11Effects of astaxanthin on antioxidant parameters in ARPE-19 cells on oxidative stress model显示文摘AIM: To observe the protective effect of astaxanthin(AST) against hydroquinone(HQ) mediated cell death in the apoptotic cascade and evaluate intracellular Ca2+ release, caspase-3, and-9 activation, reactive oxygen species(ROS) production in ARPE-19 cells.METHODS: We cultured ARPE-19 cells in special mediums and performed MTT tests to determine protective effect of AST, before exposing the cells to HQ in an incubator. We analyzed intracellular Ca2+ release experiments, mitochondrial membrane depolarization, glutathione(GSH), glutathione peroxidase(GSH-Px) and ROS experiments, and apoptosis assay.RESULTS: ROS production ranges depend on the amount of cell death. We computed the correlation between ROS ranges and cell death by 20,70-dichlorofluorescein fluorescence, and Ca2+ levels by Fura-2-AM. HQ-induced cell death found out to rise ranges of caspase-3 and-9, and mitochondrial depolarization. These three steps were delayed by AST management.CONCLUSION: ARPE-19 cells are avoided from HQinduced ROS production and caspase-3 and-9 activation by AST. AST may limit the range of caspase synthesis, Ca2+ release and excess production of ROS with antiapoptotic effect. 15玻璃体腔注射bevacizumab治疗视网膜分支静脉阻塞继发黄斑水肿(英文)显示文摘目的:通过测量随访12mo后的中央黄斑厚度(CMT)和视力,评估玻璃体腔注射bevacizumab治疗视网膜分支静脉阻塞(BRVO)继发黄斑水肿(ME)的效果。方法:被诊断为BRVO继发ME的患者行眼科检查、CMT测量、荧光素血管造影。排除荧光素血管造影出现黄斑缺血患者,其他疾病继发的其他部位新生血管化患者,有眼内治疗史(激光治疗、玻璃体腔注射或眼科手术)患者。CMT 〉250μm的32例患者给予bevacizumab(Altuzan,0.125mg/0.05mL)注射治疗,并随访12mo。分析最佳矫正视力(BCVA)logMAR数据和CMT控制参数。使用Minitab15.0软件统计分析配对t检验,P〈0.05有统计学意义。结果: BCVA logMAR数据和CMT控制参数的平均值较注射前有明显改变(P〈0.01)。平均最佳矫正视力增量为0.477±0.235,平均CMT较注射前下降257.906±88.865。10例(31%)患者对单次注射有阳性反应,平均12.6±0.66mo未复发ME。5例(15.6%)患者接受两次注射,17例(53%)3次以上。单眼平均注射量2.18±0.91(1~4)。第一组ME复发时间为2.45±0.63mo,第二组为2.58±0.66mo,第三组为3.17±0.48mo。5例(15.6%)患者需要多次注射以减轻ME,视力并未随ME的减轻而增加。结论:玻璃体腔注射bevacizumab是常规治疗BRVO继发ME的方法,有效、快速、安全。为了使疗效持久,需加强后续处理,通过激光或长效药物保持无水肿状态。视网膜静脉循环和ME须通过荧光素血管造影观察,而不能采取频繁注射。是否需再次注射必须根据ME的临床表现和视力的预测来判断。Seref Istek Ozlem Bursall Gürsoy Alagoz 2014国际眼科杂志2014,14,6:0
16Phenolic profile and investigation of biological activities of Allium scorodoprasum L.subsp.rotundum显示文摘In this study,phenolic profile of Allium scorodoprasum L.subsp.rotundum and its bioactivity in multiway approach including antioxidant,antimicrobial,antidiabetic,cytotoxic and antiinflamatory activities were investigated.The extraction of phenolic and flavonoid compounds from Allium scorodoprasum L.subsp.rotundum was optimized as a function of ethanol percentage,extraction temperature and extraction time by employing a response surface method.The optimal extraction conditions were found as 79℃extraction temperature,28%ethanol and 118 min duration of the extraction.Quercetin was determined as the major phenolic compound of the extract obtained under the optimal conditions and followed by vanillic,caffeic,2,4 hydroxybenzoic and ellagic acid.In addition to high antioxidant capacity,the obtained extract showed antimicrobial activity,against different bacteria and molds.The concentrations of the extract obtained at the optimal conditions that inhibited 50%of Aspergillus oryzae and pancreaticα-amylases andα-glycosidase activities were calculated as 11.56,14.35 and 19.35 mg extract/mL,respectively.The antiinflammatory effect of the obtained extract was also examined,and it was more inhibitory against xanthine oxidase activity than against lipoxygenase activity.The cytotoxicity of the optimized extract was observed against breast and bone cancer cells,and it showed significant cytotoxic activity especially on bone cancer cells.As a result,Allium scorodoprasum L.subsp.rotundum could serve as a promising source of natural bioactive compounds for application in food and non-food products.Tu˘gba Demir Ozlem Akpınar Haki Kara Hüseyin Güngor 2022Food Bioscience2022,46,2:0
17Estimation in the Complementary Exponential Geometric Distribution Based on Progressive Type-II Censored Data显示文摘Complementary exponential geometric distribution has many applications in survival and reliability analysis.Due to its importance,in this study,we are aiming to estimate the parameters of this model based on progressive type-II censored observations.To do this,we applied the stochastic expectation maximization method and Newton-Raphson techniques for obtaining the maximum likelihood estimates.We also considered the estimation based on Bayesian method using several approximate:MCMC samples,Lindely approximation and Metropolis-Hasting algorithm.In addition,we considered the shrinkage estimators based on Bayesian and maximum likelihood estimators.Then,the HPD intervals for the parameters are constructed based on the posterior samples from the Metropolis-Hasting algorithm.In the sequel,we obtained the performance of different estimators in terms of biases,estimated risks and Pitman closeness via Monte Carlo simulation study.This paper will be ended up with a real data set example for illustration of our purpose.Ozlem GürünlüAlma Reza Arabi Belaghi 2020Communications in Mathematics and Statistics2020,8,4:0
18Decline in subarachnoid haemorrhage volumes associated with the first wave of the COVID- 19 pandemic显示文摘Background During the COVID-19 pandemic,decreased volumes of stroke admissions and mechanical thrombectomy were reported.The study’s objective was to examine whether subarachnoid haemorrhage(SAH)hospitalisations and ruptured aneurysm coiling interventions demonstrated similar declines.Methods We conducted a cross-sectional,retrospective,observational study across 6 continents,37 countries and 140 comprehensive stroke centres.Patients with the diagnosis of SAH,aneurysmal SAH,ruptured aneurysm coiling interventions and COVID-19 were identified by prospective aneurysm databases or by International Classification of Diseases,10th Revision,codes.The 3-month cumulative volume,monthly volumes for SAH hospitalisations and ruptured aneurysm coiling procedures were compared for the period before(1 year and immediately before)and during the pandemic,defined as 1 March-31 May 2020.The prior 1-year control period(1 March-31 May 2019)was obtained to account for seasonal variation.Findings There was a significant decline in SAH hospitalisations,with 2044 admissions in the 3 months immediately before and 1585 admissions during the pandemic,representing a relative decline of 22.5%(95%CI−24.3%to−20.7%,p<0.0001).Embolisation of ruptured aneurysms declined with 1170-1035 procedures,respectively,representing an 11.5%(95%CI−13.5%to−9.8%,p=0.002)relative drop.Subgroup analysis was noted for aneurysmal SAH hospitalisation decline from 834 to 626 hospitalisations,a 24.9%relative decline(95%CI−28.0%to−22.1%,p<0.0001).A relative increase in ruptured aneurysm coiling was noted in low coiling volume hospitals of 41.1%(95%CI 32.3%to 50.6%,p=0.008)despite a decrease in SAH admissions in this tertile.Interpretation There was a relative decrease in the volume of SAH hospitalisations,aneurysmal SAH hospitalisations and ruptured aneurysm embolisations during the 19Petroselinum crispum extract ameliorates scopolamine-induced cognitive dysfunction:role on apoptosis,inflammation and oxidative stress显示文摘This study was designed to investigate whether Petroselinum crispum(PC)extract has protective effects on the brain in the scopolamine-induced Alzheimer’s disease(AD)rat model.The rats were divided into;control,scopolamine(1 mg/kg,i.p.),galantamine(1.5 mg/kg,i.p.)and PC extract(2 g/kg,p.o.)-treated scopolamine groups.On day 14,the novel object recognition test(NORT)and Morris water maze test(MWMT)were performed and then the rats were sacrificed.Scopolamine-induced cognitive impairments observed in the NORT and MWMT,significantly improved with PC extract and galantamine treatments.Scopolamine reduced M_(1) receptor expression,Bcl-2/Bax ratio,and glutathione levels in the hippocampus and frontal cortex,while malondialdehyde levels,caspase-3/9 expressions,and acetylcholinesterase(AChE)activity were found to be increased.On the other hand,PC and galantamine treatments reversed these changes.In conclusion,PC extract has shown an ameliorative effect on the spatial and recognition memory,M_(1) receptor expression,apoptosis,oxidative stress,and increased AChE activity.Thus,it was concluded that PC could prevent AD-like conditions and can be used as a functional food.However,since animal models do not completely mimic those of humans,based on the data obtained in this study,the importance of PC on human AD should be demonstrated in future studies.GökselŞener Gözde Karakadıoglu Dilek Ozbeyli Seren Ede Refiye Yanardag Ozlem Sacan Asli Aykac 2022Food Science and Human Wellness2022,11,5:0
