1High-contrast,fast chemical imaging by coherent Raman scattering using a self-synchronized two-colour fibre laser显示文摘Coherent Raman scattering(CRS)microscopy is widely recognized as a powerful tool for tackling biomedical problems based on its chemically specific label-free contrast,high spatial and spectral resolution,and high sensitivity.However,the clinical translation of CRS imaging technologies has long been hindered by traditional solid-state lasers with environmentally sensitive operations and large footprints.Ultrafast fibre lasers can potentially overcome these shortcomings but have not yet been fully exploited for CRS imaging,as previous implementations have suffered from high intensity noise,a narrow tuning range and low power,resulting in low image qualities and slow imaging speeds.Here,we present a novel high-power self-synchronized two-colour pulsed fibre laser that achieves excellent performance in terms of intensity stability(improved by 50 dB),timing jitter(24.3 fs),average power fluctuation(<0.5%),modulation depth(>20 dB)and pulse width variation(<1.8%)over an extended wavenumber range(2700-3550 cm^(−1)).The versatility of the laser source enables,for the first time,high-contrast,fast CRS imaging without complicated noise reduction via balanced detection schemes.These capabilities are demonstrated in this work by imaging a wide range of species such as living human cells and mouse arterial tissues and performing multimodal nonlinear imaging of mouse tail,kidney and brain tissue sections by utilizing second-harmonic generation and twophoton excited fluorescence,which provides multiple optical contrast mechanisms simultaneously and maximizes the gathered information content for biological visualization and medical diagnosis.This work also establishes a general scenario for remodelling existing lasers into synchronized two-colour lasers and thus promotes a wider popularization and application of CRS imaging technologies.Cihang Kong Christian Pilger Henning Hachmeister Xiaoming Wei Tom H.Cheung Cora S.W.Lai Nikki P.Lee Kevin K.Tsia Kenneth K.Y.Wong Thomas Huser 2020Light(Science & Applications)2020,9,1:5
2Structural, elemental, optical and magnetic study of Fe doped ZnO and impurity phase formation显示文摘We have prepared a series of(ZnO)1-x(Fe2O3)x≤0.10bulk samples with various concentrations of Fe dopant by ball milling and investigated their structural, compositional, optical and magnetic properties by means of X-ray diffraction(XRD), X-ray photoelectron spectroscopy(XPS),Raman spectrometer and vibrating sample magnetometer(VSM), respectively. Information about different impurity phases was obtained through Rietveld refinements of XRD data analysis. XPS results showed different valence states(Fe2+ tand Fe3+) supported by shaking satellite peaks in samples. With increasing Fe doping percentage, the crystal quality deteriorated and a shift of E2 low band(characteristic of ZnO) has been observed in Raman spectra. Energy band gap estimated from reflectance UV–vis spectroscopy showed shift for all bulk samples. The magnetic behavior was examined using a vibrating sample magnetometer(VSM), indicating ferromagnetic behavior at room temperature(300 K). 3Growth and Patterning of Pt Nanowires on Silicon Substrates显示文摘This paper describes a simple procedure for growing dense fi lms of Pt nanowires directly on silicon substrates by modifying the surface through chemical or physical means.In the former,a self-assembled monolayer of(3-mercaptopropyl)-trimethoxysilane(MPTMS)is applied which can strongly bind Pt(0)nuclei to the surface through Pt S linkages.Once attached,the Pt(0)nuclei can act as catalytic sites for the growth of Pt nanowires along the〈111〉direction.Alternately,relief features are physically created on the surface in order to generate nucleation and binding sites for Pt(0)nuclei,due to the higher free energy associated with a rough surface.Additionally,Pt nanowires have been successfully produced in well-defined patterns by scouring grooves on the silicon surface or by photochemically patterning the MPTMS monolayers with a shadow mask.We have also measured the electrochemical properties of these immobilized or patterned Pt nanowires.The results provide an effective route to producing dense fi lms of Pt nanowires with high surface areas for various electrochemical applications.Eric P.Lee Younan Xia 2008Nano Research2008,1,2:3
4Whole-blood sorting,enrichment and in situ immunolabeling of cellular subsets using acoustic microstreaming显示文摘Analyzing undiluted whole human blood is a challenge due to its complex composition of hematopoietic cellular populations,nucleic acids,metabolites,and proteins.We present a novel multi-functional microfluidic acoustic streaming platform that enables sorting,enrichment and in situ identification of cellular subsets from whole blood.This single device platform,based on lateral cavity acoustic transducers(LCAT),enables(1)the sorting of undiluted donor whole blood into its cellular subsets(platelets,RBCs,and WBCs),(2)the enrichment and retrieval of breast cancer cells(MCF-7)spiked in donor whole blood at rare cell relevant concentrations(10 mL^(−1)),and(3)on-chip immunofluorescent labeling for the detection of specific target cellular populations by their known marker expression patterns.Our approach thus demonstrates a compact system that integrates upstream sample processing with downstream separation/enrichment,to carry out multi-parametric cell analysis for blood-based diagnosis and liquid biopsy blood sampling.Neha Garg Trisha M.Westerhof Vick Liu Robin Liu Edward L.Nelson Abraham P.Lee 2018Microsystems & Nanoengineering2018,4,1:3
5Plasmonic bacteria on a nanoporous mirror via hydrodynamic trapping for rapid identification of waterborne pathogens显示文摘A rapid,precise method for identifying waterborne pathogens is critically needed for effective disinfection and better treatment.However,conventional methods,such as culture-based counting,generally suffer from slow detection times and low sensitivities.Here,we developed a rapid detection method for tracing waterborne pathogens by an innovative optofluidic platform,a plasmonic bacteria on a nanoporous mirror,that allows effective hydrodynamic cell trapping,enrichment of pathogens,and optical signal amplifications.We designed and simulated the integrated optofluidic platform to maximize the enrichment of the bacteria and to align bacteria on the nanopores and plasmonic mirror via hydrodynamic cell trapping.Gold nanoparticles are self-assembled to form antenna arrays on the surface of bacteria,such as Escherichia coli and Pseudomonas aeruginosa,by replacing citrate with hydroxylamine hydrochloride in order to amplify the signal of the plasmonic optical array.Owing to the synergistic contributions of focused light via the nanopore geometry,self-assembled nanoplasmonic optical antennas on the surface of bacteria,and plasmonic mirror,we obtain a sensitivity of detecting E.coli as low as 102 cells/ml via surface-enhanced Raman spectroscopy.We believe that our label-free strategy via an integrated optofluidic platform will pave the way for the rapid,precise identification of various pathogens.Keumrai Whang Jong-Hwan Lee Yonghee Shin Wooju Lee Young Wan Kim Dongchoul Kim Luke P.Lee Taewook Kang 2018Light(Science & Applications)2018,7,1:2
6A modular microfluidic system based on a multilayered configuration to generate large-scale perfusable microvascular networks显示文摘The vascular network of the circulatory system plays a vital role in maintaining homeostasis in the human body.In this paper,a novel modular microfluidic system with a vertical two-layered configuration is developed to generate largescale perfused microvascular networks in vitro.The two-layer polydimethylsiloxane(PDMS)configuration allows the tissue chambers and medium channels not only to be designed and fabricated independently but also to be aligned and bonded accordingly.This method can produce a modular microfluidic system that has high flexibility and scalability to design an integrated platform with multiple perfused vascularized tissues with high densities.The medium channel was designed with a rhombic shape and fabricated to be semiclosed to form a capillary burst valve in the vertical direction,serving as the interface between the medium channels and tissue chambers.Angiogenesis and anastomosis at the vertical interface were successfully achieved by using different combinations of tissue chambers and medium channels.Various large-scale microvascular networks were generated and quantified in terms of vessel length and density.Minimal leakage of the perfused 70-kDa FITC-dextran confirmed the lumenization of the microvascular networks and the formation of tight vertical interconnections between the microvascular networks and medium channels in different structural layers.This platform enables the culturing of interconnected,large-scale perfused vascularized tissue networks with high density and scalability for a wide range of multiorgan-on-a-chip applications,including basic biological studies and drug screening.Tao Yue Da Zhao Duc T.T.Phan Xiaolin Wang Joshua Jonghyun Park Zayn Biviji Christopher C.W.Hughes Abraham P.Lee 2021Microsystems & Nanoengineering2021,7,1:2

15Biodegradable magnesium materials regulate ROS-RNS balance in pro-inflammatory macrophage environment显示文摘The relationship between reactive oxygen and nitrogen species(ROS-RNS)secretion and the concomitant biocorrosion of degradable magnesium(Mg)materials is poorly understood.We found that Mg foils implanted short term in vivo(24 h)displayed large amounts of proinflammatory F4/80+/iNOS+macrophages at the interface.We sought to investigate the interplay between biodegrading Mg materials(98.6%Mg,AZ31&AZ61)and macrophages(RAW 264.7)stimulated with lipopolysaccharide(RAW 264.7^(LPS))to induce ROS-RNS secretion.To test how these proinflammatory ROS-RNS secreting cells interact with Mg corrosion in vitro,Mg and AZ61 discs were suspended approximately 2 mm above a monolayer of RAW 264.7 cells,either with or without ^(LPS).The surfaces of both materials showed acute(24 h)changes when incubated in the proinflammatory RAW 264.7^(LPS) environment.Mg discs incubated with RAW 264.7^(LPS) macrophages showed greater corrosion pitting,while AZ61 showed morphological and elemental bulk product changes via scanning electron microscopy-energy dispersive X-ray spectroscopy(SEM-EDX).X-ray photoelectron spectroscopy(XPS)analysis showed a reduction in the Ca/P ratio of the surface products for AZ61 disc incubated with RAW 264.7^(LPS),but not the Mg discs.Moreover,RAW 264.7^(LPS) macrophages were found to be more viable in the acute biodegradative environment generated by Mg materials,as demonstrated by calcein-AM and cleaved(active)caspase-3 staining(CC3).^(LPS) stimulation caused an increase in ROS-RNS,and a decrease in antioxidant peroxidase activity.Mg and AZ61 were found to change this ROS-RNS balance,independently of physiological antioxidant mechanisms.The findings highlight the complexity of the cellular driven acute inflammatory responses to different biodegradable Mg,and how it can potentially affect performance of these materials.Maria P.Kwesiga Amani A.Gillette Fatemeh Razaviamri Margaret E.Plank Alexia L.Canull Zachary Alesch Weilue He Bruce P.Lee Roger J.Guillory II 2023Bioactive Materials2023,,5:0
