2Peripartal rumen-protected methionine supplementation to higher energy diets elicits positive effects on blood neutrophil gene networks, performance and liver lipid content in dairy cows显示文摘Background: Main objectives were to determine to what extent Smartamine M(SM) supplementation to a prepartal higher-energy diet could alter neutrophil(PMN) and liver tissue immunometabolic biomarkers, and whether those responses were comparable to those in cows fed a prepartal lower-energy diet(CON).Results: Twenty-eight multiparous Holstein cows were fed CON(NEL= 1.24 Mcal/kg DM) during d-50 to d-22 relative to calving. From d-21 to calving, cows were randomly assigned to a higher-energy diet(OVE, n = 9; NEL= 1.54 Mcal/kg DM), OVE plus SM(OVE + SM, n = 10; SM = 0.07 % of DM) or remained on CON(n = 9). Al cows received the same basal lactation diet(NEL= 1.75 Mcal/kg DM). Supplementation of SM(OVE + SM) continued until 30 d postpartum. Liver biopsies were harvested at d-10, 7, and 21 relative to parturition. Blood PMN isolated at-10, 3, and 21 d relative to calving was used to evaluate gene expression. As expected, OVE increased liver lipid content postpartum; however,cows fed OVE + SM or CON had lower concentrations than OVE. Compared with OVE, cows in CON and OVE + SM had greater DMI postpartum and milk production. Furthermore, cows fed OVE + SM had the greatest milk protein and fat percentage and lowest milk SCC despite having intermediate PMN phagocytic capacity. Adaptations in PMN gene expression in OVE + SM cows associated with the lower SCC were gradual increases from-10 to 21 d in genes that facilitate migration into inflammatory sites(SELL, ITGAM), enzymes essential for reducing reactive oxygen metabolites(SOD1, SOD2), and a transcription factor(s) required for controlling PMN development(RXRA). The greater expression of TLR4 on d 3, key for activation of innate immunity due to inflammation, in OVE compared with CON cows suggests a more pronounced inflammatory state. Feeding OVE + SM dampened the upregulation of TLR4, despite the fact that these cows had similar expression of the pro-inflammatory genes NFKB1 and TNF as OVE. Cows in CON had lower overall expression of these inflammation-related genes and GSR, which generates reduced glutathione, an important cellular antioxidant.Conclusions: Although CON cows appeared to have a less stressful transition into lactation, SM supplementation was effective in alleviating negative effects of energy-overfeeding. 4An experimental validation of genomic selection in octoploid strawberry显示文摘The primary goal of genomic selection is to increase genetic gains for complex traits by predicting performance of individuals for which phenotypic data are not available.The objective of this study was to experimentally evaluate the potential of genomic selection in strawberry breeding and to define a strategy for its implementation.Four clonally replicated field trials,two in each of 2 years comprised of a total of 1628 individuals,were established in 2013–2014 and 2014–2015.Five complex yield and fruit quality traits with moderate to low heritability were assessed in each trial.High-density genotyping was performed with the Affymetrix Axiom IStraw90 single-nucleotide polymorphism array,and 17479 polymorphic markers were chosen for analysis.Several methods were compared,including Genomic BLUP,Bayes B,Bayes C,Bayesian LASSO Regression,Bayesian Ridge Regression and Reproducing Kernel Hilbert Spaces.Cross-validation within training populations resulted in higher values than for true validations across trials.For true validations,Bayes B gave the highest predictive abilities on average and also the highest selection efficiencies,particularly for yield traits that were the lowest heritability traits.Selection efficiencies using Bayes B for parent selection ranged from 74%for average fruit weight to 34%for early marketable yield.A breeding strategy is proposed in which advanced selection trials are utilized as training populations and in which genomic selection can reduce the breeding cycle from 3 to 2 years for a subset of untested parents based on their predicted genomic breeding values.Salvador A Gezan Luis F Osorio Sujeet Verma Vance M Whitaker 2017Horticulture Research2017,4,1:6
5Early-onset colorectal cancer:A separate subset of colorectal cancer显示文摘Colorectal cancer(CRC)has a great impact on the world population.With increasing frequency,CRC is described according to the presenting phenotype,based on its molecular characteristics.Classification of CRC tumors according to their genetic and/or epigenetic alterations is not only important for establishing the molecular bases of the disease,but also for predicting patient outcomes and developing more individualized treatments.Early-onset CRC is a heterogeneous disease,with a strong familial component,although the disease is sporadic in an important proportion of cases.Different molecular alterations appear to contribute to the apparent heterogeneity of the early-onset population and subgroups can be distinguished with distinct histopathologic and familial characteristics.Moreover,compared with late-onset CRC,there are characteristicsthat suggest that early-onset CRC may have a different molecular basis.The purpose of this review was to analyze the current state of knowledge about earlyonset CRC with respect to clinicopathologic,familial and molecular features.Together,these features make it increasingly clear that this subset of CRC may be a separate disease,although it has much in common with late-onset CRC.Irene Osorio Silla Daniel Rueda Yolanda Rodríguez Juan Luis García Felipe de la Cruz Vigo José Perea 2014World Journal of Gastroenterology2014,20,46:4
6Identification of quantitative trait loci and candidate genes for primary metabolite content in strawberry fruit显示文摘Improvement of nutritional and organoleptic quality of fruits is a key goal in current strawberry breeding programs.The ratio of sugars to acids is a determinant factor contributing to fruit liking,although different sugars and acids contribute in varying degrees to this complex trait.A segregating F1 population of 95 individuals,previously characterized for several fruit quality characters,was used to map during 2 years quantitative trait loci(QTL)for 50 primary metabolites,L-ascorbic acid(L-AA)and other related traits such as soluble solid content(SSC),titratable acidity(TA),and pH.A total of 133 mQTL were detected above the established thresholds for 44 traits.Only 12.9%of QTL were detected in the 2 years,suggesting a large environmental influence on primary metabolite content.An objective of this study was the identification of key metabolites that were associated to the overall variation in SSC and acidity.As it was observed in previous studies,a number of QTL controlling several metabolites and traits were co-located in homoeology group V(HG V).mQTL controlling a large variance in raffinose,sucrose,succinic acid,and L-AA were detected in approximate the same chromosomal regions of different homoeologous linkage groups belonging to HG V.Candidate genes for selected mQTL are proposed based on their co-localization,on the predicted function,and their differential gene expression among contrasting F1 progeny lines.RNA-seq analysis from progeny lines contrasting in L-AA content detected 826 differentially expressed genes and identified Mannose-6-phosphate isomerase,FaM6PI1,as a candidate gene contributing to natural variation in ascorbic acid in strawberry fruit.JoséG.Vallarino Delphine M.Pott Eduardo Cruz-Rus Luis Miranda Juan J.Medina-Minguez Victoriano Valpuesta Alisdair R.Fernie JoséF.Sánchez-Sevilla Sonia Osorio Iraida Amaya 2019Horticulture Research2019,6,1:3

12A scoping review on the field validation and implementation of rapid diagnostic tests for vector-borne and other infectious diseases of poverty in urban areas显示文摘Background:Health personnel face challenges in diagnosing vector-borne and other diseases of poverty in urban settings.There is a need to know what rapid diagnostic technologies are available,have been properly assessed,and are being implemented to improve control of these diseases in the urban context.This paper characterizes evidence on the field validation and implementation in urban areas of rapid diagnostics for vector-borne diseases and other diseases of poverty.Main body:A scoping review was conducted.Peer-reviewed and grey literature were searched using terms describing the targeted infectious diseases,diagnostics evaluations,rapid tests,and urban setting.The review was limited to studies published between 2000 and 2016 in English,Spanish,French,and Portuguese.Inclusion and exclusion criteria were refined post hoc to identify relevant literature regardless of study design and geography.A total of 179 documents of the 7806 initially screened were included in the analysis.Malaria(n=100)and tuberculosis(n=47)accounted for the majority of studies that reported diagnostics performance,impact,and implementation outcomes.Fewer studies,assessing mainly performance,were identified for visceral leishmaniasis(n=9),filariasis and leptospirosis(each n=5),enteric fever and schistosomiasis(each n=3),dengue and leprosy(each n=2),and Chagas disease,human African trypanosomiasis,and cholera(each n=1).Reported sensitivity of rapid tests was variable depending on several factors.Overall,specificities were high(>80%),except for schistosomiasis and cholera.Impact and implementation outcomes,mainly acceptability and cost,followed by adoption,feasibility,and sustainability of rapid tests are being evaluated in the field.Challenges to implementing rapid tests range from cultural to technical and administrative issues.Conclusions:Rapid diagnostic tests for vector-borne and other diseases of poverty are being used in the urban context with demonstrated impact on case detection.However,most evidence comes from malaria rapid diagnostics,with variable results.While rapid tests for tuberculosis and visceral leishmaniasis require further implementation studies,more evidence on performance of current tests or development of new alternatives is needed for dengue,Chagas disease,filariasis,leptospirosis,enteric fever,human African trypanosomiasis,schistosomiasis and cholera.Lyda Osorio Jonny Alejandro Garcia Luis Gabriel Parra Victor Garcia Laura Torres Stephanie Degroote Valery Ridde 2018Infectious Diseases of Poverty2018,7,1:2
