate tTreg precursor selection,followed by a TCR-independent phase of tTreg development in which their maturation is driven by IL-2.Therefore,we investigated whether OX40 signaling could also play a critical role in the TCR-independent phase of tTreg development.OX40−/−mice had significantly reduced numbers of CD25−Foxp3low tTreg precursors and CD25+Foxp3+mature tTregs,while OX40L treatment of WT mice induced significant proliferation of these cell subsets.Relative to tTeff cells,OX40 was expressed at higher levels in both murine and human tTreg precursors and mature tTregs.In ex vivo cultures,OX40L increased tTreg maturation and induced CAP-independent proliferation of both murine and human tTregs,which was mediated through the activation of AKT-mTOR signaling.These novel findings show an evolutionarily conserved role for OX40 signaling in tTreg development and proliferation,and might enable the development of novel strategies to increase Tregs and suppress autoimmunity.Prabhakaran Kumar Alejandra Marinelarena Divya Raghunathan Vandhana K Ragothaman Shikha Saini Palash Bhattacharya Jilao Fan Alan L Epstein Ajay V Maker Bellur S Prabhakar 2019Cellular & Molecular Immunology2019,16,2:4
9Targeting cancer testis antigens for biomarkers and immunotherapy in colorectal cancer: Current status and challenges显示文摘Colorectal cancer ranks third among the estimatedcancer cases and cancer related mortalities in United States in 2014. Early detection and efficient therapy remains a significant clinical challenge for this disease. Therefore, there is a need to identify novel tumor asso-ciated molecules to target for biomarker development and immunotherapy. In this regard, cancer testis antigens have emerged as a potential targets for developing novel clinical biomarkers and immunotherapy for various malignancies. These germ cell specific proteins exhibit aberrant expression in cancer cells and contribute in tumorigenesis. Owing to their unique expression profile and immunogenicity in cancer patients, cancer testis antigens are clinically referred as the most promising tumor associated antigens. Several cancer testis antigens have been studied in colorectal cancer but none of them could be used in clinical practice. 11Expression of annexin II in gastric carcinoma and its role in gastric cancer metastasis显示文摘AIM To investigate the expression of annexin II in gastric carcinoma and its role in the metastasis of gastric cancer.METHODS The expression of annexin II in 51 cases of gastric carcinoma and 24 cases of adjacent tissues was detected by immunohistochemistry. The relationship between annexin II and clinical features of gastric cancer was analyzed. Annexin II specific si RNA was used to inhibit the expression of annexin II in gastric cancer HGC-27 cells, and the effects of annexin II on the migration and secretion of matrix metalloproteinases(MMPs) were observed.RESULTS The positive rate of annexin II protein was 82.4% in gastric cancer tissues and 37.5% in adjacent tissues. There was significant difference between the two groups(P < 0.01); and the positive expression of annexin II was not related to the sex and age of the patients(P > 0.05). The expression of annexin IIprotein was correlated with tumor size, histological differentiation, TNM stage, Lymph node metastasis and other clinical features were significantly correlated, the difference was statistically significant(P < 0.05). Inhibition of annexin II expression, gastric cancer HGC-27 cells migration and secretion of MMPs were significantly decreased, the difference was statistically significant(P < 0.05).CONCLUSION Annexin II is highly expressed in gastric cancer tissues, annexin II protein expression is related to tumor size, histological differentiation, TNM staging, lymph node metastasis and other clinical features were significantly correlated. 12Upconversion Nanoparticles-Encoded Hydrogel Microbeads-Based Multiplexed Protein Detection显示文摘Fluorescently encoded microbeads are in demand for multiplexed applications in different fields.Compared to organic dye-based commercially available Luminex's x MAP technology, upconversion nanoparticles(UCNPs) are better alternatives due to their large antiStokes shift, photostability, nil background, and single wavelength excitation. Here, we developed a new multiplexed detection system using UCNPs for encoding poly(ethylene glycol) diacrylate(PEGDA) microbeads as well as for labeling reporter antibody. However, to prepare UCNPs-encoded microbeads, currently used swellingbased encapsulation leads to non-uniformity, which is undesirable for fluorescence-based multiplexing. Hence,we utilized droplet microfluidics to obtain encoded microbeads of uniform size, shape, and UCNPs distribution inside. Additionally, PEGDA microbeads lack functionality for probe antibodies conjugation on their surface.Methods to functionalize the surface of PEGDA microbeads(acrylic acid incorporation, polydopamine coating)reported thus far quench the fluorescence of UCNPs. Here,PEGDA microbeads surface was coated with silica followed by carboxyl modification without compromising the fluorescence intensity of UCNPs. In this study, droplet microfluidics-assisted UCNPs-encoded microbeads of uniform shape, size, and fluorescence were prepared.Multiple color codes were generated by mixing UCNPs emitting red and green colors at different ratios prior to encapsulation. UCNPs emitting blue color were used to label the reporter antibody. Probe antibodies were covalently immobilized on red UCNPs-encoded microbeads for specific capture of human serum albumin(HSA) as a model protein. 13Multiple sclerosis:integration of modeling with biology,clinical and imaging measures to provide better monitoring of disease progression and prediction of outcome显示文摘Multiple Sclerosis(MS) is a major cause of neurological disability in adults and has an annual cost of approximately $28 billion in the United States. MS is a very complex disorder as demyelination can happen in a variety of locations throughout the brain; therefore, this disease is never the same in two patients making it very hard to predict disease progression. A modeling approach which combines clinical, biological and imaging measures to help treat and fight this disorder is needed. 15中国高速铁路是否显著影响城市土地利用——来自站点、线路和典型城市土地利用变化的回答显示文摘当前中国高速铁路正处于快速、全面发展阶段,研究高速铁路对城市土地利用的影响,对城市规划、城市与高速铁路的协调发展等均具有重要意义。利用GIS等技术提取已开通高速铁路站点周边、线路两侧土地利用数据以及部分城市高铁站点周边建设用地重心,从不同空间、城市类型、开通时间等角度对提取结果进行统计分析和图形对比。结果表明:高速铁路能通过促进各类型土地向建设用地转移来影响城市土地利用;高速铁路对站点周边土地利用的影响要大于对线路两侧的影响;高速铁路对城市土地利用的影响因城市规模类型不同而不同,对站点周围土地利用的影响总体有城市规模越大影响越大的特征,对特大城市、超大城市和县线路两侧土地利用的影响大于大中城市;高速铁路对城市土地利用的影响在时间上有递延性,在空间上有衰减性。郑林昌 Shikha Thapa Magar 2022河北大学学报(哲学社会科学版)2022,47,2:2
16Highlights from the literature on risk assessment techniques adopted in the mining industry:A review of past contributions,recent developments and future scope显示文摘In this paper a comprehensive review is presented of risk assessment techniques adopted in the mining industry worldwide;those techniques applied in other hazardous industries and potential techniques which are robust,mature and holistic and can be implemented for the Indian mining industry in future to enhance workplace safety are also presented.Findings from the review are indicative of the fact that socio-technical complexity of industrial systems has increased.Recent developments in the area of risk management highlight the need for implementation of the latest robust techniques of risk assessment in the mining sector.In consideration of the present scenario,the development of a model for risk analysis having an interface between hazard identification and risk assessment,along with an interface between risk assessment and accident causation to predict if an accident will occur under given conditions,has become dire necessity.This will increase hazard awareness and enable mine management to select and prioritize problem areas and identify safety system weaknesses in both underground and surface mining.This will ultimately help decision makers,risk analysts and safety managers make a major contribution in the development of workplace safety with a near-to-zero accident rate.Verma Shikha Chaudhari Sharad 2016International Journal of Mining Science and Technology2016,26,4:2
