| 1 | Effects of thymol and carvacrol supplementation on intestinal integrity and immune responses of broiler chickens challenged with Clostridium perfringens显示文摘Background: Necrotic enteritis caused by Clostridium perfringens infection leads to serious economic losses in the global poultry production. In the present study, we investigated the protective effects of essential oils(EO, which contained 25 % thymol and 25 % carvacrol as active components) supplementation on growth performance, gut lesions, intestinal morphology, and immune responses of the broiler chickens infected with C. perfringens. A total of448 1-day-old male broiler chicks were allocated into eight treatment groups following a 4 × 2 factorial arrangement with four dietary EO dosages(0, 60, 120, or 240 mg/kg) and two infection status(with or without C. perfringens challenge from d 14 to 20).Results: The challenge did not impair the growth performance of birds, but induced gut lesions and increased crypt depth in the ileum(P ≤ 0.05). It also down-regulated the claudin-1 and occludin m RNA expression(P ≤ 0.05),up-regulated the m RNA expression of interleukin-1β(P ≤ 0.05), tended to increase the toll-like receptor(TLR)2 m RNA expression(P < 0.10) in the ileum, and enhanced the mucosal secretory Ig A production(P ≤ 0.05). In the challenged birds, dietary EO supplementation linearly alleviated the gut lesions and improved the ratio of vil us height to crypt depth(P ≤ 0.05), and the supplementation of 120 and 240 mg/kg EO increased the serum antibody titers against Newcastle disease virus(P ≤ 0.05). | 2 | Effects of Bacillus coagulans supplementation on the growth performance and gut health of broiler chickens with Clostridium perfringens-induced necrotic enteritis显示文摘Background: The poultry industry is in need of effective antibiotic alternatives to control outbreaks of necrotic enteritis(NE) due to Clostridium perfringens.Methods: This study was conducted to investigate the effects of feeding Bacil us coagulans on the growth performance and gut health of broiler chickens with C. perfringens-induced NE. Two hundred and forty 1-day-old broiler chicks were randomly assigned to a 2 × 2 factorial arrangement with two dietary B. coagulans levels(0 or 4 × 109 CFU/kg of diet) and two disease chal enge statuses(control or NE chal enged).Results: NE-induced reduction in body weight gain was relieved by the addition of B. coagulans into broiler diets compared with the NE-infected birds. NE infection damaged intestinal morphological structure, promoted intestinal C.perfringens growth and liver invasion, and enhanced anti-C. perfringens specific sI gA concentrations in the gut and specific IgG levels in serum compared with the uninfected birds. NE infection significantly(P < 0.05) decreased mucin-2(at 14 d post-infection(DPI), tol-like receptor 2(TLR2, at 7 and 14 DPI), TLR4(at 7 and 14 DPI), tumor necrosis factor super family15(TNFSF15, at 7 and 14 DPI), lysozyme(LYZ, at 14 DPI) and fowlicidin-2(at 7 and 14 DPI) mR NA levels, whereas it dramatical y(P = 0.001) increased IFN-γ mR NA levels at 7 DPI. However, chal enged birds fed diets supplemented with B.coagulans showed a significant(P < 0.01) decrease in gut lesion scores, decreased C. perfringens numbers in the cecum and liver, and an increase in fowlicidin-2 mR NA levels in compared with the uninfected birds. Using a 2 × 2 factorial arrangement of treatments, a total of 308 1-day-old male Arbor Acres broiler chicks were included to investigate the effects of Lactobacil us acidophilus(L. acidophilus) on the growth performance and intestinal health of broiler chickens chal enged with Clostridium perfringens(C. perfringens) during a 28-day trial.Results: During infection(d 14–21), C. perfringens challenge decreased the average daily gain(P < 0.05), and increased feed conversion ratio and the mortality rate(P < 0.05). However, dietary supplementation with L. acidophilus increased the body weight of C. perfringens-infected broilers on d 21(P < 0.05), and tended to decrease the mortality(P = 0.061).C. perfringens challenge decreased the villus height(P < 0.05), the ratio of villus height to crypt depth(P < 0.05) and OCLN(occludin) m RNA expression(P < 0.05), and increased the pro-inflammatory cytokine expression in the spleen and jejunum, the intestinal populations of C. perfringens and Escherichia(P < 0.05), and the serum content of endotoxin(P < 0.05), regardless of L. acidophilus supplementation. | 7 | Effect of storage time on the characteristics of corn and efficiency of its utilization in broiler chickens显示文摘Corn is one of the staple food and feed ingredients in China, therefore its storage is of particular importance. Corn is typically stored for 2 or more years in national barns before it is sold as a food or feed ingredient. However, the effects of stored corn in national barns on the animal performance and nutrient utilization have not been investigated thus far. This study attempted to determine the effects of storage time on the chemical and physical characteristics of corn and its nutritional value, broiler growth performance, and meat quality. Corn grains used in the present study were stored for 4 different periods,from 2 to 5 yr, under the same conditions in a building at the Beijing National Grain Storage Facility. A total of 240 birds in Exp. 1 and 90 birds in Exp. 2 were used to compare the effects of storage time on the utilization of nutrients of corn, the performance, and meat quality of broilers. The content of starch, crude protein, amino acids, fatty acids, and test weight generally decreased with increasing storage time. Corn stored for over 4 yr showed decreased catalase(CAT) and peroxidase(POD) activities and increased fat acidity. Body weight gain(BWG) and European production index(EPI) of broilers from 0 to 3 wk tended to decrease linearly with storage time(0.05 < P < 0.10), and the BWG and EPI of broilers from 4 to 6 wk decreased quadratically(P < 0.05), whereas feed conversion ratio(FCR) increased with storage time(P < 0.05). The FCR, performance, and EPI of broilers positively correlated with CAT activity(P < 0.05), and negatively correlated with fat acidity(P < 0.05). Drip loss of breast muscle increased linearly with corn storage time(P < 0.001); however, pH decreased linearly with corn storage time. Drip loss had a strong negative correlation with POD(P < 0.05). There were no significant differences of the storage length on metabolizable energy(ME), digestibility of crude protein, and starch(P > 0.05). The digestibility of histidine and arginine, and C18:2 and C18:3 changed quadratically with storage time(P < 0.05).Collectively, the results suggest that the use of corn stored for 4 yr in animal feed decreased the performance and meat quality of broilers. | 8 | In vitro antibacterial activity of thymol and carvacrol and their effects on broiler chickens challenged with Clostridium perfringens显示文摘Background:In the post-antibiotic era,essential oils(EO) are promising alternatives to growth-promoting antibiotics.The aim of the present study was to investigate the antibacterial activities of an EO product and its components thymol and carvacrol in vitro,and the efficacy of EO to control Clostridium perfringens challenge in broiler chickens.Results:The in vitro minimum inhibitory concentration assay showed strong antibacterial activity of the EO product,thymol,and carvacrol against pathogenic Escherichia coli,C.perfringens,and Salmonella strains,and weak activity towards beneficial Lactobacillus strains.Besides,an additive effect was observed between thymol and carvacrol.The in vivo study was carried out with 448 male broiler chicks following a 4 × 2 factorial arrangement to test the effects of EO supplementation(0,60,120,or 240 mg/kg EO in wheat-based diet),pathogen challenge(with or without oral gavage of C perfringens from day 14 to day 20) and their interactions.Each treatment consisted of eight replicate pens(seven birds/pen).The challenge led to macroscopic gut lesions,and resulted in a significant increase in ileal populations of C.perfringens and Escherichia subgroup(P< 0.05) on day 21.Dietary EO supplementation did not influence C perfringens numbers,but linearly alleviated intestinal lesions on day 21 and 28(P=0.010 and 0.036,respectively),and decreased Escherichia populations in ileum with increased EO dosages(P=0.027 and 0.071 for day 21 and 28,respectively).For caecum,EO quadratically influenced Lactobacillus populations on day 21(p=0.002),and linearly decreased the numbers of total bacteria and Escherichia on day 28(P=0.026 and 0.060,respectively).Mean thymol and carvacrol concentrations in the small intestine were 0.21 and 0.20 μg/g in intestinal digesta(wet weight),respectively,for birds fed 60 mg/kg EO,and 0.80 and 0.71 μg/g,respectively,for birds fed 240 mg/kg EO.Conclusions:These results indicated that dietary EO supplementation could affect intestinal microbiota and alleviate intestinal lesions in broilers,which may contribute in controlling C.perfringens infection in broiler chickens. | Encun Du Liping Gan Zhui Li Weiwei Wang Dan Liu Yuming Guo | 2016 | Journal of Animal Science and Biotechnology2016,7,1: | 14 |
| 9 | Landscape of DILI-related adverse drug reaction in China Mainland显示文摘Drug-induced liver injury(DILI)is a type of bizarre adverse drug reaction(ADR)damaging liver(L-ADR)which may lead to substantial hospitalizations and mortality.Due to the general low incidence,detection of L-ADR remains an unsolved public health challenge.Therefore,we used the data of 6.673 million of ADR reports from January 1st,2012 to December 31st,2016 in China National ADR Monitoring System to establish a new database of L-ADR reports for future investigation.Results showed that totally 114,357 ADR reports were retrieved by keywords searching of liver-related injuries from the original heterogeneous system.By cleaning and standardizing the data fields by the dictionary of synonyms and English translation,we resulted 94,593 ADR records reported to liver injury and then created a new database ready for computer mining.The reporting status of L-ADR showed a persistent 1.62-fold change over the past five years.The national population-adjusted reporting numbers of L-ADR manifested an upward trend with age increasing and more evident in men.The annual reporting rate of L-ADR in age group over 80 years old strikingly exceeded the annual DILI incidence rate in general population,despite known underreporting situation in spontaneous ADR reporting system.The percentage of herbal and traditional medicines(H/TM)L-ADR reports in the whole number was 4.5%,while 80.60%of the H/TM reports were new findings.There was great geographical disparity of reported agents,i.e.more cardiovascular and antineoplastic agents were reported in higher socio-demographic index(SDI)regions and more antimicrobials,especially antitubercular agents,were reported in lower SDI regions.In conclusion,this study presented a large-scale,unbiased,unified,and computer-minable L-ADR database for further investigation.Age-,sex-and SDI-related risks of L-ADR incidence warrant to emphasize the precise pharmacovigilance policies within China or other regions in the world. | Jiabo Wang Haibo Song Feilin Ge Peng Xiong Jing Jing Tingting He Yuming Guo Zhuo Shi Chao Zhou Zixin Han Yanzhong Han Ming Niu Zhaofang Bai Guangbin Luo Chuanyong Shen Xiaohe Xiao | 2022 | Acta Pharmaceutica Sinica B2022,12,12: | 12 |

| 11 | Influence of starch sources and dietary protein levels on intestinal functionality and intestinal mucosal amino acids catabolism in broiler chickens显示文摘Background: There is growing interest in carbohydrate and protein nutrition to enhance the efficiency of animal production.Reduced-crude protein diets depress environmental pollution and feeding cost,but the challenge to their adoption is maintaining digestive function and growth performance of birds.The present study was conducted to evaluate the influence of different dietary starch sources and protein levels on intestinal functionality and mucosal amino acid catabolism.Methods: Six dietary treatments,based on maize and soybean meal,were offered to 360 AA+male chicks from 6 to 35 d post-hatch as a 3 × 2 factorial array.Either waxy rice or amylose was added to a conventional maize-soy diet to provide three sources of starch with different digestion rates and relatively high and low dietary protein levels.Growth performance,parameters of intestinal functionality and concentrations of free amino acid in the portal circulation were determined.Results: In the grower phase,starch source influenced(P < 0.02) weight gain as diets containing amylose supported significantly higher weight gains than waxy rice.Significant increase of ileal ATP concentrations and Na^+/K^+-ATPase activity were found in amylose treatment.Also,amylose decreased BrdU positive cell numbers and down-regulated m RNA expression for CASP-3.GOT activity in the ileum was higher(P < 0.01) in birds offered low protein diets and there was a trend(P = 0.057) for waxy rice as a starch source to increase ileal GOT activities.There was a significant influence on the concentration of seventeen amino acids in the portal circulation with tryptophan the one exception.Waxy rice as a starch source generated 13.6% and 22.4% numerically higher concentrations of non-essential amino acids than maize and amylose,respectively.Conclusions: Amino acid catabolism in the gut mucosa is subject to nutritional regulation.Given that amino acids can be spared from catabolism in the gut mucosa by supplementation of amylose,it follows their post-enteral availability would be improved and intestinal energy would be derived more efficiently from glucose. | Dafei Yin Peter H.Selle Amy F.Moss Youli Wang Xiaoyu Dong Zhibin Xiao Yuming Guo Jianmin Yuan | 2019 | Journal of Animal Science and Biotechnology2019,10,3: | 10 |

Friction coefficients were recorded at various speeds. | 16 | Wear behavior of WC-Ni sliding against graphite under water lubrication显示文摘Wear of hard materials in contact with softer materials is a neglected area of research. However, we observed considerable wear phenomenon at a hard WC-Ni surface sliding over soft graphite under water lubrication. The influences of applied load and the application history on the wear of surface were addressed in our experimental design. Wear of both graphite and WC-Ni surfaces increased with a greater applied load and repeated sliding. The topographies of the worn surfaces showed clear micro-scratches on the hard WC-Ni surface. Scanning electron microscopy and X-ray photoelectron spectroscopy analyses revealed that the abrasive wear of the WC-Ni surface could be attributed to hard WC particles embedded in the graphite surface. | 18 | Soya saponin improves egg-laying performance and immune function of laying hens显示文摘Background:Soya saponin(SS),an active compound in soybean meals,has been widely studied in the medical field.However,it was considered as an anti-nutritional factor in poultry diets.The objective of this experiment was to measure the effects of dietary SS using three dietary treatments on egg-laying performance and immune function of laying hens.Birds were fed a low soybean meal basal diet(CON),a low-SS diet(50 SS)containing 50mg/kg SS,or a high-SS diet(500 SS)containing 500 mg/kg SS for 10 weeks.At the end of the 5th and 10th week of the trial,samples were collected for analysis.Results:Results showed that with 50 mg/kg SS supplementation,the egg production rate,feed conversion ratio(FCR),and eggshell quality tended to be improved.Serum follicle stimulating hormone(FSH)and Interleukin-4(IL-4)levels were also elevated as well as the peripheral blood LPS stimulation index,the proportion of B lymphocytes,and antibody titer of bovine serum albumin(BSA).We also found that mRNA levels of follicle stimulating hormone receptor(FSHR)in ovarian,nuclear transcription factor kappa B(NF-κB),Transforming growth factor(TGF-β)and interferonγ(IFN-γ)in spleen were up-regulated at the end of the trial.Additionally,dietary 50 mg/kg SS improved the ileal flora via up-regulating the relative abundance of Lactobacillus,Romboutsia and Lactobacillus delbrueckii.Although the immune related indicators were improved with 500 mg/kg SS supplemented,it seemed to have a negative influence on the laying-performance.Specifically,serum alanine aminotransferase(ALT),alkaline phosphatase(ALP),and the ratio of IFN-γto IL-4 were increased in the 500 SS group at the end of the trial.The m RNA levels of gonadotropin releasing hormone 1(GnRH1)in Hypothalamus,the estrogen related receptor(ERR)in ovaries were downregulated as well as the egg production rate during the trial with 500 mg/kg SS supplemented.Conclusions:The egg production performance was improved by dietary supplemented with 50 mg/kg SS via increasing ovarian FSHR transcription level and serum estrogen level.A beneficial shift in intestinal microflora was recorded,and the immune function of laying hens was also improved with 50 mg/kg SS supplementation.Surprisingly,the long-term supplementation of 500 mg/kg SS exerted a negative impact on the laying performance and physiological functions of the liver of laying hens. | Peng Li Yizhu Zhao Shaojia Yan Bocheng Song Yongfa Liu Mingkun Gao Dazhi Tang Yuming Guo | 2022 | Journal of Animal Science and Biotechnology2022,13,3: | 5 |

| 20 | Analysis and finite element simulation for mechanical response of wheat to wind and rain loads显示文摘The morphological characteristics and stalk biomechanical properties at the dough stage of wheat were determined using the variety of wheat in the breeding process.Their mechanical responses to wind and rain loads for an individual and a group of wheat were simulated using finite element method by ANSYS.The stress and displacement of each finite element can be outputted through stress nephogram and displacement nephogram,respectively.In order to judge whether the wheat could return to its original position after deformation,elastic mechanics theory was utilized to analyze the critical load of instability under both axial rain load and transverse wind load.The large displacement situation was analyzed with large displacement elastic nonlinear theory and the numerical value was obtained by ANSYS.The results show that it is possible to apply various load types on models using ANSYS and the dynamic response can be simulated well under different rain and wind loads.The location of maximum Von Mises stress can be calculated and the variation of stress can be described clearly,which are helpful to predict the wheat lodging under wind and rain loads. | Liang Li Guo Yuming | 2008 | International Journal of Agricultural and Biological Engineering2008,1,1: | 4 |
