2Accessible chromatin regions and their functional interrelations with gene transcription and epigenetic modifications in sorghum genome显示文摘Accessible chromatin regions(ACRs)provide physical scaffolds to recruit transcriptional co-regulators and displace their nearby nucleosomes in multiple plant species.Characterization of ACRs and investigation of their biological effects in Sorghum bicolor has lagged behind.Regulation of gene expression relies on the transcriptional co-regulators that are recruited to ACRs to affect epigenomic modifications of surrounding nucleosomes.In this study,we employed transposase-accessible chromatin sequencing to identify ACRs and decipher how the presence of ACRs affects gene expression and epigenetic signatures in the Sorghum genome.As a result,21077 ACRs,which are mapped to 22.9%of genes and 2.7%of repeats,were identified.The profiling of ACRs on gene structures reveals a narrow and sharp peak around the transcription start site,with relatively weak and broad signals covering the entire gene body and an explicit but wide peak from the transcription termination site to its downstream regions.We discovered that the correlations between gene expression levels and profiled ACR densities are dependent on the positions of ACRs.The occurrence of genic ACRs cumulatively enhances the transcriptional activity of intergenic ACR-associated genes.In addition,an intricate crosstalk among ACRs,gene expression,and epigenetic marks has been unveiled by integrating multiple-omics analyses of whole-genome bisulfite sequencing,6mA immunoprecipitation followed by sequencing,RNA sequencing,chromatin immunoprecipitation sequencing,and DNase I hypersensitive sites sequencing datasets.Our study provides a genome-wide landscape of ACRs in sorghum,decrypts their interrelations with various epigenetic marks,and sheds new light on their roles in transcriptional regulation.Chao Zhou Zhu Yuan Xueping Ma Huilan Yang Ping Wang Lanlan Zheng Yonghong Zhang Xiaoyun Liu 2021Plant Communications2021,2,1:2
3The Design of UWB Antenna for Mobile Phone and the Antenna Influence on Human Head Model显示文摘In order to improve the performance of the antenna at low frequency,short circuit branch and coupling feed structure are introduced into the folded broadband single antenna in this paper,then resonant frequency is merged into the antenna using method of stagger tuning,finally,a UWB antenna is designed to w ork betw een 800 MHz ~ 2700 MHz. Analysis are conducted to determine the antenna's properties. The short-circuit w ire location,feeding point location and the length of folded strip are discussed in detail. The SAR is calculated by HFSS simulation softw are w hile the antenna is close to a 3D human head model. 5Activation of lineage competence in hemogenic endothelium precedes the formation of hematopoietic stem cell heterogeneity显示文摘Hematopoietic stem and progenitor cells (HSPCs) are considered as a heterogeneous population, but precisely when, where and how HSPC heterogeneity arises remain largely unclear. Here, using a combination of single-cell multi-omics, lineage tracing and functional assays, we show that embryonic HSPCs originate from heterogeneous hemogenic endothelial cells (HECs) during zebrafish embryogenesis. Integrated single-cell transcriptome and chromatin accessibility analysis demonstrates transcriptional heterogeneity and regulatory programs that prime lymphoid/myeloid fates at the HEC level. Importantly, spi^(2+) HECs give rise to lymphoid/myeloid-primed HSPCs (L/M-HSPCs) and display a stress-responsive function under acute inflammation. Moreover, we uncover that Spi2 is required for the formation of L/M-HSPCs through tightly controlling the endothelial-to-hematopoietic transition program. Finally, single-cell transcriptional comparison of zebrafish and human HECs and human induced pluripotent stem cell-based hematopoietic differentiation results support the evolutionary conservation of L/M-HECs and a conserved role of SPI1 (spi2 homolog in mammals) in humans.
